Inhibitory Effect of Curcuma longa L. Extracts on 5-Alpha Reductase II Activity (울금 추출물의 5-Alpha Reductase II 활성 저해 효과)

  • 5-Alpha reductase II, which converts testosterone (T) to dihydrotestosterone (DHT), is a crucial enzyme II in benign prostatic hyperplasia. Inhibitory effects of Curcuma longa L. (CL) extracts on 5-alpha reductase II activity were investigated in rat prostate tissue homogenates as well as LNCaP cells expressing human 5-alpha reductase II. Hot water extract (CL-HW) of Curcuma longa L. significantly inhibited 5-alpha reductase activity by over 80% at a concentration of $100{\mu}g/mL$, whereas 20% ethanol extracts (CL-E20) of Curcuma longa L. exhibited significant inhibitory activity from $50{\mu}g/mL$. These results indicated that Curcuma longa L is a potent 5-alpha reductase II inhibitor for benign prostatic hyperplasia treatment.

Interaction of dietary Mg(II), Ca(II) and polyunsaturated fatty acids on 3-Hydroxy-3-Methylalutaryl Co-A reductase activity (불포화 지방산과 마그네슘, 칼슘과의 상호작용이 3-Hydroxy-3-Methylglutaryl Co-A reductase 활성에 미치는 영향)

  • Interaction of dietary Magnesium, Calcium and Polyunsaturated fatty acid(vegetable oils)on 3-Hydroxy-3-Methylglutaryl Co-A reductase activity was studied for a period of 30 days using isocalories and isonitrogenous as a basal diet . The subject rabbits were divided into 18 feeding groups. The results are summarized as follows: 1. The ratio of ${\alpha}_1-lipoprotein$ par lipalbumin is 0.34 for control group, 0.38 the highest group fed 0.1 Mg(II) 10ml plus perilla oil and basal diet, the lowest 0.25 group fed 0.1M Mg(II) 5ml plus sesame oil and basal diet. 2. The ratio ratio of ${\alpha}_2-lipoprotein$ per lipalbumin is 0.64 for control group. 0.95 as the highest for the group fed 0.1M Ca(II) 15ml plus sesame oil and basal diet, 0.1M Ca(II) 5ml plus perilla oil and basal diet. 3. The ratio of ${\beta}-lipoprotein$ per lipalbumin is 0.71 for control group, the highest 0.81 for the groups fed 0.1M Mg(II) 10ml plus sesame oil and basal diet, the lowest 0.37 for the group fed 0.1M Mg(II) 15ml plus soybean oil and basal diet. 4. In serum triglyceride, control group was 129.5mg%, the highest 155.4mg% for the group fed 0.1M Ca(II) 5ml plus sesame oil and basal diet, the lowest 85.7mg% for the group fed 0.1M Mg(II) 10ml plus soybean oil and basal diet. 5. In serum cholesterol, control group was 96.7mg%, the highest 152.5mg% for the group fed 0.1M Ca(II) 10ml plus sesame oil and basal diet, the lowest 80.5mg% for the group fed 0.1M Mg(II) 15ml plus soybean oil and basal diet. 6. In case of HMG-CoA reductase activity, control group was 0.95, the highest 0.98 for the group fed 0.1M Ca(II) plus soybean oil and basal diet. 7. Interaction between metal(II) ions and polyunsaturated fatty acid(vegetable oil) are soybean oil>sesame oil>perilla oil, for Mg(II). soybean oil>perilla oil>sesame oil, for Ca(II). Therefore, it is invetigated that the interaction between metal ion and polyunsaturated fatty acid is the higher, the cholesterol level is the lower, and HMG-CoA reductase activity is increased.

  • We investigated the effect of peanut sprout extract (PSE) on andropause symptoms. PSE was evaluated for impotency and benign prostatic hyperplasia via phosphodiesterase (PDE) or 5-alpha reductase II inhibition assay. Inhibition of PDE and 5-alpha reductase II activities in PSE was significantly higher than that of the non-treated control. To investigate the effects of testosterone levels by PSE, we performed cell media test using TM3 cells. Production of testosterone in TM3 cells was elevated by PSE. These results indicate that PSE was able to alleviate andropause symptoms.

