Changes in Growth Characteristics, Biological Activity and Active Compound Contents in Ginseng of Different Ages (재배년수에 따른 인삼의 생육특성, 생리활성, 성분의 변화)

  • Background: Ginseng has been used as a medicine and functional food since ancient times. It is a perennial crop, and its whose commercial valuse increases with growing period and is affected by the atmosphere and soil environment. Methods and Results: In a selected field, we measured air temperature under a shade structure and soil physicochemical properties, and determied plant and root growth as well as ginsenoside and total polyphenol content of one- to five-year-old ginsengs plants. Although air temperature above 30℃ was recored for more than 37 days, no marked growth inhibition of ginseng was detected. Among all soil physicochemical properties, except for pH, were within the allowable range the shortage increases with ginseng years. In five-year-old ginseng, the quantity is about 9.7% higher than the average weight by standard, indicating that is not affected by temperature when grown under a shade structure. Three-year-old ginseng contained the highest total ginsenoside and total polyphenol levels and exhibited the greatest DPPH radical scavenging activity. Conclusions: The total ginsenoside and protopanaxadiol/protopanaxatriol ratio were both low at five-year-old ginseng plants, which was attributed to rapid growth of the root system in five-year-oid plants. There were no significant differences in total polyphenol content and antioxidant activity between.

Anti-inflammatory Effect of Cultivated Wild Panax ginseng Extracts at Various Ages in RAW264.6 Macrophages (RAW264.7 대식세포주에서 근령별 산양삼 추출물의 항염증 효과)

  • It is well-known that cultivated wild Panax ginseng has anti-inflammatory effect. However, a comparative study on cultivation period vs biofunctionality is currently lacking. In this study, 70% ethanol extracts of 3-years (yrs)-, 5-yrs-, or 7-yrs-old cultivated wild ginseng were evaluated for their inhibitory effects on RAW264.7 murine macrophages. Specifically, the production of pro-inflammatory cytokines (interleukin-6 [IL-6] and tumor necrosis factor-alpha [TNF-${\alpha}$]), the expression of surface proteins (CD80, CD86, and MHC-II), and the phagocytic properties were investigated. RAW264.7 cells were induced by 500 ng/mL of lipopolysaccharide (LPS) and treated with 0.1, 1, and 10 ppm of samples. LPS-induced IL-6, TNF-${\alpha}$ and surface proteins in all samples were down-regulated in a dose-dependent manner. Both IL-6 and TNF-${\alpha}$ were significantly reduced at 10 ppm of the 7-yrs-old sample compared to 10 ppm of 3-yrs- and 5-yrs-old samples. CD80 and CD86 were also reduced at 10 ppm of all samples, and there was no difference among samples. The phagocytosis has no difference except in 10 ppm of 3 yr-old sample. The results suggest that cultivated wild ginseng extract has anti-inflammatory effect without decreasing phagocytosis.

Anticarcinogenic Effect of Ginseng Powders Depending on the Types and Ages using Yun's Anticarcinogenicity Test (I) (벤조피렌으로 유도된 마우스 폐선종에 대한 인삼분말의 연근별 항발암효과)

  • The authors have already shown that 6 year old red ginseng extract or its powder has remarkable anticarcinogenic effects. In this study, we further investigated whether fresh ginseng or white ginseng has similar anticarcinogenic effects and also if their anticarcinogenic effects are related to the types and ages of ginseng using Yun's anticarcinogenicity test (9 week medium term bioassay model). Dried fresh ginseng and red ginseng at 1.5, 3, 4, 5 and 6 years, and while ginseng at 3, 4, 5 and 6 years were used. The following results were obtained: 1) In the dried fresh ginseng treated groups, the incidence of lung adenoma induced by benzo(a)pyrene was 41.39) and its incidence was reduced to 31.2%, 30.0%, 31.3%, 30.7% and 27.8% after co-treatment with 1.5, 3, 4, 5 and 6 year-dried fresh ginseng, respectively. A significant effect was observed only in 6 Year-dried fresh ginseng. 2) In the white ginseng treated groups, the incidence of lung adenoma induced by benzo(a)pyrene was 45.0% and its incidence decreased to 41.3%, 38.0%, 31.6%, and 25.3% after co-treatment with 3, 4, 5 and 6 year-white ginseng, respectively. Five and 6 year-ginsengs showed significant inhibition of lung adenoma. 3) In the red ginseng treated groups, the incidence of lung adenoma induced by benzo(a) pyrene was 48.6% and its incidence diminished to 37.9%, 41.7%, 31.7%, 28.3% and 25.5% after co-treat-melt with 1.5, 3, 4, 5 and 6 year-red ginseng, respectively. In 4, 5 and 6 year-ginsengs, the anticarcinogenic effect was prominent. From the above results, we concluded that a significant anticarcinogenic effect was observed in 6 year-dried fresh ginseng, 5 and 6 year-white ginsengs, and 4, 5 and 6 year-red ginsengs.

