Structural Insights into the Interaction of Terpenoids with Streptomyces avermitilis CYP107P2

  • Streptomyces avermitilis genome includes 33 genes encoding monooxygenation-catalyzing cytochrome P450 enzymes. We investigated the structure of CYP107P2 and its interactions with terpenoid compounds. The recombinant CYP107P2 protein was expressed in Escherichia coli and the purified enzyme exhibited a typical P450 spectrum upon CO-binding in its reduced state. Type-I substrate-binding spectral titrations were observed with various terpenoid compounds, including α-pinene, β-pinene, α-terpinyl acetate, and (+)-3-carene. The calculated binding affinities (Kd) ranged from 15.9 to 50.8 µM. The X-ray crystal structure of CYP107P2 was determined at 1.99 Å resolution, with a well-conserved overall P450 folding conformation. The terpenoid compound docking models illustrated that the structural interaction between monoterpenes and CYP107P2, with the distance between heme and terpenes ranging from 3.4 to 5.4 Å, indicates potential substrate binding for P450 enzyme. This study suggests that CYP107P2 is a Streptomyces P450 enzyme capable of catalyzing terpenes as substrates, signifying noteworthy advancements in comprehending a novel P450 enzyme's involvement in terpene reactions.

Qualitative, Quantitative Analysis and Chiral Characterization of the Essential Oils of Juniperus phoenicea L. and Juniperus oxycedrus L.

  • Isolation of oils from leaves of Juniperus phoenicea and Juniperus oxycedrus was obtained by steam distillation extraction method. The compositions of essential oils (EOs) were studied by means of GC-MS and GC-FID, using the internal standard method and relative response factors. Around ninety eight compounds were determined in total, representing 98.25 g/100 g of EO of J. phoenicea and 98.48 g/100 g of EO of J. oxycedrus, respectively. The volatile leaf oils were dominated by the terpenic hydrocarbon fractions (79.87 g/100 g) and (61.27 g/100 g) characterized by high contents of α-pinene (64.6 g/100 g) and (54.0 g/100 g) in J. phoenicea and J. oxycedrus, respectively, as the main component. Also, the enantiomeric distribution of α-pinene, sabinene, camphene, δ-3-carene, β-pinene, limonene, linalool, terpinen-4-ol, bornyl acetate, and borneol in both oils is presented for the first time.

Essential Oil Analysis of Illicium anistum L. Extracts

  • Fresh japanese anise (Illicium anisatum L.) tree leaves were collected and ground after drying. The essential oils of the leaves were analyzed by gas chromatography-mass spectrometry (GC-MS) using headspace (HS) and solid phase-microextra (SPME) methods. Volatile components of the leaves were identified 21 and 65 components in HS and SPME, respectively. The main components of the essential oils obtained by HS method were eucalyptol (36.7%), (+)-sabinene (15.61%), ${\delta}$-3-carene (6.87%), ${\alpha}$-pinene (6.07%), ${\gamma}$-terpinen (5.72%), ${\alpha}$-limonene (5.26%), ${\beta}$-myrcene (4.13%), ${\alpha}$-terpinene (4.04%) and ${\beta}$-pinene (3.73%). The other components were less than 3.5%. SPME method also showed that eucalyptol (17.88%) was main. The other were 5-allyl-1-methoxy-2 (13.29%), caryophyllene (6.09%), (+)-sabinene (5.60%), ${\alpha}$-ocimene (4.89%) and ${\beta}$-myrcene (3.73%), and the rest were less amounts than 3.5%. This work indicated that many more volatile components were isolated, comparing to the previous literature data and that SPME method was much more effective than HS method in the analysis of the volatile components.

흡착튜브 - 열탈착 정량분석 기법에 기반한 과일시료로부터 자연적 휘발성유기화합물의 배출특성 연구 (Biogenic Volatile Organic Compounds (BVOC) Emissions from Fruit Samples Based on Sorbent Tube Sampling and Thermal Desorption (ST-TD) Analysis)

  • In this study, a combination of sorbent tube (ST)-thermal desorption (TD)-gas chromatography (GC)-mass spectrometry (MS) was used for quantitative analysis of liquid phase standards of 10 BVOC ((1) (+)-${\alpha}$-pinene, (2) (+)-${\beta}$-pinene, (3) ${\alpha}$-phellandrene, (4) (+)-3-carene, (5) ${\alpha}$-terpinene, (6) p-cymene, (7) (R)-(+)-limonene, (8) ${\gamma}$- terpinene, (9) myrcene, and (10) camphene). The results of BVOC calibration yielded comparatively stable pattern with response factor (RF) of 23,560~50,363 and coefficient of determination ($R^2$) of 0.9911~0.9973. The method detection limit (MDL) of BVOC was estimated at 0.03~0.06 ng with the reproducibility of 1.30~5.13% (in terms of relative standard error (RSE)). Emissions of BVOC were measured from four types of fruit samples ((1) tangerine (TO), (2) tangerine peel (TX), (3) strawberry (SO), and (4) sepals of strawberry (SX)). The sum of BVOC flux (${\sum}flux$ (BVOC) in ng/hr/g) for each sample was seen on the descending order of (1) TX=291,614, (2) TO=2,190, (3) SO=1,414, and (4) SX=2,093. If the results are compared between the individual components, the highest flux was seen from (R)-(+)-limonene (265,395 ng/hr/g) from TX sample.

