Vinylsulfilimine유도체에 대한 1-methyl-5-mercapto-1,2,3,4-tetrazole의 친핵성 첨가물에 관한 연구 (Synthetic Studies on the Nucleophilic Addition of 1-Methyl-5-mercapto-1,2,3,4-tetrazole to Vinylsulfilimines)

  • Vinylsulfilimine 유도체(H, p-$CH_3$, m-TEX>$CH_3$, p-Cl, p-Br, p-$OCH_3$, 및 p-$NO_2$)에 1-methyl-5-mercapto-1,2,3,4-tetrazole을 반응시켜 다음 7가지의 새로운 호합물을 합성하였다. S-Phenyl-S-2-(1-methyl-1,2,3,4-tetrazole-5-thio)-ethyl-N-p-tosylsulfilimine, S-p-tolyl-S-2-(1-methyl-1,2,3,4-tetrazole-5-thio)-ethyl-N-p-tosylsulfilimine, S-m-tolyl-S-2-(1-methyl-1,2,3,4-tetrazole-5-thio)-ethyl-N-p-tosylsulfilimine, S-p-chlorophenyl-S-2-(1-methyl-1,2,3,4-tetrazole-5-thio)-ethyl-N-p-tosylsulfilimine, S-p-bromophenyl-S-2-(1-methyl-1,2,3,4-tetrazole-5-thio)-ethyl-N-p-tosylsulfilimine, S-p-methoxyphenyl-S-2-(1-methyl-1,2,3,4-tetrazole-5-thio)-ethyl-N-p-tosylsulfilimine and S-p-nitrophenyl-S-2-(1-methyl-1,2,3,4-tetrazole-5-thio)-ethyl-N-p-tosylsulfilimine. 이 화합물들의 구조는 원소분석, MP, UV, IR 및 NMR 스펙트럼에 의해 확인되었다.

Analysis of a Sphingosine 1-phosphate Receptor $hS1P_3$ in Rat Hepatoma Cells

  • To examine intracellular signaling of human $S1P_3\;(hS1P_3),$ a sphingosine 1-phosphate (S1P) receptor in plasma membrane, $hS1P_3$ DNA was transfected into RH7777 rat hepatoma cell line, and the inhibition of forskolin-induced cAMP accumulation and activation of MAP kinases by S1P were tested. In $hS1P_3$ transformants, S1P inhibited forskolin-induced activation of adenylyl cyclase activity by about 80% and activated MAP kinases in dose-dependent and pertussis-toxin (PTX) sensitive manners. In oocytes expressing $hS1P_3$ receptor, S1P evoked $Cl^-$ conductance. These data suggested that PTX-sensitive G proteins are involved in $hS1P_3-mediated$ signaling, especially the positive action of S1P in cell proliferation. The potential advantages of rat hepatoma cells for the research of sphingosine 1-phosphate receptor are discussed.

Synthesis of Nucleophilic Adducts of Thiols (Ⅰ). Addition of Cysteine to $\beta$-Nitrostyrene Derivatives

  • The addition reactions of cysteine without blocking amino and carboxyl groups to substituted and unsubstituted ${\beta}$-nitro-styrene derivatives were investigated. ${\beta}$-Nitrostyrene(1a), p-methyl-${\beta}$-nitrostyrene(1b), 3,4,5-trimethoxy-$[\beta}$ -nitrostyrene(1c), $[\varpi}$-3,4-methylenedioxy-${\beta}$ -nitrostyrene(1d), o-, m- and p-chloro-${\beta}$ -nitrostyrene (1e, 1f, 1g) and o-, m- and p-methoxy-${\beta}$-nitrostyrene (1h, 1i, 1j) easily undergo addition reactions with cysteine to form S-(2-nitro-1-phenylethyl)-L-cysteine(3a), S-[2-nitro-1-(p-methyl)phenyl-ethyl]-L-cysteine(3b), S-[2-nitro-1-(3',4',5'-trimethoxy) phenylethyl]-L-cysteine(3c), S-[2-nitro-1-($[\vatpi}$ -3',4'-methylenedioxy)phenylethyl]-L-cysteine(3d), S-[2-nitro-1-(o-chloro)phenylethyl]-L-cysteine(3e), S-[2-nitro-1-(m-chloro)-phenylethyl]-L-cysteine(3f), S-[2-nitro-1-(p-chloro)phenylethyl]-L-cysteine(3g), S-[2-nitro-1-(o-methoxy)phenylethyl]-L-cysteine(3h), S-[2-nitro-1-(m-methoxy)phenylethyl]-L-cysteine(3i) and S-[2-nitro-1-(p-methoxy)phenylethyl]-L-cysteine(3j), respectively. The structure of adducts were confirmed by means of UV-spectrum, IR-spectrum, molecular weight measurement and elemental analysis. The various factors effecting the yield of cysteine adducts to ${\beta}$-nitrostyrene derivatives were also studied.

