ELECTRON MICROSCOPIC STUDY OF RESIN-DENTIN HYBRID ZONE PRODUCED BY THE MOISTENING OF ACID CONDITIONED DENTIN SURFACE (산 표면처리 후 상아질 표면의 습윤이 하이브리드층 형성에 관한 전자현미경적 연구)

  • The effect of moistening and air-drying of acid-conditioned dentin before priming on the formation of resin-dentin hybrid zone was investigated, Freshly extracted human molars were used and divided at random into 5 groups, Groups 1 - 3 consisted of specimens conditioned with 10 % phosphoric acid for 20 seconds; Group 1 served as a control in which the conditioned dentin was simply blot-dried with a damp facial tissue; Group 2 was air dried for 30 seconds ; Group 3 was air dried for 30 seconds and immediately remoistened for 10 seconds with air-water syringe. and then the specimen was blot-dried with a damp facial tissue. Groups 4-5 were not acid conditioned ; In group 4, the smear layer on the dentin was blot dried before primer placement; Group 5 was air dried only for 30 seconds, The acetone-based primer and bonding agent of All Bond 2 (Bisco. Inc., USA) and composite resin (Z-100, 3M Dental products, USA) were applied for acid conditioned dentin and non-conditioned dentin. The morphologic ultrastructure of resin-dentin hybrid zone was examined by the use of SEM and TEM. and the existence of inorganic material and analysis of Ca/P weight-percent ratio in the resin-dentin hybrid zone were revealed by the EDAX, The results were as follows : 1. In the moistened specimens from acid-conditioned groups, the resin penetrated about 3-$4{\mu}m$ into dentin and the denatured collagen smear layer was not present at the surface. The resin tag was formed to a thickeness of 3-$4{\mu}m$ at the upper part of dentinal tubule and compactively connected to each other by means of many lateral branching. 2. In the air-dried specimens from acid-conditioned groups, the resin penetrated about 2.0-$2.5\;{\mu}m$ into dentin and an upper thin black layer to a thickness of 30-35nm was identified between adhesive resin and demineralized collagen layer. The resin tag to have a diameter of $2.5{\mu}m$ was formed at the upper part of dentinal tubule. However the funnel shape of the tag was not notable compared to the moistened specimens. 3. In the remoistened specimens from acid conditioned groups, the resin penetrated about 2.0-$2.5{\mu}m$ into dentin and an upper black layer was not present. The resin tag at the upper part of dentinal tubule was formed less than $2{\mu}m$ and was weakly connected to each other by means of few lateral branching. 4. In the non-conditioned groups, the smear layer was formed to a thickness of $0.5{\mu}m$ at dentin surface. However, the resin-dentin hybrid zone was not identified by TEM. The evidence of resin penetration into intertubular and intratubular dentin did not show. 5. All the acid-conditioned groups showed that the detected calcium and phosphorus weight percent ratios at the $2{\mu}m$ upper portion from the resin-dentin interface into the resin were much higher than that at the $2{\mu}m$ lower portion from the resin-dentin interface to dentin. Mirtazapine Regulates Pacemaker Potentials of Interstitial Cells of Cajal in Murine Small Intestine (생쥐 소장 카할세포의 pacemaker potential에서 미르타자핀 효능에 관한 연구)

  • Interstitial cells of Cajal (ICCs) are the pacemaking cells in the gastrointestinal (GI) muscles that generate the rhythmic oscillation in membrane potentials known as slow waves. In the present study, we investigated the effects of mirtazapine, a noradrenergic and serotonergic antidepressant, on pacemaking potential in cultured ICCs from the murine small intestine. The whole-cell patch-clamp configuration was used to record pacemaker potential in cultured ICCs. Mirtazapine induced pacemaker potential depolarizations in a concentration-dependent manner in the current clamp mode. Y25130 (a 5-HT3 receptor antagonist), RS39604 (a 5-HT4 receptor antagonist), and SB269970 (a 5-HT7 receptor antagonist) had no effects on mirtazapine-induced pacemaker potential depolarizations. Also, methoctramine, a muscarinic M2 receptor antagonist, had no effect on mirtazapine-induced pacemaker potential depolarizations, whereas 4-diphenylacetoxy-N-methyl-piperidine methiodide (4-DAMP), a muscarinic M3 receptor antagonist, inhibited the depolarizations. When guanosine 5'-[β-thio] diphosphate (GDP-β-S; 1 mM) was in the pipette solution, mirtazapine-induced pacemaker potential depolarization was blocked. When an external Ca2+ free solution or thapsigargin, a Ca2+-ATPase inhibitor of the endoplasmic reticulum, was applied, the generation of pacemaker potentials disappeared, and under these conditions, mirtazapine induced pacemaker potential depolarizations. In addition, protein kinase C (PKC) inhibitor, calphostin C, and chelerythrine inhibited mirtazapine-induced pacemaker potential depolarizations. These results suggest that mirtazapine regulates pacemaker potentials through muscarinic M3 receptor activation via a G protein-dependent and an external or internal Ca2+-independent PKC pathway in the ICCs. Therefore, mirtazapine can control GI motility through ICCs.

