Glyoxalase I catalyzes the conversion of methylglyoxal to S-D-lactoylglutathione in the presence of glutathione. The structural gene of glyoxalase I (GLO1) was cloned from an osmotolerant yeast, Candida magnoliae, which produces a functional sweetener, erythritol, from sucrose. DNA sequence analysis revealed that the uninterrupted open reading frame (ORF) of C. magnoliae GLO1 (CmGLO1) spans 945 bp, corresponding to 315 amino acid residues, and shares 45.2% amino acid sequence identity to Saccharomyces cerevisiae Glo1. The cloned ORF in a multicopy constitutive expression plasmid complemented the glo1 mutation of S. cerevisiae, confirming that it encodes Glo1 in C. magnoliae. The responses of CmGLO1 to environmental stresses were different from those of S. cerevisiae, which only responds to osmotic stress. An enzyme activity assay and reverse transcription polymerase chain reaction revealed that the expression of CmGLO1 is induced by stress inducers such as methylglyoxal, $H_2O_2$, KCl, and NaCl. The researches related to active control systems utilizing superelastic shape memory alloys (SMA) have been recently conducted to reduce critical damage due to lateral deformation after severe earthquakes. Although Superelastic SMAs undergo considerable inelastic deformation, they can return to original conditions without heat treatment only after stress removal. We can expect the mitigation of residual deformation owing to inherent recentering characteristics when these smart materials are installed at the part where large deformation is likely to occur. Therefore, the primary purpose of this research is to develop concentrically braced frames (CBFs) with superelastic SMA bracing systems and to evaluate the seismic performance of such frame structures. In order to investigate the inter-story drift response of CBF structures, 3- and 6-story buildings were design according to current design specifications, and then nonlinear time-history analyses were performed on numerical 2D frame models. Based on the numerical analysis results, it can be comparatively verified that the CBFs with superelastic SMA bracing systems have more structural advantages in terms of energy dissipation and recentering behavior than those with conventional steel bracing systems.

The control of the deformative behaviour of pre-stressed concrete roof elements for a satisfactory service performance is a main issue of their structural design. Slender light-weight wing-shaped roof elements, typical of the European heritage, are particularly sensitive to this problem. The paper presents the results of deformation measurements during storage and of both torsional-flexural and purely flexural load tests carried out on a full-scale 40.5 m long innovative wing-shaped roof element. An element-based simplified integral procedure that de-couples the evolution of the deflection profile with the progressive shortening of the beam is adopted to catch the experimental visco-elastic behaviour of the element and the predictions are compared with normative close-form solutions. A linear 3D fem model is developed to investigate the torsional-flexural behaviour of the member. A mechanical non-linear beam model is used to predict the purely flexural behaviour of the roof member in the pre- and post-cracking phases and to validate the loss prediction of the adopted procedure. Both experimental and numerical results highlight that the adopted analysis method is viable and sound for an accurate simulation of the service behaviour of precast roof elements.

Cu/C films were prepared at room temperature under $Cu(hfac)_2-Ar-H_2$ atmosphere in order to obtain metallized polymer by using ECR-MOCVD(Electron Cyclotron Resonance Metal Organic Chemical Vapor Deposition) coupled with a DC bias system. The room temperature MOCVD on polymer substrate could be possible by collaboration of ECR and a DC bias. Structural analysis of the films by ECR was found that fine copper grains embedded in an amorphous polymer matrix with indistinctive interfacial layer. The increase in $H_2$ contents brought on copper-rich film formation with low electric resistance. On the other hand carbon-rich films with low sheet electric resistance were prepared in argon atmosphere. The electric sheet resistance of Cu/C films with good interfacial property were controlled at $10^8$~$10^0$ Ohm/sq. ranges by the $H_2$/Ar mole ratio and the shielding effectiveness of the film showed maximum up to 45dB in the our experimental range.
In meeting the technical needs for deepwater conditions and overcoming the shortfalls of single-layer pipes for deepwater applications, pipe-in-pipe (PIP) systems have been developed. While, for PIP pipelines directly laid on the seabed or with partial embedment, one of the primary service risks is lateral buckling. The critical axial force is a key factor governing the global lateral buckling response that has been paid much more attention. It is influenced by global imperfections, submerged weight, stiffness, pipe-soil interaction characteristics, et al. In this study, Finite Element Models for imperfect PIP systems are established on the basis of 3D beam element and tube-to-tube element in Abaqus. A parameter study was conducted to investigate the effects of these parameters on the critical axial force and post-buckling forms. These parameters include structural parameters such as imperfections, clearance, and bulkhead spacing, pipe/soil interaction parameter, for instance, axial and lateral friction properties between pipeline and seabed, and load parameter submerged weight. Python as a programming language is been used to realize parametric modeling in Abaqus. Some conclusions are obtained which can provide a guide for the design of PIP pipelines.
Protein modifications of recombinant pharmaceuticals have been observed both in vitro and in vivo. These modifications may result in lower efficacy, as well as bioavailability changes and antigenicity among the protein pharmaceuticals. Therefore, the contents of modification should be monitored for the quality and efficacy of protein pharmaceuticals. The interface of EPO and its receptor was visualized, and potential amino acids interacting on the interface were also listed. Two different types of modifications on the interface were identified in the preparation of rHu-EPO BRP. A UPLC/Q-TOF MS method was used to evaluate the modification at those variants. The modification of the oxidized variant was localized on the Met54 and the deamidated variants were localized on the Asn47 and Asn147. The extent of oxidation at Met54 was 3.0% and those of deamidation at Asn47 and Asn147 were 2.9% and 4.8%, respectively.

