Effect of pH on the Green Tea Extraction (침출수의 pH가 녹차 침출액 성분에 미치는 영향)

  • Green tea infusions were prepared by extracting them with aqueous solution of different pHs and their physicochemical properties were analyzed. Yellowness, chroma and total color difference values were increased as pH increased. The content of total amino acids in the green tea infusion was not influenced by pH values. Total catechin contents of the green tea infusion extracted at pH 4.0, 5.0, 6.0 and 7.0 were 32.28, 31.84, 31.14 and 14.70 mg/g, respectively. Among the four catechins investigated, epigallocatechingallate and epigallocatechin are more unstable as the pH was changed from 6.0 to 7.0. As pH increased, the extraction of caffeine was also increased. In conclusion, extraction at low pH is preferred for the preparation of green tea infusion.

A Study of the Factors Influencing on Postpartum BMI Change in Some Postpartum Women (일부 산모에서 산후 BMI 변화에 영향을 미치는 요인 분석)

  • Purpose: The purpose of this study was to examine the relationships between variety pregnancy related factors and postpartum BMI change. Methods: Analyzing the BMI of 125 postpartum women in oriental medical hospital of O O university from March, 2008 to May, 2008. Age, pregnancy period. type of delivery, parity, pregravida(PG) BMI, weight and BMI gain during pregnancy, gestational maximum(GMx) BMI were recorded. BMI also checked on each postpartum 7days(P7d), 14days(P14d), 90days(P90d), And we distinguished into Sasang Constitution by QSCC II. Results: The following results were obtained: 1. Age. pregnancy period were not correlated to postpartum BMI change. 2. PG BMI were lower in normal delivery group than caesarean delivery group. 3. PG BMI was lower in primipara group than multipara group. Weight and BMI gain during pregnancy. P90d BMI were lower in multipara group. 4. PG, GMx, P7d, P14d, P90d BMI were significally high in Taeumin. 5. PG BMI were correlated to GMx, P7d, P14d, P90d BMI. 6. Weight and BMI gain during pregnancy were correlated to GMx BMI and P7d, P14d, P90d BMI, BMI loss. Conclusion: This results suggested that weight and BMI gain during pregnancy have the best relationship with postpartum weight change.

  • Power input in stirred vessel is especially important in the design of mixers, as well as the evaluation of mixing processes. A type of baffles in mechanically agitated vessels and power employed are major factors that determine the stirring efficiency in a large scale, multi-step processes. In the present study, power input in the totally baffled agitated vesseles was compared systematically in connection with several previous studies and adequate power input correlation was found to be : $Np_{(pitch)}=({\theta}/90^{\circ})Np_{(90^{\circ})}$ Power number correlation was dependent upon the distance of among the impeller in the agitated vesseles, as follows : $$Np=7.09(n_p)^{0.7}(\frac{b_(double)}{d})(\frac{H}{D})^{0.18}$$, $${n_p}^{0.7}(\frac{b_{(double)}}d)$$<2 $$Np=8.73\{(n_p)^{0.7}(\frac{b_{(double)}}{d})\}^{0.7}(\frac{H}{D})^{0.18}$$, $${n_p}^{0.7}(\frac{b_{(double)}}d)$$>2.

Inactivation of Escherichia coli O157:H7, Salmonella Enteritidis and Listeria monocytogenes by Hydrogen Peroxide and Lactic acid (과산화수소와 유산ol Escherichia coli O157:H7, Salmonella Enteritidis 및 Listeria monocytogenes의 증식 억제에 미치는 영향)

