The Improvement of Computational Efficiency in KIM by an Adaptive Time-step Algorithm (적응시간 간격 알고리즘을 이용한 KIM의 계산 효율성 개선)

  • A numerical forecasting models usually predict future states by performing time integration considering fixed static time-steps. A time-step that is too long can cause model instability and failure of forecast simulation, and a time-step that is too short can cause unnecessary time integration calculations. Thus, in numerical models, the time-step size can be determined by the CFL (Courant-Friedrichs-Lewy)-condition, and this condition acts as a necessary condition for finding a numerical solution. A static time-step is defined as using the same fixed time-step for time integration. On the other hand, applying a different time-step for each integration while guaranteeing the stability of the solution in time advancement is called an adaptive time-step. The adaptive time-step algorithm is a method of presenting the maximum usable time-step suitable for each integration based on the CFL-condition for the adaptive time-step. In this paper, the adaptive time-step algorithm is applied for the Korean Integrated Model (KIM) to determine suitable parameters used for the adaptive time-step algorithm through the monthly verifications of 10-day simulations (during January and July 2017) at about 12 km resolution. By comparing the numerical results obtained by applying the 25 second static time-step to KIM in Supercomputer 5 (Nurion), it shows similar results in terms of forecast quality, presents the maximum available time-step for each integration, and improves the calculation efficiency by reducing the number of total time integrations by 19%.

Trends of Antimicrobial Susceptibility Test for Bacterias Isolated from Blood, Urine, Stool, and Cerebrospinal Fluid(1997~2001) (혈액 및 일반 세균배양에서 검출된 균종과 항균제 감수성 추이(1997~2001))

  • Purpose : To know the trends of antimicrobial susceptibility is critical for antimicrobial treatment. We studied the organisms isolated from blood, urine, stool, and cerebrospinal fluid from 1997 to 2001 to reveal the trends of their antimicrobial susceptibility. Methods : We conducted a retrospective study with isolates obtained from 0~18 year old outpatients and inpatients from 1997 to 2001 at Department of Pediatrics, Hanil general hospital. We gathered the data through the laboratory test files and the origin of microorganisms cultured from blood, urine, stool and cerebrospinal fluid and their antimicrobial susceptibility. Results : Microorganisms were isolated from 226(3.3%) out of 6,974 blood cultures, 365 (8.0%) out of 4,549 urine cultures, 50(1.9%) out of 2,593 stool cultures and 9(1.4%) in 655 cerebrospinal fluid cultures. The most frequently isolated organisms from blood cultures was Staphylococcus epidermidis(33.5%) which was followed by Staphylococcus aureus(19.7%), Escherichia coli(13.8%), and Burkholderia cepacia(9.0%). Among the urine cultures, E. coli was the most common(74.7%) which was followed by Group D Enterococcus(11.3%), Klebsiella pneumoniae(7.1%) and Proteus mirabilis(2.5%). The positive stool cultures all yield Salmonella species. Group D Salmonella was obtained most frequently. Among the positive cerebrospinal fluid cultures, Group B Streptococcus was isolated most frequently. Among the 40 cases of S. aureus in blood cultures, 27 cases were methicillin-resistant. The rates of susceptibility for amikacin, ceftizoxime and ceftriaxone of E. coli isolated from blood cultures were 80%, 100% and 60% in 1997 and 60%, 80% and 60% in 2001. The rates of susceptibility for amikacin, ceftizoxime and ceftriaxone of K. pnumoniae isolated from urine cultures. were 80%, 100% and 80% in 1997 and 50%, 83% and 50% in 2001 Enterococcus was isolated from 6.7% to 15.8% and vancomycin-resistant Enterococcus was observed in 17% of Group D Enterococcus isolated from urine cultures. The rates of susceptibility for amikacin, ceftizoxime and ceftriaxone of Group D Salmonella were 96%, 96% and 92% during the study period. Conclusion : Among the blood cultures S. epidermidis, S. aureus, E. coli and B. cepacia were isolated in order of frequency and among the urine cultures E. coli, Group D Enterococcus, K. pneumoniae and P. mirabilis were isolated in order of frequency. During the study period there was no big difference in major organisms isolated from blood and urine. The methicillin-resistant S. aureus was observed in 67% of S. aureus isolated from blood cultures but vancomycin-reistant S. aureus or vancomycin intermediate resistant S. aureus was not observed. The rates of susceptibility to amikacin and the third generation cephalosporin of E. coli isolated from blood cultures and K. pneumoniae from urine cultures have decreased. The isolation rates of Group D Enterococcus and vancomycin resistant Enterococcus have increased.

