Chitosan Increases the Release of Renal Dipeptidase from Porcine Renal Proximal Tubule Cells

  • Renal dipeptidase (RDPase, membrane dipeptidase, dehydropeptidase 1, EC 3.4.13.19) has been widely studied since it was first purified from porcine kidney brush border membrane. It was reported that RDPase activity in urine samples of acute and chronic renal failure patients decreases. Nitric oxide (NO) is a highly reactive free radical involved in a number of physiological and pathological processes. NO is able to act in a dual mode, leading either to induction of apoptosis or to blunted execution of programmed cell death. NO inhibited the RDPase release from porcine renal proximal tubules, which could be blocked by L-NAME. Chitosan, the linear polymer of D-glucosamine in $\beta$(1\longrightarrow4) linkage, not only reversed the decreased RDPase release by NO but also increased NO production in the proximal tubule cells. The stimulatory effect of NO on RDPase release from proximal tubules in the presence of chitosan must be different from the previously proposed mechanism of RDPase release via NO signaling pathway. Chitosan stimulated the RDPase release in the proximal tubules and increased RDPase activity to 220% and 250% at 0.1% and 1%, respectively. RDPase release was decreased to about 40% in the injured proximal tubules and was recovered in proportion to the increase of chitosan. Chitosan may be useful in recovery of renal function from $HgCl_2$injury.

Performance Improvement of IPMC(Ionic Polymer Metal Composites) for a Flapping Actuator

  • In this paper, a trade-off design and fabrication of IPMC(Ionic Polymer Metal Composites) as an actuator for a flapping device have been described. Experiments for the internal solvent loss of IPMCs have been conducted for various combinations of cation and solvent in order to find out the best combination of cation and solvent for minimal solvent loss and higher actuation force. From the experiments, it was found that IPMCs with heavy water as their solvent could operate longer. Relations between length/thickness and tip force of IPMCs were also quantitatively identified for the actuator design from the tip force measurement of 200, 400, 640, and $800{\mu}m$ thick IPMCs. All IPMCs thicker than $200{\mu}m$ were processed by casting $Nafion^{TM}$ solution. The shorter and thicker IPMCs tended to generate higher actuation force but lower actuation displacement. To improve surface conductivity and to minimize solvent evaporation due to electrically heated electrodes, gold was sputtered on both surfaces of the cast IPMCs by the Physical Vapor Deposition(PVD) process. For amplification of a short IPMC's small actuation displacement to a large flapping motion, a rack-and-pinion type hinge was used in the flapping device. An insect wing was attached to the IPMC flapping mechanism for its flapping test. In this test, the wing flapping device using the $800{\mu}m$ thick IPMC. could create around $10^{\circ}{\sim}85^{\circ}$ flapping angles and $0.5{\sim}15Hz$ flapping frequencies by applying $3{\sim|}4V$.

Synthesis and Properties of New Phthaloperinone Dyes containing Anthraquinone Moiety (Anthraquinone을 포함하는 신규 Phthaloperinone 색소의 합성과 특성에 대한 연구)

  • We have synthesized five novel phthaloperinone dyes via a condensation reaction to be applied as yellow colorants for liquid crystal display(LCD) color filters. The reaction between 1,8-naphthalic anhydride(1a), 4-chloro-1,8-naphthalic anhydride(1b), 4-bromo-1,8-naphthalic anhydride(1c), 3-nitro-1,8-naphthalic anhydride(1d), 4-nitro-1,8-naphthalic anhydride(1e) and 1,2-diaminoanthraquinone(2) proceeded readily giving a product in 72-88% yields. The synthesized dyes were characterized by UV-Vis, mass spectrometry and elemental analysis. The spectral properties and thermal stability of the dyes were examined. The dyes absorb at around 400-450nm. All five dyes showed satisfactory thermal stability: the dyes retain 99-100% of its original weight at $300^{\circ}C$, 98-100% at $350^{\circ}C$, 92-98% at $400^{\circ}C$, and 84-92% at $450^{\circ}C$. We have quantitatively evaluated the reaction mechanism and reactivity of dye molecules by means of Pariser-Parr-Pople Molecular Orbital Method(PPP-MO).

