목적 : 이 연구는 Vipera lebetina turanica의 사독약침액(蛇毒藥鍼液)(Snake venom toxin, SVT)이 인간 신경아세포종의 암세포주인 SK-N-MC 세포에서 암세포성장의 억제 및 그 기전에 대하여 살펴보고자 하였다. 방법 : SVT를 처리한 후 SK-N-MC의 성장억제를 관찰하기 위해 CCK-8 assay와 LDH assay를 시행하였고, apoptosis 평가에는 세포형태의 관찰과 DAPI, TUNEL, Annexin V-PI double staining assay 및 cell detachment assay를 시행하였다. 세포자멸사 관련 세포기전을 보기 위하여 세포주기, 세포내 칼슘량, 세포내 활성산소량 및 미토콘드리아의 세포막전위 변화를 측정하였고, DNA fragmentation assay를 시행하였으며, 세포자멸사 조절 단백인 Bax, Bcl-2, caspase-3, -9의 발현 변화 관찰에는 western blot analysis를 시행하였다. 결과 : SK-N-MC 세포에 SVT를 처리한 후, 신경아세포종 세포의 성장, Apoptosis의 유발 및 기전에 영향을 관찰하여 다음과 같은 결과를 얻었다. 1. SVT를 처리한 SK-N-MC 세포 관찰에서 $20{\mu}g/m{\ell}$ SVT 처리가 암세포성장의 유의한 억제를 나타내었다. 세포독성 관찰에서 SVT처리는 처리하지 않은 것에 비하여 증가를 나타내었다. 2. 세포자멸사 평가에서 SVT를 처리한 SK-N-MC 세포는 세포자멸사의 특징적 형태를 나타내었다. TUNEL assay에서는 세포자멸사 활성세포가 미약하게 나타난 반면 cell detachment assay와 Annexin V-PI double staining에서는 각각 세포박리와 세포자멸사 활성세포의 유의한 증가를 나타내었다. 3. 세포자멸사 관련 세포기전연구에서 SVT를 처리한 SK-N-MC 세포의 세포주기, 세포내 칼슘량 및 DNA fragmentation에는 유의한 변화가 관찰되지 않은 반면 세포내 활성산소 양은 유의한 증가를 나타내었고, 그에 따른 미토콘드리아 세포막 전위의 유의한 변동이 관찰되었다. 4. SVT를 처리한 SK-N-MC는 세포자멸사 관련 단백 발현에서 caspase-9에 대해 유의한 증가를 나타내지 않았으나 Bax 및 caspase-3의 유의한 증가와 Bcl-2의 유의한 감소를 나타내었다. 결론 : 이상의 결과는 SVT가 세포내 활성산소를 증가시키므로 미토콘드리아의 세포막전위에 변화를 일으켜 인간 신경아세포종 세포주인 SK-N-MC의 세포박리와 유관한 세포자멸사를 유발하므로 증식억제 효과가 있음을 입증한 것이다.
Effect of insecticide carbofuran and phenobarbital sodium(PB) or 3-methylcholanthrene(3-MC), they were orally administered by the chemicals, alone or in combination, on activities of several enzymes in rats were investigated. In in vivo test for the effect of this chemicals on activity of enzyme in rat, activities of acetylcholinesterase(AChE) and butyrylcholinesterase(BuCheE) were inhibited by $20{\sim}70%$ for 48 hrs after the oral administration of carbofuran alone of 3.8mg/kg, whereas those were lowered at the beginning, but recovered to the control level after 24 hrs, in case of the mixed administration of carbofuran+PB or carbofuran+3-MC. The activity of glutathione S-transferase(GST) was inhibited by more than 15 to 35% for an early period of 0.5 to 6 hrs, in the case of the administration of carbofuran alone, whereas that was slightly inhibited at the beginning, recovered almost to the control level after 3 hrs, and raised by mere than 20% above the control after 6 hrs, in case of the mixed administration of carbofuran+PB or carbofuran+3-MC. When carbofuran was administered alorig with PB or 3-MC, the activities of UDP-glucuronosyltransferase(UDPGT) and cytochrome P-450 were more than 2.6 to 2.8 times higher than that in the case of the administration of carbofuran alone for 6 hrs. These results suggest that a simultaneous application of carbofuran and PB or 3-MC is critical for the enhancentment of activity of GST, UDPGT and cytochrome P450 and the protection of rat from carbofuran toxicity.