A Study on High-resolution Numerical Simulation with Detailed Classification of Landuse and Anthropogenic Heat in Seoul Metropolitan area (수도권지역의 지표이용도 및 인공열 상세적용에 따른 고해상도 수치실험 연구)

  • In this study, the high-resolution numerical simulation results considering landuse characteristics are analyzed by using single layer Urban Canopy Model (UCM) in Weather Research Forecast (WRF). For this, the impact of urban parameters such as roughness length and anthropogenic heat in UCM is analyzed. These values are adjusted to Seoul metropolitan area in Korea. The results of assessment are verified against observation from surface and flux tower. Forecast system equipped with UCM shows an overall improvement in the simulations of meteorological parameters, especially temperature at 2 m, surface sensible and latent heat flux. Major contribution of UCM is appreciably found in urban area rather than non-urban. The non-urban area is indirectly affected. In simulated latent heat flux, applying UCM is possible to simulate the change similarly with observations on urban area. Anthropogenic heat employed in UCM shows the most realistic results in terms of temperature and surface heat flux, indicating thermodynamic treatment of UCM could enhance the skills of high resolution forecast model in urban and non-urban area.

  • Tegillarca granosa, is one of the most important fishery resources throughout Asia. However, due to industrialization factories, marine environmental pollution, and global warming, the marine fishery production has drop sharply. In order to understand the genetic factors of the blood clam, which is a major fishery resource on the southern coast of Korea, the whole genome of blood clam was studied. The assembled genome of T. granosa was 915.4 Mb, and 19 chromosomes were identified. 25,134 genes were identified, and 22,745 genes were functionally annotated. As a result of performing gene gain and loss analysis between the blood clam genome and eight other types of shellfish, it was confirmed that 725 gene groups were expanded, and 479 gene groups were contracted. The homeobox gene cluster of blood clam showed a well-preserved genetic structure within lophotrochozoan ancestor. T. granosa genome showed high similarity between three hemoglobin genes with Scarpharca broughtonii. The blood clam genome will provide information for the genetic and physiological characteristics of blood clam adaptation, evolution, and the development of aquaculture industry.

A New Rice Variety with Good Qualilty and Multiple Diseases Resistance "Sangok" (중생 고품질 복합내병성 신품종 벼 "상옥")

  • "Sangok", is a new japonica rice variety (Oryza sativa L.), which is a midium maturing ecotype developed by the rice breeding team of National Yeongnam Agricultural Experiment Station (NYAES) in 2003. This variety was derived from the cross of Milyang 101/YR8697Acp97 (in 1988/1989 winter) and selected by combination of the bulk and pedigree breeding. The pedigree of Sangokbyeo, designated as Milyang 182 in 2000, was YR12950-B-B-B-19-2-4-2-2. It has about 79cm stature in culm length and is medium maturing. This variety is resistant to bacterial blight ($K_1$, $K_2$, and $K_3$), stripe virus and moderately resistant to leaf blast disease. Milled rice kernels of "Sangok" is translucent, clear in chalkness and good at eating quality in the panel test. The yield potential of "Sangok" in milled rice is about 5.16MT/ha at ordinary fertilizer level of local adaptability test. This cultivar would be adaptable to the southern plain of Korea below the Chungnam province by latitude from ordinary transplanting to transplanting after barley harvest.

