질소산화물의 선택적 환원 제거시 염화수소기체가 촉매에 미치는 영향 (The Effect of HCl Gas on Selective Catalytic Reduction of Nitrogen Oxide)

  • 본 연구에서는 소각로용 SCR(Selective Catalytic Reduction) 촉매개발의 일환으로, 소각로 배기가스 중에 다량 함유되어 있는 염화수소 (HCl) 기체가 촉매활성에 미치는 영향에 관하여 고찰하였다. 연구에 사용된 촉매는 상용 $V_2O_5-WO_3/TiO_2$ 촉매와 구리이온이 교환된 모더나이트형 제올라이트, CuHM 촉매로 실험은 수분의 유무에 따라 건조가스 조건과 습윤가스 조건으로 나누어 수행하였다. 건조가스 조건에서는 염화수소농도가 증가함에 따라 CuHM 촉매의 NO 제거활성이 가역적으로 증가하는 반면 $V_2O_5-WO_3/TiO_2$ 촉매는 비가역적으로 저하되는 상반되는 결과를 보였다. 그리고 수분이 포함된 습윤가스 조건에서는 CuHM와 $V_2O_5-WO_3/TiO_2$ 촉매 모두 비가역적으로 활성이 감소됨이 관찰되었으나, 상대적으로 CuHM이 보다 안정적인 활성을 나타내었다. 이러한 활성변화는 $NH_3$ TPD(Temperature Programmed Desorption) 탈착곡선의 증가, 감소로부터 HCl에 따른 일시적인 산량 증가 또는 영구적인 산점의 변화와 관계됨을 알 수 있었다. 그리고 BET 및 ICP 분석을 통하여 염화수소기체에 의한 촉매의 표면적과 $Cu^{{+}{+}}$$V_2O_5$ 함유량의 변화를 관찰하였다. 이상의 연구결과로부터 HCl과 같은 산가스를 함유하고 있는 배기가스 중의 질소산화물을 제거할 경우 CuHM 촉매에 대한 선택적 촉매 환원공정의 적용 가능성을 확인할 수 있었다.

세포배양에서 Cryptosporidium parvum의 발육 (Development of Cryptosporidium parvum in cell culture)

  • The purpose of this study was to establish a method for in vitro culture of C parvum isolated in Korea by determination of suitable cell model to complete development of this parasite. The result obtained were summerized as follows: 1. To determine the most suitable cell line, six types of cell line were examined by microscopy. All cell lines were infected with C parvum and showed the highest infection score in HmLu cells. 2. The staining methods including DMSO-modified acid-fast(A-F) stain, hematoxylin-eosin(H & E) stain and immunofluorescence antibody(IFA) stain were applied to examine the infection of C parvum in cell culture. These staining methods were possible to examine the infection of C parvum in cell culture. The most sensitive one was IFA staining technique. 3. Developmental stages of C parvum in HmLu cell were observed. After the initial 8 hour incubation period, some trophozoites were observed. The meronts and gametes were appeared at 24-48 hour post inoculation(PI), and oocysts were observed firstly at 48-72 hour PI. 4. In H & E stain, the parasite appeared as basophilic within parasitophorous vacuole membrane(PVM) and lying in cytoplasm at near the nucleus of the host cells. It was able to distinguish the type I, type II meronts and gametes. 5. In DMSO-modified acid-fast stain, specific stained parasites were appeared firstly after 48 hour PI. The parasites were showed with different degrees of staining bright red color within PVM. 6. The endogenous stages of parasites in HmLu cell recovered at 48, 96, 120 and 144 hour after inoculation were reacted with rabbit immunized serum in immunofluorescence antibody and avidin-biotin complex peroxidase staining technique.

Shear-induced structure and dynamics of hydrophobically modified hydroxy ethyl cellulose (hmHEC) in the presence of SDS

  • The interaction between hydrophobically modified hydroxyethyl cellulose (hmHEC), containing approximately 1 wt% side-alkyl chains of $C_{16}$, and an anionic sodium dodecyl sulphate (SDS) surfactant was investigated. For a semi-dilute solution of 0.5 wt% hmHEC, the previously observed behaviour of a maximum in solution viscosity at intermediate SDS concentrations, followed by a drop at higher SDS concentrations, until above the cmc of surfactant when the solution resembles that of the unsubstituted polymer, was confirmed. Additionally, a two-phase region containing a hydrogel phase and a water-like supernatant was found at low SDS concentrations up to 0.2 wt%, a concentration which is akin to the critical association concentration, cac, of SDS in the presence of hmHEC. Above this concentration, SDS molecules bind strongly to form mixed micellar aggregates with the polymer alkyl side-chains, thus strengthening the network junctions, resulting in the observed increase in viscosity and elastic modulus of the solution. The shear behaviour of this polymer-surfactant complex during steady and step stress experiments was examined In great detail. Between SDS concentrations of 0.2 and 0.25 wt%, the shear viscosity of the hmHEC-polymer complex network undergoes shear-induced thickening, followed by a two-stage shear-induced fracture or break-up of the network. The thickening is thought to be due to structural rearrangement, causing the network of flexible polymers to expand, enabling some polymer hydrophobic groups to be converted from intra- to inter-chain associations. At higher applied stress, a partial local break-up of the network occurs, while at even higher stress, above the critical or network yield stress, a complete fracture of the network into small microgel-like units, Is believed to occur. This second network rupture is progressive with time of shear and no steady state in viscosity was observed even after 300 s. The structure which was reformed after the cessation of shear is found to be significantly different from the original state.

