This experiment was done to determine the optimum conditions for the induction of tetraploidy in Panax ginseng C. A. Meyer using bud length, temperature and plant growth regulator pretreatments. Highest callus formation was obtained when the medium was inoculated with flower bud in the size of 2-3 mm in length. The optimum temperature for the callus formation was high when treated at $4^{\circ}C$ for 4-5 days. Among the treatments of growth regulators and different concentration, highest callus formation was observed in combination of 5 mg/L 2,4-D and 1 mg/L kinetin for P. ginseng. As a result of flow cytometer analysis, all 7 adventitious roots were confirmed as tetraploidys. Cytological analysis revealed that the chromosome number of tetraploid roots was 96, while that of diploid roots was 48. Tetraploid ginseng roots were inoculated to flower bud size of 2-3 mm in length. The callus formation was optimum when treated with 1 mg/L 2,4-D at $4^{\circ}C$ for 5 days. Compared with control roots, tetraploid roots were thicker and longer and had few lateral branches. Fresh weight of tetraploid roots was relatively higher than the control roots.
Using recombinant Vaccinia virus(vSC8) that express ${\beta}$-galactosidase, a model heterologous protein, conditions for virus and protein production were investigated in tissue culture flask. As host animal cells HeLa and HeLa S3 were used. It was demonstrated that cells infected during the exponential growth phase gave higher protein yield than those infected during the stationary growth phase and calf serum concentration after virus infection did not significantly alter protein yield. Pretreatment of cell layer with hypotonic solution enhanced the virus infectivity. Optimum cell growth and recombinant protein production was achieved at $37^{\circ}C$. But, during 2 hours of virus infection period incubation temperature must be lowered to 20∼$30^{\circ}C$ for maximum recombinant protein yield. To enhance virus replication, the effects of adrenal glucocorticoid hormone (Dexamethasone) and silkworm hemolymph were evaluated. Only dexamethasone increased about 20% of ${\beta}$-galactosidase yield in HeLa S3 cells when added with 10-7∼10-5M concentration 24 hours before infection.

This study evaluates the pretreatment for analysis of benzidine metabolites in urine by measuring the recovery rates according to the temperature and periods of storage of the urine. By the solid phas e extraction, the recovery rates of basic hydrolysis are benzidine 67.4 %, monoacetylbenzidine 105.1 %, and diacetylbenzidine 115.8 %, respectively. By the liquid extraction, the recovery rates of back-extraction into 0.1 M perchloric acid are benzidine 105.7%, monoacetylbenzidine 94.2 %, diacetylbenzidine 72.8 %, respectively. The difference of the recovery rates between the back-extraction into 0.1 M HCl and 0.1 M perchloic acid after basic hydrolysis are 101 % and 98.8 %, respectively. When the recovery rates of the urinary s amples of pH 3, pH 7, pH 12 at $25^{\circ}C$ and $-76^{\circ}C$ are compared for four weeks, there are no differences according to the temperature and the periods of storage. The above results show that the solid phase extraction and back-extraction by 0.1 M perchloric acid after basic hydrolys is are suitable for the analysis of benzidine metabolites. There are no difference of the recovery rates of the urinary samples stored at $25^{\circ}C$ and $-76^{\circ}C$ at pH 3, pH 7, pH 12, respectively for 28 days.

To enable off-season peach canning, peaches were halved and processed into solid packs. Following variables were compared in relation to storage stability of solid packs. First as pretreatment-dipping in citric and ascorbic solution after lye peeling; second as pasturization in pouch and hot fill process; third as packaging method-2 ply(PET/PE) and 3 ply(PET/Al/PE) plastic film pouch and metal container; fourth as storage condition $-20^{\circ}C\;and\;-5^{\circ}C$. After 25 weeks storage test, all peaches in different packs were processed into canned peaches just like commercial products, and then quality was organoleptically evaluated from the viewpoint of processing and storage variable of solid packs. The results are follows; 1. Ascorbic acid dipping in 3 ply pouch and citric acid dipping in 2 ply pouch had better quality retention effect than any other pretreatments in respective packing. 2. Pasteurization method gave great effect on the color and texture of the solid pack during storage, but not on the quality of the final canned peach. 3. Can and 3 ply packing were better than 2 ply pouch in storage of solid pack and quality of final canned peach. 4. The $5^{\circ}C$ storage gave better quality than $20^{\circ}C$.
PURPOSE. To evaluate the effects of hydrogen peroxide pretreatment and heat activation of silane on the shear bond strength of fiber-reinforced composite posts to resin cement. MATERIALS AND METHODS. The specimens were prepared to evaluate the bond strength of epoxy resin-based fiber posts (D.T. Light-Post) to dual-curing resin cement (RelyX U200). The specimens were divided into four groups (n=18) according to different surface treatments: group 1, no treatment; group 2, silanization; group 3, silanization after hydrogen peroxide etching; group 4, silanization with warm drying at $80^{\circ}C$ after hydrogen peroxide etching. After storage of the specimens in distilled water at $37^{\circ}C$ for 24 hours, the shear bond strength (in MPa) between the fiber post and resin cement was measured using a universal testing machine. The fractured surface of the fiber post was examined using scanning electron microscopy. Data were analyzed using one-way ANOVA and post-hoc analysis with Tukey's HSD test (${\alpha}=0.05$). RESULTS. Silanization of the fiber post (Group 2) significantly increased the bond strength in comparison with the non treated control (Group 1) (P<.05). Heat drying after silanization also significantly increased the bond strength (Group 3 and 4) (P<.05). However, no effect was determined for hydrogen peroxide etching before applying silane agent (Group 2 and 3) (P>.05). CONCLUSION. Fiber post silanization and subsequent heat treatment ($80^{\circ}C$) with warm air blower can be beneficial in clinical post cementation. However, hydrogen peroxide etching prior to silanization was not effective in this study.