  • It was our working hypothesis that introduction of 11-keto groups to 12-oleanene/ursene series of triterpenoids should endow them with corticoid-like activities, since pharmacological actions of glycyrrhetinic acid (GA) are known to be caused by inhibition on $corticoid-{\delta}^4-reductase$. 11-Keto-triterpenoids derived artificially in these studies, such as 11, 19-diketo-18, 19-secoursolic acid methyl ester(I), $11-keto-{\beta}-boswellic$ acid derivatives (IIa-IIc), 11-Keto-presenegenin dimethyl ester (III), II-keto-oleanolic acid derivatives (IVa-IVd) and 11-keto-hederagenin (V) possess the fundamental functions of ${\alpha},\;{\beta}-unsaturated$ ketone on C-11 and hydroxyl group on C-3, as like GA (VI). Additionally, they involve the carboxyl groups on rings A (II, III), D (I, III, IV, V) and E (VI), and the hydroxyl groups on rings A (III, V) and C (III). All the compounds competitively inhibited $corticoid-5{\beta}-reductase$, and the highest inhibitory potency appeared in I. All of them except $3,\;11-diketo-{\beta}-boswellic$ acid methyl ester (IIc) were more effective about five times to twice than GA. On carrageenin-induced edema test, compounds I and IVa-IVd showed anti-inflammatory activities, but III enhanced rather edema. Structure-activity relations were found in the aspects of hydrophilicity of ring A and hydrophobicity of rings C/D. The more they were hydrophilic in ring A and hydrophobic in rings C/D, the more they inhibited the enzyme. And the more they were hydrophobic in rings C/D, the more they exhibited antiiflammatory activities. However, the increased hydrophilicity in ring A resulted in increasing edema, probably due to a nonspecific inhibition on $aldosterone-5{\beta}-reductase$.

Cosmetic Potency of Puerariae Radix in Dermal Fibroblasts

  • Interaction between epidermis and dermis plays an important role in wound healing and hair follicle formation. This study focused on investigating the potency of ethanol extract of Puerariae Radix (EPR) as cosmetic ingredient using human dermal fibroblasts (hDFn). Our results revealed that EPR suppressed collagenase activity dose-dependently. EPR inhibited activity of $5{\alpha}$-reductase I and II at the final concentration of $25{\mu}g/ml$ in hDFn cells. Also, EPR promoted the proliferation and the ERK activation of cells. ERK phosphorylation by EPR was blocked by specific inhibitor of ERK, PD98059. EPR-induced cell proliferation was blocked by PD98059. This means that EPR could promote the proliferation of hDFn cells via the activation ERK. Collectively, these results suggest that EPR may be used as a new cosmetic ingredient.

The Experimental Study on the Effect of Herbal Exrtacts on Hair Growth and Acnes (복분자(覆盆子), 석창포(石菖蒲), 상침자 및 숙지황(熟地黃)이 모발성장(毛髮成長)과 면포에 미치는 실험적(實驗的) 연구(硏究))

  • The effects of four extracts of medicinal herbs, Rubus coreanum, Acorus calamus, Morus alba and Rehmannia glutinosa on hair growth activity and acnes control were investigated. In the course of screening natural extracts for hair growth, we found that the extract of dried root of Rubus coreanum has the hair growth promoting effect. After topical application of these extracts to the back of C57BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. The growth of dermal papilla cells and mouse vibrissae hair follicle cultured in vitro, however, was not affected by treatment of these extracts. Furthermore these extracts do not possesspotent inhibitory effect on $5{\alpha}-reductase$ I and II activity and anti-bacterial effect on Escherichia coli , Propionibacterium acnes, Pityrosporum ovale, Staphylococcus aureus, Staphylococcus epidemidis, and Candida albicans. RT-PCR analysis showed that these extracts did notinduce mRNA levels of growth factors such as insulin-like growth factor-I, keratinocyte growth factor, hepatocyte growth factor and vascular endothelial growth factor in dermal papilla cells. These results suggest that Rubus coreanum has hair growth promoting effect. However, the effects of these materials on the hair growth promotion are not mediated through inhibition of $5{\alpha}-reductase$ I and II activity, stimulation of hair follicle cells and expression of growth factors in the dermal papilla cells.

