  • A cold-adapted porcine reproductive and respiratory syndrome virus (CA-VR2332) was generated from the modified live virus strain VR2332. CA-VR2332 showed impaired growth when cultured at 37℃ with numerous mutations (S731F, E819D, G975E, and D1014N) in the hypervariable region of the NSP2, in which the mutation S731F might play a vital role in viral replication at 30℃. Conserved amino acid sequences of the GP5 protein suggests that CA-VR2332 is a promising candidate for producing an effective vaccine against PRRSV infection. Further studies on replication and immunogenicity in vivo are required to evaluate the properties of CA-VR2332.

Assessment of Contamination Level of Foodborne Pathogens in the Main Ingredients of Kimbab during the Preparing Process (김밥제조단계에서의 김밥 주원료에 대한 위해미생물의 오염도 평가)

  • Contamination levels of total aerobic bacteria, coliforms, Esherichia coli, Staphylococcus aureus, Bacillus cereus, and Listeria monocytogenes of Kimbab and its main ingredients sampled from general and specialized restaurants were compared. Total aerobic bacteria and coliform counts of Kimbab samples from both restaurant types were not significantly different (p>0.05), showing approximately $4\;to\;6\;log_{10}CFU/g$. E. coli counts were significantly higher in kimbab from general restaurants ($65%:\;0.7-2.6\;log_{10}CFU/g$) than those from specialized ones ($8.33%:\;0.70log_{10}CFU/g$), whereas those of S. aureus and B. cereus were not significantly different (p>0.05). L. monocytogenes was not detected in all Kimbab samples. These results indicate hygiene of Kimbab and its main ingredients are deleterious. Contamination levels of pathogens determined in the present study may be used as primary data for microbial risk assessment.

  • Extracellular amylase from tile filtrate of the submerged culture of Ganoderma lucidum was partially purified by ammonium sulfate precipitation and its properties were studied. The optimum pH and temperature of the enzyme activity were 5.5 and 5$0^{\circ}C$. respectively. This enzyme was most stable at pH 5.0 and stable up to 3$0^{\circ}C$, but it lost completely the activity when it was treated at 6$0^{\circ}C$ for 10 min. The enzyme was activated by the addition of M $n^{++}$, $C^{++}$ and C $u^{++}$, but inhibited by H $g^{++}$, A $g^{++}$ And various enzyme inhibitors and chemical reagents did not affect the enzyme activity. The enzyme hydrolyzed the boiled amylaceous polysaccharides, but it hydrolyzed raw starches very slowly. The activation energy of the enzyme for soluble starch was calculated and found to be 7.06 Kcal per mole. The Km values of the enzyme for soluble starch, amylose, amylopectin and glycogen were 0.16, 0.37, 0.19, and 0.16mg/$m\ell$, respectively. Maltose was found to inhibit the enzyme activity and kinetic analysis revealed a competitive type of inhibition.n.n.n.n.n.

A Growth Inhibition Effect of Saponin from Red Ginseng on Some Pathogenic Microorganisms (홍삼사포닌의 일부 병원성미생물에대한 생육억제 효과)

  • Saponin isolated from red ginseng was added to cultures of Staphylococcus aureus and Candida albicans in order to investigate saponin's influence on the growth of some pathogenic bacteria and yeasts. S. aureus and C. albicans were incubated at $38^{\circ}C\;and\;28^{\circ}C$ for 5 days with 100 rpm after addition of 0.013, 0.125, 0.500 and 1.000% (w/v, final concentration) of saponin, respectively. After incubated for 1 day, 2 days or 5 days, pH and viable cell counts of the cultures were investigated. The both of pH of S. aureus and C. albicans were decreased in concentration-dependent manner. Viable cell counts after incubation of 5 days were $1.0\;{\times}\;10^8,\;9.4\;{\times}\;10^7,\;1.0\;{\times}\;10^3$ and 0 CFU/ml, respectively, when compared with $1.8{\times}10^8\;CFU/ml$ of saponin non-treated group. Especially, 1.0% concentration of saponin inhibited completely the growth of S. aureus. While, viable cell count in C. albicans somewhat lower values than that of saponin non-treated group, but the values not significant. These results suggest that ginseng saponin inhibit the growth of S. aureus in a concentration-dependent manner, but not the growth of C. albicans.

