  • The ryanodine-receptor $Ca^{2+}$ release channel protein in the sarcoplasmic reticulum membrane of rabbit skeletal muscle plays an important role in muscle exitation-contraction (E-C) coupling. Various types of detergents were tested, including Chaps, cholate, octylglucoside, Zwittergents, Mega-9, Lubrol PX, and Triton X-100 for solubilization of this protein. Among these, Chaps and Triton X-100 were found to optionally solubilize the channel complex. Optimum conditions for this solubilization were pH 7.4 with a salt concentration of 1 M. The addition of phospholipid in the solubilization step helped in stabilizing the protein. The purification of the receptor was performed using sucrose density gradient centrifugation. Various methods [dilution, freeze-thaw, adsorption (Biobeads), and dialysis] were investigated to incorporate the Chaps-solubilized and purified $Ca^{2+}$ release channel protein into liposomes made from different types of phospholipids. Of these, a combined method consisting of a dialysis, freeze-thaw and sonication steps yielded the best results. Reconstituted vesicles produced by this method with 95% phosphatidylcholine (from soybean extract) had good function.

  • Mature acetylcholinesterase (AChE) gene (gm, 1,836 bp) was cloned from the housefly and successfully expressed in the E. coli CodonPlus (DE3) RIL system (GM-E, 72 kDa) with a yield of 1,630 mU/g fresh cells. Using the gm, 10 kinds of mutants were constructed and expressed for enhancing sensitivity to insecticides. The sensitivity of these mutants to five kinds of organophosphate (OP) and three carbamate insecticides was investigated by measuring the apparent bimolecular inhibition constant ($k_i=k_2/K_d$). Surprisingly, the sensitivity of quadruple mutant IGFT was enhanced as much as 7-fold for acephate, 164-fold for demeton-S-methyl, 484-fold for dichlorvos, 523-fold for edifenphos, 30-fold for ethoprophos, 30-fold for benfuracarb, 404-fold for carbaryl, and 107-fold for furathiocarb, compared with that of GM-E, although the sensitivity of each single point mutant was slightly increased. These mutational studies indicated that (i) contradictory to Walsh et al. [39], the residue 327 is the important key residue for enhancing sensitivity as much as the residue 262, (ii) the residue 82 and additional residues of 234, 236, and 585 are also important, and (iii) sensitivity was cooperatively accelerated as the number of strategic mutations increased.

  • A scanning stereo-PIV system was developed to measure the three-dimensional distribution of three-component velocity in a turbulent round jet. A laser light beam produced by a high repetition rate YLF pulse laser was expanded vertically by a cylindrical lens to form a laser light sheet. The light sheet is scanned in a direction normal to the sheet by a flat mirror mounted on an optical scanner, which is controlled by a programmable scanner controller. Two high-speed mega-pixel resolution C-MOS cameras captured the particle images illuminated by the light sheet, and stereoscopic PIV method was adopted to acquire the 3D-3C-velocity distribution of turbulent round jet in an octagonal tank filled with water. The jet Reynolds number was set at Re=1000 and the streamwise location of the measurement was fixed at approximately x = 40D. Time evolution of three-dimensional vortical structure, which is identified by vorticity, is visualized. It revealed that the existence of a group of hairpin-like vortex structures was quite evident around the rim of the shear layer of the jet. Turbulence statistics shows good agreement with the previous data, and divergence of a filtered (unfiltered) velocity vector field was $7\%\;(22\%)$ of root-me an-squared vorticity value.

  • Like other major cities in the world, Bangkok and some other cities in Thailand are expanding rapidly. The Office of Transports and Traffic Policy and Planning had selected three cities for a study of urban change; namely, Bangkok Metropolis and vicinity ( including Nontha buri, Samut Prakan , Prathum Thani, Samut Sakhon and Nakhon Pathom), Chiang Mai and Nakhon Nayok. The main objective of the study is to monitor urban development and urban extension as well as the change of landuse from farm land to urban area during two periods by using available satellite data. LANDSAT-5 TM and SPOT-HRV panchromatic were used for the first period and LANDSAT-7 ETM+ and IRS-lD panchromatic were used for the second period with a lapsed time of 14 years. I was found that during this period Bangkok Metropolis and vicinity extended 1,222 square kilometer; Chiang Mai 68.3 square kilometer, Nakhon Nayok 5.97 square kilometer. Most landuse categories which had been changed to urban were rice paddy and mixed orchard. Some suggestion for sound urban development based on satellite data was also included.

