Antioxidant Activities and Monacolin K Production of Fermented Chinese Yam by Monascus sp. MK805 (Monascus sp. MK805 균주를 이용한 발효마의 Monacolin K 생산과 항산화 활성)

  • This study was carried out to investigate the characteristics of fermented chinese yam and rice (Dioscorea japonica, Dioscorea batatas, Dioscorea opposita, Rice, FCYR) using Monascus sp. MK805. The extracts from FCYR were measured to examine pigments, antioxidant activities were investigated through DPPH radical scavenging activity, total polyphenol and flavonoid contents, reducing power. Also it was investigated monacolin K productivity by FCYR. Pigments productivity (yellow, orange and red) were 26.2, 13.9, 17.3 at Dioscorea japonica, 41.9, 22.6, 53.2 at Dioscorea batatas, 12.5, 7.5, 9.7 at Dioscorea opposita and 10.1, 7.7, 10.2 at rice, respectively. DPPH radical scavenging activity of FCYR was about 69.7, 79.6, 57.8, and 42.3%, total polyphenol contents of FCYR was about 480.6, 658.7, 379.3, and 212.9 mg/kg, total flavonoid contents of FCYR was about 342.5, 448.4, 235.2, and 168.7 mg/kg, reducing power of FCYR was about 1.57, 2.14, 1.14, and 0.35 (OD at 700 nm), respectively. And then monacolin K productivity of FCYR was about 453.8, 509.5, 332.2, and 263.2 mg/kg, respectively.

Characteristics on the Crytocaryon irritans of Rock bream, Oplegnathus fasciatus in the embankment fish farm (축제식양식장에서 사육한 돌돔, Oplegnathus fasciatus의 백점충, Crytocaryon irritans 감염 특성)

  • Crytocaryon irritans infection of rock bream was investigated in embankment fish farm from July to December 2009. Prevalence variation was 20.0~88.0% that was high in september and was low in July. While no mortality was found in July and August, it occurred after mid september. Environmental conditions during the survey period were water temperature $19.3{\sim}24.3^{\circ}C$, dissolved oxygen $5.0{\sim}7.1\;mg\;L^{-1}$ and salinity 31.9~33.7 psu, and the water temperature during mortality season was $24.0^{\circ}C$. External symptom of the rock bream was secretion mucus but swimming showed fine in the early infected period. While, the symptoms in the severe outbreak season were excess of mucus, falling off tail and congestion, bleeding spot the surface and weakened swimming. We observed free living and parasitic stages in the gills and body surface of rock bream. Hepato somatic index (HSI) was $1.9{\pm}1.1{\sim}3.5{\pm}1.7%$, which was low in the September being mortality season but was high in November. The percentages of hematocrit infected with Crytocaryon irritans were 37.3% and 41.0% in July and August, respectively while they gradually decreased to 32.1% and 24.2% in september and October, respectively. Total cholesterol and Triglyceride values rapidly decreased by October. After mortality, AST and ALT were 7 and 5 folds higher compared to non-mortality season.

  • Friedel-Crafts alkylation reaction of an isomeric mixture of 1,2,3,4,5,6,7,8- (2) and 1,2,3,4,5,6,7,8-octahydrophenanthrene (2') with excess vinylchlorosilanes such as vinyl(methyl)dichlorosilane (1a) and vinyltrichlorosilane (1b) in the presence of aluminum chloride catalyst at 80 $^{\circ}C$ gives only one dialkylated products, 9,10-bis[2-(chlorosilyl)ethyl]-1,2,3,4,5,6,7,8-octahydroanthrenes [(Cl2XSiCH2CH2)2C14H16: X = Me (4a), Cl (4b)] in good yields, but 9,10-bis[2-(chlorosilyl)ethyl]-1,2,3,4,5,6,7,8-octahydrophenanthrenes are not obtained. However, monoalkylation of 2 with 1 affords a mixture of both isomeric compounds, 9-[2-(chlorosilyl)ethyl]-1,2,3,4,5,6,7,8-octahydroanthracenes 3 and -phenanthrenes 3'. The yield of product 3' is always higher than that of 3. When a mixture of 3 and 3' is alkylated again with 1, only product 4 without phenanthrene type compounds is obtained, indicating that the isomerizations between 2 and 2', or 3 and 3'occur under the alkylation condition. The alkylation with dimethylvinylchlorosilane or trimethylvinylsilane did not proceed. The structure of 4a is determined by X-ray single crystal diffraction analysis.