Pro-Inflammatory Role of S1P3 in Macrophages

  • Sphingosine kinase 1 and its product, sphingosine 1-phosphate (S1P), as well as their receptors, have been implicated in inflammatory responses. The functions of receptors $S1P_1$ and $S1P_2$ on cell motility have been investigated. However, the function of $S1P_3$ has been poorly investigated. In this study, the roles of $S1P_3$ on inflammatory response were investigated in primary peritoneal macrophages. $S1P_3$ receptor was induced along with sphingosine kinase 1 by stimulation of lipopolysaccharide (LPS). LPS treatment induced inflammatory genes, such iNOS, COX-2, $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$. TY52156, an antagonist of $S1P_3$ suppressed the induction of inflammatory genes in a concentration dependent manner. Suppression of iNOS and COX-2 induction was further confirmed by western blotting and NO measurement. Suppression of $IL-1{\beta}$ induction was also confirmed by western blotting and ELISA. Caspase 1, which is responsible for $IL-1{\beta}$ production, was similarly induced by LPS and suppressed by TY52156. Therefore, we have shown $S1P_3$ induction in the inflammatory conditions and its pro-inflammatory roles. Targeting $S1P_3$ might be a strategy for regulating inflammatory diseases.

Luminescence of CaS:Bi

  • Luminescence of bismuth activated CaS, CaS:Bi, prepared in sodium polysulfide is studied. Excitation spectrum of CaS:Bi shows a band at 350 nm due to the recombination process between holes in Na+Ca2+ and electrons in conduction bands, in addition to bands at 260 nm from band gap of CaS, and at 320 nm (1S0→1P1) and at 420 nm (1S0→3P1) from electronic energy transitions of Bi. Emission band at 450 nm is from 3P1→1S0 transition of Bi3+, bands at 500 nm and 580 nm correspond to recombinations of electron donors (Bi3+Ca2+ and VS2-) with acceptors (VCa2+ and Na+Ca2+). Emission band of 3P1→1S0 transition is shifted to longer wavelength from CaS:Bi to BaS:Bi, due to the increase of the Stokes shift by the decrease of the crystal field parameter from CaS:Bi to BaS:Bi.

The Formation of Small Satellites near the Saturnian A-Ring

  • The origin of five small satellites (1980 S1, 1980 S3, 1980 S26, 1980 S27 and 1980 S28) and F-ring of Saturn which were recently discovered inside the orbit of Mimas, is proposed in terms of the resonant effects by external satellites in the Saturnian primordial extended ring. From the calculations of resonance locations and strengths, the following conclusion could be derived. Assuming that present two objects 1980 S1 and 1980 S3 were originally a single object 1980 S1+S3, 1980 S1+S3 had been formed due to Enceladus' 2/1 resonance and 1980 S26 due to Mimas' 3/2 resonance at first and then 1980 S28. 1980 S27 and F-ring were formed in the next due to 1980 S1+S3's 8/7, 9/8 and 10/9 resonances, respectively.

Optimal Timing to Assess Drain Amylase Concentration after Elective Gastrectomy

  • Purpose: While the amylase concentration of the drainage fluid (dAmy) has been reported to be a predictor of postoperative pancreas-related complications (PPRC), the optimal timing for its measurement has not been fully investigated. Materials and Methods: The clinicopathological data of 387 patients who underwent elective gastrectomy for gastric cancer were reviewed. Laboratory data, including dAmy on postoperative days 1 (dAmy1) and 3 (dAmy3), and serum C-reactive protein (sCRP) concentrations on postoperative days 1 (sCRP1) and 3 (sCRP3) were compared between patients with PPRC and without PPRC. Results: Nineteen of the 387 patients (4.9%) developed PPRC. The optimal cutoff values of dAmy1, dAmy3, sCRP1, and sCRP3 were 1514 IU/L, 761 IU/L, 8.32 mg/dL, and 15.15 mg/dL, respectively. The area under the curve of dAmy1 was greater than that of dAmy3 (0.915 vs. 0.826), and that of sCRP3 was greater than that of sCRP1 (0.820 vs. 0.659). In the multivariate analysis, dAmy1 (P<0.001) and sCRP3 (P=0.004) were significant predictors of PPRC, while dAmy3 (P=0.069) and sCRP1 (P=0.831) were not. Thirteen (41.9%) of 31 patients with both dAmy1 ≥1,545 IU/L and sCRP3 ≥15.15 mg/dL had PPRC ≥Clavien-Dindo II. In contrast, among 260 patients with both dAmy1 <1,545 IU/L and sCRP3 <15.15 mg/dL, none developed PPRC. Conclusions: dAmy1 was more useful than dAmy3 in predicting PPRC. The combination of dAmy1 and sCRP3 may be a useful criterion for the removal of drains on postoperative day 3.

