Synergistic effects of dietary $\beta$-1,3 glucan and feed stimulants in juvenile olive flounder (Paralichthys olivaceus)

  • The present study was conducted to investigate the effects of dietary supplementation of $\beta$-1,3 glucan and feed stimulants(BAISM) as a feed additive for juvenile olive flounder (Paralichthys olivaceus). Eight experimental diets supplemented with $\beta$-1,3 glucan and feed stimulants at 0%, $\beta$-1,3 glucan 0.05% + Baism 0.45%, $\beta$-1,3 glucan 0.05% + Baism 0.95%, $\beta$-1,3 glucan 0.10% + Baism 0.90%, $\beta$-1,3 glucan 0.10% + Baism 1.90%, $\beta$-1,3 glucan 0.15% + Baism 1.35%, $\beta$-1,3 glucan 0.15% + Baism 2.85% and $\beta$-1,3 glucan 0.30% + Baism 2.70% of diets as a dry-matter(DM) basis were prepared. Three replicate groups of fish averaging 9.2 $\pm$ 0.1g (Mean $\pm$ SD) were randomly distributed in each aquarium as a group of 15 fish and fed one of eight experimental diets for seven weeks. After the feeding trial, $\beta$-1,3 glucan 0.10% + Baism 0.90%, $\beta$-1,3 glucan 0.10% + Baism 1.90% diets had a higher weight gain (WG), feed efficiency(FE), specific growth rate(SGR) and protein efficiency ratio(PER) than did fish fed 0%, $\beta$-1,3 glucan 0.05% + Baism 0.45%, $\beta$-1,3 glucan 0.05% + Baism 0.95%, $\beta$-1,3 glucan 0.15% + Baism 2.85% and $\beta$-1,3 glucan 0.30% + Baism 2.70% (P<0.05). however, there was no significant difference among fish fed $\beta$-1,3 glucan 0.05% + Baism 0.45%, $\beta$-1,3 glucan 0.05% + Baism 0.95%, $\beta$-1,3 glucan 0.15% + Baism 2.85% and $\beta$-1,3 glucan 0.30% + Baism 2.70%(P>0.05). and $\beta$-1,3 glucan 0.10% + Baism 0.90% diets had a higher peak value of CL(Chemiluminescence) and lysozyme activity, than did fish fed the other diets (P<0.05). These results indicated that dietary sipplementation of $\beta$-1, 3 glucan and Baism affected growth, feed efficiency, specific growth rate, protein efficiency ratio, Peak value of CL and Lysozyme activity, and the optimum dietary supplementation level of $\beta$-1, 3 glucan and Baism as a feed additive could be approximately $\beta$-1, 3 glucan 0.10% + Baism 0.90% of diet in juvenile olive flounder (Paralichthys olivaceus).

Effect of Nitrogen Source on the Change of UDP-glucose and ${\beta}$-1,3-glucan Concentration

  • UDP-glucose는 ${\beta}$-1,3-glucan 합성의 중요한 전구체로 이를 측정함으로써 세포 내의 glucan synthesis 대사의 활성도를 추정할 수 있는 중요한 지표가 됨을 본 연구 결과를 통하여 알 수 있었다. UDP-glucose는 세포 성장기에 다량 생산되다가 ${\beta}$-1,3-glucan 합성하는 시기에 일정한 농도가 되며 ${\beta}$-1,3-Glucan 합성 메카니즘에서 glucose를 운반하는 중요한 역할을 하는 것으로 나타났다. 2단 연속 발효조를 이용하여 세포 성장 발효조와 ${\beta}$-1,3-glucan 생산 발효조에서 UDP-glucose 변화를 관찰하여 ${\beta}$-1,3-glucan 생산시 농도가 높음을 관찰할 수 있었다. ${\beta}$-1,3-Glucan 생산 발효조의 pH를 5.5로 조절함으로써 UDP-glucose의 농도를 증가시킬 수 있을 뿐만 아니라 ${\beta}$-1,3-glucan의 생산 속도를 최적화할 수 있었다.

  • ${\beta}$-Glucan은 효모나 버섯의 세포벽이나 곡물의 섬유에 들어있는 천연 화합물이며, 면역 시스템을 활성화 한다고 알려져 있다. Aureobasidium pullulans 수용성의 ${\beta}$-1,3/1,6-glucan을 생성한다. 본 연구에서는 A. pullulans 로부터 생성된 ${\beta}$-1,3/1,6-glucan의 면역활성에 관한 효과를 조사하기 위하여, 면역활성에 관여하는 NK 세로의 활성과 대식세포에 미치는 영향을 관찰하였다. NK 세포에 의한 Yac-1 세포의 사멸율은 ${\beta}$-1,3/1,6-glucan을 처리한 군은 처리하지 않은 대조군에 비하여 63-39% 증가하는 것으로 나타났고, 대식세포의 탐식작용은 zymosan을 처리한 군보다 15-21% 증가함을 보였다. LP-BM5 바이러스 복제 억제능을 확인하기 위하여 SC-1/LP-BM5 세포주에 ${\beta}$-1,3/1,6-glucan을 처리한 결과 $200{\mu}g/mL$ 처리에서 LP-BM5 바이러스 복제능이 최대 74% 유의적으로 감소하였다. 이 결과는 ${\beta}$-1,3/1,6-glucan이 면역활성에 관여하는 NK 세포와 대식세포를 활성화하여 면역체계가 민감하게 반응할 수 있도록 만들고, LP-BM5 바이러스 복제를 억제하여 면역력을 증진할 수 있다는 것을 보여준다.

