Characterization of the din (damage-inducible) and tin (temperature-inducible) Genes Isolated from Escherichia coli (대장균에서 분리된 din (damage-inducible)과 tin (temperature-inducible) 유전자들의 특성)

  • Mu d1(Ap lac) bacteriophage can be used to search for genes which are members of a common regulatory network without having to know the functions of the genes in advance. Aim was for obtaining the loci in the SOS network as well as temperature inducible loci. For this purpose, recA441 allele was used. This allele encodes a thermosensitive recA gene product; thus, the recA441 allele can be activated upon temperature upshift without by external DNA damage. Approximately 10, 000 colonies were screened, and then searched for the colonies which expressed .betha.-galactosidase higher level at 42.deg.C than at 30.deg.C. The strains identified fell into two dlasses; (i) ones in which the increased expression was $recA^{+}$ $lexA^{+}$ -dependent, that is, din(damage-inducible) genes which were due to the activation of recA441 allele and (ii) ones in which the increased expression was $recA^{+}$ $lexA^{+}$ -independent and only temperature-inducible, tin genes. Rough mapping position was obtained for these genes.

  • 국내 저수지 17,477개소 중 30~40 여년전에 축조된 곳은 16,646개소로 95.2%가 노후화로 인한 지속적인 유지보수가 필요하며, 기후변화에 따른 대응능력이 현저하게 낮다. 하구둑, 방조제, 대형 농업용저수지 같은 대규모 농업기반시설들은 축조 당시보다 설계홍수량이 2~3배 이상 증가함에 따라 기후변화에 대비하기 위해 조속한 치수능력 증대가 필요하다. 하지만 대형 농업용저수지의 치수능력 증대를 위해서는 지형적 입지조건, 시공 가능성, 경제성 등을 고려하여 여수로의 위치, 규모, 형식 변경 등이 수반된다. 따라서 본 연구에서는 무한천 상류 예당저수지의 여수로 위치변경에 따른 저수지 하류 하천 흐름변화에 대하여 2차원 수치모의 연구를 수행하였다. 본 연구에서는 TELEMAC-2D 모형을 적용하여 저수지 하류 하천의 흐름을 모의하였고, RMA-2 모의결과와 비교검토 하였다. TELEMAC-2D 모형은 상류(subcritical flow)와 사류(supercritical flow) 흐름 모의가 가능하며, 홍수범람 뿐만아니라 동일 격자망으로 유한요소법과 유한체적법 모두 수치모의가 가능한 특징이 있다. 본 연구에서는 시간과 공간에 대하여 2차 정확도를 가지는 WAF(Weighted Average Flux) 유한체적기법과 $k-{\varepsilon}$ 난류모형을 적용하였다. 수치모의시 유역종합치수계획의 30년, 50년, 100년 빈도 계획홍수량에 대하여 여수로 위치변경 전(CASE-1)과 위치변경 후(CASE-2) 저수지 하류하천 흐름변화에 대해 모의하였다. 본 연구는 국내 하천 특성을 반영한 기설 저수지의 하류하천의 흐름에 대한 수치모의의 기초자료로 활용되어, 저수지 방류 흐름이 하류하천에 미치는 영향 분석 등에 기여할 것으로 기대된다.

Effects of oscillation parameters on aerodynamic behavior of a rectangular 5:1 cylinder near resonance frequency

  • Large Eddy Simulation (LES) is used to explore the influence of vibration frequency and amplitude on the aerodynamic performance of a rectangular cylinder with an aspect ratio of B/D=5 (B: breadth; D: depth of cylinder) at a Reynolds number of 22,000 near resonance frequency. In smooth flow conditions, the research employs a sequence of three-dimensional simulations under forced vibration with diverse frequency ratios fe / fo = 0.8-1.2 (fe : oscillation frequency; fo : Strouhal frequency when the rectangular cylinder is stationary ) and oscillation amplitudes Ah/D = 0.05 - 0.3. The individual influences of fe / fo and Ah/D on the characteristics of integrated and distributed aerodynamic forces are the focal points of discussion. For the integrated aerodynamic force, particular emphasis is placed on the analysis of the dependence of velocity-proportional component C1 and displacement-proportional component C2 of unsteady aerodynamic force on amplitude and frequency ratio. Near the resonance frequency, the dependencies of C1 and C2 on amplitude are stronger than that of frequency ratio. For the distributed aerodynamic force, the increase in frequency and amplitude promotes the position of the main vortex core and reattachment to the leading edge in the streamwise direction. In the spanwise direction, vibration enhances the spanwise correlation of aerodynamic force to weaken the three-dimensional effect of the flow field, and a lower frequency ratio and larger amplitude amplify this effect.

