흰쥐 측뇌실의 Metallothionein 유사단백질과 Stress Protein의 유도 (Induction of Metallothionein-like Protein in the Rat Brain by Intracerebroventricular Cadmium Treatment)

  • 쥐 뇌에·처 카드뮴에 대한 metallothionein-like protein(MTLP)의 유도 생성 능력을 알아보기 위하여 Stereotaxic Bpparatus를 이용하여 측뇌실에 카드뮴을 주사하여 다음과 같은 결과를 얻었다. 대조군과 생리식염수 처리군에서는 MTLP의 양적 변화가 없었고. 카드를 처리군은 대조군과 식염수 처리군에 비하여 MTLP가 2배 이상 유도되었다. MTLP의 분자량은 6.000-6,500 Oa 정도 였으며. 흡광도가 254 nm에서 높게 나타나고. 280 nm에서 낮게 나타나는 것으로 보아 thiol 함량이 높고 방향족 아미노산이 적은 단백질임을 알 수 있었다 또한 카드뮴 처리군에서는 MTLP 이외의 여러 종류의 protein-30. 64, 68, 80, 108 kDa-들이 유도되었다 이와 같은 결과로 카드뮴은 흰쥐 뇌에서도 MTLP의 유도 능력이 있음을 알 수 있었다.

High Expression of Lung Resistance Protein mRNA at Diagnosis Predicts Poor Early Response to Induction Chemotherapy in Childhood Acute Lymphoblastic Leukemia

  • Background: Treatment failure in leukemia is due to either pharmacokinetic resistance or cell resistance to drugs. Materials and Methods: Gene expression of multiple drug resistance protein (MDR-1), multidrug resistance-related protein (MRP) and low resistance protein (LRP) was assessed in 45 pediatric ALL cases and 7 healthy controls by real time PCR. The expression was scored as negative, weak, moderate and strong. Results: The male female ratio of cases was 2.75:1 and the mean age was 5.2 years. Some 26/45 (58%) were in standard risk, 17/45(38%) intermediate and 2/45 (4%) in high risk categorie, 42/45 (93%) being B-ALL and recurrent translocations being noted in 5/45 (11.0%). Rapid early response (RER) at day 14 was seen in 37/45 (82.3%) and slow early response (SER) in 8/45 (17.7%) cases. Positive expression of MDR-1, LRP and MRP was noted in 14/45 (31%), 15/45 (33%) and 27/45 (60%) cases and strong expression in 3/14 (21%), 11/27 (40.7%) and 8/15 (53.3%) cases respectively. Dual or more gene positivity was noted in 17/45 (38%) cases. 46.5 % (7/15) of LRP positive cases at day 14 were in RER as compared to 100% (30/30) of LRP negative cases (p<0.05). All 8 (100%) LRP positive cases in SER had strong LRP expression (p=<0.05). Moreover, only 53.3% of LRP positive cases were in haematological remission at day 30 as compared to 100% of LRP negative cases (p=<0.05). Conclusions: Our study indicated that increased LRP expression at diagnosis in pediatric ALL predicts poor response to early treatment and hence can be used as a prognostic marker. However, larger prospective studies with longer follow up are needed, to understand the clinical relevance of drug resistance proteins.

Development of Predictive Growth Models for Staphylococcus aureus and Bacillus cereus on Various Food Matrices Consisting of Ready-to-Eat (RTE) Foods

  • We developed predictive growth models for Staphylococcus aureus and Bacillus cereus on various food matrices consisting primarily of ready-to-eat (RTE) foods. A cocktail of three S. aureus strains, producing enterotoxins A, C, and D, or a B. cereus strain, were inoculated on sliced bread, cooked rice, boiled Chinese noodles, boiled bean sprouts, tofu, baked fish, smoked chicken, and baked hamburger patties at an initial concentration of 3 log CFU/g and stored at 8, 10, 13, 17, 24, and $30^{\circ}C$. Growth kinetic parameters were determined by the Gompertz equation. The square-root and Davey models were used to determine specific growth rate and lag time values, respectively, as a function of temperature. Model performance was evaluated based on bias and accuracy factors. S. aureus and B. cereus growth were most delayed on sliced bread. Overall, S. aureus growth was significantly (p<0.05) more rapid on animal protein foods than carbohydrate-based foods and vegetable protein foods. The fastest growth of S. aureus was observed on smoked chicken. B. cereus growth was not observed at 8 and $10^{\circ}C$. B. cereus growth was significantly (p<0.05) more rapid on vegetable protein foods than on carbohydrate-based foods. The secondary models developed in this study showed suitable performance for predicting the growth of S. aureus and B. cereus on various food matrices consisting of RTE foods.