Varietal and Annual Variations of β-Glucan Contents in Korean Barley (Hordeum vulgare L.) and Oat (Avena sativa L.) Cultivars

  • Varietal and annual variations in the contents of ${\beta}$-glucan fractions per weight grain samples were examined in sixteen covered and eighteen naked barley and five oat cultivars developed in Korea. Also, the effect of pearling on ${\beta}$-glucan content was investigated. Average contents of total, soluble and insoluble ${\beta}$-glucan fractions were 5.25, 3.72, and 1.53%, respectively, in covered barley, and 5.86, 3.51, and 2.35%, respectively, in naked barley. Soluble ${\beta}$-glucan content was higher in covered barley, though total ${\beta}$-glucan content higher in naked barley. The total and insoluble ${\beta}$-glucan contents were higher in pearled grains. Total ${\beta}$-glucan content was higher in waxy barley than in non-waxy barley. Duwonchapssalbori, a two-rowed and waxy naked barley cultivar, was highest in total, soluble and insoluble ${\beta}$-glucan contents. Highly significant positive correlations were observed between total ${\beta}$-glucan and soluble ${\beta}$-glucan contents both in covered and naked barley. There were significant annual variations in total ${\beta}$-glucan content in barley. Average contents of total, soluble and insoluble ${\beta}$-glucans of oat cultivars were 4.33, 3.44, and 0.89%, respectively. Contents of all fractions of ${\beta}$-glucans were higher in barley than in oat. These results would be useful for the breeding of high ${\beta}$-glucan variety and also for the use barley and oat as valueadded food ingredients.

  • A soluble dietary fiber, $\beta$-glucan, contained in oat and barley has nutritional benefits such as hypocholesterolemic effects and influences blood glucose regulation. The contents of $\beta$-glucan in both cereals range from 3 to 7% with the exception of a certain barley genotype which contains up to 16% $\beta$-glucan. $\beta$-Glucan is distributed mainly in the cell walls of endosperm and the distal (bran) portion of kernel. Various procedures have been developed for increasing the extraction yield of $\beta$-glucan. Oat gum prepared with weak alkali extractionand alcohol proecipitation following protein removal usually contains 80% $\beta$-glucan.The most commonly used method for $\beta$-glucan quantitiation is an enzymatic procedure combining lichenase plus $\beta$-glucosidase followed by measuring the amount of glucos released by glucose oxidase-peroxidase treatment. The increase in foam-and emulsion-stabilizing capacity of $\beta$-glucan is due to the increase in viscosity of the aqueous phase. Therefore, $\beta$-glucan shows great potentials as a thickener and a stabilizer.

색소에 접합된 $\beta$-glucan을 이용한 $\beta$-glucan 분해효소 생산 균주의 분리 및 동정 (Isolation and identification of $\beta$-glucan degrading enzyme producing bacterium using coloured $\beta$-glucan)

  • A bacterium K-4-3, producing $\beta$-glucan hydrolyzing enzyme, was isolated from soil and identified to be Bacillus subtilis by its morpholohical and physiological characteristics. $\beta$-glucan was coloured using cibacron blue 3G-A and cross linded by the addition of 1, 4-butanedioldiglycidyl ether. This substrate was used for the isolation of $\beta$-glucanase producing microorganism. The $\beta$-glucan hydrolyzing enzyme actibity from isolated K-4-3 strain was also measured using the modified substrate. Bacillus subtilis K-4-3 produced the highest extracellular $\beta$-glucan hydrolyzing activity in the basal medium containing $\beta$-glucan as a carbon source, peptone and tryptone as a nitrogen source, and magnesium sulfate as an inorganic salt. The optimum temperature and initial pH for $\beta$-glucanase production by Bacillus subtilis K-4-3 were $37^{\circ}C$ and pH6. The highest enzyme activity was obtained at the culture age of 54 hrs with rotary shaking at $37^{\circ}C$. The crude enzyme showed the highest activity at pH 7.5-8.0 and $65^{\circ}C$.