  • Dietary lipids are rapidly hydrolysed and biohydrogenated in the rumen resulting in meat and milk characterised by a high content of saturated fatty acids and low polyunsaturated fatty acids (PUFA), which contributes to increases in the risk of diseases including cardiovascular disease and cancer. There has been considerable interest in altering the fatty acid composition of ruminant products with the overall aim of improving the long-term health of consumers. Metabolism of dietary lipids in the rumen (lipolysis and biohydrogenation) is a major critical control point in determining the fatty acid composition of ruminant lipids. Our understanding of the pathways involved and metabolically important intermediates has advanced considerably in recent years. Advances in molecular microbial technology based on 16S rRNA genes have helped to further advance our knowledge of the key organisms responsible for ruminal lipid transformation. Attention has focused on ruminal biohydrogenation of lipids in forages, plant oils and oilseeds, fish oil, marine algae and fat supplements as important dietary strategies which impact on fatty acid composition of ruminant lipids. Forages, such as grass and legumes, are rich in omega-3 PUFA and are a useful natural strategy in improving nutritional value of ruminant products. Specifically this review targets two key areas in relation to forages: i) what is the fate of the lipid-rich plant chloroplast in the rumen and ii) the role of the enzyme polyphenol oxidase in red clover as a natural plant-based protection mechanism of dietary lipids in the rumen. The review also addresses major pathways and micro-organisms involved in lipolysis and biohydrogenation.

Measurement of Carbon Nanotube Agglomerates Size and Shape in Dilute Phase of a Fluidized Bed (유동층 반응기 희박상 내 탄소나노튜브 응집체의 크기 및 형상 측정)

  • Size and shape of carbon nanotube (CNT) agglomerates in the dilute phase of a bubbling fluidized bed ($0.15m\;i.d{\times}2.6m\;high$) have been determined by the laser sheet technique. Axial solid holdup distribution of the CNT particles showed S curve with dense phase and dilute phase in bubbling fluidization regime. Heywood diameter and Feret diameter of the CNT agglomerates in the dilute phase of bubbling fluidized bed increased with increasing gas velocity. The CNT particle number in the agglomerates increased with increasing of gas velocity. Aspect ratio increased and circularity, roundness and solidity decreased with increasing of gas velocity. A possible mechanism of agglomerates formation was proposed based on the obtained information.

Improvement of Electronic Properties and Amplification of Electron Trapping/Recovery through Liquid Crystal(LC) Passivation on Amorphous InGaZnO Thin Film Transistors

  • 본 연구에서는 nematic 액정의 종류 중 하나인 5CB (4-Cyano-4'-pentylbiphenyl) 물질을 박막 트랜지스터 (TFT)의 passivation 층으로 사용했을 때 그 전기적 특성향상을 확인하였다. RF-magnetron sputtering법으로 증착된 비정질 InGaZnO 박막을 활성층으로 사용한 TFT를 제작하여 그 활성층 위에 drop형식으로 passivation 하였다. 그 결과, drain current (I_DS)가 약 10배 정도 증가하고, linear region(V_D=0.5V)에서 mobility와 subthreshold slope(SS)이 각각 6.7에서 12.2, 0.3에서 0.2로 향상되는 것이 보였다. 이것은 gate bias가 인가되었을 때 freedericksz 전이를 통한 액정의 배향과 이때 형성된 dipole 형성에 의한 것으로 보이며, 이러한 LC의 배향은 편광현미경을 통하여 표면과 수직으로 배향한다는 사실을 확인 할 수 있었고 이 LC-passivation된 a-IGZO TFT의 전기적 특성의 향상에 대한 mechanism을 제시하였다. 그리고 배향한 LC가 가지는 dipole에 의해 bias stress 상황에서 독특한 electron trapping과 recovery의 증폭효과가 나타났다. V_G=+20V의 positive gate bias stress를 1000s동안 가했을 때, passivation되지 않은 a-IGZO TFT의 경우 +4V의 threshold voltage shift(${\Delta}V$_TH)가 발생되었고, 바로 -20V의 negative gate bias를 30s간 가해주었을 때 -2.5V의 ${\Delta}V$_TH가 발생하였다. 반면 LC-passivation된 a-IGZO TFT의 경우 각각 +5V와 -4V의 ${\Delta}V$_TH로 더 큰 변화를 가져왔다. 이러한 LC에 의한 electron trapping/recovery 증폭효과에 대한 model을 제시하였다.

Influence of stiffeners on the performance of blind-bolt end-plate connections to CFST columns

  • The paper aims to investigate the mechanical mechanism and seismic effect of stiffeners in blind bolt endplate connection to CFST column. A precise 3D finite element model with considering the cyclic properties of concrete and steel materials was established, and the efficiency was validated through monotonic and cyclic test data. The deforming pattern and the seismic performance of the unstiffened and stiffened blind bolt endplate connections were investigated. Then a parametric analysis was conducted to analyze the contribution of stiffeners and the joint working behaviors with endplate under cyclic load. The joint stiffness classifications were compared and a supplement stiffness classification method was proposed, and the energy dissipation ability of different class connections were compared and discussed. Results indicated that the main deformation pattern of unstiffened blind bolt endplate connections was the local bending of end plate. The vertical stiffeners can effectively alleviate the local bending deformation of end plate. And influence of stiffeners in thin endplate and thick endplate was different. Based on the stiffness of external diaphragm welded connection, a more detailed rigidity classification was proposed which included the pin, semi-rigid, quasi-rigid and rigid connection. Beam was the main energy dissipation source for rigid connection. For the semi-rigid and quasi-rigid connection, the extended endplate, stiffeners and steel beam would all participate in the energy dissipation.