곰취와 한대리곰취의 휘발성 향기성분 분석 (A Comparison the Volatile Aroma Compounds between Ligularia fischeri and Ligularia fischeri var. spiciformis Leaves)

  • The volatile aroma of fresh leaves is one of main factor in taste of all the edible green plants. The volatile aroma in almost edible green leaves are suggested as essential oil compounds. Ligularia fischeri, Synurus deltoides, Ligularia fischeri var. spiciformis and Aster scaber are one of the favourable edible green plants in Korea. In this study, volatile aroma compounds from Ligularia fischeri and Ligularia fischeri var. spiciformis species were analyzed by the SPME/GC/MSD method. Ligularia fischeri had 78 volatile aroma compounds such as D-limonene(20.28%), ${\alpha}$-pinene(dextro, 14.15%), L-${\beta}$-pinene(12.85%), 3-carene, ${\beta}$-cubebene(10.39%), etc. Ligularia fischeri var. spiciformis had 83 volatile aroma compounds such as D-limonene(36.97%), ${\beta}$-cubebene(13.95%), L-${\beta}$-pinene(13.38%), ${\alpha}$-pinene(dextro, 4.76%), caryophylle-ne(3.33%) etc. Conclusively, the commom volatile aroma compounds in Ligularia fischeri and Ligularia fischeri var. spiciformis leaves were D-limonene, ${\alpha}$-pinene, L-${\beta}$-pinene, ${\beta}$-cubebene, Caryophyllene, ${\alpha}$-farnesene, terpinolen. However, the composition and amount of volatile aroma compounds were very different between the two species.

참마의 휘발성 풍미성분 (Volatile flavor components of Dioscorea japonica)

  • 야생식용식물인 참마뿌리의 성숙도에 따른 휘발성 풍미성분을 분석하기 위해 SDE 방법으로 정유성분을 분리한 다음 GC 및 GC-MS를 이용하여 성분을 확인하였다. 어린 참마뿌리에서는 탄화수소류 35종, 알데히드류 5종, 케톤류 1종, 알콜류 9종, 에스테르류 2종, 유기산류 3종 및 기타 4종이 확인되었고, 성숙한 참마뿌리에서는 탄화수소류 23종, 알데히드류 2종, 알콜류 7종, 에스테르류 1종, 유기산류 8종 및 기타 1종이 확인되었다. 어린 참마에서 탄화수소류, 알데히드류, 알콜류 및 에스테르류의 함량이 더 많았고 풋풋한 내음이 더 강한 것으로 나타났으며 성숙한 참마에서는 고급 지방산의 함량이 상당히 많은 것으로 나타났다.

Anti-nociceptive and anti-inflammatory activities of the essential oil isolated from Cupressus arizonica Greene fruits

  • Background: Cupressus arizonica Greene is a coniferous tree with great importance in fragrance and pharmaceutical industries. Essential oils from C. arizonica (EC) have shown potential antioxidant, and anti-microbial activities. This study aimed at investigating the anti-nociceptive and anti-inflammatory effects/mechanisms of EC. Methods: The EC was evaluated for anti-nociceptive and anti-inflammatory activities on male Wistar rats using a formalin test and carrageenan-induced paw edema, respectively. Also, we pre-treated some of the animals with naloxone and flumazenil in the formalin test to find out the possible contributions of opioid and benzodiazepine receptors to EC anti-nociceptive effects. Finally, gas chromatography/mass spectrometry (GC/MS) analysis was used to identify the EC's constituents. Results: EC in intraperitoneal doses of 0.5 and 1 g/kg significantly decrease the nociceptive responses in both early and late phases of the formalin test. From a mechanistic point of view, flumazenil administration 20 minutes before the most effective dose of EC (1 g/kg) showed a meaningful reduction in the associated anti-nociceptive responses during the early and late phases of the formalin test. Naloxone also reduced the anti-nociceptive role of EC in the late phase. Furthermore, EC at the doses of 1, 0.5, and 0.25 g/kg significantly reduced paw edema from 0.5 hours after carrageenan injection to 4 hours. GC/MS analysis showed that isolated EC is a monoterpene-rich oil with the major presence of α-pinene (71.92%), myrcene (6.37%), δ-3-carene (4.68%), β-pinene (3.71%), and limonene (3.34%). Conclusions: EC showed potent anti-nociceptive and anti-inflammatory activities with the relative involvement of opioid and benzodiazepine receptors.