Outbreak of Shigellosis Occurred in a Preschool and Two Elemetary Schools in Mapo-Gu, Seoul (서울시 마포구 관내 어린이집 및 초등학교에서 집단 발병한 세균성 이질)

  • Purpose : Shigellosis is still an important cause of acute food-borne diarrheal diseases throughout the world. We investigated the transmission routes and clinical course through the outbreak of shigellosis in Mapo-Gu, Seoul. Methods : From October 7th to November 19th in 2005, Mapo-Gu public health center had referred 32 patients with confirmed or suspected shigellosis to the National Medical Center. We had investigated source of infection, epidemiology, laboratory findings, and clinical course of the cases occurred during this outbreak. Results : Among 32 patients, 24 patients had been confirmed with shigellosis, 8 patients had been diagnosed with suspected shigellosis. They ranged in age from 5 months to 12 years old and their mean age was 6.5 years. The clinical manifestations were as follows; diarrhea, fever, abdominal pain and asymptomatic condition. Symptoms had sustained for 3.7 days on the average. S. sonnei were cultured by rectal swab and founded to be resistant to ampicillin and TMP/SMX except to 3rd generation cephalosporin. After treatment with antibiotics such as cefixime and ceftriaxone or imipenem and conservative treatment with electrolyte and fluid replacement for 5~7 days, Stool cultures of the rectal swab grew no Shigella in these cases except 3 cases. Conclusion : An outbreak of shigellosis had occurred in a preschool and elementary school children. From the same results of antimicrobial susceptibility and pulsed-field gel electrophoresis patterns in this study, we suggest that the outbreak of shigellosis in this report had been originated from a single strain. According to all negative results about suspected food and water cultures, we couldn't find out source of infection. Through materials offerred by Mapo-Gu Public Health Center, we presumed the trasmission routes probably were person-to-person.