  • 건조방법을 달리한 께묵(Hololeion maximowiczii) 뿌리의 색도변화에 대해서 조사하였다. Hunter value에서 분말의 경우에는 HADB가 더 진한 편이었으나 용액의 경우에는 HADB가 제일 연한 색깔을 띄었다. 수용액 색소의 420nm에서의 흡광도는 FD group이, 자외부에서 흡광도는 HADB group이 가장 강하였다. 께묵의 색소는 분말상태에서보다 용액상태에서 더 불안정하였으며, 색소분해에 대한 활성화에너지는$(90{\sim}100^{\circ}C)$ 약 20.9kcal/mole이었다.

  • 무당벌레(Harmonia axyridis)성충 더듬이에 분포하는 감각기를 주사전자현미경으로 관찰하였다. 무당벌레 성충 더듬이에서 형태적으로 크게 차이를 보이는 5종류의 감각기, $B\ddot{o}hm$, Basiconic, Chetiform, Trichoid 그리고 Coeloconic sensilla를 관찰할 수 있었다. $B\ddot{o}hm$ sensilla는 자루마디와 팔굽마디 기부안쪽의 등쪽면과 배쪽면에서만 관찰되었고, 이것은 분포된 위치로 미루어 더듬이의 움직임과 위치를 확인하는 자기반응감응성 감각기로 보여진다. Basiconic sensilla와 Chetiform sensilla는 형태적 차이에 의하여 3가지 subtype으로 나눌 수 있었으며, Trichoid sensilla는 암 수컷 더듬이 모두에서 더듬이 말단의 2마디에서만 확인되었다. 무당벌레 성충 더듬이에서 관찰된 sexual dimorphism은 더듬이 감각기의 분포와 종류가 있었는데 이것은 암컷과 수컷간의 더듬이 감각기의 분포가 다르게 나타났으며, 감각기의 다양성이 가장 높게 나타난 말단마디의 배쪽면에서 관찰된 Coeloconic sensilla는 수컷에서만 관찰되었고, Trichoid sensilla의 경우 수컷보다 암컷에서 더 많이 관찰되었다. 이러한 sexual dimorphism은 동종의 성상호간 통신에 이용되는 것으로 보여진다.

Pseudomonas sp. S-47로부터 5-Chloro-2-Hydroxymuconic Semialdehyde Dehydrogenase를 암호화하는 xylG 유전자의 클로닝 및 염기서열 분석 (Cloning and Nucleotide Sequence Analysis of xylC Gene Encoding 5C-2HMS Dehydrogenase from Pseudomonas sp. S-47.)

  • Pseudomonas sp. S-47은 xylXYZLTE 유전자에 의하여 암호화되는 효소군에 의하여 4CBA를 분해하여 5-chloro-2-hydroxymuconic semialdehyde(5C-2HMS)를 생성하는데, 본 연구에서는 이 5C-2HMS의 다음 분해과정을 확인하였다. xylXYZLTE 유전자와 5-chloro-2-hydroxymuconic semialdehyde dehydrogenase(5C-2HMSD)를 암호화하고 xylG 유전자를 포함하는 재조합 균주인 pCSS202로부터, xylG 유전자를 포함하는 재조합 플라스미드 pENV5를 만들었다. 이 플라스미드는 2-hydroxymuconic semialdehyde, 3-chloro-muconate, 2-hydroxy-6-oxohepta-2,4-dienoate, 2-hydroxy-5-methylmuconic semialdehyde와 같은 aromatic compound 에서 분해능을 나타냈으며, 그 중 5C-2HMS에서 가장 높은 분해능을 나타내었다. 또한 5C-2HMSD를 암호화하는 유전자인 xylC의 염기서열을 분석한 결과, 약 1,600 bp의 염기와 486개의 amino acid residue를 갖고있는 것을 확인하였다. P. sp. S-47의 xylG 유전자를 비교 분석한 결과 P. putida CF600, P. putida G7과 P. putida mt-2 등의 5C-2HMS dehydro-genase와 85% 이상의 amino acid homology를 보여주었다.

