TNF-$\alpha$에 의해 유도된 세포부착분자의 발현에 대한 Delphinidin chloride의 억제 효과 (Delphinidin Chloride Effects on the Expression of TNF-$\alpha$ Induced Cell Adhesion Molecules)

  • 안토시아닌(Anthocyanin)은 플라보노이드계 화합물의 한 부류로 항산화, 항암 및 항궤양, 항당뇨, 중금속해독, 시력보호, 콜레스테를 저하 등의 다양한 생리활성을 가지는 것으로 보고되어 있다. 죽상경화과정은 염증성 사이토카인의 분비 또는 혈관손상으로 인한 백혈구의 부착과 이동을 통해 시작된다. 본 연구는 이러한 죽상경화의 초기과정에서 안토시아닌 혼합물 중 single compound인 delphinidin chloride (DC) 인간혈관 내피세포주(HUVEC, human umbilical vein endothelial cell line)에서 백혈구 부착과 관련이 있는 ICAM-1 (Intraceliular Adhesion Molecule-1)과 VCAM-1 (Vascular Adhesion Molecule-1) 발현에 미치는 영향에 대해 조사하였다. 세포독성이 없는 농도에서 TNF-$\alpha$에 의해 유도된 혈관 내피세포에 대한 단핵구의 부착정도를 측정하기 위해 monocyte-endothelial cell adhesion assay와 광학현미경을 이용한 형태학적 관찰을 한 결과 DC가 처리농도 의존적으로 부착을 억제하였다. 내피세포로부터 TNF-$\alpha$에 의해 유도된 세포부착 분자인 VCAM-1과 ICAM-1의 발현에 대한 영향을 western blot analysis 및 RT-PCR방법으로 비교 분석한 결과 VCAM-1과 ICAM-1의 단백질과 mRNA수준에서의 발현이 농도 의존적으로 감소되었다. 이러한 결과들을 종합해 볼 때 안토시아닌 중에서 DC를 실험한 결과 DE는 TNF-$\alpha$에 의해 유도된 내피세포의 ICAM-1과 VCAM-1 발현 억제효과를 확인할 수 있었다.

BMI와 사망과의 관련성 - 강화 코호트 연구 - (Association between BMI and Mortality - Kangwha cohort study -)

  • Objectives : To investigate the association between BMI and Mortality. Methods : This study was based on the analysis and assembly of the 'Kangwha Cohort Study', previously conducted by the Department of Preventive Medicine, Yonsei University. A total of 2,696 males and 3,595 females were followed for almost ten years and ten months from March 1985 to January 1996, a total of whom 2,420 died during this period. The Cox's proportional hazards regression model was used to analyze this data. Results : We found a U-shaped relationship between BMI and mortality among the aged men in the Kangwha cohort. The hazard ratio of dying was adjusted for age, marital status, occupation, self cognitive health level, chronic disease, smoking, and alcohol frequency, then sorted by body mass index into the following groups; less than 10.5, 18.5 to less than 21.0, 21.0 to less than 23.5, 23.5 to less than 26.0 and greater than or equal to 26. The corresponding ratios for men were 1.81(1.50-2.19, 95%CI), 1.31(1.14-1.51, 95%CI), 1.0(referent), 1.05(0.87-1.26, 95%CI) and 1.39(1.09-1.76, 95%CI), respectively. And for women, 1.46(1.19-1.78), 1.12(0.95-1.31, 95%CI), 1.0(referent), 1.00(0.84-1.20, 95%CI) and 1.09(0.89-1.34, 95%CI), respectively. Conclusions : The risk of death among aged men in Kangwha increased in the under and overweight groups. The relationship between BMl and mortality has been well studied in Western populations, but little is known about the association between BMI and mortality in our country. So, on the basis of this study, it is apparent that more studies of the relationship between BMI and mortality will be needed for future work.