  • The inhibitory effect of the food processing agent on growth of Escherichia coli O157:H7, Salmonella Enteritidis, and Listeria monocytogenes was performed with hydrogen peroxide and lactic acid, and combination of hydrogen peroxide and lactic acid. The minimun inhibitory concentration (MIC) of hydrogen peroxide in E coli O157:H7 was 100 ppm at pH 5.0, 6.0, 6.5 and 7.0, while in Listeria monocytogenes 25 ppm at PH 5.5, 6.0 and 50 ppm at PH 6.5, 75ppm at pH 7.0. MIC of lactic acid in E coli O157:H7 was 2500 ppm at pH 5.0, 6.0, 6.5 and 7.0. MIC of lactic acid in S. Enteritidis was 1250 ppm at pH 5.0, 2500 ppm at pH 5.5, 6.0, 5.5 and 7.0, while in L monocytogenes 625 ppm at pH 5.5 and 125 ppm at pH 6.0, 6.5 and 7.0. MIC of combined hydrogen Peroxide and lactic acid in E. coli O157:H7, S. Enteritidis, and L. monocytogenes was 75 ppm of hydrogen peroxide with 2500 ppm of lactic acid at pH 6.5. The correlations between MICs of hydrogen peroxide and lactic acid in E. coli O157:H7, S. Enteritidis and L. monocytogene were obtained through the coefficient of $determination(R^2)$. $R^2$ value were 0.9994, 0.9935 and 0.9283, respectively. The inhibitory effect of hydrogen peroxide and lactic acid in E. coli O157:H7, S. Enteritidis and L. monocytogenes could be confirmed from the result of this experiment. Therefore, it was expected that the food process would increase or maintain by using lactic acid together with hydrogen peroxide.

$\alpha$- and $\beta$-Amylase Isozyme Expresser Native Proteins in Tropical Silkworm Bombyx mori L.

  • Amylase isozyme based three multivoltine viz., N+p, Np, N+ $p^{cho}$ and two bivoltine-D6+p, D6p syngenic lines (Syn. L) were developed from germplasm (GP) stocks Nistari (N) and D6 respectively. haemolymph isozyme pattern at pH 7.0 and 8.5 depicted a total 11 number (Am $y_{1 to 6}$ at pH 7.0 and Am $y^{l to 5}$ at pH 8.5) of native proteins (NP) of various sizes are amylase isozyme expressers. Among eleven NPs, two NPs of 770 kDa (Am $y^{6}$ at pH 7.0) and 376 kDa (Am $y^3$ at pH 8.5) are $\alpha$-amylase expressers and remaining NPs of 370, 364, 350, 329 and 274 kDa at pH 7.0 and 206, 292, 416, 725 kDa at pH 8.5 are $\beta$-amylase expressers. Accordingly, digestive juice amylase isozyme pattern at aforesaid pH also depicted a total number of 10 NPs (Am $y^{1 to 5}$) at each pH 7.0 and 8.5 are amylase expressers of which NP of 387 kDa (Am $y^4$ at pH 7.0) and 780 kDa (Am $y^{5}$ at pH 8.5) are a-amylase expresser. Remaining NPs of 338,297 & 216 kDa at pH 7.0 and 370, 341, 329 &302 kDa at pH 8.5 are $\beta$-amylase expresser. Recurrent backcross lines (RBL) viz., N+pRBL and NpRBL were developed through introgression of high shell weight character (a multigenic trait) to be used further for congenic line (Con. L) development and to understand any association with introgressed character. Isozyme pattern in haemolymph of RBLs depicted only one $\alpha$-amylase of 770 kDa at pH 7.0 and 376 kDa at pH 8.0 with three and four respective $\beta$-amylase bands but in bivoltine lines numbers of $\beta$-amylase bands vary between 1 to 2 at aforesaid pH. Variability was also observed in digestive juice of multivolitine and its RBLs but bivoltine lines express null activity at both pH except appearance of one very week $\alpha$-amylase band D6+p at pH 8.5. Overall study suggests that not a single NP at both pH is common for expression of any band of amylase isozyme i.e., a totally different set of proteins are the amylase isozyme expresser at specific pH and no molecular factor of amylase is associated in developed RBLs which showed improvement on survival, single cocoon shell weight (SCSW) and single filament length over receptor parents.s.s.s.