Microflora of the Lower Part of the Nagdong River (낙동상 하류 수역의 Microflora)

  • This experiment was carried out to evaluate general characteristics of the water quality, total coliform, fecal coliform and microflora at the St. 1 to St. 3 and St. 4 to St. 7. Eighty-four water samples were collected from the 7 stations from July 1988 to April 1990 (see Fig. 1) Range and mean value of the samples were as follows water temperature 2.0-29.9$^{\circ}$C, 16.3$^{\circ}$C pH 6.86-9.08, 7.62: electrical conductivity 54.85$\mu \mho$/cm, 4.300 m$\mho$/cm, 911.93 $\mu \mho$/cm : turbidity 0.9-36 NTU, 6.8 NTU, respectively. pH and electrical conductivity at St. 4 to St. 7 were higher than those at St. 1 to St. 3, but turbidity at St. 1 to St. 2 was 7 times higher than that at Sonagdong river area. The bacterial density of the samples ranged 91-110,000/100ml for total coliform. 21-15,000/100ml for fecal coliform. Specially, the geometric mean value of the St. 3 was 11,836 / 100ml for it leveled heavy contaminaiton. Predominant species among the 3,874 strains isolated form the samples were 30.6% Enterobacteriaceae, 14.7% Acinetobacter, 9.0% Aeromonas. 8.9% Neisseria, and 7.5% Vibrio.

Changes in Bioactive Compounds Contents of 'Maehyang' and 'Seolhyang' Strawberry Fruits by UV Light Illumination

  • The net photosynthetic rate of 'Seolhyang' strawberry plants was measured daily for 7 days after treatment at three UV illumination dosages (0, 9.8, and 29.5 $kJ{\cdot}m^{-2}$). The net photosynthetic rates of the strawberry plants with 9.8 and 29.5 $kJ{\cdot}m^{-2}$ UV light illumination decreased by 20.2 and 61.4%, respectively, at 7 days after UV light treatments. UV treatments with two illumination dosages (7.9 and 15.7 $kJ{\cdot}m^{-2}$) altered the phenolic compounds contents during the cultivation period when compared to those in the control fruits. The anthocyanin content with 7.9 $kJ{\cdot}m^{-2}$ UV light illumination of 'Seolhyang' increased by 18.7% compared with those in control fruits at the 11 DAT. However, the anthocyanin content of 'Maehyang' was not significantly different among treatments during experiment period. The highest level of ellagic acid was found with 7.9 $kJ{\cdot}m^{-2}$ UV light illumination in both cultivars at the 11 DAT. Our results showed that strawberries illuminated with UV light during cultivation period had higher bioactive compounds contents than control fruits. These results suggest that UV light treatments may be a useful non-chemical way of promoting strawberry fruits quality.

The Isolation of Bacillus sphaericus 366M-9 Producing New Cephalosporin-C Deacetylase (CAH) and its Enzymatic Characterization (신규 Cephalosporin-C Deacetylase(CAH) 생산 균주인 Bacillus sphaericus 366M-9의 선발 및 그 효소학적 특성)

  • Several microorganisms (esterase-producing group) were isolated by the solid selective media containing-naphtylacetate. Among them, strain 366M-9 having a high activity of cephalosporin-C deacetylase (CAH; EC 3.1.1.41) was selected. The strain 366M-9 was identified as Bacillus sphaericus on the basis of morphological, physiological, and biochemical characteristics. The production of CAH reached at maximum value after 32 hrs, when cultivated in the optimal medium containing dextrin 2.5%, peptone 2.5%, sodium chloride 0.5%, dipotassium phosphate 0.25%, ferrous sulfate 0.02%, and 7-ACA 0.1% at $30^{\circ}C$ with initial pH 6.0. The CAH was purified by 3 steps with ammonium sulfate precipitation, adsorption chromatography on hydroxyapatite column, and Sephadex G-200 gel chromatography. The final enzyme preparation was homogeneous as judged by the analysis of SDS-PAGE and HPLC. Optimum temperature and pH for CAH activity were $50{\circ}C$ and around 7.0, respectively. And the enzyme was stable at pH 6.0~8.0, up to $50^{\circ}C$. The Michaelis-Menten constants ($K_{m}$ ), $V_{max}$ were 0.87 mM and 1.22 unit/ml, respectively.

