Cultural Condition of Pseudomonas mendocina for Polysaccharide Production (Pseudomonas mendocina 에 의한 Polysaccharide 생산)

  • The cultural condition of Pseudomonas mendocina for polysaccharide production was examined. The optimal medium contains following composition per liter of distilled water: Sucrose 23.75g, $(NH_4)_2SO_4$ 1.57g, Yeast extract 0.5g, $KH_2PO_4\;2.9g,\;MgSO_4.\;7H_2O\;1.0g,\;CaCO_3$ 2.5g. The optimum temperature and pH were $30^{\circ}C$ and 6.5. At the condition. Ps mendocina produced 5.98g/l of polysaccharide. The culture viscosity after 3 days was 191mPa.s at $70sec^{-1}$. The product yield $(Y_{p/s})$ and specific productivity $(Q_p)$ were 25.18% and 32.83mg/g-cell/h.

Classification of One-Crewmen Coastal Fishing Boats by the Fish Species caught with A Multivariate Analysis (어획어종의 다변량분석에 의한 1인승 연안어선의 분류)

  • On the basis of the seven species of fish caught by fishing boats with one crewmen belonging to the Iwawada Fisheries Cooperative of Chiba Prefecture, the fishing boats were classified by species with high market values, and the results obtained were reclassified by discriminant function. As a result, the fishing boats were classified into six groups. These six groups are : G1 featuring the main catches of yellowtails ; G2 flounders ; G3 skipjack tunas, G4 squids ; G5 demersal fish, and G6 other miscellaneous species. Furthermore, principal component analysis were carried out on fish catches of the seven species in terms of the value obtained from a catch from the scores of the first, second, third and fourth principal components. The results of analysis show that fishing boats with one crewman can be broadly classified into three groups ; i.e., Groups G1/G2, Groups G3/G4/G5 and Group G6.

  • Smart applications based on the Network of Everything also known as Internet of Everything (IoE) are increasing popularity as network connectivity requires rise further. As a result, there will be a greater need for developing 6G technologies for wireless communications in order to overcome the primary limitations of visible 5G networks. Furthermore, implementing neural networks into 6G will bring remedies for the most complex optimizing networks challenges. Future 6G mobile phone networks must handle huge applications that require data and an increasing amount of users. With a ten-year time skyline from thought to the real world, it is presently time for pondering what 6th era (6G) remote correspondence will be just before 5G application. In this article, we talk about 6G dreams to clear the street for the headway of 6G and then some. We start with the conversation of imaginative 5G organizations and afterward underline the need of exploring 6G. Treating proceeding and impending remote organization improvement in a serious way, we expect 6G to contain three critical components: cell phones super broadband, very The Web of Things (or IoT and falsely clever (artificial intelligence). The 6G project is currently in its early phases, and people everywhere must envision and come up with its conceptualization, realization, implementation, and use cases. To that aim, this article presents an environment for Presented Distributed Artificial Intelligence as-a-Services (DAIaaS) supplying in IoE and 6G applications. The case histories and the DAIaaS architecture have been evaluated in terms of from end to end latency and bandwidth consumption, use of energy, and cost savings, with suggestion to improve efficiency.

Management of Critical Control Points to Improve Microbiological Quality of Potentially Hazardous Foods Prepared by Restaurant Operations (외식업체에서 제공하는 잠재적 위험 식품의 미생물적 품질향상을 위한 중점관리점 관리방안)

