Production and Characterization of Monoclonal Antibodies to Bacillus thuringiensis subsp. canadensis

  • 30 monoclonal antibodies (mAbs) were produced against Bacillus thuringiensis subsp. canadensis. Out of the these, 6 mAbs were selected for further studies. SDS-PAGE analyses of sonicated antigens of 10 8. thuringiensis strains showed that they generally had both predominant protein antigens of molecular weights of 45 kilodalton (kd) except for shandogiensis and konkukian, and 37kd except for israelensis, tochigiensis, and shandogiensis, respectively. These results indicate that 4kd and 37kd may be important for demonstrating common antigens except for a few strains of B. thuringiensis. In comparing the result of the westem blot using mAbs with that of using polyclonal antibodies to canadensis, we found that immunoreactive proteins of 99 and 39 kd were identified as common antigens, which might act as antigenic determinants, and might be surface or flagella antigens. Reactivities of mAbs with 41 strains of 8. thuringiensis demonstrated that mAbs of C-1, C-3, C-4, C-S and C-6 except C-2 did not recognize epitopes of thuringiensis, but that all of the mAbs recognized epitopes of galleriae, kurstaki, dakota, tolrJokuensis, silo, toguchini, and leesis. The potential applications of the mAbs we produced would be useful tools for the clarification of taxonomy, investigation of antigenic relationship between B. thuringiensis strains, and localization of specific surface and flagella antigens.

프랑스 A-71고속도로변에 설치된 침전조에서 채취된 간극수와 퇴적물에 함유된 Pb, Zn, Mn 및 Cd의 지화학적 행동 및 오염에 관한 연구 (The Geochemical Behaviour and Environmental Pollution of Pb, Zn, Mn and Cd in Interstitial Waters and Sediments from a Retention Pond along the A-71 Highway, France)

  • Retention ponds have been dug along some of the motorways in France to minimize environmental pollution by keeping pollutants from spreading over the surrounding area. In order to study heavy metal pollution and diagenetic behaviour of sediments, eight core samples were collected from the bottom of a retention pond located along the A-71 motorway in Sologne. The metal concentrations in interstitial waters and extractable metal concentrations in sediment layers using sequential chemical extraction method were determined. The depth distributions of Pb, Zn and Cd concentrations in interstitial water and particulate sediments were studied, and distribution coefficients (KD) were also determined to investigate the environmental mobility of these elements. In addition. the index of geoaccumulation and the Fe-normalized enrichment factor were calculated to differentiate the natural accumulation from the anthropogenic pollution. The vertical concentration profiles of heavy metals in core sediments indicate that surface enrichments (0~2 cm) of Pb, Zn, Cd and organic carbon were always observed at each core sample, due to the early diagenesis. However, the major factor contributing to the accumulation of Cd at the sediment surface is attributed to the dissolution of Cd from polluted roadside soil during periods of rainstorms and its subsequent redeposition on the sediment surface after being carried to the retention pond. A comparison of the KD values indiactes that a decrease in the KD values for Pb and Zn was observed with depth while KD values for Cd increase. According to the KD values. the relative mobility of studied metals was determined as following: Mn>Zn>Cd>Pb, for the upper layer, and Mn>Cd>Zn>Pb, for the lower layers.

유기물함량이 다른 감귤원 토양에서 8 개 농약의 흡착상수 (Adsorption Coefficients of Eight Pesticides for Citrus Orchard Soils with Different Soil Organic Matter)

  • 토양 유기물함량이 다른 감귤원 토양 19개 토양통에서 30개의 토양시료를 채취하여 8개 농약에 대한 흡착특성을 분석하였다. 흡착형태 alachlor와 chlorpyrifos는 C형, diuronn과 diniconazole은 L형, metribuzin, metolachlor 및 linuron은 유기탄소 함량이 증가함에 따라 S, C 및 L형이 복합적으로 나타났으며, chlorothalonil은 유기탄소 함량에 따라 C, S, L 및 H형이 모두 나타났다. metribuzin, metolachlor 및 alachlor의 분배계수 값은 $10\;L\;kg^{-1}$ 이하로서 유기탄소 함량이 증가함에 따라 직선적인 형태로 높아졌다. linuron, diuron 및 chlorothalonil의 분배계수 값은 60, 200 및 $400\;L\;kg^{-1}$ 이하에서 지수 형태로 증가했으며, diniconazole과 chlorpyrifos는 로그 형태로 증가하였다. Freundlich식의 Kf 값도 유기탄소 함량이 많아짐에 따라 분배계수 값과 거의 유사한 형태로 증가되었으나 분배계수 값에 비해 직선성이 컸다.