Study on the Atomization Characteristics of a Counter-swirling Two-phase Atomizer with Variations of Swirl angle (역선회 이류체 미립화기의 선회각 변화에 따른 미립화 특성연구)

  • Experimental and analytical researches have been conducted on the twin-fluid atomizers for better droplet breakup during the past decades. But, the studies on the disintegration mechanism still present a great challenge to understand the drop behavior and breakup structure. In an effort to describe the aerodynamic behavior of the sprays issuing from the internal mixing counter-swirling nozzle, the spatial distribution of axial (U) radial (V) and tangential (W) components of droplet velocities are investigated across the radial distance at several axial locations of Z=30, 50, 80, 120 and 170mm, respectively. Experiments were conducted for the liquid flow rates which was kept constant at 7.95 g/s and the air injection pressures were varied from 20 kPa to 140 kPa. Counter-swirling internal mixing nozzles manufactured at angles of $15^{\circ},\;30^{\circ},\;45^{\circ}$ and $60^{\circ}$ the central axis with axi-symmetric tangential-drilled holes was considered. The distributions of velocities and turbulence intensities are comparatively analyzed. PDPA is installed to specify spray flows, which have been conducted along the axial downstream distance from the nozzle exit. Ten thousand of sampling data was collected at each point with time limits of 30 second. 3-D automatic traversing system is used to control the exact measurement. It is observed that the sprays with all swirl angle have the maximum SMD for on air injection pressure of 20 kPa and 140 kPa with centerline, respectively. The nozzle with swirl angle of $60^{\circ}$ has vest performance.

The Molecular Mechanism of Safrole-induced DNA Adducts and its Role to Oral Carcinogenesis

  • IARC classified areca quid as a human carcinogen. Areca quid chewed in Taiwan includes Piper betle inflorescence, which contains high concentrations of safrole (15 mg/fresh weight). Safrole is a documented rodent hepatocarcinogen, and chewing areca quid may contribute to human exposure (420 $\mu$m in saliva). The carcinogenicity of safrole is mediated through 1'-hydroxysafrole formation, followed by sulfonation to an unstable sulfate that reacts to form DNA adducts. Using human liver microsomes and Escherichia coli membranes expressing bicistronic human P450s, CYP2E1 and CYP2C9 were identified as the main P450s involved in the activation of safrole. We have demonstrated the presence of stable safrole-dGMP adducts in human oral tissues following areca quid chewing using $^{32}$ P-postlabeling and HPLC mass spectrometry methods. By studying 88 subjects with a known AQ chewing history and 161 matched controls, we have demonstrated that the presence of safrole-DNA adducts in peripheral blood cells was correlated to AQ chewing, and CYP2E1 seemed to play an important role in the modulation of safrole-DNA adduct formation. We have also shown that safrole can form stable safrole-DNA adducts as well as oxidative damages in rodent liver. However, the stable safrole-DNA adducts may represent a more significant initial lesion as compared to the rapidly repaired safrole-induced 8-hydroxy-2'-deoxyguanosine. This oxidative DNA damage is mediated through the formation of hydoryxchavicol, the major safrole metabolite in human urine. Hydroxychavicol may have gone through two-electron oxidation to the o-quinone; then via one-electron reduction to semiquinone radicals to generate oxidative DNA damage. However, these reactive metabolites can be efficiently conjugated by GSH. These data suggest that safrole may contribute to the initiation of oral carcinogenesis through safrole-DNA adduct and not oxidative DNA damage. In addition, CYP2E1 may modulate this adduct formation.

  • Hot workability of the AA5083 alloy ws investigated by torsion test at temperature ranges of $350{\sim}520^{\circ}C$ and strain rates of 0.5, 1.0, and 3.0/sec. The flow stress and hot ductility of the AA5083 alloy as a function of deformation variables such as temperature and train rate were studied. The microstructural evolution of the AA5083 alloy was studied in relation to Zener-Hollomon parameter (Z=exp( /RT) Also the hot restoration mechanism of the AA5083 alloy was small when Z val-ues were higher than $1.73{\times}1016/sec(370^{\circ}C,\;0.5/sec)$ In addition the difference microstructures during hot deformation. It was found that the increase of flow curves and deformed microstructures during hot deformation. It was found that the increase of flow stress of the AA5083 alloy was small when Z val-ues were higher than $1.73{\times}1016/sec(370^{\circ}C.\;0.5/sec)$. However under the low Z values less than $1.73{\times}1016/sec(370^{\circ}C,\;0.5/sec)$ the flow stress increase with increasing the Z values. The large dispersoid particles in the matrix grain decreased the flow strain of the AA5083 alloy because it caused the stress concentration during hot deformation.