Development of a Low-cost Monocular PSD Motion Capture System with Two Active Markers at Fixed Distance (일정간격의 두 능동마커를 이용한 저가형 단안 PSD 모션캡쳐 시스템 개발)

  • In this paper, we propose a low-cost and compact motion capture system which enables to play motion games in PS2(Play Station 2). Recently, motion capture systems which are being used as a part in film producing and making games are too expensive and enormous systems. Now days, motion games using common USB camera are slow and have two-dimension recognition. But PSD sensor has a few good points, such as fast and low-cost. In recently year, 3D motion capture systems using 2D PSD (Position Sensitive Detector) optic sensor for motion capturing have been developed. One is Multi-PSD motion capture system applying stereo vision and another is Single-PSD motion capture system applying optical theory ship. But there are some problems to apply them to motion games. The Multi-PSD is high-cost and complicated because of using two more PSD Camera. It is so difficult to make markers having omni-direction equal intensity in Single-PSD. In this research, we propose a new theory that solves aforementioned problems. It can measure 3D coordination if separated two marker's intensity is equal to. We made a system based on this theory and experimented for performance capability. As a result, we were able to develop a motion capture system which is a single, low-cost, fast, compact, wide-angle and an adaptable motion games. The developed system is expected to be useful in animation, movies and games.

Studies on Corrected RGP Lens Fitting and Use of Care System (올바른 RGP렌즈의 처방과 관리용품의 사용에 대한 결과 연구)

  • We performed refraction, keratometry, slit lamp biomicroscopy. We selected 58 current spherical RGP lens wearers for this three-month study. All patients exhibits at least 0.75D of corneal astigmatism measured with the keratometer, and 37 patients had corneal astigmatism of 1.50D or greater. At least follow-up visit, we measured Snellen acuity with lenses, and performed overrefraction, overkeratometry and slit lamp biomicroscopy. We charted lens position, movement and surface quality. During the three month, biomicroscopy revealed no corneal edema and neovascularization on any patients. Fluorescein staining were 52 patients case of grade 0.5 patients case of grade 1, and 1 patient case of grade 2. In evaluating post-fit residual cylinder, on overrefraction as a percentage of refractive cylinder. By the initial visit, one-week visit, one-month visit, and two-month visit are 41%, 34%, 29%, respectively. In this data, we knew no change after one month. The average overrefraction for these eyes in absolute diopters is 0.26D(initial visit), 0.22D(one-week visit, 0.17D(one-month visit), and 0.16D(two-month visit). The use of a regimen containing a dedicated daily cleaner was more effective in maintaining patient comfort and lens cleanliness than was the use of a regimen containing only a multipurpose solution.

  • This paper proposes the motion generation method in which the movement of the 3-links leg subsystem in constrained to slider-link and a singular posture can be easily avoided. This method is the realization of jumping control moving in a vertical direction, which mimics a cat's behavior. To consider the movement from the point of the constraint mechanical system, a robotics system for realizing the motion will change its configuration according to the position. The effectiveness of the proposed scheme is illustrated by simulation and experimental results.

  • This paper presents the characteristic analysis by changing the shape and position of auxiliary pole core for Automotive stater motor. The brush contact resistance and mechanical loss calculated from the experimental data are used in characteristic analysis. The Finite Element Analysis Method(FEM) and 3D Equivalent Magnet Circuit Network(EMCN) are used for analysis. The validity of the proposed methods are verified by comparison of analysis results and experimental ones.

  • LiDAR(Light Detection and Ranging) is considered to be a very accurate and useful tool for detection and reconstruction of ground objects. LiDAR data has information about both intensity and x,y,z position of the ground objects. LiDAR data can be collected from both first and last-return, which are called multi-return, with up to 5 different returns simultaneously. In this paper, an approach to reconstruct buildings in urban area using LiDAR multi-return data is presented. The reconstructed buildings are combined with DEM(Digital Elevation Model) produced from DSM(Digital Surface Model) in given area to implement 3D modeling. As a result, it is shown that buildings in urban area can be reconstructed and classified by the integration of the multi-return and intensity data of LiDAR.

  • The digital map expresses natural topography and artificial things with 3D position coordinates in the computer such as the road, railway, building, river, mountain, paddy and dryland. Therefore, those should contribute to the information-oriented society by maintaining information and providing it to users quickly. However it is difficult to maintain the most recent topographic information all the time because of restricted budget and time. The purpose of this study is to investigate and analyze the updating area of the digital map using remotely sensed data, and to furnish the useful information reducing cost and time. To predict updating area of the digital map, we applied the urban changes analysis method to Landsat TM images from produced date of the digital map to up-to-date. Classification method for urban change analysis applied single band process algorithm. This study presents that updating area of the digital map is predicted by only the rate of 40% on total research area.

  • A controller design procedure for an electro-hydraulic positioning systems has been developed using $H{\infty}$ control. The generalized plant models and weighting function for multiplicative uncertainty modelling error was presented along with $H{\infty}$ controller designs in order to investigate the robust stability and performance. Both disturbance rejection and command tracking performances were improved with the $H{\infty}$ controller, and the better uniformity of time response is achieved across wide range of operating conditions than the PID, LQR and LQG control scheme. The multiplicative uncertainty case was specifically suited for the design of an electro-hydraulic positioning control systems using $H{\infty}$ control.