품종에 따른 강낭콩 전분의 이화학적 성질비교 (Comparison on Physicochemical Properties of Korean Kidney Bean Starch according to Varieties)

  • 품종별 강낭콩 전분의 성질에서 전분입자는 타원형으로 표면은 매끄러웠고 품종에 따른 차이는 보이지 않았다. 아밀로그램의 호화개시 온도는 농도$(7{\sim}9%)$가 증가함에 따라 높아졌고, 최고 점도는 나타나지 않았으며, $92.5^{\circ}C$에서의 점도 대수값과 농도는 모두 직선적인 관계를 보였다. 물결합 능력은 $94.7{\sim}102.0%$, $70{\sim}90^{\circ}C$에서의 팽윤력과 용해도는 각각 $3.79{\sim}12.21%$$1.84{\sim}17.38%$, 아밀로오스 함량은 $31.1{\sim}32.8%$, Blue value는 $0.64{\sim}0.66%$, 비교점도는 $2.27{\sim}2.61{\;}mlg^{-1}$였고, $50{\sim}95^{\circ}C로 가열시 광투과도는 온도가 높아짐에 따라 증가하는 호화패턴으로 강낭콩 품종간에 차이를 보였다. 시차주사 열량기에 의한 호화양상에서 호화개시온도와 호화정점은 분홍색종과 적색종은 비슷하였으나 백색종이 차이를 보였고, 호화엔탈피는 $1.9{\sim}3.5{\;}cal/g$으로 분홍색종>적색종>백색종의 순으로 품종간에 차이를 보였다.

Gender-independent efficacy of mesenchymal stem cell therapy in sex hormone-deficient bone loss via immunosuppression and resident stem cell recovery

  • Osteoporosis develops with high prevalence in both postmenopausal women and hypogonadal men. Osteoporosis results in significant morbidity, but no cure has been established. Mesenchymal stem cells (MSCs) critically contribute to bone homeostasis and possess potent immunomodulatory/anti-inflammatory capability. Here, we investigated the therapeutic efficacy of using an infusion of MSCs to treat sex hormone-deficient bone loss and its underlying mechanisms. In particular, we compared the impacts of MSC cytotherapy in the two genders with the aim of examining potential gender differences. Using the gonadectomy (GNX) model, we confirmed that the osteoporotic phenotypes were substantially consistent between female and male mice. Importantly, systemic MSC transplantation (MSCT) not only rescued trabecular bone loss in GNX mice but also restored cortical bone mass and bone quality. Unexpectedly, no differences were detected between the genders. Furthermore, MSCT demonstrated an equal efficiency in rectifying the bone remodeling balance in both genders of GNX animals, as proven by the comparable recovery of bone formation and parallel normalization of bone resorption. Mechanistically, using green fluorescent protein (GFP)-based cell-tracing, we demonstrated rapid engraftment but poor inhabitation of donor MSCs in the GNX recipient bone marrow of each gender. Alternatively, MSCT uniformly reduced the $CD3^+T$-cell population and suppressed the serum levels of inflammatory cytokines in reversing female and male GNX osteoporosis, which was attributed to the ability of the MSC to induce T-cell apoptosis. Immunosuppression in the microenvironment eventually led to functional recovery of endogenous MSCs, which resulted in restored osteogenesis and normalized behavior to modulate osteoclastogenesis. Collectively, these data revealed recipient sexually monomorphic responses to MSC therapy in gonadal steroid deficiency-induced osteoporosis via immunosuppression/anti-inflammation and resident stem cell recovery.