Effect of prenatal different auditory environment on learning ability and fearfulness in chicks

  • Objective: Early environmental enrichment in life can improve cognition in animals. The effect of prenatal auditory stimulation on learning ability and fear level in chick embryos remained unexplored. Therefore, this study investigated the effect of prenatal auditory stimulation on the learning ability and fear level of chicks. Methods: A total of 450 fertilized eggs were randomly divided into 5 groups, including control group (C), low-sound intensity music group (LM), low-sound intensity noise group (LN), high-sound intensity noise group (HN) and high-sound intensity music group (HM). From the 10th day of embryonic development until hatching, group LM and group LN received 65 to 75 dB of music and noise stimulation. Group HN and group HM received 85 to 95 dB of noise and music stimulation, and group C received no additional sound. At the end of incubation, the one-trial passive avoidance learning (PAL) task and tonic immobility (TI) tests were carried out, and the serum corticosterone (CORT) and serotonin (5-HT) concentrations were determined. Results: The results showed that compared with the group C, 65 to 75 dB of music and noise stimulation did not affect the PAL avoidance rate (p>0.05), duration of TI (p>0.05) and the concentration of CORT (p>0.05) and 5-HT (p>0.05) in chicks. However, 85 to 95 dB of music and noise stimulation could reduce duration of TI (p<0.05) and the concentration of CORT (p<0.05), but no significant effect was observed on the concentration of 5-HT (p>0.05) and PAL avoidance rate (p>0.05). Conclusion: Therefore, the prenatal auditory stimulation of 85 to 95 dB can effectively reduce the fear level of chicks while it does not affect the learning ability.

폐유리의 가수분해 반응에 의한 발포유리의 제조(III) - 안정화 열처리공정 및 Scale-up Test - (Production of Foamed Glass by Using Hydrolysis of Waste Glass(III) - Heat Treatment for Stabilization and Scale-up Test -)

  • 가수분해시킨 폐 유리를 사용하여 발포유리블록의 제조시 발포유리블록의 안정화를 위한 열처리 공정을 조사하였으며 이를 바탕으로 상업적 생산을 위한 scale-up 실험을 시도하였다. 최적의 열처리 조건은 발포소성 후 $550{\sim}600^{\circ}C$로 급냉시켜 안정화시키고 이후 $200^{\circ}C$에서 추가적으로 열응력을 해소하고 $0.3^{\circ}C/min$의 속도로 실온까지 냉각시키는 것이다. 이러한 열처리 조건을 거침으로서 기계적으로 안정성을 지닌 $250mm{\times}250mm{\times}90mm$ 크기의 발포유리가 생산 가능하였으며 이 경우 발포유리블록의 물성은 밀도 $0.28{\pm}0.06g/cm^3$, 열전도도 $0.048{\pm}0.005kcal/hm^{\circ}C$, 흡수율 $0.5{\pm}0.09vol%$, 선팽창계수$(8.6{\pm}0.2){\times}10^{-6}m/m^{\circ}C$($400^{\circ}C$ 기준), 휨강도 $15.0{\pm}0.6kg/cm^2$, 압축강도 $39.5{\pm}0.6kg/cm^2$의 특성을 갖는 것으로 확인되었다.

  • This study was performed to evaluate the mechanical properties of light-cured restorative composites by compression and indentation tests. Five commercially available light-activated composites (Heliomolar : HM, Aelitefil : AF, Amelogen : AG, Clearfil AP-X : CF, Z100 ZH) were used Disc specimens of 12mm in diameter and 1 mm in thickness and Cylindrical specimens of 3mm in diameter and 6mm in length were prepared for the indentation and compressive test, respectively. All specimens were immersed in distilled water at $37^{\circ}C$ for 30 days. An indentation test of 68-degree trigonal diamond pyramid was made under 10 g load for 15 seconds and an Knoop indentation test was made under 50 g load for 15 seconds. Hardness numbers, characteristic indentation depths and permanent deformation were measured during indentation of the 68-degree trigonal diamond pyramid compressive test was carried out at a crosshead speed of 0.5mm/min. The results obtained were summarized as follows, 1. The highest hardness value was obtained in the CF group and the lowest value was obtained in the HM group. Hardness values showed no significant, difference between AG group and HM group but other groups showed the significant differences in each group(p<0.05). 2. Hardness number by Knoop pyramid were higher than those of 68-degree trigonal diamond pyramid. 3. Plastic deformation during the indentation of 68-degree trigonal diamond pyramid was the lowest in the CF group and the largest in the HM group. Results of Tukey test showed the significant difference between CF group and others; also between ZH and AF groups and AG and HM groups(p<0.05) 4. The highest compressive strength was obtained in the CF group and the lowest compressive strength was obtained in the ZH group. Compressive strength values showed no significant difference between CF group and ZH group but other groups showed the significant differences in each group(p<0.05).

  • 1-methylnaphthalene(1-MN)과 2-methylnaphthalene(2-MN) 사이에서의 트랜스메틸화 반응을 제올라이트, H-mordenite(HM), H-Beta$(H{\beta})$, H-USY(HUSY) 촉매를 이용한 고정층 반응기에서 수행하였다. $H{\beta}_{25}(SiO_2/Al_2O_3=25)$ 촉매는 다음과 같은 조건, 반응온도 $350^{\circ}C$, 반응압력 1.5 Mpa, 전체 액상 반응물의 WHSV $2.7g_{feed}/g_{cat}{\cdot}h$, 반응물 1-MN과 2-MN의 몰비 1:1, 반응 1시간에서 2-MN/1-MN 비=2.3와 2,6-DMN/2,7-DMN 비=1.3을 보이면서 다른 촉매보다 높은 전환율과 안정성을 나타내었다. 촉매의 세공구조, 산성도와 관련하여 촉매성능을 해석하였다.