Comparison of Photosynthesis between Treatment and Non-treatment of Lime Bordeaux Mixture in 3 Year Old Root in Panax ginseng C. A. Meyer

  • Korean ginseng has been used for thousands of years as an important medicinal plant. Lime-Bordeaux mixture (LBM) was made with copper sulfate and quicklime, which was sprayed instead of pesticides in ginseng field. Net photosynthesis ($P_N$) was compared between Treatment and Non-treatment of LBM in 3 Year Old Ginseng. $P_N$ in control plot recorded $2.94{\mu}mol\;(CO_2)\;m^{-2}s^{-1}$ at the first day of experiment, which was similar until the last day of experiment. However, The $P_N$ in LBM recorded $2.23{\mu}mol\;(CO_2)\;m^{-2}s^{-1}$, which was lower than that in control plot. As time goes by, The $P_N$ in LBM was gradually increased up to $3.21{\mu}mol\;(CO_2)\;m^{-2}s^{-1}$ and finally, it was similar with that in control plot at 7th day as a $3.20{\mu}mol\;(CO_2)\;m^{-2}s^{-1})$.

Effects of light intensity and temperature on photosynthesis and respiration of Panax ginseng leaves (인삼엽(人蔘葉)의 광합성(光合成)과 호흡(呼吸)에 미치는 광도(光度) 및 온도(溫度)의 영향(影響))

  • Effects of temperature and light intensity on photosynthesis, respiration and chlorophyll content of ginseng (Panax ginseng C. A. Meyer) were as follow. 1. Optimum light intensity for apparent photosynthesis at $25^{\circ}C$ was 25Klux($1.35mgCO_2/dm^2{\cdot}hr$) for two years old ginseng grown in pot and 30Klux($1.94mgCO_2/dm^2{\cdot}hr$) for the six years old palmate cut leaves. Optimum temperature at 25Klux was $15^{\circ}C$ ($1.40mgCO_2$) for the 2 years old and 20 to $22^{\circ}C$ ($2.03mgCO_2$) for the 6 years old. 2. Dark respiration increased almost linearly with the increase of air temperature till $25^{\circ}C$ (2.6times between $16^{\circ}C$ to $25^{\circ}C$ for the 6 years old and 1.8 times between $15^{\circ}C$ to $25^{\circ}C$ for the 2 years old). Dark respiration was 11.1 % of net photosynthesis at $16^{\circ}C$, 17.8% at $25^{\circ}C$ for the 6 years old and 40% at $15^{\circ}C$, 64.7% at $25^{\circ}C$ for 2 years old. 3. Stomata appeared only in abaxial surface (lower epidermis) and stomatal frequency was $37per\;mm^2$. 4. Above results together with other informations quoted here strongly suggest that air temperature is much better criteria than light intensity for the improvement of shading roof material and shading construction. That is to promise maximum light intensity unless air temperature is above $25^{\circ}C$.

  • The variation of the amount of starch, size and shape of the starch granules, amylose content, and blue value of the starch in the Korean ginseng roots from one year old to five year old cultivated at Kumsan was studied. The results obtained were as follows; 1) The starch content of the ginseng root(dried) was increased with the age of the root; that is, 9.62% for one-year-old roots, 10.35% for two-year-old root, 15.50% for three-year-old root, 17.05% for four-year-old root, and 18.32% for five-year-old root. 2) The shape of the ginseng starch granules was round or short oval, and in the latter case the ratio of minor axis to major axis was 1 to 1.1. Diameter of the starch granules was in the range of $1.48\;{\mu}\;to\;8.14\;{\mu}$ and the most frequent granule size was $3\;{\mu}\;(32.1{\sim}35.7%)$. The number of big size starch granules was increased during the five years of growing, while, the number of small size granules was decreased. 3) The amylose content in the ginseng starch was varied with the age of the root, in the range of $53.6{\sim}70.5%$. 4. The blue value of the ginseng starch was in the range of 0.60 to 0.71.