  • The purpose of this study was to present management guidelines for critical control points by analyzing microbiological hazardous elements through screening Potentially Hazardous Foods (PHF) menus in an effort improve the microbiological quality of foods prepared by restaurant operations. Steamed spinach with seasoning left at room temperature presents a range of risk temperatures which microorganisms could flourish, and it exceeded all microbiological safety limits in our study. On the other hand, steamed spinach with seasoning stored in a refrigerator had Aerobic Plate Counts of $2.86{\pm}0.5{\log}\;CFU/g$ and all other microbiological tests showed that their levels were below the limit. The standard plate counts of raw materials of lettuce and tomato were $4.66{\pm}0.4{\log}\;CFU/g$ and $3.08{\pm}0.4{\log}\;CFU/g$, respectively. Upon washing, the standard plate counts were $3.12{\pm}0.6{\log}\;CFU/g$ and $2.10{\pm}0.3{\log}\;CFU/g$, respectively, but upon washing after chlorination, those were $2.23{\pm}0.3{\log}\;CFU/g$ and $0.72{\pm}0.7{\log}\;CFU/g$, respectively. The standard plate counts of baby greens, radicchio and leek were $6.02{\pm}0.5{\log}\;CFU/g$, $5.76{\pm}0.1{\log}\;CFU/g$ and $6.83{\pm}0.5{\log}\;CFU/g$, respectively. After 5 minutes of chlorination, the standard plate counts were $4.10{\pm}0.6{\log}\;CFU/g$, $5.14{\pm}0.1{\log}\;CFU/g$ and $5.30{\pm}0.3{\log}\;CFU/g$, respectively. After 10 minutes of chlorination treatment, the standard plate counts were $2.58{\pm}0.3{\log}\;CFU/g$, $4.27{\pm}0.6{\log}\;CFU/g$, and $4.18{\pm}0.5{\log}\;CFU/g$, respectively. The microbial levels decreased as the time of chlorination increased. This study showed that the microbiological quality of foods was improved with the proper practices of time-temperature control, sanitization control, seasoning control, and personal and surface sanitization control. It also presents management guidelines for the control of potentially hazardous foods at the critical control points in the process of restaurant operations.

G-Proteins Expressed in the Ocellus of the Hydromedusan, Spirocodon saltatrix.

  • We have cloned a hydromedusan opsin cDNA and showed that the deduced amino acid sequence of the cytoplasmic loop between helices 5 and 6 (loop 5-6) was clearly different from that reported so far. The amino acid sequence of the loop 5-6 is important on determination of the specificity for the coupled G- protein. To clarify which class of G-protein mediates the phototransduction system in the ocellus of the hydromedusan, we investigated G-proteins expressed in the ocellus. By PCR against the cDNA of the ocellus with primers designed according to the conserved amino acid sequence in G-protein a subunit, we obtained three kinds of cDNA fragments. Based on the sequence similarities, ttwo of them (JGI and JG3) were classified as $G_{i}$ and $G_{q}$, respectively. The other one (JG2) was a new subtype within $G_{*}$ class. Electron microscopic immunocytochemistry with the antiserum against the C-terminal sequence of $G_{q}$ or $G_{t}$ revealed the presence. of the both classes in the ocellus. The similarity of the C-terminal sequence of the JG2 with that of bovine $G_{t}$ suggests that the anti- $G_{t}$ antiserum would bind to JG2. These results suggest the possibility that the hydromedusan rhodopsin decides the specificity for the coupled G-protein by the other domain than the loop 5-6.oop 5-6.5-6.

Algae removal by Loess and coagulant (황토와 응집제에 의한 조류 제거)

  • The removal of algae was conducted by loess, aluminum sulfate and PAC on a laboratory scale. The loess was consists of organic matter 1.4%, T-N $289{\mu}g/g$, T-P $17{\mu}g/g$, $Al841.2{\mu} g/g$, Fe $592.7{\mu}g/g$, Ca $10.6{\mu}g/g$, Mg $85.5{\mu}g/g$ and Mn $6.6{\mu}g/g$. Test water was dominated by Microcystis aeruginosa. When test water was mixed with 0.01, 0.05. and 0.1 g/$\ell$ of the loess in 5 minutes, after settled in 1 hour, the removal of chl-a was 2, 22, and 36% respectively. The removal of chl-a was 69%, 70% in pH 4.5 and 9.0 and above 92% between pH 5.0 and 8.0 after the $2mg/{\ell}$ of aluminum sulfate was added. When the $2mg/{\ell}$ of PAC and $0.05g/{\ell}$ of loess were added to test water together, the removal of chl-a was 95-99% in pH 4.0-8.0, 60% in pH 9.0, and 18% in pH 10.0. The removal of chl-a was higher when loess, aluminum sulfate and PAC was used together than used alone.

  • The cultural condition for polysaccharide production by Pseudomonus detafietdii was studied. The optimal medium contains the following composition per liter of distilled water: glucose (25g/$\ell$), peptone (2.06g/$\ell$), KH$_2$PO$_4$(2g/$\ell$), MgSO$_4$.7$H_2O$ (2g/$\ell$), yeast extract (0.5g/$\ell$), CaCO$_3$(2.5g/$\ell$). The temperature and pH optimum were 3$0^{\circ}C$ and 6.5. The agitation speed was 300 rpm. 5.91g of polysaccharide was produced at the condition in flask culture.