Synthesis and Characterization of Poly(9,9-dioctylfluorene-2,7-vinylene) for Light Emitting Diode Application

  • Fluorenevinylene-based EL polymers, poly(9,9-dioctylfluorene-2,7-vinylene) (PFV) and poly[(9,9-dioctylfluorene-2,7-vinylene)-co-{2-methoxy-5-(2 ethylhexyloxy)-1,4-phenylenevinylene}] (PFV-co-MEH-PPV), have been synthesized by Gilch polymerization method. The resulting polymers were soluble in common organic solvents and easily spin cast onto the indium-tin oxide (ITO) substrate. The weight average molecular weight and polydispersity of PFV and PFV-co-MEH-PPV were in the range of 22.2 - 43.2 x $10^4$ and 1.9 - 3.0, respectively. Double-layer light-emitting diodes with ITO/PEDOT/Polymer/Al configuration were fabricated. PFV-co-MEH-PPV showed better EL properties than those of PFV and MEH-PPV The turn-on voltage of poly(9,9dialkylfluorene) derivatives were dramatically decreased to the 2.5 V compared to fluorene-based EL polymers. The maximum brightness and luminescence efficiency were up 7 to 1350 cd/$m^2$ and 0.51 Cd/A.

Chemical Components and Pulverization conditions of the Pollens of Pinus rigida and Pinus koraiensis (리기다소나무와 잣나무 화분의 화학적 성분과 파쇄 조건에 관한 연구)

  • This study was to analyze the chamical components of the pollens of Pinus rigida and P. koraensis and to investigate the optimal conditions for pollen pulverization. The results are as follow: (1) The contents of moisture, crude ash, crude protein, crude fat, crude filber and carbohydrate in the pollen of P. rigida were 9.9%, 2.5%, 13.1% 2.9% 7.5% 64.1% respectively, while those of P. koraiensis were 9.8%, 2.1%, 11.1% 2.8% 8.1% 66.1% respectively. All the contents of P. rigida and P. koraiensis were much higher than those of general crop grains. (2) Eighteen different amino acids were detected in the pollen of P. rigida and P. koraiensis. Among them, ten essential amino acids were indentified, which showed high nutritions values. (3) The contents of vitamin A, B1, B2, C, and E in the pollen of P. rigida were 8.7 mg, 10.1 mg, 15.9 mg, 32.9 mg, 1.9 mg, respectively, while those of in the pollen of P. koraiensis were 7.9 mg, 9.7 mg, 14.5 mg, 34.2 mg, and 2.5 mg respectively. Vitamin C among them was abundant. (4)When the pollen grains of P. prigida was pulverized for 50minutes at 5,000rpm and P. koraiensis for 40 minutes at 5,000rpm by Overhead stirrer, the 100% of pollen was pulverized.

Comparison of IVF Outcomes in Patients with Endometriosis According to Severity (자궁내막증이 있는 불임 여성에서 중등도에 따른 체외 수정의 결과 비교)

  • Objective: To evaluate the impact of endometriosis on IVF-ET cycles and to compare IVF outcomes between stage I/II and stage III/IV endometriosis. Methods: We analyzed 697 patients (1,199 cycles) with endometriosis (stage I-II:638 cycles, stage III-IV: 561 cycles) and 325 pts (459 cycles) with tubal factor as controls between January 1994 and April 2004. Pts with endometriosis were diagnosed by laparoscopy and medical and surgical treatment were done in 353 cycles (55.3%) and 466 cycles (83.1%) of stage I-ll/stage III-IV endometriosis. Cycles with age>35 years or FSH>20 miU/mL or severe male factor infertility were excluded. Results: The number of retrieved oocytes ($9.97{\pm}7.2$ vs. $13.4{\pm}7.9$ (p<0.0001 )), total number of embryos ($6.5{\pm}4.8$ vs. $9.1{\pm}5.6$ (p<0.0001)), and good quality embryos ($2.43{\pm}1.6$ vs. $2.74{\pm}1.7$ (p=0.013)) significantly decreased in stage III-IV endometriosis than in control. But pregnancy rate of stage III-IV endometriosis was comparable with control (35.7% vs. 36.8%). Fertilization rate and number of total embryos were lower in stage I-II endometriosis than in control ($64.8{\pm}22.9$ vs. $70.8{\pm}20.8$ (p<0.0001), $7.6{\pm}5.0$ vs. $9.1{\pm}5.6$ (p<0.0001)). In patients with medical and surgical treatment of endometriosis, pregnancy rate and live birth rate was significantly lower in stage I-II than in stage III-IV endometriosis (29.2 vs. 36.2 (%), p=0.045, 23.9 vs. 31.5 (%), p=0.043). There was no difference in the mean age, but the duration of infertility was significantly longer ($56.5{\pm}26.3$ vs. $46.9{\pm}25.8$ (mon), p<0.0001) and fertilization rate was lower ($64.7{\pm}23.3$ vs. $70.5{\pm}22.7$ (%), p=0.001) in stage I-II than stage III-IV endometriosis. Conclusion: We suggest that IVF should be considered earlier in patients with minimal to mild endometriosis because of significantly decreased fertilization rates.