Finasteride Increases the Expression of Hemoxygenase-1 (HO-1) and NF-E2-Related Factor-2 (Nrf2) Proteins in PC-3 Cells: Implication of Finasteride-Mediated High-Grade Prostate Tumor Occurrence

  • A number of naturally-occurring or synthetic chemicals have been reported to exhibit prostate chemopreventive effects. Synthetic $5{\alpha}$-reductase (5-AR) inhibitors, e.g. finasteride and durasteride, gained special interests as possible prostate chemopreventive agents. Indeed, two large-scale epidemiological studies have demonstrated that finasteride or durasteride significantly reduced the incidence of prostate cancer formation in men. However, these studies have raised an unexpected concern; finasteride and durasteride increased the occurrence of aggressive prostate tumor formation. In the present study, we have observed that treatment of finasteride did not affect the growth of androgen-refractory PC-3 prostate cancer cells. Finasteride also failed to induce apoptosis or affect the expression of proto-oncogenes in PC-3 cells. Interestingly, we found that treatment of finasteride induced the expression of Nrf2 and HO-1 proteins in PC-3 cells. In particular, basal level of Nrf2 protein was higher in androgen-refractory prostate cancer cells, e.g. DU-145 and PC-3 cells, compared with androgen-responsive prostate cancer cells, e.g. LNCaP cells. Also, treatment of finasteride resulted in a selective induction of Nrf2 protein in DU-145 and PC-3 cells, but not in LNCaP cells. In view of the fact that upregulation of Nrf2-mediated phase II cytoprotective enzymes contribute to attenuating tumor promotion in normal cells, but, on the other hand, confers a selective advantage for cancer cells to proliferate and survive against chemical carcinogenesis and other forms of toxicity, we propose that finasteride-mediated induction of Nrf2 protein might be responsible, at least in part, for an increased risk of high-grade prostate tumor formation in men.

The Study about the Comparison of Korean-Western Medicine on Hair (모발에 대한 동서의학적 고찰)

  • Objectives : The purpose of this study is to compare hair and alopecia of Korean Medicine with those of western medicine. Methods : We studied relationships between hair and essense(精), qi(氣), blood(血), five vicera(五臟) and meridians(經絡) through literature review about hair and alopecia. We compared Korean medicine with western medicine on physiology of hair and treatment of alopecia based on the study. Results & Conclusion : 1. Congenital essense(先天之精) is related with genetic factor and acquired essence(後天之精) is connected with nutritional factor. Defending function of Defense qi(衛氣) is related with immune reaction and qi stagnation(氣鬱) is associated with stress reaction. Atrophy of vascular tissues observed in alopecia scalp means deep relationship between blood(血) and alopecia, further deficiency of blood can cause telogen effluvium. 2. Kidney qi(腎氣) is related with inhibiting combination of Androgen receptor and Dihydrotestosterone(DHT) or activating hair growth factior. Pi(脾) is connected with alopecia seborrheica caused by damp-heat(濕熱) and alopecia areata caused by excessive prudence(思慮過度). Heart(心) is associated with atrophy in vascular tissue of scalp and liver(肝) is connected with metabolism. 3. Armpit hair and pubic hair as secondary sex characteristics are realated with Yangming Meridian(陽明經) and beard and hair at crown part where Type II $5{\alpha}$-reductase is activated much are associated with Taiyang Meridian(太陽經). 4. Juglandis Semen pharmacopuncture and Ganoderma lucidum pharmacopucture have better effects on inhibit $5{\alpha}$-reductase than Finasteride. Minoxidil and PRP are similar with promoting blood flow and removing stasis(活血祛瘀). Seven-star needling(七星針) is similar with microneedling. 5. Alopecia can be caused by due to lack of circulation Views we need solution to improve circulation coincide in opinion with Korean Medicine and Western medicine.