  • 현대의 건축시기는 다원성의 시기 또는 복합성의 시기라고 불리운다. 모더니즘과 포스트모더니즘의 시기를 거치며 다양한 형태적 실험의 과정을 겪었고 수많은 이데올로기와 이론들로 무장된 논쟁의 역사도 갖게 되었다. 이러한 현대건축의 다양성은 건축디자인이라는 전문직의 사회에 기여하는 역할을 반추(反芻)함과 동시에 다변하는 인간사고의 역동성을 반영하고 있다. 건축관련 책자나 저널 등에서 흔히 발견할 수 있는 Neo-Classicism, Deconstructivism, Minimalism, High-Tech, New Moderns 등등의 건축양식들은 스타일을 지칭하는 언어라기보다는 건축적 사고를 지칭하는 언어라고 할 수 있다. 결국 건축디자인이라는 작업은 인간을 둘러 싼 건조환경의 생성에서 출발하는 것이므로 건축적 사고방식은 그 시기의 사회현상과 밀접한 관계를 맺게 된다. 20세기 말부터 불어닥친 디지털 문화의 사회변혁은 전 세계를 하나의 채널로 엮게 되었고, 이러한 영향들은 지역과 민족, 그리고 종교적인 요소들까지도 혼합하여 국경을 초월한 미래의 새로운 이상들을 꿈꾸게 되었다. 19세기 산업혁명과 함께 급속히 벨라진 인류문명의 진보는 필수적으로 세계화를 동반하는 것이고, 세계화와 지역성사이의 괴리를 통해 나타나는 문화적 비판과 충돌은 아직도 진행상태인 것이다. 세계화라는 통염은 문명의 보편성을 기반으로 하며 전통문화의 파괴가 뒤따를 수밖에 없는데, 현재와 같이 IT혁명을 통해 급속도로 빨라진 세계문화의 교류는 한 국가가 뿌리를 내리고 있는 문화적 유산이라는 토양을 포기하여야 할 단계에 와 있다고 논평할 정도이다. 여기서 건축은 문화적 상징으로 작용하게 되는데, 그것이 심미적이든 관습적이든 또는, 경제적이든 건축가는 해답의 제시라는 임무에 직면하게 된다. 20세기에 진행된 모더니즘의 건축은 구조와 공간에 대한 고전건축의 숙원을 고덕건축이 이룬 것처럼 15세기 르네상스 시대로부터 진행된 형태와 공간, 기능, 그리고 아름다움과 기술(technology) 또는 경제성 등에 대한 건축적 의문과 탐구를 일단락지었다고 할 수 있다. 이러한 모더니즘의 한복판에 Mies van der Rohe라는 거장이 깊은 발자국을 남기며 한 시대를 이루었고 그가 남긴 시대정신의 이념과 가치를 Miesianism이라고 부른다. 실용성과 도덕성을 근본으로 하는 Miesianism은 대중에 호소하는 미적 표현주의를 부정하고 지역정서를 중시하는 문맥적 접근방식을 경시함으로써 보편화라는 현상을 불러일으킨 국제주의 양식의 한 부류로 비판을 받아왔다. 즉, Miesianism의 단순하고 강렬한 외형적 요소는 그것이 내포하는 기술적 합리성이나 공간적 완결성을 무시한 채 전 세계에 영향을 미쳤고 동시에 지역적 저항을 받게 되었다. 시카고 및 전 세계의 Miesian들, 즉 Mies van der Rohe의 제자들이나 그로부터 영향을 받은 수많은 건축가들은 이러한 저항과 비판에 직면하게 되며 새로운 사고의 시대적 요구 앞에 고뇌하게 된다. 한국에서는 1978년 Mies van der Rohe의 제자인 김종성이 미국에서 서울로 돌아와 '서울건축컨설탄트'를 설립하며 본격적으로 Miesianism의 규범적 건축론을 설파하기 시작하였다. 이른바 시카고 국제주의학파의 건축전수라고 할 수 있는데 '서울건축컨설탄트'를 통하여 배출된 김종성의 제자들은 명쾌하고도 간결한 건축해법의 경험을 토대름대로의 정체성을 갖고자 노력하였으나 결국 다원적 가치를 요구하는 시대적 흐름 속에 혼란을 겪고 있는 것 또한 사실이다. 본 연구는 Miesianism의 기원을 밝히고 그것의 실수와 오류를 밝힘과 동시에 현대의 여러 가지 건축유형들과 비교하여 봄으로써 Post-Miesianism의 실체와 그 미래에 대하여 고찰하여 보고자 한다. 미스의 유산이라고 할 수 있는 Miesianism의 요체는 첫째, Schinkel로부터 이어받은 시대정신의 사명감, 둘째, Berlage가 전해준 전문가로서의 도덕성을 바탕으로 하여 이루어진 기술적 합리성의 실현, 그리고 마지막으로 그 당시의 추상예술에 대한 이해로써 받아들인 nee-Suprematism의 아방 가르데적인 자세라고 할 수 있다. 그러나 이것들을 전파하고 수용하는 과정에서 미스적인 단순하고 명쾌한 외관이 더욱 강한 인상을 남기게 되었고 또한 미스가 현대성의 본질이라고 믿는 기술의 전수는 모든 외관을 동일하게 만드는 International Style로서의 보편적 획일성을 초래하였다. Mies van der Rohe의 강력하고 규범적인 dogma는 그의 제자들에게는 강렬한 카리스마로 각인되었고, Mies가 원한 것이 아닐지라도 그들을 하나의 틀 속에 가두는 영향력을 행사하였다. 2세대이든 3세대이든 기술적 합리성과 추상적 단순미라는 믿음을 맹목적으로 추종할 때 그들은 Miesian으로 남아있게 되며, 거기서 벗어났을 때 non-Miesian, 또는 배신자로 취급하기까지 하였다. 이것은 미스의 제자들에게 강한 족쇄가 되어 형태적 또는 개념적 변화의 시도를 어렵게 하였고 시대적 요구에 부응하지 못한다는 평가에 이르게 되었다. 문화의 다양성과 역동성을 인정함과 동시에 Miesianism에 대한 새롭고 시대에 적합한 해석을 기대하게 되는데, 이러한 기대 속에서 우리는 Post-Miesianism이라는 새로운 유형을 엿볼 수 있게 된다. Post-Miesianism의 징후를 현대의 다양한 건축유형에서 발견할 수 있는데, High-Tech의 구축적이고 정밀한 건축 기술적 해법과 Minimalism의 반복성을 통한 추상미의 표현 등에서 뿐만이 아니라 Post-Modernism의 인간성의 추구나 문맥적 고려, 또는 해체주의의 형이상학적이고 아방가르데적인 실험주의 정신까지조차 Post-Miesianism이 공유할 수 있는 부분이 있다고 보여진다. 또한, 미스의 영향이 2세대, 3세대의 건축가들에게 미치면서 여러 방향으로 변질되어 가는 것을 우리는 발견할 수 있으며 그러한 시대성에 적응하는 시도들이야말로 새로운 미래를 향한 Post-Miesianism의 실체인 것이다. Mies가 말하였듯이 한 시대의 끝은 그 시대가 완전히 이해되었을 때이며, 모더니즘의 숙제는 아직 해결되지 않았으므로 Miesianism의 탐구는 Post-Miesianism이라는 새로운 주제로 계속될 것이다. 이를 통하여 21세기 디지털 문명의 시대 속에서 현대건축이 안을 수밖에 없는 보편성과 지역성의 충돌이라는 문제의 해결에 한 발걸음 다가갈 수 있기를 기대하여 본다.