Effects of Sodium Alginate, Gum Karaya, and Gum Arabic on the Foaming Properties of Sodium Caseinate (Sodium Alginate, Gum Karaya 및 Gum Arabic이 Sodium Caseinate의 거품성에 미치는 효과)

  • To investigate the effects of sodium alginate, gum karaya and gum arabic on the foaming properties of sodium caseinate, surface tension, specific viscosity, turbidity, foaming ability and foam stability of the caseinate solution with added gums were examined. Surface tensions of the 5%(w/v) protein solutions containing gums at pH 7.0 and 8.0 were $43.7{\sim}44.7dyne/cm$ and $43.6{\sim}44.0 dyne/cm$, respectively. Specific viscosities of the solutions with 0.2 and 0.3% sodium alginates were 15.6 and 39.1 at pH 7.0 (control, 2.8), and 12.1 and 8.2 at pH 8.0 (control, 2.6), respectively. Turbidities were $69.5{\sim}74.0$ at pH 7.0 and $68.0{\sim}72.5$ at pH 8.0. The optimum conditions for foaming ability of the solutions were 0.1% conc. and 15 min whipping in addition of sodium alginates; 0.2% conc. and 20 min whipping in gum karaya; 0.1% conc. and 10 min whipping in gum arabic. For foam stability optimal concentrations were 0.3% in sodium alginate and gum karaya at pH 7.0 and 0.2% at pH 8.0. Addition of sodium alginates was most effective to increase foam stability of the solution, but was not effective to increase foaming ability. At same pH, surface tensions and turbidity of the solutions were related to foaming ability and specific viscosities were related to foam stability.

Distribution and Characteristics of Acidotolerant Heterotrophic and Naphthalene­Degrading Bacteria in Acidic Soils (산성토양에서 내산성 종속영양세균과 나프탈렌분해세균의 분포 및 특성)

  • The distribution and characteristics of acidotolerant heterotrophic and naphthalene-degrading bacteria were investigated in two forest areas, one near Ulsan petrochemical industrial complex (Sunam) and the other in countryside (Daeam). Average values of soil pH at Sunam and Daeam were 3.8 and 4.6, respectively. When het­erotrophic and naphthalene-degrading bacteria were enumerated by most probable number (MPN) procedures at Sunam, the median values of heterotrophs growing at pH 7.0 and pH 4.0 were $5.3{\times}10^7\;and\;3.3{times}10^7$ MPN/g, whereas those of naphthalene-degraders were $5.6{\times}10^4\;and\;4.0{times}10^5$ MPN/g, respectively. While the medians of heterotrophs at Daeam were larger than those at Sunam, the concentrations of naphthalene-degraders were higher at Sunam compared to those at Daeam. From the MPN tubes and enrichment cultures, we obtained 17 isolates of naphthalene-degraders which were identified as Sphingomonas paucimobilis, Brevundimonas vesic­ularis, Burkholderia cepacia, Ralstonia pickettii, Pseudomanas fluorescens, and Chryseomonas luteola. Among them, 6 isolates showed higher naphthalene-degrading activity on minimal media of pH 4 compared to pH 7, whereas the extent of growth was not greater at pH 4 than at pH 7 when they were inoculated on nutrient-rich media. It is plausible that the pH may affect naphthalene-degrading activity of the isolates by changing fatty acid composition of bacterial membrane.

Phospholipid Production by Pseudomonas sp.CH-414 under Various Culture Conditions (배양 조건에 따른 Pseudomonas sp.CH-414의 Phospholipid 생산능의 변화)