1-히드록시카르바페넴의 합성에 관한 연구 (Studies on the Synthesis of 1-Hydroxycarbapenems)

  • 4-styryl-2-azetidinone 7b의 bromoacetate에 의한 N-알킬화, 가오존분해, 산화, 말론산 에틸 마그네슘과의 축합 및 디아조 전달반응의 일련의 과정을 거처서 (3S,4S)-1-(t-butoxycarbonylmethyl)-3-[(S)-1-(t-butyldimethylsilyloxy)ethyl]-4-(2-diazo-2-ethoxycarbonyl-1-oxoethyl)-2-azetidinone(14)을 합성하였다. 또한 4-formyl-2-azetidinone 5b 의 Wittig 반응, ethoxycarbonylformonitrile oxide와의 1,3 이중극성 고리화 첨가반응, 촉매 수소화 및 디아조화 반응의 일련의 과정을 거쳐서 (3S,4S)-3-[(S)-1-(t-butyldimethylsilyloxy)ethyl]-4-(3-diazo-3-ethoxycarbonyl-1-hydroxypropyl)-2-azetidinone(21)을 합성하였다. 그러나 화합물 14 또는 21을 $Rh_2(OAc)_4$ 로 처리하여 1-hydroxycarbapenem 또는 1-hydroxycarbapenam으로 고리화하는 것을 성공하지 못하였다.

ZnO의 과잉첨가가 Ba(${Zn}_{1/3}{Ta}_{2/3}$)$O_3$세라믹스의 마이크로파 유전특성에 미치는 영향 (Effect of Excess ZnO on Microwave Dielectric Characteristics of Ba(${Zn}_{1/3}{Ta}_{2/3}$)$O_3$ Ceramics)

  • Dielectric properties of Ba(ZnS11/3TTaS12/3T)OS13T+x ZnO(x=0, 0.4, 0.8, 1.0 wt%) ceramics have been investigated at microwave frequencies. With excess ZnO, the sinterability was improved and the dielectric constant($\varepsilon$S1rT) and the unloaded quality factor(QS1UT) were increased. The structure changed into hexagonal from pseudocubic as being annealed at 140$0^{\circ}C$ in excess ZnO composition. Also, the temperature coefficient of the resonant frequency ($\tau$S1fT) turned into (-)ppm/$^{\circ}C$ when sintered at 155$0^{\circ}C$ for 2 hours. But the specimen sintered in ZnO muffling showed increased density and $\varepsilon$S1rT but lowerde QS1uT. Among the specimen investigated, expecially the composition added with 0.4wt% excess ZnO showed the most optimum dielectric values ($\varepsilon$S1rT=28, QS1uT x f=120000GHz) better than those of original Ba(ZnS11/2T TaS12/3T)OS13T ceramics.

  • M(S-S,ph)(N-N,H) (M = Ni(II), Pd(II), Pt(II); (S-S,ph) = 1,2-diphenylethylenedithiolate; (N-N,H)=1,10-phenanthroline) 착물의 친전자성 및 친핵성 리간드 반응을 조사하였다. norbornadiene과의 반응성은 중심금속의 역결합의 정도에 의존하며 2,5-dithia-3,4-diphenyl-tricyclo[4,4,1,0]-undeca-3,8-diene을 생성하였다. methyl iodide와의 반응은 (N-N,H) 리간드의 이탈 능력에 지배되며 메틸화된 $M(S-S,ph)_2$ 착물을 생성하였다. 이 반응의 주생성무리은 열 분해에 의해 얻어진 ${\alpha},{\alpha}{\prime}$,-bismethylthiostibene $(CH_3S-SCH_3,ph)$의 구조로부터 M(S-S,CN)(N-N,H)((S-S,CN) = 1,2-dicyanoethylenedithiolate)의 새로운 혼합 리간드 착물을 합성하였다.