This study was carried out to investigate the effects of phenobarbital sodium(PB) and 3-methylcholanthrene(3-MC) on carbofuran cytotoxicity and to develop antitoxic agents based on the effectivness. Experimental groups for carbofuran cytotoxicity were divided into five groups ; medium alone and four treatments of carbofuran (1, 25, 50 and $100{\mu}M)$, and those for compensation effects were divided into six groups ; medium alone, $IC_{50}$ carbofuran and four combinations of carbofuran and PB or 3-MC($IC_{50}$ carbofuran plus 1, 25, 50, $100{\mu}M$ of PB and 3-MC, respectively). After incubation for 48 hrs under the same conditions, MTT(Tetrazolium MTT), NR(Neutral red) and SRB(Sulforhodamine B protein) assay were performed. Fifty percentage inhibition of MTT, NR, and SRB against carbofuran in rat fibroblast cell were 60.7, 82.5 and $87.0{\mu}M$, respectively. At the combination treatments of $IC_{50}$ of carbofuran and $100{\mu}M$ of PB, the significant compensation effects were observed from the results of MTT and NR but not from that of SRB absorbance. And at the combination treatments of $IC_{50}$ of carbofuran and 3-MC, the relatively significant compensation effects were found at $50{\mu}M$ 3-MC from the results of MTT and at $100{\mu}M$ 3-MC from that of NR and SRB absorbances, respectively. From the results of light microscopy, combination treatments of $carbofuran(IC_{50})$ and PB or 3-MC showed good regeneration in carbofuran toxicity of rat fibroblast cells. These results suggest that PB or 3-MC can compensate the cytotoxity of carbofuran insecticide in rat NIH3T3 fibroblast cells.
Unique cartilage matrix-associated protein (UCMA) is an extrahepatic vitamin K-dependent protein rich in γ-carboxylated (Gla) residues. UCMA has been recognized for its ability to promote osteoblast differentiation and enhance bone formation; however, its impact on osteoblasts under hyperglycemic stress remains unknown. In this paper, we investigated the effect of UCMA on MC3T3-E1 osteoblastic cells under hyperglycemic conditions. After exposure to high glucose, the MC3T3-E1 cells were treated with recombinant UCMA proteins. CellROX and MitoSOX staining showed that the production of reactive oxygen species (ROS), which initially increased under high-glucose conditions in MC3T3-E1 cells, decreased after UCMA treatment. Additionally, quantitative polymerase chain reaction revealed increased expression of antioxidant genes, nuclear factor erythroid 2-related factor 2 and superoxide dismutase 1, in the MC3T3-E1 cells exposed to both high glucose and UCMA. UCMA treatment downregulated the expression of heme oxygenase-1, which reduced its translocation from the cytosol to the nucleus. Moreover, the expression of dynamin-related protein 1, a mitochondrial fission marker, was upregulated, and AKT signaling was inhibited after UCMA treatment. Overall, UCMA appears to mitigate ROS production, increase antioxidant gene expression, impact mitochondrial dynamics, and modulate AKT signaling in osteoblasts exposed to high-glucose conditions. This study advances our understanding of the cellular mechanism of UCMA and suggests its potential use as a novel therapeutic agent for bone complications related to metabolic disorders.
The preparation and the functional properties of methylcellulose (MC) and hydroxypropyl-methylcellulose (HPMC) edible films with and without calcium were investigated. All the prepared films exhibited transparent and whitish color with $2.38{\sim}3.55$ haze intensity. Tensile strength of MC films were stronger than HPMC films with and without calcium, and moreover addition of calcium increased tensile, but elongation of HPMC film was specially lower than the other films. Solubility of films did not differ with calcium addition but decreased with increasing viscosity in HPMC films. water vapor transmission rate (WVTR) of HPMC and MC film were not affected by calcium, but viscosity of film's raw material was important to determine WVTR in HPMC. Oxygen permeabilities of MC films were lower than those of HPMC films, and became lower with calcium addition. According to scanning electron microscope (SEM) observation on the surface characteristics, MC film with calcium had relatively uniform and smooth surface than HPMC films.

MC-DS-CDMA is technique where a single data symbol is transmitted at multiple subcarriers which are orthogonal to each other. With this technique, frequency diversity can be achieved. Time variant channels lead to interchannel interference which increases the bit error rate for MC-DS-CDMA systems. The performance of PSAM MC-DS-CDMA system over time variant channels is analyzed. The BER performance of this system over multipath fading environment is evaluated, considering the channel estimation error, carrier frequency offset.

Purpose : This study was aimed at investgating the effect of McKenzie exercise on temporomandibular joint disorder (TMJD) and body balance. Methods : Thirty subjects with TMJD were selected. They were randomly assigned to one of two groups (15 in each group) : namely the conservative treatment group and McKenzie exercise group. The conservative treatment group were treated physical therapy for 6 weeks (3 times a week). The McKenzie exercise group performed McKenzie exercise for 6 weeks (3 times a week). We measured mouth opening, pain, muscle activation and body balance before and after exercise by using the maximum mouth opening MMO), visual analog scale (VAS), electromyography and Biodex Balance System, respectively. Results : We found statistically significant differences in pain and mouth opening in the conservative treatment group and McKenzie exercise group before and after (p<.05). Conclusion : We confirmed the effect of McKenzie exercise on TMJD. Thus we thought these results could be used as basic data and reference for TMJD. But we need more study effect of McKenzie exercise on TMJD and body balance.
