애기땅빈대의 열수 추출 및 에탄올 추출물의 항산화 활성 비교 (Comparison on the Antioxidative Activity of Ethanol and Hot Water Extracts of Euphorbia supina Rafinesque)

  • Objectives : In this study, the antioxidant activities of the 80% ethanol and hot water extracts of Euphorbia supina Rafinesque were investigated. Methods : We measured total phenol contents, flavonoid contents, 2,2-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging assay. The production of reactive oxygen species was measured in LPS-induced RAW 264.7 cells using flow cytometry system. Results : The content of phenol in the hot water extract was $65.529{\pm}0.462mg/g$ and $126.932{\pm}1.894mg/g$ in the 80% ethanol extract, and that of flavonoid in the hot water extract was $16.063{\pm}0.471mg/g$ and $29.159{\pm}1.963mg/g$ in the ethanol extract. The 80% ethanol extract also showed higher DPPH and ABTS radical scavenging activities ($90.8{\pm}1.0%$ and $92.5{\pm}0.7%$) than the hot water extract ($81.5{\pm}0.5%$ and $91.5{\pm}0.2%$). The production of reactive oxygen species(ROS) was reduced dose-dependently by 80% ethanol and hot water extract at concentration of 1, 10 and $100{\mu}g/m{\ell}$ of RAW 264.7 cells. Conclusion : According to these results, the 80% ethanol extract of Euphorbia supina Rafinesque has a good anti-oxidative effects than the hot water extract. Thus, the 80% ethanol extract of Euphorbia supina Rafinesque may serve as useful natural antioxidants.

탈지들깨박 Ethanol 추출물의 항산화 효과 (Studies on Antioxidant Activity of Ethanol Extracts from Defatted Perilla Flour)

  • The antioxidant activity of ethanol extracts from defatted perilla flour was investigated by measuring peroxide value of perilla oil during storage at $45^{\circ}C$. The antioxidant activity of ethanol extracts was also compared with BHA, BHT and tocopherol. Anti-oxidant activity of ethanol extracts was also examined in corn oil and lard. The ethanol extracts contents of defatted perilla flour and the original perilla seed were 7.69 and 4.56% respectively. The antioxidant activity of ethanol extracts was superior to that of 0.02% BHT, BHA and tocopherol in the perilla oil substrate, merely in concentration of one-twentieth as much as that contained in original perilla oil seeds. The fractions of non-polar solvent (hexane and chloroform) obtained from silicic acid column chromatography are less effective than that of polar solvent as an antioxidant. Antioxidant activity of partially purified ethanol fraction is slightly inferior to that of original crude ethanol extracts. Ethanol extracts were also effective in corn oil and lard almost same as in perilla oil. The total phenolic compound contents of crude ethanol extracts and partially purified ethanol fraction were 9.3, 6.4%, respectively.

원형질 융합에 의한 전분으로부터 에탄올 발효효모균주의 개량 (Construction of Starch-assimilating and Ethanol-fermenting Yeast by Protoplast Fusion)

  • 에탄올 내성 및 고 알콜 생성 균주인 S. cerevisiae BU-1 와 glucoamylase 생성 우수 균주인 S. diastaticus A15 를 시험 균주로 하여 genetic manipulation 에 의하여 haploid 인 S. cerevisiae BUI.alpha.26($alc^r thr^-$) 와 S. diastaticus A15a6($STA^+ hom$) 균주를 분리하였다. 이등 효모 균주의 원형질체 형성을 위하여 0.8 M sorbitol 의 존재하에서 zymolyase $200\mu$g/ml 와 $400\mu$g/ml 을 2 시간 동안 처리시 두 균주 모두 95% 이상 원형질체를 얻었다. PEG 6000 을 90 분간 반응시킬 경우 융합 빈도는 $ 3.25 {\times} 10^{-3}$ 이었다. 원형질체 융합으로 생성된 융합주의 유전적 분석, 에탄올 내성, glucoamylase 생성을 측정한 결과, 이들 융합주의 유전자형은 STA $alc^r$ thr hom. '/STA' $alc^r$ thr hom로 예상되었으며, glucoamylase 활성도는 A15a6 의 경우 2.7 units 이었으나, 융합주 F7, F10 은 각각 4.2 와 8.4 units 로 나타났다. 에탄올 내성 시험을 하여 선별한 결과, 가장 우수한 균주인 융합주 7개 중 F7 과 F10 을 선별하였다. 5% 의 녹말이 포함된 발효배양액에서 $30^{\circ}C$ 5일간 배양시 생성된 에탄올 생성은 융합주 F7 과 F10 은 각각 2.0% 와 1.85 이였다.

