Long Noncoding RNA HOXA11-AS Modulates the Resistance of Nasopharyngeal Carcinoma Cells to Cisplatin via miR-454-3p/c-Met

  • To elucidate the mechanism of action of HOXA11-AS in modulating the cisplatin resistance of nasopharyngeal carcinoma (NPC) cells. HOXA11-AS and miR-454-3p expression in NPC tissue and cisplatin-resistant NPC cells were measured via quantitative reverse transcriptase polymerase chain reaction. NPC parental cells (C666-1 and HNE1) and cisplatin-resistant cells (C666-1/DDP and HNE1/DDP) were transfected and divided into different groups, after which the MTT method was used to determine the inhibitory concentration 50 (IC50) of cells treated with different concentrations of cisplatin. Additionally, a clone formation assay, flow cytometry and Western blotting were used to detect DDP-induced changes. Thereafter, xenograft mouse models were constructed to verify the in vitro results. Obviously elevated HOXA11-AS and reduced miR-454-3p were found in NPC tissue and cisplatin-resistant NPC cells. Compared to the control cells, cells in the si-HOXA11-AS group showed sharp decreases in cell viability and IC50, and these results were reversed in the miR-454-3p inhibitor group. Furthermore, HOXA11-AS targeted miR-454-3p, which further targeted c-Met. In comparison with cells in the control group, HNE1/DDP and C666-1/DDP cells in the si-HOXA11-AS group demonstrated fewer colonies, with an increase in the apoptotic rate, while the expression levels of c-Met, p-Akt/Akt and p-mTOR/mTOR decreased. Moreover, the si-HOXA11-AS-induced enhancement in sensitivity to cisplatin was abolished by miR-454-3p inhibitor transfection. The in vivo experiment showed that DDP in combination with si-HOXA11-AS treatment could inhibit the growth of xenograft tumors. Silencing HOXA11-AS can inhibit the c-Met/AKT/mTOR pathway by specifically upregulating miR-454-3p, thus promoting cell apoptosis and enhancing the sensitivity of cisplatin-resistant NPC cells to cisplatin.

Multifrequency monitoring of a flaring gamma-ray blazar 3C454.3 at 22 and 43GHz

  • We report the results of the monitoring of a flaring gamma-ray blazar, 3C454.3 in total flux density at 22 and 43GHz and in polarization at 22GHz with KVN Ulsan 21-m radio telescope every 3-4 days from 19 November 2010 to 31 January 2011. After an extraordinary 5-day gamma-ray outburst in November 2010, the radio total flux density at 22/43GHz and the linear polarization at 22GHz has been decreased with a variation of a short time scale. In this paper, we also discuss a spectral change of 3C454.3 at 22 and 43GHz after the extraordinary gamma-ray outburst.

A SEARCH FOR AGN INTRA-DAY VARIABILITY WITH KVN

  • Active galactic nuclei (AGN) are known for irregular variability on all time scales, down to intra-day variability with relative variations of a few percent within minutes to hours. On such short timescales, unexplored territory, such as the possible existence of a shortest characteristic time scale of activity and the shape of the high frequency end of AGN power spectra, still exists. We present the results of AGN single-dish fast photometry performed with the Korean VLBI Network (KVN). Observations were done in a “anti-correlated” mode using two antennas, with always at least one antenna pointing at the target. This results in an effective time resolution of less than three minutes. We used all four KVN frequencies, 22, 43, 86, and 129 GHz, in order to trace spectral variability, if any. We were able to derive high-quality light curves for 3C 111, 3C 454.3, and BL Lacertae at 22 and 43 GHz, and for 3C 279 at 86 GHz, between May 2012 and April 2013. We performed a detailed statistical analysis in order to assess the levels of variability and the corresponding upper limits. We found upper limits on flux variability ranging from ~1.6% to ~7.6%. The upper limits on the derived brightness temperatures exceed the inverse Compton limit by three to six orders of magnitude. From our results, plus comparison with data obtained by the University of Michigan Radio Astronomy Observatory, we conclude that we have not detected source-intrinsic variability which would have to occur at sub-per cent levels.

A Study on the Distribution of Summer Water Temperatures of the Central Coast of the Southern Sea of Korea Using Numerical Experimentation (수치실험을 이용한 남해 중부 연안의 하계 수온 분포 연구)

  • To understand the spatial-temporal distribution of seawater in Korea's South Sea, seawater movement and the distribution of water temperature has been analyzed using a hydrodynamic model (the Princeton Ocean Model). The directions of tidal currents were generally westward during flood tides and eastward during ebb tides. Northeastward Tsushima Warm Currents in the open sea, which is deeper than 50m were stronger than in coastal areas. Analysis of data from the hydrodynamic model showed that the water temperature in the semi-closed bay was relatively higher ($26{\sim}28^{\circ}C$) than in the open sea ($18{\sim}22^{\circ}C$). The exchange volume of semi-closed seawater was $10,331m^3/sec$ in Gwangyang Bay, $16,935m^3/sec$ in Yeosu-Gamag Bay and $13,454m^3/sec$ in Geoje-Hansan Bay. Therefore, it was shown that the lower seawater exchange volume is, the higher seawater temperature will be.