담배나방 세포질다각체병 바이러스의 동정 및 병원성에 관한 연구 (A Cytoplasmic Polyhedrosis Virus Isolated from the Oriental Tobacco Budworm, Heliothis assulta Guenee (Lepidoptera: Noctuidae))

  • 담배나방 유충에서 분리한 세포질다각체병 바이러스의 형태, 다각체 단백질 및 핵산의 전기영동상과 바이러스의 병원성을 조사하여 본 바이러스를 이용한 담배나방의 생물적 방제 이용성을 검토하고자 본 실험을 수행하였다. 다각체의 형태는 외관상 6각형으로 0.5~3.7 ${\mu}m$ 크기이고 바이러스 입자는 정 20면체로 55nm였다. SDS-PAGE에 의한 다각체 단백질은 단일 롤리?타이드인 24.3 Kd와 5개의 작은 구성분으로 이루어졌다. 바이러스입자는 7개의 폴리?타이드로 구성되어 있으며 분자량은 28.0~133.6 Kd였다. 바이러스 게놈은 10개의 조각으로 된 총 분자량 18.08 Md인 이본쇄 RNA로 각 조각의 분자량 범위는 0.65~2.79 Md이였다. 3령 유충에 대한 담배나방 세포질 다각체병바이러스의 $LC_{50}$$2.895{\times}10^5PIBs/ml$이었으며 $5.0{\times}10^{6}PIBs/ml$의 농도에서 $LT_{50}$에서 16.4일이었다.

한국산 구름버섯의 균주간(菌株間) 생리적(生理的) 특성(特性) 비교(比較) (Comparisons of Physiological Characteristics in Coriolus versicolor Intraspecific Strains)

  • 구름버섯에 있어 3계통(系統)의 한국산(韓國産)과 1종(種)의 일본산(日本産) 균주간(菌株間) 생리유전적(生理遺傳的) 유연관계(類緣關係)를 알아보기 위하여 각(各) 균주(菌株)의 생리적(生理的) 특징(特徵)과 전기영동법(電氣泳動法)에 의한 생화학적(生化學的)인 차이 및 인공재배(人工栽培)에 의한 자실체(子實體)의 형태(形態)를 비교(比較)한 결과(結果)를 요약(要約)하면 다음과 같다. 각(各) 균주(菌株)의 생육조건중(生育條件中) 배지(培地)의 선택성에는 대체로 동일한 경향(傾向)을 보였는데 고체배지상(固體培地上)에서는 PDA, CVT-I 그리고 MES가 양호했으며, 액체배지(液體培地)에서는 927과 CVT-III가 가장 양호하였다. 또 최적(最適) pH는 5.6이었으며 온도(溫度)는 $25{\sim}30^{\circ}C$가 적당한 것으로 나타났다. 전기영동법(電氣泳動法)에 의한 단백질(蛋白質) 및 동위효소(同位酵素)들의 pattern에 있어서는 16001과 KD88001이 대체로 유사하였으나 다른 균주(菌株)들 간(間)에는 각각 다르게 나타났다. 각(各) 균주(菌株)를 인공재배(人工栽培)하여 자실체(子實體)의 형태(形態)를 조사한 결과(結果) 16001과 KD88001은 형태(形態)나 색상(色相)에서 매우 유사함을 보여 동일 계통(系統)으로 나타났으나 16002와 CVT-80은 현저한 차이를 보여 서로 다른 계통(系統)인 것으로 사료된다.

