Skin Protective Effect of Methylated Marliolide through Induction of NRF2/ARE (메틸말리올라이드의 NRF2/ARE 유도를 통한 피부 세포 보호 효과)

  • In the present study, we have investigated whether methylated marliolide could induce NRF2 thereby exerting anti-oxidant effects. MTT assay showed that methylated marliolide did not exhibit cytotoxicity on HaCaT cells. Methylated marliolide induced a higher ARE-dependent luciferase activation in HaCaT ARE-GFP-luciferase cells, compared with resveratrol. In addition, exposure of methylated marliolide to HaCaT cells significantly induced NRF2 and transcriptionally activated HO-1 and NQO1, both of which are target genes of NRF2. Finally, methylated marliolide protected HaCaT cells against TPA-induced oxidative damages on nucleotides and lipids. Together, results shows that methylated marliolide could suppress oxidative damages through induction of NRF2 which implies that methylated marliolide might serve as a good candidate for novel cosmetic ingredient with anti-oxidant effects.

Antimutagenic and Cytotoxic Effects of Aster scaber Root Ethanol Extract (참취뿌리 에탄올추출물의 항돌연변이성 및 암세포 성장억제효과)

  • This study was performed to determine the antimutagenic and cytotoxic effect of Aster scaber root ethanol extract on Salmonella typhymurium TA98, TA100 and cancer cell lines using Ames test and cytotoxicity assay, respectively. Cancer cell lines include chronic myelogenous leukemia(K562), human gastric carcinoma(KATOIII), human hepatocellular carcinoma(Hep3B) and human breast adenocarcinoma(MCF-7). Futher fractionations with hexane, chloroform, ethyl acetate, butanol and water from ethanol extract of Aster scaber root were performed to obtain effective fraction. Ethanol extract and ethyl acetate fraction showed 79% and 82% inhibitory effect on the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) against TA100, while 48% and 60% inhibition was observed on the mutagenesis induced by 4-nitroquinoline-l-oxide(4NQO) against TA98. In the meanwhile, ethyl acetate fraction showed 78% and 85% inhibitory effect on the mutagenesis induced by benzo(${\alpha}$)pyrene[B(${\alpha}$)P] against TA98 and TA100, respectively, while 83% inhibition was observed on the mutagenesis induced by 3-amino-l,4-dimethyl-5H-pyrido(4,3-b) indole(Trp-P-1) against TA98. Ethyl acetate fraction (0.125 mg/ml) showed the strongest cytotoxic effect against K562, KATOIII, Hep3B and MCF-7 at the same concentration compared to those of other fractions. Ethanol extract and water fraction showed the least inhibitory effect.

Dough Characteristics and Biological Effects of Mixed Flour of Buckwheat and Wheat (메밀 혼합분의 반죽특성과 생리활성 검색)

  • This study was to investigate the mixed buckwheat flour quality by observing antimutagenic and cytotoxic effects of mixed buckwheat flour extracts using Ames test and SRB (sulforhodamine B) assay. Samples were prepared to the ratio of 100% (B1), 90% (BF1), 80% (BF2), 70% (BF3) and 60% (BF4) (w/w) flour buckwheat based on wheat flour weight. The initial pasting temperature in an amylograph was increased according to the increase of the buckwheat flour. The water absorption in farinograph decreased with the addition of buckwheat flour. The inhibition rates of B1, BF3 and BF4 extract (160 g/plate) were 45%, 37.3% and 42% against the mutagenesis of Salmonella Typhimurium TA100 induced by MNNG $(0.4{\mu}g/plate)$, respectively. In addition, the B1 at the same concentration showed 64% and 44.3% inhibition on the mutagenesis of Salmonella Typhimurium TA98 and TA100 induced by 4NQO $(0.15{\mu}g/plate)$, respectively. In SRB assay, human breast adenocarcinoma (MCF 7), human hepatocellular carcinoma (Hep3B), human stomach adonocarcinoma (AGS), human lung carcinoma (A549) and human cervical adenocarcinoma (HeLa) proliferations were inhibited by the increase in the sample concentration.