  • 본 연구의 목적은 곰취(Ligularia fischeri)와 한대리곰취(Ligularia fischeri var. spicifoprmis) 정유의 향취 및 화학성분을 비교하는데 있다. 곰취와 한대리곰취의 정유 함량은 각각 0.12%와 0.04%였으며, 역치수준은 각각 0.01%와 0.1%였다. 향수개발전문가에 의해 평가된 곰취 정유의 향취는 citrus, coniferous, green, oily, smoky, wet하였던 반면 한대리곰취 정유의 향취는 balsam, earthy, oily, wet하여 매우 상이하였다. 곰취 정유에는 총 19종의 화학성분이 검출되었는데 탄화수소가 14종(75%), 알코올이 4종(20%), 아세테이트가 1종(5%)이었으며, 주된 화학성분은 L-${\beta}$-pinene(36.02%), D-limonene(25.64%), ${\alpha}$-pinene(24.85%), ${\beta}$-phellandrene(5.39%), 3-carene(3.77%), ${\alpha}$-phellandrene(2.05%)이었다. 한대리곰취 정유에는 총 25종의 화학성분이 검출되었는데 탄화수소 17종(68%), 알코올 4종(16%), 아세테이트 3종(12%), 질소화합물 1종(4%)이었으며, 주된 화학성분은 D-limonene(39.80%), L-${\beta}$-pinene(35.43%) ${\alpha}$-pinene(11.94%), 3-carene(3.70%), ${\beta}$-cubebene(3.51%), ${\beta}$-phellandrene(2.38%)이었다. 한대리곰취 정유에서는 ${\rho}$-cymene, ${\gamma}$-muurolene, ${\gamma}$-cadinene, germacrene D, ingol 12-acetate, butyl 9,12,15-octadecatriene과 nimorazole이 미량 검출되었으나, 곰취 정유에서는 이러한 화학성분들이 검출되지 않았다. 본 연구의 결과 각각 독특한 향취를 갖는 곰취와 한대리곰취 정유는 향장산업제품 개발을 위한 소재로 활용될 수 있을 것이라 판단된다.

잣송이 부산물로부터 수증기 증류법에 의한 피톤치드의 추출 (Production of Phytoncide from Korean Pine Cone Waste by Steam Distillation)

  • 수증기 증류법을 이용하여 잣나무의 손상 없이 잣송이 부산물로부터 피톤치드 성분의 대량 추출 가능성을 조사하였다. 또한 수증기 증류법에 의해 얻어진 잣나무 피톤치드 oil을 대상으로 column chromatography법에 의한 피톤치드의 구성 성분의 분리 가능성을 타진하였다. 잣송이 부산물로부터 물 수증기 증류법과 수증기 증류법에 의해 피톤치드의 대량생산이 가능하며, 수증기 증류법의 경우가 물-수증기 증류법의 경우에 비해 수율이 2배가량 높을 정도로 효과적이었다. 수증기 증류법의 경우 $100^{\circ}C$, 30 min간 증류 시 잣나무 피톤치드 oil의 수율이 최고점이 얻어졌다. 얻어지는 잣나무 피톤치드 oil의 주성분은 ${\alpha}$-pinene, ${\beta}$-pinene, D-limonene ${\beta}$-myrcene 및 3-carene 등 12가지 이상의 성분으로 이루어지고 있음을 확인하였다. 또한 수용성의 성분이 함유되어 있는 다량의 유효성분도 함께 생산되어지면 이들 성분은 oil상에서 확인된 물질과는 동일한 성분은 전혀 없는 verbenone, ${\alpha}$-terpinieol, fenchol, 등의 성분으로 이루어짐을 확인하였다. 얻어지는 잣나무 피톤치드 oil의 다양한 구성 성분들은 column chromatography법에 의해 그 분리가 가능함을 확인하였다.

Antibacterial Activity of Essential Oil from Abies holophylla against Respiratory Tract Bacteria

  • We extracted essential oils from four species (Pinus densiflora, Larix kaempferi, Pinus koraiensis, and Abies holophylla) in the family Pinaceae to investigate their antibacterial activities against respiratory tract bacteria (Klebsiella pneumoniae, Haemophillus influenzae, Streptococcus pyogenes, Streptococcus pneumonia, and Neisseria meningitides). Among the tested oils, that from A. holophylla was showed strong activity based on disc diffusion and broth medium dilution (minimum inhibitory concentration, MIC) assays. Qualitative analysis of A. holophylla oil was carried out by GC-MS; ${\alpha}$-pinene, camphene, ${\beta}$-pinene, 3-carene, limonene, bornyl acetate, borneol, ${\beta}$-caryophyllene, ${\alpha}$-caryophyllene, caryophyllene oxide, and ${\alpha}$-bisabolol were identified as its major constituents. Fractionation by silica gel chromatography was performed to analyze the active constituents of the crude oil. In particular, one fraction containing caryophyllene oxide as the major constituent showed stronger antibacterial activity than the crude oil of A. holophylla. Growth rates of bacterial strains exposed to fraction D were explored by optical density (OD600) measurements while morphology was examined by optical microscopy observations ( ${\times}1000$). OD600 of K. pneumoniae decreased from 0.2582 to 0.005 in response to treatment with fraction D at a MIC value of $0.31{\mu}{\ell}/m{\ell}$.