Cardiovascular risk factors of early atherosclerosis in school-aged children after Kawasaki disease

  • Purpose: The aim of this study was to determine whether school-aged children with Kawasaki disease (KD) have an increased risk for early atherosclerosis. Methods: The study included 98 children. The children were divided into the following groups: group A (n=19), KD with coronary arterial lesions that persisted or regressed; group B (n=49), KD without coronary arterial lesions; and group C (n=30), healthy children. Anthropometric variables and the levels of biochemical markers, including total cholesterol, high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), apolipoprotein A, apolipoprotein B, homocysteine, highsensitivity C-reactive protein (hs-CRP), and brachial artery stiffness using pulse wave velocity were compared among the three groups. Results: There were no significant differences in blood pressure and body index among the three groups. Additionally, there was no sex-specific difference. Moreover, the levels of triglyceride, HDL-C, apolipoprotein A, and hs-CRP did not differ among the three groups. However, the levels of total cholesterol (P =0.018), LDL-C (P =0.0003), and apolipoprotein B (P =0.029) were significantly higher in group A than in group C. Further, the level of homocysteine and the aortic pulse wave velocity were significantly higher in groups A and B than in group C (P=0.0001). Conclusion: School-aged children after KD have high lipid profiles and arterial stiffness indicating an increased risk for early atherosclerosis.

Surgical Outcomes of Kommerell Diverticulum

  • Background: We aimed to assess the clinical outcomes of patients who underwent surgical repair of Kommerell diverticulum (KD) with individualized surgical methods. Methods: A retrospective analysis was performed of adult patients (aged ≥17 years) who underwent surgery to treat KD between June 2008 and October 2019. Results: Nine patients (median age, 45 years; range, 19-67 years; 7 men) underwent surgical repair. The indications for surgical therapy were acute aortic dissection in 2 patients, the presence of compressive symptoms due to dilated KD in 4 patients, and aneurysm growth in 3 patients. Various surgical techniques were used: (1) resection of the diverticulum stump and revascularization of the aberrant subclavian artery (n=3), (2) one-stage total-arch replacement including the diverticulum segment (n=3), and (3) hybrid repair (n=3). Early mortality occurred in 1 case of hybrid repair. Transient paraparesis occurred in a patient who underwent total arch repair as part of complicated acute aortic dissection. During follow-up (median duration, 30 months; range, 7-130 months), no late death or associated aortic complications were documented. All survivors were free from symptoms and had no abnormal findings on follow-up computed tomography. Conclusion: With a customized surgical approach and appropriate consideration of patient-specific anatomy and associated comorbidities, KD can be repaired with favorable outcomes.

Clinical outcome of patients with refractory Kawasaki disease based on treatment modalities

  • Purpose: Although a significant number of reports on new therapeutic options for refractory Kawasaki disease (KD) such as steroid, infliximab, or repeated intravenous immunoglobulin (IVIG) are available, their effectiveness in reducing the prevalence of coronary artery lesions (CAL) remains controversial. This study aimed to define the clinical characteristics of patients with refractory KD and to assess the effects of adjuvant therapy on patient outcomes. Methods: We performed a retrospective study of 38 refractory KD patients from January 2012 to March 2015. We divided these patients into 2 groups: group 1 received more than 3 IVIG administration+steroid therapy, (n=7, 18.4%), and group 2 patients were unresponsive to initial IVIG and required steroid therapy or second IVIG (n=31, 81.6%). We compared the clinical manifestations, laboratory results, and echocardiographic findings between the groups and examined the clinical utility of additional therapies in both groups. Results: A significant difference was found in the total duration of fever between the groups ($13.0{\pm}4.04days$ in group 1 vs. $8.87{\pm}2.30days$ in group 2; P=0.035). At the end of the follow-up, all cases in group 1 showed suppressed CAL. In group 2, coronary artery aneurysm occurred in 2 patients (6.4%). All the patients treated with intravenous corticosteroids without additional IVIG developed CALs including coronary artery aneurysms. Conclusion: No statistical difference was found in the development of CAL between the groups. Prospective, randomized, clinical studies are needed to elucidate the effects of adjunctive therapy in refractory KD patients.