Characterization of the drug receptors responsible for intestinal contraction in Israeli carp (이스라엘잉어 장관 수축에 관여하는 약물수용체에 관한 연구)

  • In order to elucidate the characterization of receptors involved in inestinal motility of Israeli carp, spontaneously contracting Israeli carp intestinal preperations were prepared and mounted in the organ chambers for contraction traicings using a polygraph. Various contractile agonists were treated and their dose-response curves were constructed. $EC_{50}$ values$(pD_2)$ of each agonist on specific receptors, $pA_2$ values of competitive antagonists against some agonists, and $K_1$, values of noncompetitive antagonists against some agonists were analyzed for characterization of receptors related with the intestinal contraction. Results obtained through the experiments were summarized as follows: 1. Acetylcholine(ACh) exhibited biphasic dose-response curves: initial ACh-induced dose dependent contractions were observed in pM levels but followed by decreased response in in-between concentration levels. Dose dependent contractions reappeared in ${\mu}M$ level. The peaks in pM and ${\mu}M$ levels appeared in $10^{-13}M$ and $3{\times}10^{-5}M$, respectvely. 2. Carbachol(CaCh) exhibited dose dependent contractions from $10^{-9}M$ to $10^{-5}M$, and its $pD_2$ values were higher than those of ACh($5.60{\pm}0.11$). ACh and CaCh exhibited equiactive contractions. Nicotine had no effects on contractile responses of Israeli carp intestine. 3. ACh-induced responses were inhibited by atropine($K_1:7{\times}10^{-8}M$), a muscarinic antagonist, in a non-competitive manner. But CaCh-induced responses were inhibited by both antimuscarinic atropine($pA_2:9.52{\pm}0.14$) and selective $M_2$ antagonistic 4-DAMP($pA_2:8.16{\pm}0.09$), in competitive manners. Nicotine receptor antagonistic decamethonium and hexamethonium had no effects on ACh-and CaCh-induced contractions. Therefore, the cholinergic receptor related to intestinal motility of Israeli carp was assumed as $M_2$ type. 4. In Israeli carp intestine, 5-HT (serotonin) exhibited dose dependent contractions in concentration range from $10^{-8}M$ to $10^{-5}M$. The maximal responses, however, were corresponded to about 50% of those of ACh or CaCh. 5-HT induced contractions were inhibited by $5-HT_2$ antagonistic ketanserin ($K_1: 7.8{\times}10^{-4}M$) in a non-competitive manner, but not by both of anti $5-HT_1$, spiperone and anti $5-HT_3$, MDL-72222. Hence, $5-HT_2$ receptors are suggested to be existed in Isreli carp intestine.

  • By process of treatment for a case which diagnosed as spinal cord infarction and admitted from the 18th, April, 2000 to the 29th, July, 2000, the result are as foilows. Method and Results : In the earlier days of admission, this patient was diagnosed as damp-heat(濕熱) and was dosed with Cheongjo-tang. In medication, weakness in upper and lower limbs of the patient was improving. After diagnosed as Taeyangin, the patient was dosed with Ogapijangchuk-tang and punctured with Taeyangin Taegeuk-chim and Sagae-chim. As the result, Weakness in upper and lower limbs are improved remarkably. Conclusion : The prognosis of patients diagnosed spinal cord infarction is not so good. But, in the case of this patient, power and sense of four limbs improved remarkably through oriental medical treatment with rehabilitative therapy.