  • The 65th Medical Brigade and Public Health Command District-Korea, in collaboration with the Migratory Bird Research Center, National Park Research Institute, conducted migratory bird tick surveillance at Sogugul and Gaerin Islands (small rocky bird nesting sites), Jeollanam-do (Province), Republic of Korea (ROK), on 30 July and 1 August 2009. Breeding seabirds captured by hands in their nesting burrows were banded, identified to species, and carefully examined for ticks during the nesting season. A total of 9 Ornithodoros sawaii larvae were removed from 4 adult Hydrobates monorhis (Swinhoe's storm petrel). The identification of the larvae of O. sawaii collected from migratory seabirds were molecularly confirmed using mitochondrial 16S rDNA primer sets.

Anti-obesity and Cholesterol-lowering Effects of Germinated Brown Rice in Rats Fed with High Fat and Cholesterol Diets (발아현미의 섭취에 의한 흰쥐의 비만 억제 및 콜레스테롤 저하 효과)

  • To investigate the anti-obesity and cholesterol-lowering effects of germinated brown rice (GBR), male Sprague-Dawley rats were randomly divided into the following 4 groups and fed with high fat and cholesterol diets for 5 weeks; control group fed with experimental diet, rice group fed with diet containing 50% rice, brown rice group fed with diet containing 50% brown rice and GBR group fed with diet containing 50% GBR. All groups showed no significant difference in body weight, but the GBR group showed the lowest value in body weight, $471.2{\pm}17.8\;g$. Body weight gain and FER of GBR group, $310.6{\pm}14.7\;g$ and $41.8{\pm}2.0%$, respectively, were also lower than those of other groups. Blood triglyceride and cholesterol levels of the GBR group were $54.0{\pm}23.4\;mg/dL$ and $64.8{\pm}14.7\;mg/dL$, respectively, which were significantly lower than those of rice groups and were the lowest values among the experimental groups. The weights of epididymal and kidney fat of GBR group also showed the lowest values compared to other groups. The liver total lipid and total cholesterol of the GBR group, $216.3{\pm}35.7\;mg/g$ liver and $16.5{\pm}0.7\;mg/g$ liver, respectively, were lower than those of other groups, and total lipid, total cholesterol and triglyceride of GBR group showed the same results. These results suggest that GBR diet reduces body weight and fat gain, and has cholesterol-lowering effect.

  • Automated driving car is drawing attention as a seminal service representing 4th industrial revolution era based on 5G network, AI, IOT and sensor technology. automated driving car is expected to evolve into the final level which does not require driver's input. Drivers are able to consume new additional time in private space. Many industries started to compete to control these time and space. Media industry is expecting quite big change due to the introduction of automated driving cars. This research examines the impact of the media industry and social & institutional issues of automated driving cars based on depth interviews of experts. The introduction of automated driving cars is giving new opportunity for media industry as contents provider. Telcos and IT corporations are expected to compete each other to get the control of infotainment systems of automated driving cars. The reform of current regulations regarding car driving is pointed as important task to protect private information and the introduction of automated driving cars.

Characteristics of Fertility on Strawberry Cultivated Soil of Plastic Film House in Chungnam Province in Korea (충남지역 시설 딸기재배 토양 비옥도 특성)