Suppressive Effects of Defatted Green Tea Seed Ethanol Extract on Cancer Cell Proliferation in HepG2 Cells (HepG2 Cell에서 녹차씨박 에탄올 추출물의 암세포 증식 억제효과)

  • Defatted green tea seed was extracted with 100% ethanol for 4 hr and then fractionated with petroleum ether, ethyl acetate and butanol. The ethanol and butanol extracts showed greater increases in antiproliferation potential against liver cancer cells than petroleum ether, ethyl acetate, $H_2O$, and hot water extracts did. Thus, this study was carried out to investigate the anti-proliferative actions of defatted green tea seed ethanol extract (DGTSE) in HepG2 cancer cells. The DGTSE contained catechins including EGC ($1039.1{\pm}15.2\;g/g$), tannic acid ($683.5{\pm}17.61\;{\mu}g/g$), EC ($62.4{\pm}5.00\;{\mu}g/g$), ECG ($24.4{\pm}7.81\;{\mu}g/g$), EGCG ($20.9{\pm}0.96\;{\mu}g/g$) and gallic acid ($2.4{\pm}0.68\;{\mu}g/g$), but caffeic acid was not detected when analyzed by HPLC. The anti-proliferation effect of DGTSE toward HepG2 cells was 83.13% when treated at $10\;{\mu}g$/mL, of DGTSE, offering an $IC_{50}$ of $6.58\;{\mu}g$/mL. DGTSE decreased CYP1A1 and CYP1A2 protein expressions in a dose-dependent manner. Quinone reductase and antioxidant response element (ARE)-luciferase activities were increased about 2.6 and 1.94-fold at a concentration of $20\;{\mu}g$/mL compared to a control group, respectively. Enhancement of phase II enzyme activity by DGTSE was shown to be mediated via interaction with ARE sequences in genes encoding the phase enzymes. DGTSE significantly (p<0.05) suppressed prostaglandin $E_2$ level, tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) protein expressions, and NF${\kappa}$B translocation, but did not affected nitric oxide production. From the above results, it is concluded that DGTSE may ameliorate tumor and inflammatory reactions through the elevation of phase II enzyme activities and suppression of NF${\kappa}$B translocation and TNF-${\alpha}$ protein expressions, which support the cancer cell anti-proliferative effects of DGTSE in HepG2 cells.

Hepatoprotective Activity of Dandelion (Taraxacum officinale) Water Extract against D-Galactosamine-Induced Hepatitis in Rats (D-Galactosamine으로 유발된 간손상에 대한 민들레 열수추출물의 예방효과)

  • This study aimed to investigate the protective effect of dandelion water extract (DWE) on liver injury induced by D-galactosamine (GalN) in Sprague-Dawley rats. Fifty rats were divided into 5 groups; normal control (C), DWE-control (DWE-C: saline injection after feeding 3% DWE diet), GalN-control (GalN-C: GalN injection after normal diet), DWE I (GalN injection after feeding 1.5% DWE diet), and DWE II (GalN injection after feeding 3% DWE diet). After 2 weeks, the acute hepatitis was induced by GalN (650 mg/kg, i.p.) and 24 hrs later, all rats were sacrificed. The DWE supplement ameliorated the serum alanine and aspartate aminotransferase (AST, ALT) as well as alkaline phosphatase (ALP) and tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$. Hepatic antioxidative enzyme activities, such as catalase, GSH peroxidase, GSH reductase, and Mn-superoxide dismutase (SOD) were slightly or significantly elevated by the treatment of DWE. Moreover, the histological examination corresponded with these biochemical observations. According to these findings, dandelion could be used as a potential therapeutic material for treating chemically induced acute hepatitis.