  • In "EuiMoonBubRyool" chapter 4, Yuchang criticized previous doctors since "damaged by wet in autumn" written in "Somun" must be corrected as "damaged by dry in autumn". The reason why the sentence must be corrected is that dry and damp are different atmosphere. He tried to explain with changes of season and the rapid pulse in autumn is said to be a contraindiction in "Naekyung" and it supports Autumn dry theory. He criticized previous doctors about many incorrect points of dry disease throu호 various aspects. Criticism on Dongwon's treatment, Dangye's medical theory and not having a treatment, and Mujungsoon's treatment and etc. are the examples. Symtoms and treatment of Dry disease that Yuchang commented helped in deepening the contents and widening the general understanding of the disease.

  • The Radix of Gentiana macrophylla (Gentianaceae) has been used for dispelling wind-damp, relaxing the sinews, and clearing heat from deficiency. In order to evaluate its quality, the method for the quantitative determination of gentiopicroside as standard compound has been developed. Gentiopicroside was a major compound of Gentianae Macrophyllae Radix. It was analyzed by HPLC using 30% MeOH as a solvent system. The amount of gentiopicroside from Gentianae Macrophyllae Radix was in the range of $2.22{\pm}1.22%\;(n=18)$.

  • This paper includes a study on the performance analysis of SDINS by a simulator using Matlab/Simulink. The performance model is considered gravity and Coriolis force, and a barometer is included to damp down diversity of a perpendicular axis error. Using the simulator, the performance included gyro sensor errors was analyzed in various maneuvering patterns. Also, the performance is virtually presented for the variation of error parameters of gyro and accelerometer under GUI.

  • A general review is made on Insamyangwitang(人蔘養胃湯). Following conclusions are drawn from the clinical cases of Insamyangwitang in Hyungsang medicine. 1. Insamyangwitang is composed of four different prescriptions of Huisaentang, Sakoonjatang, Eajintang and Pyungwisan. Huisaentang is usually prescribed for the intestinal convulsion. Sakoonjatang for the deficiency of Ki. Eajintang for retention of phlegm. 2. Insamyangwitang is effective in strengthening the spleen, drying the dampness, warning the middle-warmer to stop vomiting, regulating the flow of Ki, and eliminating phlegm. 3. Insamyangwitang is applicable to malaria caused by cold, intestinal convulsion, abdominal mass, edema, tympanites, Yin syndrome of exogenous febrile disease, distension, lack of appetite, stomachache, and diarrhea. 4. Persons with the following characteristic in Hyungsang are more susceptible to Insamyangwitang : Jung type, Hyul type, fish type, Taium meridian type, white fat damp constitution, person with big mouth, and woman rather than man.

Multimachine Stabilizer using Sliding Mode Observer-Model Following including CLF for Measurable State Variables

  • In this paper, the power system stabilizer(PSS) using the sliding mode observer-model following(SMO-MF) with closed-loop feedback (CLF) for single machine system is extended to multimachine system. This a multimachine SMO-MF PSS for unmeasureable plant state variable is obtained by combining the sliding mode-model following(SM-MF) including closed-loop feedback(CLF) with the full-order observer(FOO). And the estimated control input for unmeasurable plant sate variables is derived by Lyapunov's second method to determine a control input that keeps the system stable. Time domain simulation results for the torque angle and for the angular velocity show that the proposed multimachine SMO-MF PSS including CLF for unmeasurable plant sate variables is able to damp out the low frequency oscillation and to achieve asymptotic tracking error between the reference model state at different initial conditions and at step input.

Modeling and Control of VSI type FACTS controllers for Power System Dynamic Stability using the current injection method

  • This paper describes modeling Voltage Sourced Inverter (VSI) type Flexible AC Transmission System (FACTS) controllers and control methods for power system dynamic stability studies. The considered FACTS controllers are the Static Compensator (STATCOM), the Static Synchronous Series Compensator (SSSC), and the Unified Power Flow Controller (UPFC). In this paper, these FACTS controllers are derived in the current injection model, and it is applied to the linear and nonlinear analysis algorithm for power system dynamics studies. The parameters of the FACTS controllers are set to damp the inter-area oscillations, and the supplementary damping controllers and its control schemes are proposed to increase damping abilities of the FACTS controllers. For these works, the linear analysis for each FACTS controller with or without damping controller is executed, and the dynamic characteristics of each FACTS controller are analyzed. The results are verified by the nonlinear analysis using the time-domain simulation.