Effect of Herbicide Quinclorac on Ginseng Growth (제초제 Quinclorac이 인삼생육에 미치는 영향)

  • In this experiment, germination test of ginseng seed and growth test of two year old ginseng in soil incorporated with quinclorac (3,7-Dichloro-8-quinoline carboxylic acid) were conducted to evaluate phytotoxicity caused by quinclorac residue in soil. Ginseng seed germiilation was hardly affected, but growth of ginseng seedling was retarded by quinclorac. The phytotoxicity of quinclorac on ginseng seedling was expressed at the lowest level tested, 3.75 g a.i./10a (118 of recommended rate for rice cultivation). The representative phytotoxicity in ginseng were decreased root weight, shortened leaf width and leaf length, and weak lateral root Quinclorac residue decreased by 1/4 in pot soil after 2 months and below 2 ppm in the paddy soil applied at usual rate (30 g a.i./10a) after 1∼ 2 years. Below 2 ppd ginseng did not show any phytotoxicity symptom.

Component Profile Analysis of Irradiated Korean White Ginseng Powder (방사선 조사 인삼의 성분변화에 관한 분석)

  • Currently, some food materials are disinfected by $\gamma$-irradiation (using Co-60) or ethylene oxide treatment. These treatments were applied to ginseng powder and the ginseng components such as ginsenosides, polyacetylenes and phenolic acids were analyzed by HPLC to determine any compositional changes due to irradiation. No appreciable difference was observed in the HPLC pattern of ginsenosides, polyacetylenes of ginseng powder after 10 key irradiation or ethylene oxide treatment (EO $CO_2$= 3 : 7, w/wfb) from those of untreated fresh ginseng powder when they were analyzed soon after treatments. When the ginseng powders were stored at room temperature for three years after the same treatment, the HPLC patterns of polyacetylenes and phenolic acid fraction showed appreciable change from those of fresh ginseng powder, however, the HPLC patterns of three year old samples did not show any appreciable difference.

A Study on the Analysis of Amino Acids in Korean Ginseng (韓國人蔘의 年根別 및 貯藏期間別 아미노酸分析)

  • The contents of amino acids were examined in the 3, 4, 5, and 6 year-old roots of fresh ginseng and the 1979, 1980, 1981, and 1982 years' products of white and red ginsengs. Samples extracted with 75% ethanol for free amino acids and hydrolyzed with 6N-HCL for total amino acids were analyzed by Amino Acid Analyzer (Hitachi model KLA-5). The results were summarized as follows: 1. Amino acids from extracted samples were 18 kinds of Tryptophan, Lysine, Histidine, Arginine, Aspartic acid, Threonine, Serine, Glutamic acid, Proline, Glycine, Alanine, Cystine, Valine, Methionine, Isoleucine, Leucine, Tyrosine, and Phenylalanine. 2. Amino acids detected in hydrolyzed samples were 17 kinds execpt Tryptophan of extracted ones. 3. Arginine was the highest quantity of amino acids in ginseng. 4. The content of Tryptophan was 0.5690 mg/g in the 6 year-old fresh ginseng and trace quantities in other samples. 5. The contents of amino acids were increased in fresh ginseng according to cultivation year. 6. The contents of amino acids in white ginseng were slightly decreased but those in red ginseng were not changed during the storage time. 7. The content ratio of free amino acids to total amino acids were 1:3.

  • The best environments such as crown density, temperature, light intensity and humidity have to be identified because these factors are strongly related to the growth and several physiological characteristics. The purposes of this study are \circled1 to collect basic data fer growth, \circled2 to identify the best growth environments. to achieve these purposes, oak, pine and mixed forest stands have been selected in this study. forest ginseng seeds were sown in these forest four years ago. Several environmental and growth factors have been surveyed. In general, mean tree age, DBH and average height are 20-25 years old, 14-17 cm and 7-9 m, respectively. The growths of forest ginseng grown in oak stand are better than those of pine and mixed stands.