Activities of esterase and acetylcholinesterase on the diamond backmoth, Plutella xylostella (Lepidoptera : Yponomeutidae) and beet armywarm, Spodoptera exigua (Lepidoptera : Noctuidae) and inhibitions of acetylcholinesterase with flupyrazofos (배추좀나방과 파밤나방의 효소활성 및 flupyrazofos 에 의한 AChE 활성 저해)

  • The activities of esterase and acetylcholinesterase(AChE) on the Diamond backmoth (DBM), Plutella xylostella (Lepidoptera: Yponomeutidae) and Beet armywarm (BAW), Spodoptera exigua (Lepidoptera: Noctuidae) and inhibitions of AChE with flupyrazofos were clarified from the results of a series of experiments. These findings are described in brief as follows. The AChE activities of DBM and BAW in heads were $1.5{\sim}11.1{\mu}$mol/g/min in $1st{\sim}4th$ instar larvae of DBM and $1.7{\sim}45.2{\mu}$mol/g/min in $1st{\sim}6th$ instar larvae of BAW, respectively. Those were $25{\sim}30$ times higher in above 4th instar larvae of BAW than that of the 1st instar larvae of DBM. The activities of aliesterase in heads were $1.7{\sim}4.7$ times higher in $2nd{\sim}4th$ instar larvae of DBM and $8{\sim}55$ times higher in $3rd{\sim}6th$ instar larvae of BAW than 1st instar larvae of DBM. In abdomens, those were $3{\sim}17$ times higher in $2nd{\sim}4th$ instar larvae of DBM and $12{\sim}30$ times higher in $3rd{\sim}6th$ instar larvae of BAW than 1st instar larvae of DBM. Median AChE inhibition concentration $(I_{50})$ of flupyrazofos to the 2nd instar larvae of DBM and BAW were 92 nM and $15{\mu}M$, respectively, and those to the 4th instar larvae of DBM and BAW were $1.8{\mu}M$ and 3.1 mM, respectively. Insensitivity ratio of flupyrazofos in the 2nd instar BAW larvae showed ca. 162 times higher than that in the 2nd instar larvae of DBM, and that of the 4th instar BAW larvae showed ca. 1,720 times higher insensitivity to flupyrazofos than that of the 4th instar DBM larvae. AChE activities in the 2nd instar larvae of DBM and BAW at 32 h after applicaton of flupyrazofos decreased from 67.6% to 32.4% of the activity of the untreated control. That of the 4th instar larvae of DBM increased for 0.5 h after application flupyrazofos up to 75% of the untreated control, and after that it decreased to 34.5% of the untreated control at 32 h. In contrast, in the 4th instar larvae of BAW AChE activities increased for 8 h gradually up to 102 % of the activity of the untreated control, and then the activity decreased to 97% of the untreated control at 16 h after treatment.