A Study on the Effect of Indomethacin on Distribution of Langerhans Cells and T-lym-phocytes in 4-Nitroquinoline-1-oxide(4NQO) induced Carcinogenesis at the Palate and Tongue of Albino Rat (4-Nitroquinoline-1-oxide(4NQO)유도 백서 구강암 발암과정에서 Indomethacin 이 Langerhans 세포와 T-임파구의 발현에 미치는 영향에 관한 연구)

  • This study was undertaken to investigate the effect of indomethacin on the distribution of Langerhans cells and T-lymphocytes related with immune response of 4-Nitroquinoline-1-oxide induced carcinogenesis at the palate and tongue of albino rat. 54 Sprague-Dawley strain 10 weeks old albino rats, about 150gm weighted, divided into a normal group of 6 rats without treatment, a control group of 12 rats given indomethacin, a carcinogenesis group of 18 whose palatal mucosa were appiled with 4-Nitroquinoline-1-oxide three times a week, and experimental group of 18 rats were treated with indomethacin and whose palatal mucosa were applied 4-Nitroquinoline-1-oxide. All these 54 rats were subjected to be observed as being ATPase stained specimens, specimens for the observation of light and electron microscope, and T-lymphocyte stained specimens. The obtained results were summarized as follows; 1. In carcinogenesis group, proliferation of epithelial layer and rete peg were observed early period of the experiment and showed parakeratosis, individual cell keratinization, acanthosis, and lymphocyte infiltration from 13th week of the experiment on lightmicroscopically, while experimental group showed less reaction than that of carcinagenesis group. 2. The number of Langerhans cells in normal group rarely changed until 21st week of the experiment, while the Langerhans cells increased markedly from 3rd week of the experiment in control group. 3. The number of Langerhans cells were decreased markedly and persistantly until 21st week of the experiment both in carcinogenesis and experimental groups. 4. Appearance of the T-helper cells and T-suppressor cells were minimal and irregullar in number both in normal and control groups. Thus it is assumed that administration of indomethacin and distribution of Langerhans cells showed close relation. 5. In carcinogenesis and experimental groups, the number of the T-helper cells was apparently inereased than that of the T-suppressor cells, but increasing pattern in experimental group was less than in carcinogenesis group. These cells increased most in the 21st week, decreased from the 23rd week and the appearance of these cells were irregular in general throughout the experiment.

  • Antimutagenic effect of Doenjang (Korean soy paste) on various carcinogens in Salmonella typhimurium strains of TA98 and TA100 were studied. By the addition of methanol extract of Doenjang to aflatoxin B₁(AFB₁)in the experimental system, the mutagenicity of AFB₁ on the strains of TA98 and TA100 was com-pletely inhibited. The methenol extract of the Doenjang also inhibited the mutagencities induced by direct mutagens such as N-methy1-N'-nitro-N-nitroguanidine(MNNG)and 4-nitroquinoline-1-oxide(4-NQO), and another indirect mutagens of benzo(a) pyrene(BaP) and dimethylnitrosamine(DMN). From the solvents and thin layer chromatographic(TLC) fractionations, free fatty acid(s), especially linoleic acid in Doenjang seemed to be one of the active antimutagenic compounds.

Studies on Antimutagenic and Cytotoxic Effects of Seaweeds Protein-Polysaccharides

  • Polysaccharide content in protein-polysaccharides (PPS) extracted from sea mustard, sea tangle and fusiform were 40.61, 38.42 and 52.80% , respectively. 5% of sea tangle PPS showed highest inhibitory activity on 4-nitorquinoline -1-oxide(4-NQO) against Salmonella typhimurium TA100 compared to the other seaweed PPS. 5% of sea mustard PPS showed highest inhibition ration of 62% on 2-nitrofluorene(2-NF)against Salmonella typhimurium TA98. These PPS extracts showed cytotoxic activity against human colon cancer cell (SW-480), and showed mild cytotoxic activity on human stomach cancer cell(SNU-1) and human hepatic cancer cell(HepG 2).

  • The regulation of DNA repair genes expression was investigated using fused genes, in which the promoter of repair genes was hybridized with the lacZ structural gene. The activities of beta-galactosidase expressed from the fused gense were highly increased when the host cells were exposed to methylating agents, such as methyl methansulfonate (MMS), N-methyl-N'-nitro-nitrosoguanidine (MNNG) and methyl nitrosourea (MNU). On the other hand, the enzyme activities from the fused genes were not induced when the cells were treated with ethylating or nonalkylating agents, such as ethyl methansulfonate (EMS), 4-nitroquinoline-1-oxide (4NQO), Bleomycin, and Benzo(a)pyrene (BP).