가와사끼 병에 합병된 간기능 이상소견에 대한 임상적 및 병리조직학적 소견 (Clinical and Histopathologic Findings on the Abnormal Liver Function Complicated with Kawasaki Disease)

  • Purpose: The aim of this research is to examine the abnormal liver function complicated with Kawasaki disease (KD) and to analyze its clinical characteristics and complications. Methods: Ninty eight cases were diagnosed as having KD, among which thirty four cases had abnormal alanine aminotransferase (ALT) (>30 IU/L). These abnormal ALT cases were evaluated in terms of age and sex distribution, major symptoms, complications, laboratory and histopathologic findings. Results: Male to female ratio was 2.4:1 and most patients (91.2%) were under 5 years of age. Cervical lymphadenopathy was observed in 3 cases (8.8%); and recurred cases were two (5.8%). Average fever duration was 8 days; average length of hospitalization was 9 days; and average recovery period was 13 days. Immediately after admission, positive CRP was observed in 31 cases (91.2%), leukocytosis (> $10,000/mm^3$) in 26 cases (76.0%), thrombocytosis (> $450,000/mm^3$) in 7 cases (20.6%), and anemia (<10 gm/dl) in 7 cases (20.6%), respectively. GB hydrops or cholecystitis were noted in 3 cases (8.8%), abnormal ECG finding in 1 case (2.9%), coronary dilatation or aneurysm in 2 cases (5.9%). Liver biopsy was done in four cases and revealed mild infiltration of lymphocytes on the portal area and mild bile duct proliferations. Conclusion: The abnormal liver function was noted in 34.7% of KD patients, and subsided all within one month. But the liver function test should be checked closely in patients of the abnormal liver function test complicated with KD despite of its good prognosis.

가와사키병에서의 TNF-alpha 유전자의 다형성 및 관상동맥 합병증과의 연관성 (Polymorphisms of tumor necrosis factor-alpha promotor gene in Kawasaki disease and relation to the risk of coronary artery lesion)

  • 목 적 : 저자들은 가와사키병 환아에서 TNF-alpha 유전자의 다형성을 조사함으로써 가와사키병과 유전자 다형성의 관련 여부를 알아보고, 또한 관상동맥 병변의 발생과 연관이 있는지를 살펴보려 하였다. 방 법 : 2003년 1월부터 2007년 1월까지 가와사키병 환아 51명과 대조군 48명을 대상으로 TNF-alpha 촉진자의 단일 유전자 다형성을 살펴보았으며, 가와사키병 환아 중 관상동맥 병변이 있는 24명(관상동맥병변군)과 관상동맥 이상이 없었던 27명(정상관상동맥군)에서의 유전자 다형성을 또한 비교하였다. 말초 혈액에서 DNA를 추출하여 TNF-alpha 유전자 -308 부위의 촉진자에 위치한 2개의 단일 염기 서열 G/A에 대한 대립 유전자의 다형성을 Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) 방법으로 분석하였다. 결 과 : 가와사키병 환아군에서의 -308번 TNF-alpha 유전자의 다형성은 51명 중 9명으로 17.6%였고, 정상 대조군에서는 48명 중 3명으로 6.8%로 가와사키병 환아군에서 높았으나 통계학적으로 유의성은 없었다. 가와사키병 환아 중 관상동맥병병군 24명 중 3명인 12.5%에서 유전자 다형성이 있었고 정상관상동맥군은 27명 중 6명인 22.2%로 정상관상동맥군에서 더 높은 빈도 이었으나 통계학적으로 유의하지 않았다. 결 론 : 본 연구에서는 가와사키병 환아의 TNF-alpha의 다형성과 가와사키병의 발병과의 관련성이 통계학적으로 유의한 수준은 아니지만 가와사키병 환아에서 G/A 빈도수가 17.6%으로 대조군에서 6.8% 보다 다소 높게 나타난 결과를 얻었고 앞으로 많은 수의 환아를 대상으로 한다면 유의한 차이가 있을 것으로 생각되므로 향후 대규모의 지속적인 연구가 필요할 것이다.