Properties of Chemical Compositions of Safflower Seeds (산지별 홍화씨의 화학적 성분 특성)

  • This study was carried out to investigate the chemical properties of safflower seeds cultivated at some areas in Korea. Safflower was used as one of the functional foods and medicinal plants for many centuries. Porximate compositions of safflower seeds were moisture, 4.8~8.1%, crude protein 16.5~19.5%, crude ash 2.7~3.3%, curde fiber 38.5~43.4%, and crude fat 16.6~24.4%, respectively. Glutamic acid, aspartic acid and arginine were the major amino acids and their contents were 2,587.4~3,143.5 mg%, 1,315.8~1,654.8 mg%, and 1,171.9~1,484.2 mg%. K, P, Ca, and Mg were major minerals and their contents were 611.6~886.3 mg%, 501.5~596.7 mg%, 208.5~641.2 mg%, and 530.6~639.5 mg%, respectively. The free sugars that were identified include raffinose, sucrose, glucose, fructose. ${\alpha}$-tocopherol content was 0.14~3.82 mg%. Contents of vitamin C was 0.43~3.39 mg%. The safflower seeds fatty acids were palmitic acid, stearic acid, oleic acid, linoleic acid and linoleic acid. The major fatty acid was linoleic acid it's content was 74.8~82.9%.

Study on the Sintering Temperature and Electrical Properties of CuO Doped (Ba0.5,Sr0.5)TiO3 Ceramics (CuO를 첨가한 (Ba0.5,Sr0.5)TiO3 세라믹의 소결온도와 전기적 특성의 연구)

  • The influence of CuO addition on what of the $(Ba,Sr)TiO_3$ ceramics was studied. The sintering temperature of $(Ba,Sr)TiO_3$ ceramics was lowered by the addition of CuO additives. The 1 - 5 wt% CuO were selected and employed as the sintering aids. Low-Temperature Co-fired Ceramic technologies are popular technologies used in the manufacture of microwave devices. In this study, crystalline and electrical properties of CuO doped $(Ba,Sr)TiO_3$ ceramics were investigated to determine the low temperature sintering properties. The addition of CuO to $(Ba,Sr)TiO_3$ lowered the sintering temperature from $1350^{\circ}C$ to $1150^{\circ}C$. The dependence of the sintering temperature shrinkage rate and mechanism of CuO doped $(Ba,Sr)TiO_3$ ceramics are investigated and discussed. Also, the crystalline structure of CuO - doped $(Ba,Sr)TiO_3$ ceramics is discussed by the X-ray diffraction (XRD) method.

Impact fracture behavior on particle volume fraction of nano silica composite materials (입자 함유율의 변화에 따른 나노 실리카 복합재료의 충격파괴거동)

  • The present study is undertaken to evaluate the effect of volume fraction on the results of Charpy impact test for the rubber matrix filled with nano sized silica particles composites. The Charpy impact tests are conducted in the temperature range $0^{\circ}C$ and $-10^{\circ}C$. The range of volume fraction of silica particles tested are between 11% to 25%. The critical energy release rate $G_{IC}$ of the rubber matrix composites filled with nano sized silica particles is affected by silica volume fraction and it is shown that the value of $G_{IC}$ decreases as volume fraction increases. In regions close to the initial crack tip, fracture processes such as matrix deformation, silica particle debonding and delamination, and/or pull out between particles and matrix which is ascertained by SEM photographs of Charpy impact fracture surfaces.

The long-term mm/radio activity of active galactic nuclei

  • I present an analysis of the long-term evolution of the fluxes of six active galactic nuclei (AGN) - 0923+392, 3C 111, 3C 273, 3C 345, 3C 454.3, and 3C 84 - in the frequency range 80 - 267 GHz using archival calibration data of the IRAM Plateau de Bure Interferometer. Our dataset spans a long timeline of ~14 years with 974 - 3027 flux measurements per source. We find strong (factors ~2-8) flux variability on timescales of years for all sources. The flux density distributions of five out of six sources show clear signatures of bi- or even multimodality. Our sources show mostly steep (alpha~0.5-1), variable spectral indices that indicate outflow dominated emission; the variability is most probably due to optical depth variations. The power spectra globally correspond to red-noise spectra with five sources being located between the cases of white and flicker noise and one source (3C 111) being closer to the case of random walk noise. For three sources the low-frequency ends of their power spectra appear to be upscaled in spectral power by factors ~2-3 with respect to the overall powerlaws. We conclude that the source emission cannot be described by uniform stochastic emission processes; instead, a distinction of "quiescent" and (maybe multiple) "flare" states of the source emission appears to be necessary.

MOGABA: Monitoring of Gamma-ray Bright AGN with KVN 21-m radio telescopes at 22 and 43GHz

  • We introduce an ongoing project for monitoring total flux density at 22 and 43GHz, linearly polarized flux, and polarization angle at 22GHz of Gamma-ray bright AGN (Active Galactic Nuclei) with KVN (Korean VLBI Network) 21-m radio telescopes. The project started in May, 2011 with an effective monitoring cycle of 4 days, observing four main objects (3C 454.3, BL Lac, 3C 273, and 3C 279). More objects were included in the source list when they had flared in Gamma-ray. In this paper, we report the current status of the project and preliminary results for the monitoring observations.

A study on the hardening characterstics of the TiC layer formed by the reactive deposition technique (반응석출법에 의해 피복된 TiC의 경화거동에 관한 연구)

  • In this study, lattice parameter, binding energy and microstructures of TiC layer according to the addition of Fe, Cr were investigated in the reactive deposition coating. From the results, the lattice parameters of the TiC layers by using ferro-titanium as a precursor were 4.329~4.339A but the lattice parameters of the TiC layers formed by ferro-titanium and ferro-chromium decreased to 4.316~4.330A. The hardness of the former's was HV(100g) 3,000~3,400kg/mm and the hardness of the latter's was HV (100g) 3,800~3,900. But, regardless of Cr and Fe, the binding energy of TiC layers were 454.75 eV for $Ti2p_{3/2}$ and were 281.85 eV for Cls. Meanwhile, the TiC layers were densified by addition of Fe, Cr and internal defects were reduced Therefore. it can be concluded that the remarkable hardness increment was obtained by the improvement of microstructures of TiC rather than the increase of bond strength or Peierls stress.