온천수의 생리활성 검색

  • 예로부터 류마티스 및 습진과 같은 피부병 등에 치료 효과가 있어 민간요법으로 널리 이용하여 왔던 천연수 중 하나인 온천수에 대하여 성분 분석 및 각종 생리활성에 대한 연구를 통하여 그 효과를 검증하고자 하였다. 943m에서 취수된 경산 사이판 온천수에 함유되어 있는 각종 성분을 분석한 결과 17종류의 성분이 검출되었으며 그중 무기물이 11종이 분석이 되었고 다른 온천수와 비교하여 K이나 Na가 비교적 많이 함유되어 있는 것이 특징이었다. 각종 발암성 물질에 대하여 경산 사이판 온천수의 돌연변이 억제활성을 검토하기 위해서 Samonella typhimurium TA98을 이용하여 Ames test를 행하였다. 그 결과 직접변이원인 4NQO에서는 40%의 억제 효과를 나타내었고 간접변이원인 Trp-P-1과 B(a)P은 각각 52.4%, 43.1%의 비교적 낮은 억제활성을 보였다. 또한 경산 사이판 온천수의 항암활성 규명을 위해 인간 암세포주를 이용한 SRB assay를 행하였는데 폐암세포인 A549에서는 66%, 위암세포인 AGS에서 45.6%, 유방암세포인 MCF-7에서 37%, 자궁암세포인 HeLa에서는 47.5%, 그리고 간암세포인 Hep3B에서는 52.7%의 억제율을 보였다.

Antimutagenic Effects of Enzymatic Browning Reaction Products of polyphenol Compounds by polyphenoloxidase derived from Mushroom(Agaricus bisporus) (양송이 유래 Polyphenoloxidase에 의한 Polyphenol 화합물의 효소적 갈변생성물의 돌연변이 억제효과)

  • The antimutagenic effects of enzymatic browning reaction products (MEBRPs) of polyphenol compounds (catechol, homocatechol, hydroxyhydroquinone, pyrogallol) by enzyme extracted from mushroom (Agaricus bisporus) were demonstrated through spore rec-assay using B. subtilis $H17(rec^+)$ and $M45(rec^-)$, Ames test using S. typhimurium TA98 and TA100 and SOS chromotest using E. coli PQ37/plasmid pKM101. In spore rec-assay, the MEBRPs showed antimutagenic effects by decreasing of the inhibition zone induced by MNNG. In Ames test with S-9mix in both TA98 and TA100, all of MEBRPs showed strong antimutagenic effects of about 21 to 99% against mutation by $B({\alpha})P$ and Trp-P-1, as adding $300\;{\mu}l$ of the MEBRPs. In SOS chromotest, MEBRPs showed antimutagenic effects by inhibiting the SOS-inducing function induced by 4NQO and MMC, as increasing in concentration of the MEBRPs. But they did not showed mutagenicity in these bacterial assays.

Enhanced production of cellulase by a mutant strain of aspergillus phoenicis (Aspergillus phoenicis의 한 돌연변이주에 의한 cellulase의 생성 및 그 특성)

  • Mutational experiments were performed to imporve the cellulase productivity of Aspergillus phoenicis KU175, isolated from the southern part of Korea, as a high cellulase producer. By treatment ultra-violet light nad 4-NQO(4-Nitroquinoline-N-Oxide), mutation waas induced, and treatment ultra-violet light and 4-NQO (4-Nitroquinoline-N-Oxide), mutation was induced, and A.phoenicis KU175-115 was finally selected for its highest avicelase production. Avicelase production of the mutant was increased about 2 times compared with those of the wild strain. However, activities of other hydrolytic enzymes, such as amylase, protease and nuclease, of the mutant strain didn't show a marked difference compared with those of the nuclease, of the mutant strain didn't show a marked difference compared with the wild strain, except slight increase in ribonuclease activity and slight decrease in glucoamylase activity. Avicelases from the mutant strain selected were purified from wheat bran culture by successive salting out, followed by dialysis and column chromatography, and their charcteristics were compared with thosw of the wild strain. Avicelase was separated into three peaks in the mutant strain as well as in the case of wild strain. Avicelase II activity of the mutant strain was prominently higher than that of the wild strain, while avicelase I and III activities of those were equivalent. The optimal pH ranges and stability of avicelase II from the mutant strain were pH4-5 and pH3.5-6.0, respectively, as well as in the case of the wild strain. The optimal temperature and thermal stability of avicelase II from the mutant strain were $40{\sim}50^{\circ}C\;and\;20{\sim}55^{\circ}C$, respectively. These results were same as those of the wild strain. By the using of Eadie-Hofastee plot, $K_m\;and\;V_{max}$ of avicelase II from the mutant and the wild strain were calculated to be 2.29mg/ml and $4.84{\mu}g$ reducing sugar as glucose per min equally, from the line fitted to the data by the least square method. Activity of avicelase II from the mutant strain was slightly activated by $Mg^{++}\;but\;inhibited\;by\;Cu^{++}, \;Mn^{++}\;and\;Zn^{++}$, as well as in the case of the wild strain. Therefore, it was concluded that the mutant didn't induce the formation of another avicelase isozyme, or the changes in the properties of avicelase, but induce the changes in the productively of the same avicelase II by the action of regulatory gane.