버섯균사체 배양액에 함유된 Glucan의 정량 및 항생제 내성억제 효과 (Determination of Glucan from Mycellium and Its Activity against Methicillin-Resistant Staphylococcus Aureus)

  • 항암, 항콜레스테롤, 항산화 및 면역증가 효과와 피부재생 효과 등 여러 가지 생리활성이 밝혀지고 있는 ${\beta}-1,3/1,6-glucan$은 크게 식물성 유래, 효모 및 곰팡이, 버섯유래의 것으로 등으로 분류할 수 있다. 본 연구에서는 이처럼 다양한 효능을 나타내는 버섯균사체로부터 ${\beta}-glucan$를 다량함유하고 보다 쉽게, 대량으로 생산하기 위한 일환으로 연구를 하였다. 이때 고분자 중합체인 ${\beta}-glucan$을 분리하여 핵자기 공명분석기로 분석한 결과 ${\beta}-1,3-$${\beta}-1,6-$결합이 혼재되어 있는 ${\beta}-1,3/{\beta}1,6-glucan$의 전형적인 구조임을 확인 할 수 있었다. 또한 ${\beta}-1,3/{\beta}1,6-glucan$의 함량을 gel permeation chromatography으로 분석한 결과 $766{\pm}0.19$ mg/g 함유되었음을 확인 할 수 있었다. 또한 항생재 내성균주인 MRSA의 항균효과를 측정한 결과 MIC가 18mg/mL임을 확인 할 수 있었다. 이 연구결과는 통하여 효과적인 생산방법과 효능을 접목시켜 보다 저렴한 생산비로 대량 생산할 수 있는 방법을 개발할 수 있을 것으로 사료된다.

보리로부터 분리한 ${\beta}-Glucan$의 이화학적 특성 (Physicochemical Characteristics of ${\beta}-Glucan$ Isolated from Barley)

  • 찰성(waxy) 및 메성(non-waxy) 보리로 부터 ${\beta}-glucan$을 분리, 정제하였고 정제한 ${\beta}-glucan$의 표면구조, DSC특성, 고유점도를 비교하였으며 ${\beta}-glucan$의 첨가가 보리전분의 호화특성에 미치는 영향을 조사하였다. 보리의 총 ${\beta}-glucan$ 함량은 찰성보리가 6.5%, 메성보리가 5.3%였다. 알칼리 추출 후 1차 정제한 crude ${\beta}-glucan$${\beta}-glucan$ 함량은 찰성보리가 62.9%, 메성보리가 59.2%였으며, 수율은 찰성보리가 5.54%로 메성보리의 수율 3.34%보다 높았다. 2차 정제한 ${\beta}-glucan$의 수율도 찰성이 4.46%로 메성의 2.59%보다 높았다. SEM으로 관찰한 정제 ${\beta}-glucan$의 표면구조는 섬유상의 fibril들이 서로 얽힌 망상의 배열을 이루고 있었다. DSC로 측정한 융점은 찰성보리 ${\beta}-glucan$$184.6^{\circ}C$로 메성의 $180.3^{\circ}C$ 보다 높았고 겔화에 의한 상전이는 $68^{\circ}C$$84^{\circ}C$ 부근에서 2개의 흡열피크로 나타났으며 찰성 ${\beta}-glucan$의 엔탈피 값이 작았다 정제한 ${\beta}-glucan$의 고유점도는 메성보리보다 찰성보리 ${\beta}-glucan$이 더 컸다. 보리전분에 ${\beta}-glucan$을 첨가하여 호화시켰을 때 상승작용으로 비스코그램의 점도를 증가시켰으며 그 효과는 메성전분에서 현저하였다.

  • Assay conditions for screening of $\beta$-1,3-glucan synthase inhibitor were evaluated. Cells in the beginning of mid-log phase showed the highest activity of the $\beta$-1,3-glucan synthase. Cells permeabilized with 1% digitonin treatment could be used as a good crude enzyme source for convenient screening of the $\beta$-1,3-glucan synthase inhibitors. Calcofluor white (0.125% in final) and papulacandin B (25 $\mu$g/ml) inhibit 90% and more than 50% of the $\beta$-1,3-glucan synthase activity, respectively. Cells grown at 37$\circ$C showed higher enzyme activity than those of 25$\circ$C. Catalytic factor of the $\beta$-1,3-glucan synthase was solubilized from particulated membrane preparations, holoenzyme, by extracting with 0.00938% CHAPS.

영지 (G. lucidum)의 액체배양에 의한 β-1,3-Glucan 생산에 미치는 서로 다른 pH Process의 영향 (Effect of Different pH Processes on Branched β-1,3-Glucan Production from Submerged Culture of Ganoderma lucidum)

  • A submerged cultivation of Ganoderma lucidum was carried out in an air-lift fermenter system, and the effects of different pH processes on extracellular branched ${\beta}$-1,3-glucan(EPS) production and mycelial growth(MDW) were investigated. The controlled pH process improved the production of branched ${\beta}$-1,3-glucan and biomass in comparison to the uncontrolled pH process. However, the maximum production of branched ${\beta}$-1,3-glucan were obtained by the bi-staged pH process. From these results, we confirmed that the bi-staged pH process was the most effective for improving the production of branched ${\beta}$-1,3-glucan from submerged culture of G. lucidum.
