Role of Surface Protective Antigen A in the Pathogenesis of Erysipelothrix rhusiopathiae Strain C43065

  • To clarify the role of surface protective antigen A (SpaA) in the pathogenesis of Erysipelothrix rhusiopathiae C43065 (serotype 2), the spaA deletion mutant of E. rhusiopathiae ${\Delta}spaA$ was constructed by homologous recombination. The virulence of the ${\Delta}spaA$ mutant decreased more than 76-fold compared with that of the wild-type strain C43065 in mice. The mutant strain was sensitive to the bactericidal action of swine serum, whereas the wild-type strain was resistant. The adhesion of wild-type strain to MEF cells was inhibited significantly by treatment with rabbit antiserum against recombinant SpaA (rSpaA) as compared with the treatment with normal rabbit serum, but the mutant strain was not affected. The mutant strain was readily taken up by mouse peritoneal macrophages in the normal rabbit serum, whereas the wild-type strain was resistant. Whereas the rabbit antiserum against rSpaA promoted the phagocytosis of wild-type strain by macrophages, the mutant strain was not affected. In addition, mice vaccinated with the formalin-killed mutant strain were provided 40% protection against challenge by the homologous virulent strain as compared with those with wild-type strain, NaOH-extracted antigen, or rSpaA, which provided more than 80% protection against the same infection. These suggested that SpaA has an important role in the pathogenesis of E. rhusiopathiae infection and could be a target for vaccination against swine erysipelas.

Inhibitory Effect of SPA0355, a Thiourea Analogue, on Inflammation and Alveolar Bone Loss in Rats with Ligature-Induced Periodontitis

  • It has been documented that SPA0355 exerts antiinflammatory effects via the inhibition of nuclear factor-kappaB activation. In present study, we investigated the inhibitory effects of SPA0355 on periodontitis in an animal model. Periodontitis was induced by ligation of the cervix of the 1st molar in the left mandible in rats. After ligature, the rats were randomly divided into four groups and topically applied with SPA0355 (0.5, 1, and 2%) or the vehicle alone once daily for 10 days. Body weight and food intake were measured daily throughout the experimental period. At day 10 post-ligature, the infiltration of inflammatory cells and distance of the cementoenamel junction (CEJ) to the alveolar bone crest (ABC) in the distal area of ligatured tooth were estimated histopathologically. No changes in body weight or food intake were found between the control and SPA0355 groups. The degree of inflammation was decreased in all three SPA0355 application groups. A decrease CEJ-ABC distance was observed in the 0.5% and 1% SPA0355 groups. These results indicate that SPA0355 inhibits the infiltration of inflammatory cells and alveolar bone resorption and suggests its potential as a therapeutic agent for periodontitis.

Stimulatory Effect of Staphylococcal Protein A on Inflammatory Response in Human HaCaT Keratinocytes (사람의 피부상피세포에서 황색포도상구균의 독소인자인 Staphylococcal Protein A의 염증반응 촉진효과)

  • Staphylococcus aureus is a major human pathogen that is associated with various types of local and systemic infection. Staphylococcal protin A (SPA), a highly expressed surface component of S. aureus, may have a role in virulence such as activating inflammation and interfering with immune clearance. We examined the effect of recombinant SPA on inflammatory response in human HaCaT keratinocytes. The recombinant SPA protein was prepared using the pET-28a Vector System in Escherichia coli. The expression of pro-inflammatory related adhesion molecules and cytokines in HaCaT cells incubated for 6, 12, and 24 h with SPA (2 ${\mu}g$/ml) was analyzed by comparative RT-PCR or ELISA. The expression of E-selectin, ICAM-1, MCP-1, IL-6 and IL-8 was significantly increased in HaCaT from 6 to 24 h after treatment with SPA. SPA showed the effect on the adhesion-promoting ability of U937 monocytes to HaCaT cells. Our data demonstrate that SPA stimulates inflammatory response of HaCaT cells, implicating an important factor for exacerbation of skin inflammation of immunologic disease.