This study investigated whether or not the major bioactive compounds of Noni (Morinda citrifolia) are involved in anti-inflammatory activity through the JAK/STAT upper signaling pathway in RAW 264.7 cells. The experimental results show that the M. citrifolia ethyl acetate fraction (Mc-EtOAc) obtained by sequential fractionation with organic solvents from the plant's dried fruits exhibits the highest antioxidant activity. In addition, the cytoprotective effects of Mc-EtOAc against H2O2-induced oxidative stress in the RAW 264.7 cells suppressed cytotoxicity in a dose-dependent manner. The group pretreated with Mc-EtOAc at a concentration of 240 ㎍/ml showed higher cell viability of 84.5%, compared to 71.6% in the LPS-treated group, and LPS-induced NO production decreased to half the amount in the positive control group. Mc-EtOAc treatment also led to a significant dose-dependent reduction in iNOS expression. Although COX-2 expression was increased by 300% following LPS induction, it was significantly decreased in a dose-dependent manner by pretreatment with Mc-EtOAc at concentrations of 120 and 240 ㎍/ml. An inhibition of the mRNA expression of pro-inflammatory cytokines IL-1β and TNF-α was observed. The investigation also revealed that the phosphorylation levels of pJAK1 and pSTAT3 in LPS-induced RAW 264.7 cells were significantly reduced by Mc-EtOAc treatment.
Osteoporosis is a metabolic bone disease that is characterized by low bone mass resulting from an increase in bone resorption relative to bone formation. The most current therapies for osteoporosis have focused on inhibiting bone resorption by osteoclasts. The purpose of this study is to develop new anabolic agents for treatment of osteoporosis that have fewer risks compared to conventional therapies. We searched the natural products that were derived from the traditional Asian medicines which have been used for treatment of bone related diseases. Icaritin is a flavonoid glycoside derived from the herb Epimedium which has beneficial effects on bone formation. To determine the effect of icaritin on bone formation, we examined the effect of icaritin on MC3T3-E1 cell proliferation and differentiation. For determining the effects of icaritin on proliferation, we performed the MTT assay using MC3T3-E1 cells. To evaluate whether icaritin could promote the osteogenic differentiation of MC3T3-E1 cells, alkaline phosphatase (ALP) activity and mRNA expressions of Runx2, osteocalcin (OCN), RANKL, and osteoprotegerin (OPG) were determined. Icaritin increased MC3T3-E1 cell proliferation. Icaritin increased the ALP activity of MC3T3-E1 cells on 72 hour culture in osteogenic media. mRNA expression of Runx2 was increased after 24 hour culture with icaritin. mRNA expression of osteocalcin was increased after 72 hour culture with icaritin. In addition, icaritin increased the mRNA expressions of OPG and RANKL. However, icaritin increased the mRNA expression of OPG much more than that of RANKL, and then, it increased the OPG/RANKL ratio. These results suggest that icaritin promotes osteogenic differentiation of osteoblasts and decreases osteoclast formation regulated by osteoblasts.

The purpose of this research is to suggest diverse directions for eco-friendly knit designing through analyzing the characteristics of Stella McCartney's knit designs. The first stage of the research was to explore the characteristics of eco-friendly fashion design based on literature review. The next stage was to categorize the characteristics of eco-friendly design found in Stella McCartney' knit fashion based on the precedent analysis. The data collected showed 274 examples from 40 Stella McCartney collections, including Spring, Resort, Pre-Fall, and Fall Ready-to-wear collections between 2013 and 2022. All information was collected using www.vogue.com. As a result, the characteristics of Stella McCartney's knit design were classified into four directopms: 1) Perpetual Naturalism, 2) Cultural Complexity, 3) Practical Functionality, and 4) Subcultural Reproducibility. The first characteristic, 'Perpetual Naturalism' values the continuous symbiosis between animals and human beings, thereby protecting global environmental value. Stella McCartney's knit design continuously showed a longing for nature's beauty through fashion design, which allowed people to enjoy the meaning of symbiosis between human beings and animals. Second, 'Cultural Complexity' is the characteristic that appears in Stella McCartney's knit fashion design when collaborating with various artists and/or mixing traditional knit motives inspired by traditional cultures and retro moods. Third, 'Practical Functionality' is the design characterisitic that allows items to be worn for a long time as it is comfortable, simple, and practical. Stella McCartney's knit designs pursue easy-to-wear designs that are comfortable and have practical designs with simple details. Lastly, 'Subcultural Reproducibility' showed in Stella McCartney's knit designs reflect Hippie culture, which pursued a rejection of conventional values, and promoted sustainable energy. This culture peaked in the 1960s and 1970s, when 'Beatles' were mainly working. Additionally, retro styled knit designs inspired by other various music genres from the 1980s and 1990s appeared in this category as well.