Selection of (Ac/Ds) insertion mutant lines by abiotic stress and analysis of gene expression pattern of rice (Oryza sativar L.) (비생물학적 스트레스 관련 벼 Ac/Ds 삽입 변이체의 선발 및 유전자 발현 분석)

  • Transposon-mediated insertional mutagenesis is one of powerful strategy for assessing functions of genes in higher plants. In this report, we have selected highly susceptible and tolerance plant by screening about high salt (3% NaCl) and cold stresses ($4^{\circ}C$) from F2 seeds of 30,000 Ac/Ds insertional mutagenesis lines in rice (Oryza sativa L. cv. Dongjin). In order to identify the gene tagging, insertion of Ds element was analyzed by Southern blot and these results revealed that 19 lines were matched genotype of selected lines with phenotype from the first selected 212 lines, and 13 lines have one copy of Ds elements. The Franking Sequence Tags (FSTs) of selected mutant lines showed high similarities with the following known function genes: signal transduction and regulation of gene expression (transpoter, protease family protein and apical meristem family protein), osmotic stress response (heat shock protein, O-methyltransferase, glyceraldehyde-3-phosphate dehydrogenase and drought stress induce protein), vesicle trafficking (SYP 5 family protein) and senescence associated protein. The expression pattern of 19 genes were analyzed using RT-PCR under the abiotic stresses of 9 class; 250mM NaCl, osmotic, drought, 3% $H_2O_2$, $100{\mu}M$ ABA, $100{\mu}M$ IAA, 0.1 ppm 2,4-D, $4^{\circ}C$ cold and $38^{\circ}C$ high temperature. Isolated knock-out genes showed the positive response about 250 mM NaCl, drought, $H_2O_2$, PEG, IAA, 2,4-D, ABA treatment and low ($4^{\circ}C$) and high temperature ($38^{\circ}C$). The results from this study indicate that function of selected knock-out genes could be useful in improving of tolerance to abiotic stresses as an important transcriptional activators in rice.

Studies on the Behavioral response in Silkworm larvae, Bombyx mori. Genetic Analysis and Correlated Response to Economic Characters of Walking Distance (가잠유충의 행동반응성에 관한 연구)

  • The objective of the present study was to investigate the behavioral responses of sillkworm (Bombyx mori) larvae to: the dominant effect of walking distace: and the relationships between walking distance and the various economic characters on yellow sheets. The means of walking distances of progeny from F2 and F1 F1 and F2 generations, which was a hybrid between the long and the short walking distances, were shorter than those of parents. This may indicate that the genes for the short walking distances were partially dominant over those for the longwalking. Estimated broad sense heritability of larval walking distance was different for each larval instar, which fell into the range of 52∼84%. Genetic correlation was apparent between walking distances of all larval stages and some economic characters. The number of cocoons per liter and the period from incubation to emergence have shown the positive genetic correlation with walking distances. The cocoon yield, the sing cocoon weight, and the cocoon shell weight have shown to be in the negative correlation with walking distances. No correlation was observed between walking distances of the mature larvae and any economic characters. The analysis by the path coefficient showed negative direct effect of walking distances on the cocoon yield, and positive indirect effect on the single cocoon weight and the cocoon shell weight.