고위험산모와 일반산모의 산후 6주간 증상에 대한 전향적 관찰 연구 - 일개 한방병원 산후조리원을 이용한 산모를 중심으로 (A Prospective Observational Study on Symptoms of the High Risk Group and Normal Group Used Postpartum Care Center during Six Weeks after Childbirth in Korean Medicine Hospital)

  • Objectives: The aim of this study was to observe the changes of women's postpartum symptoms, the quality of life and depression scale over the first six weeks after childbirth. Methods: Twenty seven mothers who received Korean medical treatment in the outpatient department treatment (from September 27th, 2017 to January 5th, 2018) were evaluated for Verbal numerical rating scale (VNRS), edema index, EuroQol Visual Analogue Scale (EQ-VAS), and Edinburgh Postnatal Depression Scale (EPDS). Results: There were 17 high risk participants (63.0%) and 10 normal participants (37.0%). The VNRS of edema is the highest in the first week, and the VNRS of joint pain is the highest from the second week to the sixth week in all patients. The Extra Cellular Water/Total Body Water (ECW/TBW) of high risk group significantly decreased from $0.403{\pm}0.011$ to $0.387{\pm}0.006$(p<0.05) in the first 2 weeks. The ECW/TBW of normal group significantly decreased from $0.393{\pm}0.070$ to $0.383{\pm}0.011$ (p<0.05) in the first 2 weeks. The EQ-VAS of high risk group increased from $64.12{\pm}13.941$ to $69.35{\pm}18.155$ (p<0.05) in the first 2 weeks. But this difference was not significant statistically (p=0.234). The EQ-VAS of normal group significantly increased from $62.50{\pm}21.763$ to $74.00{\pm}9.661$ (p<0.05) in the first 2 weeks. The difference of EPDS was not statistically significant between the first week and the sixth week in every participants. Conclusions: VNRS was the highest in edema in the first week, joint pain was the highest from the second week to six week. The edema index of high risk groups was higher than that of the normal group in the first week (p<0.05). The EQ-VAS of normal group significantly increased (p<0.05) in the first 2 weeks but high risk group didn't. In the EPDS, the ratio of nine or more points of high risk group was more than twice than normal group in the first 2 weeks.

Thinning and drilling laser-assisted hatching in thawed embryo transfer: A randomized controlled trial

  • Objective: In frozen and thawed embryos, the zona pellucida (ZP) can be damaged due to hardening. Laser-assisted hatching (LAH) of embryos can increase the pregnancy rate. This study compared thinning and drilling of the ZP before frozen embryo transfer (FET). Methods: Patients were randomly allocated into two groups for LAH using thinning or drilling on day 2 after thawing. Twenty-five percent of the ZP circumference and 50% of the ZP thickness was removed in the thinning group, and a hole $40{\mu}m$ in diameter was made in the drilling group. Results: A total of 171 in vitro fertilization/intracytoplasmic sperm injection FET cycles, including 85 cycles with drilling LAH and 86 cycles with thinning LAH, were carried out. The thinning group had a similar ${\beta}$-human chorionic gonadotropin-positive rate (38.4% vs. 29.4%), implantation rate (16.5% vs. 14.4%), clinical pregnancy rate (36.0% vs. 25.9%), miscarriage rate (5.8% vs. 2.4%), ongoing pregnancy rate (30.2% vs. 23.5%), and multiple pregnancy rate (7.0% vs. 10.6%) to the drilling LAH group. There were no significant differences in pregnancy outcomes between subgroups defined based on age (older or younger than 35 years) or ZP thickness (greater or less than $17{\mu}m$) according to the LAH method. Conclusion: The present study demonstrated that partial ZP thinning or drilling resulted in similar outcomes in implantation and pregnancy rates using thawed embryos, irrespective of women's age or ZP thickness.

Morie Topography를 이용한 피부 탄력 평가와 Cutometer 결과와의 상관성 연구 (Correlation between Cutometer and Quantitative Evaluation Using Moire Topography in Age-related Skin Elasticity)