  • To reduce the dose of fertilizers is very important to sustainable production of many horticultural crops, including strawberry. In order to practice the environment friendly agriculture of strawberry cultivation in plastic film house, soil chemical properties of 435 soil samples (232 for loam, 83 for sandy loam, and 120 for silt loam) in Chungnam Province from2008 to 2009 were determined. The average of pH, EC, OM, Av. $P_2O_5$, Ex. $K^+$, Ex. $Ca^{2+}$, Ex. $Mg^{2+}$, and Ex. $Na^+$ was 6.5, 2.28 dS $m^{-1}$, 26 g $kg^{-1}$, 910 mg $kg^{-1}$, 1.09 $cmol_c\;kg^{-1}$, 8.3 $cmol_c\;kg^{-1}$, 2.5 $cmol_c\;kg^{-1}$, and 0.58 $cmol_c\;kg^{-1}$, respectively. The content of Av. $P_2O_5$ in sandy loam soil was significantly higher than silt loam soil, whereas other properties showed no difference between soil texture. The kinds of strawberry cultivars showed no difference in soil chemical properties. The frequency distribution within optimum range of soil chemical properties was 30.6%, 35.4%, 37.0%, 5.3%, 8.5%, 8.5%, and 17.9% for pH, EC, OM, Av. $P_2O_5$, Ex. $K^+$, Ex. $Ca^{2+}$, and Ex. $Mg^{2+}$, respectively. Especially, excessive portion of Av. $P_2O_5$, and Ex. $Ca^{2+}$ were high 86.9%, and 86.0%, respectively. EC values of soil samples were significantly positive correlatoin with all chemical properties except soil pH. In principle component analysis of chemical properties in soil samples, the percentage of variance explained by PC 1 was 38.8%, while PC 2 explained 17.8%of the variance, for a cumulative total of 56.6%. These results were able to distinguish between soil textures and strawberry cultivars. Also, these results considered that understanding of soil chemical properties under using principal component analysis be able to improve amounts of fertilizers for sustainable agriculture in plastic film house.

Isolation and Characterization of Lactic Acid Bacteria with Angiotensin-Converting Enzyme Inhibitory and Antioxidative Activities (안지오텐신 전환효소 저해 활성 및 항산화 활성을 가진 젖산균의 분리 및 특성)

  • In this study, we isolated and characterized plant-associated lactic acid bacteria which are able to produce angiotensin-converting enzyme (ACE) inhibitory and antioxidative activities. Five lactic acid bacteria were isolated from plants (grape and leek), a plant-associated fermentative product (Kimchi) and Korean traditional alcohol (Dongdongju). Strains K-1 and K-21 from Kimchi, strain L-5 from leek, strain G-3 from grape, and strain D-3 from Dongdongju were identified as Pediococcus pentosaceus, Lactobacillus plantarum, Weissella cibaria, L. plantarum, and L. brevis, respectively, by 16S rRNA gene analysis. ACE inhibitory activities of isolated strains ranged from 44.3 to 71.9% in the MRS broth. G-3, L-5 and K-1 strains especially showed high ACE inhibitory activities (59.8-98.69%) in the MRS broth containing skim milk. DPPH radical scavenging activities of the strains were in the range of 42.5-82.7%. All strains showed varying levels of resistance in artificial gastric fluid (pH 2.5), retaining viability ranging from 42.2 to 88.1% after 3 hr of incubation. All strains showed high resistance to 0.3% oxgall after 24 hr of incubation; survival rates were in the range of 55.4-112.8%. Isolated strains were found to be antagonistic to some pathogens including Pseudomonas aeruginosa.

A Study on UV Filters, Preservatives and Heavy Metals Contained in the Children's and Inorganic Sunscreens Distributed in Korea (국내 유통 어린이 및 무기 자외선차단제에서 자외선 차단성분, 보존제 및 중금속 실태 조사)

  • In this study, 27 inorganic sunscreens and 23 sunscreens for children were collected to investigate the use and incorporation of 17 types of sunscreen agents, 13 types of preservatives, and 5 types of heavy metals. As a result, sunscreen agents were detected in the order of titanium dioxide (41 cases), zinc oxide (29 cases), bis-ethylhexyloxyphenyltriazine (10 cases), ethylhexyl salicylate (8 cases), and ethylhexylmethoxynamate (8 cases), and preservatives were detected in order of phenoxyethanol (6 cases), benzoic acid (1 case), and dihydroacetic acid(1 case). All of the identified sunscreen agents were suitable for labeling, but phenoxyethanol, a preservative component other than labeling, was detected at a concentration of 0.1%. Heavy metals such as lead, cadmium, arsenic, antimony, and nickel were not detected or were detected at various concentrations of less than 3.6 ㎍/g. All the detected UV filters, preservatives, and heavy metals were less than the allowed maximum amount stipulated by the Regulations on Cosmetic Safety Standards in Korea.