  • Objective: To understand the domestication and spread of horses in history, genetic information is essential. However, mitogenetic traits of ancient or medieval horses have yet to be comprehensively revealed, especially for East Asia. This study thus set out to reveal the maternal lineage of skeletal horse remains retrieved from a 15th century archaeological site (Gongpyeongdong) at Old Seoul City in South Korea. Methods: We extracted DNA from the femur of Equus caballus (SNU-A001) from Joseon period Gongpyeongdong site. Mitochondrial (mt) DNA (HRS 15128-16116) of E. caballus was amplified by polymerase chain reaction. Cloning and sequencing were conducted for the mtDNA amplicons. The sequencing results were analyzed by NCBI/BLAST and phylogenetic tool of MEGA7 software. Results: By means of mtDNA cytochrome b and D-loop analysis, we found that the 15th century Korean horse belonged to haplogroup Q representing those horses that have historically been raised widely in East Asia. Conclusion: The horse is unique among domesticated animals for the remarkable impact it has on human civilization in terms of transportation and trade. Utilizing the Joseon-period horse remains, we can obtain clues to reveal the genetic traits of Korean horse that existed before the introduction of Western horses.

  • In this study, we analyzed the effects of seismic displacements due to the mega thrust earthquake occurred near Tohoku-Oki area on Mar. 11, 2011 with Mw 9.0 magnitude in the context of evaluation of position change by the earthquake on the Korean and Asian GPS permanent stations. For this, two weeks GPS data observed around the event of Tohoku-Oki earthquake (Mar. 4 ~ Mar. 18, 2011) were obtained from 22 GPS permanent stations in the vicinity of epicenter (Korea, Japan, Russia, China and Taiwan) and 284 IGS global stations. All available GPS data were processed and adjusted by GAMIT/GLOBK software to estimate the co-seismic horizontal displacements at each station. As the results of GPS analysis, the co-seismic displacements due to Tohoku-Oki earthquake were clearly revealed in the GPS stations of Asian region, Japan and its neighboring countries, and even to affect the horizontal position of GPS station (WUHN in China) which are located about 2,702km away from the epicenter. In conclusion, it was found that the Tohoku-Oki earthquake had resulted in the horizontal displacements ranging from 14.9 mm to 58.3 mm in Korea. So, these displacements can cause the position error of GPS geodetic survey up to 20 mm without updating the coordinates of Korean geodetic network.

  • Due to the continuous expansion of the highway, the opening of the high-speed railway(KTX), etc, the demands of domestic air transport decreasing. In the circumstances, airports and local governments are spending themselves in taking a measure to activate local airports. In this aspect, this study seeks the smooth flow of human movement through the activation of Daegu International Airport by analyzing the present condition of Daegu International Airport in relation to the appearance of the high-speed railway, deducing plans for activating airports, and presenting the roadmaps for long-, mid- and short-term roles for the development of Daegu Airport, and Presents substantial fulfilling plans to secure the competitive power of Daegu International Airport by speeding up the flow of air goods of the area for the long run. It is suggested in the study that by setting up an organization exclusively responsible for activating Daegu International Airport in the city of Daegu, supporting local travel agencies to promote consistent demands, supporting aviation companies to employ Daegu International Airport as a major air base, establishing airlines with priority for airports that open newly in local cities of Japan and establishing and expanding airlines in China areas be promoted actively. Ministry of Construction & Transportation makes it a means of main airport Policy to only support Incheon International Airport to be a hub. However not only Incheon International Airport should be an amok representing the country, but also the wide Metropolitan area should supply the demands, and airports based in the West and East Coast areas need to be developed in the future. It is suggested that since air demands of domestic local airports are based on the demands of the of gin destination in neighboring region, multiple bases need to be developed. It is suggested that by developing not 1 in mega hub (Incheon International Airport) but 1 in mega hub. 2 in regional hub (the West and East Coast) and Jeju foy the tourism hub in our developing airports system.

  • Background The use of acellular dermal matrix (ADM) in implant-based immediate breast reconstruction has been increasing. The current ADMs available for breast reconstruction are offered as aseptic or sterile. No published studies have compared aseptic and sterile ADM in implant-based immediate breast reconstruction. The authors performed a retrospective study to evaluate the outcomes of aseptic versus sterile ADM in implant-based immediate breast reconstruction. Methods Implant-based immediate breast reconstructions with ADM conducted between April 2013 and January 2016 were included. The patients were divided into 2 groups: the aseptic ADM (AlloDerm) group and the sterile ADM (MegaDerm) group. Archived records were reviewed for demographic data and postoperative complication types and frequencies. The complications included were infection, flap necrosis, capsular contracture, seroma, hematoma, and explantation for any cause. Results Twenty patients were reconstructed with aseptic ADM, and 68 patients with sterile ADM. Rates of infection (15.0% vs. 10.3%), flap necrosis (5.0% vs. 7.4%), capsular contracture (20.0% vs. 14.7%), seroma (10.0% vs. 14.7%), hematoma (0% vs. 1.5%), and explantation (10.0% vs. 8.8%) were not significantly different in the 2 groups. Conclusions Sterile ADM did not provide better results regarding infectious complications than aseptic ADM in implant-based immediate breast reconstruction.