  • 나이가 들어감에 따라 피부에 주름이 생기고 건조해지며 탄력이 감소하게 된다. 피부 탄력은 주름과 같은 다른 변화항목처럼 노화에 따른 변화를 극명하게 눈으로 확인하기 힘들어 평가 방법에 타당성을 부여하는 과정이 더욱 중요하다. 본 연구에서는 연령과 기기측정 결과인 Cutometer parameters 값 간의 상관관계를 확인하였으며, 각각의 parameters가 노화에 의한 피부 탄력 감소를 반영해준다는 사실을 확인하였다(Ur/Uf, Ur/Uf, Ur/Ue, Ua, $r=0.687{\sim}0.725)$. 또한 눈으로 보여지는 변화와 직접 피부를 만져보았을 때 느껴지는 변화를 정량평가하기 위한 평가법을 개발하였다. 50명의 건강한 여성을 대상으로 얼굴 부위에서 Moire 사진을 획득하였으며 이중 피부 탄력을 설명해줄 수 있는 다섯 단계의 Moire topographic photo scale을 선정하였다. 그 다음 동일한 조건으로 $20{\sim}61$세의 여성을 대상으로 Moire 사진을 촬영하고, 제시된 scale을 기준으로 피부 탄력정도를 평가하였다. 본 연구결과, Cutometer를 이용한 기기평가$(r=-0.687{\sim}-0.725)$와 Moire topography score (r=-938)에 의한 결과는 연령과 매우 높은 음의 상관관계를 가짐을 확인할 수 있었다. 또한 Cutometer 값과 Moire topography 점수간의 상관성도 비교 두 평가법 간의 상관관계가 높음을 알 수 있었다(r=0.711). 따라서 안면부 Moire topography를 이용한 평가는 기기측정으로 확인할 수 있었던 피부 탄력 변화를 시각적으로 나타내어 줄 수 있었다.

Pediocin 용액침지가 두부의 저장성에 미치는 영향 (Effect of Pediocin Treatment on Soybean curd Quality during Storage)

  • 본 연구는 pediocin 처리에 따른 두부의 저장성 연장 효과를 확인하고자 pediocin을 첨가한 침지액에 침지시킨 두부를 $10^{\circ}C$에서 저장하면서 두부의 품질변화를 측정하였다. pH는 무처리구의 경우 저장 5일 이후 pH가 감소하는 경향을 보였으나 첨가구에서는 pH 감소가 억제되었다. 산도의 변화는 무처리구의 경우 저장 5일 후 급격한 증가를 보인 반면에 300 ppm처리구는 저장 6일, 500 ppm 처리구는 7일 이후 빠른 증가를 보였으나 1,000 ppm 처리구에서는 급격한 증가를 보이지 않았다 저장 중 침지액 탁도의 변화는 무처리구의 경우 저장 6일 이후부터 급격한 증가를 보였고, 300, 500ppm 처리구의 경우 저장 6일 이후 서서히 증가한 반면에 1,000ppm처리구에서는 저장기간 내에 큰 증가를 보이지 않았다. 두부 침지액의 초기 일반세수는 2.50 log CFU/mL으로 처장기간이 경과함에 따라 서서히 증가하다가 저장 5일 이후부터는 빠르게 증가하는 양상을 보였다. 그러나 1,000ppm처리구의 경우 2일까지는 균수가 감소하였고, 300, 500 ppm 처리구의 경우에서는 저장 1.5일까지 균수가 감소하였다가 다시 증가함을 보였다. 대장균군의 검출을 확인 해 본 결과 무처리구는 저장 2.5일에서 검출되었고, 300 ppm과 500 ppm 처리구에서는 각각 3.5일, 5일에서 검출되었으며 1,000 ppm처리구에서는 저장 7일 이후 검출되었다. 따라서 pediocin처리는 두부의 저장성 연장에 효과가 있음을 확인하였다.