  • Recently, mobile communication technology has new eras with supply of 5G commercial communication. 5G mobile communication service is currently supplied with city area, services will be quickly expanded to entire area of country. This mmWave based 5G mobile communication is under spreading for human communication with voice and Internet service. After completion of construction of this human communication, this technology will be expanded to industrial communications. Railway communication system is an example of this industrial communications. In this, performance of traffic transmission for railway communication network based on this 5G railway communication networks will be analyzed with computer simulations. Construction requirements of 5G railway communication networks will be suggested with this analysis results.

Analysis of Nucleotides and Their Derivatives in Renal Tissue of Rat during Ischemia by HPLC (흰쥐의 신허혈에서 HPLC를 이용한 핵산대사산물의 분석)

  • In rat kidney, the changes in concentrations of nucleotides and their derivatives during ischemia induced by renal artery ligation was measured quantitatively with high performance liquid chromatography(HPLC). After the ligation of renal artery for 60minutes, the concentrations of the nucleotides and derivatives were measured. In ischemic tissue, IDP was significantly decreased from $217.4{\pm}12.68{\mu}g$ in control to $80.7{\pm}18.39{\mu}g$ (p<0.01) ; ATP, $307.2{\pm}56.63{\mu}g$ to $47.6{\pm}5.95{\mu}g$ (p<0.01) ; ADP+AMP, $227.1{\pm}7.98{\mu}g$ to $61.4{\pm}3.92{\mu}g$(P<0.01); $NAD^+$, $217.9{\pm}4.49{\mu}g$ to $126.6{\pm}10.44{\mu}g$(P<0.01) ; GTP, $202.5{\pm}23.76{\mu}g$ to $117.7{\pm}14.24{\mu}g$ (P<0.05) ; GMP, $54.5{\pm}9.03{\mu}g$ to $23.7{\pm}0.46{\mu}g$(p<0.05), and inosine, $16.6{\pm}3.45{\mu}g$ to $7.8{\pm}0.87{\mu}g$ (P<0.05). But hypoxanthine and xanthine were significantly increased from $113.0{\pm}15.58{\mu}g$ to $159.7{\pm}12.07{\mu}g$ (P<0.05) and from $87.7{\pm}6.77{\mu}g$ to $173.1{\pm}12.52{\mu}g$ (P<0.01). In ischemic kidney, concentration of ATP was decreased to 39.9% of control at 10 minutes, 19.8% at 30 minutes, and 15.5% at 60 minutes, and ADP+AMP were decreased to 70.3% of control at 10 minutes, 67.3% at 30 minutes, and to 27.0% at 60 minutes, but hypoxanthine and xanthine were increased to 121.5% and 127.1% at 10 minutes, 126.0% and 174.4% at 30 minutes, and 141.4% and 197.3% at 60 minutes Total adenosine nucleotides were decreased to 20.3% of control during 60 minutes of ischemia, but hypoxanthine and xathine were increased to 157.5 % of control. These results suggest that the changes in the concentration of nucleotides and their metabolic derivatives are useful indices of the extents of tissue ischemia in rat kidney.

Bioethanol Production Using By-product of VPP (Value Prior to Pulping) (VPP (Value Prior to Pulping) 부산물을 이용한 바이오에탄올 생산)

  • In this study, we evaluated optimal conditions for ethanol production of the spruce hydrolysate (SH) obtained from diethyl oxalate pretreatment. Fermentable sugar concentration in SH was 29.04 g/${\ell}$ except arabinose. Monosaccharides obtained from the oligomer degradation were mainly mannose (39.26 g/${\ell}$) and galactose (12.83 g/${\ell}$). Concentration of 5-HMF and furfural which are inhibitors on ethanol fermentation were 0.09 g/${\ell}$ and 0.04 g/${\ell}$ respectively. Concentration of acetic acid and total phenolic compounds in SH were 1.4 g/${\ell}$ and 2.83 g/${\ell}$. Ethanol production using hydrolysate was 11.7 g/${\ell}$ at optimal pH 6.0 after 48 h. Specific ethanol production was 0.15 (g/(${\ell}^*h$)) at pH 5.0 and 5.5. while that was 0.24 (g/(${\ell}^*h$)) at pH 6.0. Specific ethanol production has difference depend on initial pH for fermentation. Ethanol production was 14.3 g/${\ell}$ after 48 h when xylanase 20 IU was added in SH for degradation of oligomer during fermentation. It implied that ethanol production increased by 22.2% compare with control (without xylanase).