대두올리고당 생산을 위한 한외여과 대두침출액의 이온교환 (Ion Exchange of Ultrafiltrated Soybean Cooking Water for the Production of Soy-oligosaccharides)

  • 대두침출액으로부터 대두올리고당을 생산하기 위하여 한외여과한 대두침출액의 정제를 목적으로 이온교환공정을 검토하였다. 차단분자량 20,000인 막을 사용하여 한외여과한 대두침출액을 이온교환처리할 경우 회분은 양이온교환수지에 의하여 대부분 제거되었으며 단백질 및 색소제거에는 음이온교환수지의 효과가 높았다. 회분 및 단백질 제거효과가 높은 수지는 각각 양이온교환수지 SK1B 및 음이온교환수지 WA30이었다. 이온교환시 ERR이 낮을 경우 단백질과 회분의 제거는 효과적이었으나 상당량의 올리고당이 수지에 흡착되어 소실되었다. 차단분자량 20,000인 막을 사용하여 한외여과한 대두침출액을 차단분자량 5,000인 막으로 2차 한외여과하여 단백질을 추가로 제거하고 혼합수지를 사용하여 ERR 5.0으로 이온교환처리한 결과, 단백질, 회분, 색소가 거의 전량 제거되었고 올리고당의 회수율도 높아 적정 이온 교환조건으로 결정되었다.

한국 여성의 Lactadherin 유전자 Cloning과 발현 연구 (Cloning and Expression of Lactadherin Gene from Korean Women)

  • Lactadherin은 모유의 유지방구막의 당단백질의 하나로 mucin, butyrophilin과 결합된 복합체이다. 특히 모유중의 mucin과 lactadherin은 출생 직후 면역력이 약한 유아를 병균의 침입으로부터 효율적으로 방어하여 초기 유아의 생존과 성장 및 발달에 매우 중요한 역할을 수행한다. Lactadherin은 유아 설사의 원인이 되는 rotavirus의 번식과 성장을 억제한다. 아울러 이 단백질은 새로운 혈관의 형성을 촉진하는 주요한 단백질로 알려져 있으며, 이 단백질의 결핍이 치매의 발생과 관련되는 것으로 보고되고 있다. 본 연구는 이처럼 중요성이 강조되는 lactadherin에 대한 생화학적 및 생리학적인 연구를 하기 위한 기초연구를 진행하였다. 한국 여성의 유선조직에서 mRNA를 분리하였고, 1.2 kb lactadherin cDNA 유전자를 cloning하여 염기서열과 아미노산 배열을 결정하였다. 이 cDNA를 pET vector에 삽입하여 E. coli에서 43 kD 단백질을 발현시켰으며 Western blot으로 확인하였으며, 이 단백질을 정제하여 토끼에서 항체를 생산하여, 한국 여성의 모유에서 발현되는 70, 55, 46, 30 kD의 band를 확인하였다. 아울러 백인 여성의 lactadherin 유전자와 한국 여성의 정상 및 유방암 조직의 유전자 비교에서 다양한 SNP가 관찰되었고 변이의 다형성이 관찰되었다.

Isolation of Two cDNAs Encoding a Putative Peptidohlycan Recognition Protein Gene from the Domestic Silkworm, Bombyx mori

  • Peptidohlycan recognition protein (PGRP) is one of the pattern recognition proteins in innate immunity of insect. We isolated differentially expressed two cDNAa, BTL-LPI and BTL-LP2, in the fat body of Bombyx mori larvae injected with bacteria by subtractive hybridization method. These two clones showed amino acid sequence divergence of 30.4%. In the comparison with other insect PGRP genes, BTL-LP2 showed 48.8% and 45.2% of sequence homology to the known PGRP genes of Bombyx mori and Tricoplusia ni, respectively, and BTL-LP2 was 31.8% and 30.9% , respectively. Phylogenetic analysis showed relatively close relationship of the BTL-LP2 to the known insect PGRP, unlike BTL-LPI, which was equidistant both to insect and mammals, suggesting a divergent relationships of the two newly cloned B. mori PGRP genes. Northern blot analyses confirmed an induction of the expression of BTL-LP2 by the bacterial infection in the Int body of B. mori, suggesting the involvement of the gene in the insect immunity.