주름조개풀(Oplismenus undulatifolius) 추출물의 생리활성 효소 억제 효과 (Inhibitory activities on biological enzymes of extracts from Oplismenus undulatifolius)

  • 주름조개풀에 함유되어 있는 phenolic contents는 water, 70% ethanol을 용매로 하여 추출하였을 때 각각 7.7, 10.1 mg/g의 함량을 나타내었다. 추출물의 phenolic 농도를 $50-200{\mu}g/mL$로 조절하여 항산화 효과를 측정한 결과, DPPH에서는 water, ethanol 추출물 $50{\mu}g/mL$ phenolics 농도에서 각각 78, 82%였고, ABTS radical decolorization는 $100{\mu}g/mL$ phenolics 농도에서 각각 92, 76%의 radical 소거능을 나타내었다. PF는 $200{\mu}g/mL$ phenolics 농도에서 water, ethanol 추출물에서 각각 1.51, 2.08 PF로 측정되었으며, TBARs값은 water, ethanol 추출물 $50{\mu}g/mL$ phenolics 농도에서 각각 84, 99%의 활성을 나타내었다. 기능성 식품 활성을 측정하는 ${\alpha}-Glucosidase$ 저해활성을 측정한 결과 ethanol 추출물 $200{\mu}g/mL$ phenolics 농도에서 44%의 억제력을 나타내었고, ${\alpha}-amylase$ 저해활성은 water 추출물 $50-200{\mu}g/mL$ phenolics 농도에서 37-88%의 효소 억제력을 나타내었다. 미용 식품 활성을 나타내는 tyrosinase 저해활성을 측정한 결과 ethanol 추출물에서 82%의 tyrosinase 저해효과를 나타내었다. 또한 elastase 저해활성을 측정한 결과 water, ethanol 추출물에서 각각 4, 61%로 나타내었고, collagenase 저해활성을 측정한 결과 water, ethanol 추출물 $200{\mu}g/mL$ phenolics 농도에서 각각 39, 67%로 ethanol 추출물에서 높은 억제력을 나타내어 우수한 저해효과를 확인하였다. Astringent 효과를 측정한 결과 water, ethanol 추출물 $200{\mu}g/mL$ phenolic 농도에서 각각 13, 32%의 astringent 효과를 나타내었다. Hyaluronidae 저해활성을 측정한 결과 ethanol 추출물 $200{\mu}g/mL$ phenolic 농도에서 46%의 Hyaluronidae 저해효과를 나타내었다.

A Study on Emissions and Catalytic Conversion Efficiency Characteristics of an Electronic Control Engine Using Ethanol Blended Gasoline as Fuels

  • In this paper, the effects of ethanol blended gasoline on emissions and their catalytic conversion efficiency characteristics were investigated in a multiple-point EFI gasoline engine, The results show that with the increase of ethanol concentration in the blended fuels, THC emissions were drastically reduced by up to thirty percent, And brake specific fuel consumption was increased, but brake specific energy consumption could be improved. However, unburned ethanol and acetaldehyde emissions increased. Pt/Rh based three-way catalysts were effective to reduce acetaldehyde emissions, but had low catalytic conversion efficiency for unburned ethanol. The effect of ethanol on CO and NOx emissions and their catalytic conversion efficiency had close relation to the engine's speed, load and air/fuel ratio. Furthermore fuels blended with thirty percent ethanol by volume could dramatically reduced THC CO and NOx emissions at idle speed.

Ethanol Fermentation in Lactose Medium Using a Fusant Strain of Saccharomyces cerevisiae and Kluyveromyces fragilis

  • The fermentative characteristics in ethanol production from lactose, with increased ethanol tolerance, of a fusant yeast strain constructed by protoplast fusion of Saccharomyces cerevisiae and Kluyveromyces fragilis were studied. The ethanol tolerance of this strain was increased to 8.0%, compared with the parent K. fragilis. During batch ethanol fermentation the optimal cultivation conditions for this fusant yeast were an initial pH of 4.5, a culture temperature $30^\circ{C}$. stirring at 100 rpm without aeration in 10% lactose medium (supplied with 1.0% yeast extract). Using this fusant strain in whey fermentation to ethanol, maximum ethanol production reached 3.41% (w/v) (theoretical yield; 66.7%) after a 48 hour cultivation period.

Rat에 있어서 ethanol투여가 체조직의 무기질 함량에 미치는 영향 (The effect of ethanol on zinc, calcium and copper concentrations of tissues in rats)

  • This study was designed in order to clarify the effect of ethanol drinking on the mineral contents on liver, kidney, muscle and hair. Forty-five rats were divided into 2 groups and a control group. The control group received tap water and the other 2 groups were given 8% and 40% ethanol as drinking source. Liver, kindney, muscle and hair samples were taken and analyzed for zinc, calcium and copper contents by atomic absorption spectrophotometric methods. The results obtained are summrized as follows; 1. The zinc content of muscle showed significant (p<0.01) decrease in both groups. 2. The calcium content of hair showed significant (p<0.1) increase in 8% ethanol group. 3. The copper contents of kidney and muscle in 8% ethanol group and liver in 40% ethanol group showed significant (p<0.1) decrease.