In order to develop low salt fermented hen clam sauce with high stability, the effects of citric acid pretreatment and fermentation temperature were examined. The VBN contents of surf clam sauce treated with citric acid (SCA) were lower than those of control and the same sauces added alcohol at two temperature conditions ($5^{\circ}C$ and room temperature) during fermentation. The maximum $NH_2-N$ contents of control and SCA during fermentation at $5^{\circ}C$ were 501.3 and $618.4{\sim}691.6\;mg/100g$, respectively, and the pH of those showed $5.61{\sim}6.24$ and $2.43{\sim}3.21$. The total creatine contents of control and SCA, respectively, were $36.8{\sim}27.6\;mg/100g$ ranges. As the time of treatment with citric acid was longer, the degradation of ATP, ADP and AMP in the SCA was faster. In the control, the Ala content was $19.6{\sim}23.02%$ and was highest level among all free amino acids. As fermenting term was longer, among the free amino acids, Ala and Gly were large amounts in control and SCA, respectively, and Gly was slowly increased in SCA during fermentation. As the results of organoleptic test, the tastes of SCA showed good score than control.
In this paper, the dyeing ability of the volcanic ash dyeing on cotton knitted fabrics were investigated. Acrylic copolymer was used to improve the depth of fabric color in the dyeing process. K/S values of dyed fabrics were measured to examine the dyeing properties. Two different classes of dyeing process were tested; (1) volcanic ash dyeing after pretreatment and (2) simultaneous co-treatment with volcanic ash and acrylic copolymer. In the first process, the effects of parameters such as the concentration of volcanic ash, concentration of Na2S04, dyeing time, dyeing temperature and pH of dyebath were noted. In the second process, the effects of parameters such as concentration of acrylic copolymer, dyeing time and temperature, and drying temperature were noted. Experimental results showed that the co-treatment of acrylic copolymer improved the dyeing properties of cotton knitted fabrics with volcanic ash. For the first dyeing experiment, concentration of dispersing agent was 0.1%, concentration of volcanic ash was 4%, treatment time was 20minutes, concentration of $Na_2SO_4$ was 2%, treatment temperature was $60^{\circ}C$ and treatment pH of dyebath was neutral. In the second dyeing experiment, concentration of acrylic copolymer was 2%, treatment temperature was $80^{\circ}C$, treatment time was 40 minutes, and treatment drying temperature was $150^{\circ}C$.
Effect of nitric oxide (NO) treatment on physiology and quality characteristics of peaches (Prunus persica L. Batsch) was determined. Peaches were treated for 4 hr with NO (0, 10, 100 ppm) gas under oxygen-free atmosphere at $10^{\circ}C$, packaged with 0.05-mm LDPE film, and stored at $10^{\circ}C$ for 15 days. Treatment with 100 ppm NO reduced the ethylene production and the loss of flesh firmness in peaches, but did not affect soluble solids, titratable acidity, and surface color. Oxygen-free atmosphere induced the reduction of respiration rate and ethylene production. Modified atmosphere packaging retarded the loss of flesh firmness and surface greenness, but decreased soluble solids and titratable acidity regardless of NO treatment.
We examined the effect of pretreatments on seed germination, seedling growth, optimal seedling density of Phellodendron amurense. The seeds were carefully purified after collection in late Oct. from natural population at Mt. Chiri. For the evaluation of pretreatment effects, seeds were sowed in an experimental nursery after being stored in open ground($2{\sim}8^{\circ}C$) with soaking in 20% $H_2O_2$, 50-fold pon-pon solution, cold moist stratification in refrigerator($2{\sim}4^{\circ}C$) and dry storage in roam temperature($4{\sim}18^{\circ}C$), respectively, For the effect of growth density of seedlings, seedlings germinated in nursery were precisely controlled with 53, 104, 220, 380 seedlings per $m^2$. Among pretreatments for the promotion of germination rate, pon-pon treatment(68.8%) was highest, the others were higher than that in dry storage(2.3%), and these seed pretreatment methods were significantly different at 1% level. The height growth of seedlings showed the most vigor from Jun.12 to Jul. 8, and from Jul.20 to Aug. 2, and it reached to 72.8% of height growth. In case of 1-0 seedling, although seedling density per $m^2$ was increased, seedling growth was decreased. The optimal seedling density was 104 seedlings per $m^2$. The standard seedlings for plantation were above height 55cm, root collar diameter 8.1mm, and root length 21cm. These results demonstrated that seed of P. amurense requires stratification for the germination promotion.
Wet air oxidation (WAO) of oily sludge was carried out using $Fe^{3+}$ as catalyst, placed in a 0.5 L batch autoclave in the temperature range of $250-330^{\circ}C$. Experiments were conducted to investigate the effects of temperature, the initial COD, reaction time, concentration of catalyst and $O_2$ excess (OE) on the oxidation of the oily sludge. The results showed that in the WAO 88.4% COD was achieved after 9 min reaction at temperature of $330^{\circ}C$, OE of 0.8 and the initial COD of 20000 mg/L. Temperature was found to have a significant impact on the oxidation of oily sludge. Adding a catalyst significantly improved the COD removal. Homogenous catalyst, $Fe^{3+}$, showed effective removal for pollutants. COD removal was 99.7% in the catalytic wet air oxidation (CWAO) over $Fe^{3+}$ catalyst. The results proved that the CWAO was an effective pretreatment method for the oily sludge.