Effect of [6]-Gingerol, a Pungent Ingredient of Ginger, on Osteoblast Response to Extracellular Reducing Sugar

  • Diabetes is marked by high glucose levels and is associated with decreased bone mass and increased fracture rates. To determine if [6]-gingerol could influence osteoblast dysfunction induced by 2-deoxy-D-ribose (dRib), osteoblastic MC3T3-E1 cells was treated with dRib and [6]-gingerol and markers of osteoblast function and oxidized protein were examined. [6]-Gingerol ($10^{-7}\;M$) significantly increased the growth of MC3T3-E1 cells in the presence of 30 mM dRib (p<0.05). [6]-Gingerol ($10^{-7}\;M$) caused a significant elevation of alkaline phosphatase (ALP) activity, collagen content, and osteocalcin secretion in the cells. We then examined the effect of [6]-gingerol on the production of osteoprotegerin and protein carbonyl in osteoblasts. Treatment with [6]-gingerol ($10^{-9}$ and $10^{-7}\;M$) increased osteoprotegerin secretion in osteoblastic cells. Moreover, [6]-gingerol ($10^{-9}$ and $10^{-7}\;M$) decreased protein carbonyl contents of osteoblastic MC3T3-E1 cells in the presence of 30 mM dRib. Taken together, these results demonstrate that [6]-gingerol inhibits dRib-induced damage and may be useful in the treatment of diabetes related bone diseases.

Recombinant Expression, Isotope Labeling, and Purification of Cold shock Protein from Colwellia psychrerythraea for NMR Study

  • Cold shock proteins (Csps) are a subgroup of the cold-induced proteins on reduction of the growth temperature below the physiological temperature. They preferentially bind to single-stranded nucleic acids to translational regulation via RNA chaperoning. Csp plays important role in cold adaptations for the psychrophilic microorganism. Recently, Cold shock protein from psychrophilic bacteria, Colwellia psychrerythraea (CpCsp) has been identified. Three dimensional structures of a number of Csps from various microorganisms have been solved by NMR spectroscopy or X-ray crystallography, but structures of psychrophilic Csps were not studied yet. Therefore, cloning and purification protocols for further structural study of psychrophilic Csp have been optimized in this study. CpCsp was expressed in E. coli with pET-11a vector system and purified by ion exchange, size exclusion, and reverse phase chromatography. Expression and purification of CpCsp in M9 minimal media was carried out and $^{15}N$-labeled proteins with high purity over 90% was obtained. Further study will be carried out to investigate the tertiary structure and dynamics of CpCsp.

Immunological Assays of Freezing Tolerance in Barley using Antifreeze Proteins Antisera

  • In order to measure an antifreezing tolerance, antifreeze proteins accumulated upon cold acclimation in apoplast were analyzed. As Dongborilho were cold-acclimated for 3 to 74 days there was an abrupt increase in apoplastic proteins up to 30 days and then decrease to the similar levels. Among the known antifreeze proteins, CLP produced in E. coli. and TLP purified from apoplast were used to generate antisera that allow to measure and localize the proteins in leaves of barley. The CLP of 27.7 kDa and TLPs of 6, 26, 27, 30, and 31 kDa were increased in their amounts in apoplast as cold treatment being longer. There was a correlation among the amounts of those proteins accumulated in apoplast and freezing tolerance as shown in field and ion leakage tests for five cultivars. The deposit of CLP was localized in the marginal area and the area adjacent to leaf vescular bundle cells in an increasing manner according to duration of cold acclimation but no variation was observed in terms of it's distribution. Based on the close correlation between levels of antifreeze proteins and degrees of freezing tolerance, the immunological methods was to develop to estimate a freezing tolerance in barley.