In this study, in order to investigate a modeling technique of the structure with bolted joints, four kinds of finite element model are introduced; a solid bolt model, a coupled bolt model, a spider bolt model, and no bolt model. All proposed models take account on prestrained effect and contact behavior of flanges to be joined. Among these models, a solid bolt model, which is modeled by using a 3-D solid element and a surface-to-surface contact element between the head/nut and the flange interfaces, has the best accurate responses compared with the experimental results. In addition, coupled bolt model, which couples the degree of freedom between the head/nut and the flange, shows the best effectiveness and usefulness in view of computational time and memory usage. Finally, the bolt model proposed here is adopted for structural analysis of a large diesel engine of a ship consisting of several parts which is connected by long stay bolts.
A new finite shear wall element model and a method for calculation of 3D multi-storied only shear walled or shear walled - framed structures using finite shear wall elements assumed ideal elasto - plastic material are developed. The collapse load of the system subjected to factored constant gravity loads and proportionally increasing lateral loads is calculated with a method of load increments. The shape functions over the element are determined as a cubic variation along the story height and a linear variation in horizontal direction because of the rigid behavior of the floor slab. In case shear walls are chosen as only one element in every floor, correct solutions are obtained by using this developed element. Because of the rigid behavior of the floor slabs, the number of unknowns are reduced substantially. While in framed structures, classical plastic hinge hypothesis is used, in nodes of shear wall elements when vertical deformation parameter is exceeded ${\varepsilon}_e$, this node is accepted as a plastic node. While the system is calculated with matrix displacement method, for determination of collapse safety, plastic displacements and plastic deformations are taken as additional unknowns. Rows and columns are added to the system stiffness matrix for additional unknowns.
Purpose : This study is to define the structural patterns of gender roles of women as perceived by co-eds. Method : Q-methodology was used on 174 statements collected through interviews with women aged between 20 an d 50. 38 Q-samples were selected from 330 Q-population. The Q-samples were administered to 29 co-eds. Result : Analysis of Q-type obtained by QUANAL program revealed three types of subjectivity in the perception of gender roles. Type 1, role is characterized by the tendency to play down the female gender role and think that women's body is an essential factor in forming the female gender role. Type 2, rejects motherhood image obedience and chastity dictated by male chauvinism. Type 3, shows a tendency to believeth at female gender cannot be separated from maternity and that women's basic role is housekeeping. The three types were common in believing in self-development and equal rights through economical in dependence.
In a super-large underground with "large span and high side wall", it is buried in mountains with uneven lithology, complicated geostress field and developed geological structure. These surrounding rocks are more susceptible to stability issues during the construction period. This paper takes the left bank of Baihetan hydropower station (span is 34m) as a case study example, wherein the deformation mechanism of surrounding rock appears prominent. Through analysis of geological, geophysical, construction and monitoring data, the deformation characteristics and factors are concluded. The failure mechanism, spatial distribution characteristics, and evolution mechanism are also discussed, where rock mechanics theory, $FLAC^{3D}$ numerical simulation, rock creep theory, and the theory of center point are combined. In general, huge underground cavern stability issues has arisen with respect to huge-scale and adverse geological conditions since settling these issues will have milestone significance based on the evolutionary pattern of the surrounding rock and the correlation analyses, the rational structure of the factors, and the method of nonlinear regression modeling with regard to the construction and development of hydropower engineering projects among the worldwide.