  • Using Pseudomonas sp. CH-414, the optimum culture conditions were investigated for the cell growth and the phospholipid production in batch culture by varying pH and aeration rate. With starting the cultivation under the conditions of pH 7.0 and 1vvm, pH was controlled to 6 or 8 at 30 hours of culture time. In the case of changing into pH 6.0, the phospholipid production was increased by ca. 20% with comparison to the case of pH 7.0. However, the biomass and the phospholipld concentration were rapidly decreased after 30 hours of culture time when pH was controlled to 8.0. As the aeration rate was increased, the biomass was increased while the phospholipid concentration was considerably varied and unstable. Especially, the concentration of phospholipid was rapidly decreased with 3vvm of aeration rate. Finally, under the culture conditions of pH 7.0 and 3vvm until 30 hours for the cell growth, which were controlled to pH 6.0 and 1vvm for the stable production of phospholipid beyond that time, the dry cell weight was $18.5g/\ell$ and the phospholipid concentration was $\0.83g/ell$ (45mg/g cell).

Studies on Blood Gas Values in Non-Human Primates Reared in Korea (國內詞育 원숭이의 血液가스値에 관한 硏究)

  • Blood gas values such as pO$_2$ were studied in common marmosets, crab-eating macaques and Japanese macaques, rhesus macaques and celebes macaque reared in Korea. Blood pH and blood gas values were evaluated in both arterial and venous blood. pH, p$CO_2$, and pO$_2$, of arterial blood in common marmosets were measured as 7.4$\pm$0.1, 29.2$\pm$3.6 mmHg and 81.5$\pm$8.9 mmHg, respectively. Corresponding values in one crab-eating macaque were 7.3, 41.3 mmHg and 46.5 mmHg, respectively. In case of venous blood, pH, p$CO_2$, and pO$_2$, in common marmosets were observed as 7.2$\pm$0.2, 64.9$\pm$18.3 mmHg and 23.5$\pm$5.4 mmHg, respectively. On the while, pH, p$CO_2$, and pO$_2$, of venous blood in crab-eating macaques showed 7.2$\pm$0.2, 49.9$\pm$8.0 mmHg and 38.3$\pm$8.8 mmHg, respectively. Venous pH, p$CO_2$, and pO$_2$, in Japanese macaques were 7.1$\pm$0.2, 56.4$\pm$5.3 mmHg and 40.1$\pm$9.3 mmHg, respectively. Those values in one rhesus macaque were 7.2, 61.1 mmHg and 24.9 mmHg, and in celebes macaque were 7.1, 54.3 mmHg and 31.8 mmHg, respectively.

Soil buffer capacities from the differrent host rocks by the treatment of artificial acid precipitation

  • To investigate the weathering soil buffering capacities of the artificial acidic precipitation, the weathering soils and their leachate solutions were sampled from the host rocks(granite;GR, rhyolite;RH, gabbro;GA, basalt;BA, two serpentinite;SE1, SE2 and limestone;LI) and analyzed for pH and chemical properties. 1n the soil pH of the GR and RH ,the acidic rocks, were 5.02 and 5.95, respectively. And the GA and BA, basic rocks, were 6.52 and 7.57. The SE1 and SE2 were 8.90 and 8.89. While the LI was 7.84. These results means the typical soil pH properties by host rocks. After the artificial acidic precipitation input 5OOml, the average changes of soil leachate solutions treated by pH levels(pH 5.0, 4.0 and 3.0), were pH 5.73, 5.00 and 4.40. in GR soil, and pH 6.19, 5.99 and 5.57 in RH. GA were pH 6.31, 6.04 and 5.86, BA were pH 7.05, 6.85 and 6.56 and SE1 were pH 8.31, 8.26 and 7.71. SE2 were pH 8.29, 8.24 and 7.96. LI were pH 7.55, 7.46 and 6.79. The soil leachate pHs from volcanic rocks were higher than those from the plutonic rocks and GR soils showed greater response than other soils. With increasing 100ml input-solution, the soil leachate pHs were mainly decreased. Cation concentrations, CEC, EC and total nitrogen concentrations of RH and BA soils, the volcanic rocks, were higher than those of GR and GA soil, the plutonic rocks. On the contrary, Al concentrations of the GR and GA soils were higher than those of RH and BA soils, partly because of high quartz content in GR and Al content in the biotite and plagioclase in GA.