숙지황(Rehmannia glutinosa) 추출물의 생리활성 (Charactrization of Biological Activities of Rehmannia glutinosa Extracts)

  • 숙지황(Rehmannia glutinosa)으로부터 유용성분 추출은 40% 에탄올 추출물에서 $5.1{\pm}0.2mg/g$으로 가장 높은 phenolic compound 함량을 나타내었다. 숙지황 추출물의 항산화활성은 전자공여능의 경우 물 추출물과 40% ethanol 추출물 모두에서 85~93%의 높은 전자공여능이 확인되었으며, ABTS radical cation decolorization의 경우 물 추출물과 40% ethanol 추출물이 55~62%의 비슷한 수준의 항산화효과를 효과를 나타내었고, antioxidant protection factor (PF)는 $200{\mu}g/ml$의 농도에서 물 추출물과 40% ethanol 추출물 모두에서 1.6~1.9 PF의 높은 항산화력이 확인되었고, TBARs는 물 추출물과 40% ethanol 추출물이 비슷한 수준의 항산화효과를 효과를 나타내는 것으로 측정되었다. 숙지황 추출물의 고혈압억제효과는 물 추출물에서 87.2%, 40% ethanol 추출물에서 81.1%의 저해활성을 나타내었다. 관절염억제효과는 매우 낮게 측정 되었으며, 피부상재균에 대한 항균효과는 확인되었다. 숙지황 추출물의 tyrosinase 저해 활성은 40% ethanol 추출물에서 70%의 높은 저해효과가 확인되어 미백효과를 기대할 수 있었다. 주름 생성과 관련된 elastase 저해 효과는 물 추출물에서 76%, ethanol 추출물 57%의 높은 elastase 억제효과를 관찰할 수 있었다. 숙지황 추출물에 대한 hyaluronidase 저해활성은 40% ethanol 추출물에서만 $200{\mu}g/ml$ phenolic의 농도에서 약 5%의 매우 낮은 hyaluronidase 저해활성을 나타내었다.

알코올의 농도 및 투여 경로에 따른 위산분비 변동 (Effects of Ethanol on Gastric Acid Secretion in Anesthetized Rat)

  • It has been known that ethanol stimulates the secretion of gastric acid regardless of its route of administration. Recently, however, some studies have challenged this view and claimed that ethanol inhibits the gastric acid secretion. This study was undertaken to investigate the effects of ethanol on the gastric acid secretion in anesthetized rat in respect to the route of administration and the concentration of alcohol. Normal saline (pH adjusted to 6.0) was used as standard perfusion solution and ethanol was mixed as 0.8, 1.7, 5, 10 and 20%. Four ml of perfusion fluid was given into stomach via gastric tube and drained from duodenal tube every 5 min. Acid secretion was measured by back titration to pH 6.0 with N/20 NaOH and expressed as ${\mu}Eq/5$ min. Low concentration of ethanol up to 1.7% in perfusion solution caused little changes in acid secretion, but moderate concentration such as perfusion of 5% or 10% ethanol solution inhibited both the basal and histamine-induced gastric secretion. Moreover, loss of perfused acid was seen by 20% ethanol, which means back diffusion of hydrogen ions into the gastric mucosa. However, intravenous administration of ethanol, maintained at the level of 0.1% alcohol in blood, caused significant stimulation of gastric acid. We, therefore, conclude that in anesthetized rat ethanol has dual effects on acid secretion, i.e., inhibiting and enhancing by oral and intravenous administration, respectively, but further investigation is necessary to clarify these effects.

  • Rhodotorula sp. Y -55 효모의 중식특성을 etha nol, acetic acid 또는 acetaldehyde 상에서 batch culture로 조사하였다. 기질농도가 중가함에 따라 중식수율이 감소하고 유도기가 연장되었다. 비성장속도(${\mu}$)는 지수기에서 ethanol과 acetaldehyde의 초기농도에 따라 변화하였으나 acetic acid는 이와 상관이 없었으며 그 값은 $O.107h^{-1}$로 나타났다. 최대 비성장속도는 20g/L의 ethanol에서 $O.270h^{-1}$, O.2g/L의 acetaldehyde에서 $O.041h^{-1}$이었다. 또 최대 중식수융은 109/L의 ethanol에서 32.6%, 20g/L의 a acetic acid에서 25.6%와 O.2g/L의 acetaldehyde 에서 45%였다. 세포내 조단백질과 핵산 함량은 e­t thanol상에서 각각 41.5wt%와 4.9wt %였고 acetic acid상에서는 각각 40.2wt %와 4.7wt%로 나타났다.