Temporomandibular joint disc plication with MITEK mini anchors: surgical outcome of 65 consecutive joint cases using a minimally invasive approach

  • Background: The purpose of this study is to introduce our modified disc plication technique using MITEK mini anchors and to evaluate the clinical outcome for patients with internal derangement (ID) of the temporomandibular joint (TMJ). Patients and methods: We evaluated 65 joints in 46 patients, comprised 32 women and 14 men, who first visited the Asan Medical Center from December 2012 to December 2016. The age of the patients ranged from 14 to 79 years, with a mean age of 36.6 years. The patients presented with joint problems including pain, joint noise, and mouth opening limitation (MOL). Patients who met our inclusion criteria underwent unilateral or bilateral disc repositioning surgery with our minimally invasive disc plication technique using MITEK mini anchors and No. 2-0 Ethibond® braided polyester sutures. The variables taken into account in this study were the range of maximum mouth opening (MMO), painful symptoms (evaluated with the visual analog scale, VAS), and the type of noise (click, popping, crepitus) in the TMJ. Results: Preoperative examination revealed painful symptoms in 50.7% (n = 35) of the operated joints (n = 69) and the presence of clicks in 56.5% (n = 39). Postoperative examination revealed that 4.3% (n = 3) of the operated joints had painful symptoms with lower intensity than that in the preoperative condition. Additionally, 17.4% (n = 12) had residual noise in the TMJ, among which two were clicking and the other 10 had mild crepitus. The intensity of the postoperative residual noise was significantly decreased in all cases compared to that in the preoperative condition. Among patients with MOL below 38 mm (n = 18), the mean MMO was 31.4 mm preoperatively and 44.2 mm at 6 months postoperatively, with a mean increase of 13.8 mm. A barely visible scar at the operation site was noted during the postoperative observation period, with no significant complications such as facial palsy or permanent occlusal disharmony. Conclusion: Subjective symptoms in all patients improved following the surgery. TMJ disc plication using MITEK mini anchors with our minimally invasive approach may be a feasible and effective surgical option for treating TMJ ID patients who are not responsive to conservative treatment.

감초 신품종과 약전 수재 감초 추출물의 항산화 활성 및 암세포 독성 비교 연구 (Antioxidant Activity and Cytotoxicity against Human Cancer Cells of Glycyrrhiza New Varieties : A Comparison with Glycyrrhiza Official Compendia)

  • Objectives : The Glycyrrhiza new varieties, WONGAM and SINWONGAM, were developed through interspecific cross between Glycyrrhiza glabra and Glycyrrhiza uralensis by the National Institute of Horticultural and Herbal Science, Rural Development Administration in Korea. This in vitro study was undertaken to compare the antioxidant and cytotoxic effects between Glycyrrhiza new varieties (WONGAM and SINWONGAM) and official compendia (Glycyrrhiza glabra and Glycyrrhiza uralensis). Methods : Antioxidant activity was determined by DPPH (1,1-diphenyl-2-picrylhy drazyl), ABTS (2,2-azino-bis (3-rthylbenz-thiazoline-6-sulfonic acid)) diammonium salt, Nitrite radical scavenging assay, and Reducing Power assay. Cytotoxicity was determined by MTT assay and cell morphology was observed by an inverted microscope. Results : The DPPH, ABTS, Nitrite radical scavenging activities and reducing power of Glycyrrhiza glabra, Glycyrrhiza uralensis, WONGAM, and SINWONGAM were evaluated at different concentrations (0, 10, 50, 100, 500, 1000 ㎍/㎖). Glycyrrhiza glabra, Glycyrrhiza uralensis, WONGAM, and SINWONGAM showed similar dose-dependent increase in antioxidant activities. The cytotoxic effects with increasing doses of Glycyrrhiza new varieties and official compendia did not differ in HCT116, HT29, A549, MDA-MB231, PC3, ACHN, and HeLa cells. However, significant difference in cytotoxicity were observed in AGS, MCF7 and Hep3B cells by Glycyrrhiza glabra, Glycyrrhiza uralensis, WONGAM, and SINWONGAM. Conclusions : These results showed that Glycyrrhiza new varieties and official compendia acts as a potent antioxidant. Also, the finding that equivalent cytotoxic potency was observed in a cell dependent manner. Our study suggests that Glycyrrhiza new varieties may offer a wide-variety of health benefits.