Flagship Store Experience of Luxury and SPA Brands -Effect on Store Emotion and Loyalty- (럭셔리와 SPA 플래그십 스토어 체험 -점포 감정 및 충성도에 미치는 효과-)

  • This study estimated a structural model for examining causal relationships among flagship store experiences, store emotion and loyalty, and to compare the structural models between luxury and fast fashion SPA brands. A total of 416 responses were collected from consumers who had experience with a variety of luxury and SPA flagship stores. Findings confirmed that flagship store experience consisted of four factors, such as sensory, affective, intellectual and behavioral experiences. Sensory and behavioral experiences had positive effects on store emotion that influenced loyalty. Affective and cognitive experiences influenced loyalty. All aspects of experiences explained store emotion or loyalty for luxury flagship stores; however, two of experiences (i.e., sensory and affective) were important to determine store emotion or loyalty for SPA flagship stores. The study discussed managerial implications for fashion brands to develop and manage flagship stores.

Combined Spa-Exercise Therapy for Vital Sign, Body Composition, Skin Status in Participants of Middle-Aged Women: A Before and After Study (12주간의 온천요법 전후 갱년기 환자의 혈압, 맥박, 체성분, 피부 변화에 관한 임상연구)

  • Objectives The aim of this clinical study was to find out the changes of vital sign, body composition, skin status before and after combined spa-exercise therapy in participants of middle-aged women. Methods In this study 24 participants of middle-aged women went through 12 weeks of combined spa-exercise therapy. Each week vital sign, body composition, skin status were measured before exercise, which consisted of warming-up, whole body exercise and final exercises bathing in spa ($32{\sim}36^{\circ}C$). Results 1. The average of systolic blood pressure significantly decreased from $126.38{\pm}19.33mmHg$ to $120.46{\pm}11.18mmHg$ after 11 weeks of spa-exercise therapy (p<0.05). 2. The average of moisture of skin significantly increased from $36.83{\pm}3.18$ to $44.21{\pm}7.43$ after 11 weeks of spa-exercise therapy (p<0.05). 3. The average of elasticity of skin significantly increased from $59.75{\pm}3.50$ to $62.63{\pm}2.29$ after 11 weeks of spa-exercise therapy (p<0.05). 4. The average of diastolic blood pressure, pulse, et cetera. didn't significantly change after spa-exercise therapy. Conclusions The results showed that 12 weeks of spa-exercise therapy had significant effect on reducing systolic blood pressure, increasing moisture, elasticity of skin in participants of middle-aged women, which means spa-exercise therapy may be used as a treatment on high blood pressure and cutaneous diseases. Further studies are anticipated to find out other various effects of spa-exercise therapy.

Factors Relating to Social Physique Anxiety in Adolescent Girls (청소년기 여학생의 사회적 체격불안 관련 요인)

  • Purpose: This descriptive study was to explore the factors relating to Social Physique Anxiety(SPA) in the adolescent girls(N=700). These factors included the biological(level of school[age], period of menstruation, and BMI), the sociocultural(Influence of Mass Media, [IMM], Stereotype of Ideal Female Body[SIFB], type of school, perceived figure, and ideal figure) and the BW control-related factors(experience of bodyweight [BW] control, satisfaction at the result of BW control, and motive of BW control). Method: The sample was 700 girls of the middle and high schools in Jeju-do. The data was collected by the self-reported questionnaire and processed with the SPSS Win 12.0 program. Result: 26% of the variance in SPA was explained by the biological factors(level of school) and the sociocultural factors (perceived figure, ideal figure, type of school, and IMM). SPA, IMM, and SIFB were significantly correlated with each others(r=1,65-2.92, p<.05). The mean SPA scores according to BW control-related factors were significantly different. Conclusion: This results suggest that media education is necessary to reduce or eliminate the adolescent girl's SPA. In addition, SPA needs to be considered as the important concept for the future nursing researches and interventions related to BW control.

Influence of SPA Brands' Sustainable Activities on the Consumer's Ethical Consciousness, Brand Loyalty and Purchase Intention: Focused on H&M Brand (Korea vs. Russia) (SPA 브랜드의 지속가능 활동이 윤리의식과 브랜드 충성도 및 구매의도에 미치는 영향 - 한국과 러시아의 H&M 비교 중심으로 -)

  • Today, the fashion industries, which are appearing in the world, can satisfy their economic needs, as well as provide and distribute their products in order to introduce sustainable methods and avoid becoming a bad influence on future generations. Recently, SPA brands have contributed to the sustainable development of society with different kinds of products, such as organic, recycled and environmental products. For this research, among different SPA brands, we chose H&M as the most representative one. Using Russian and Korean consumers as examples, we analyzed how sustainable activities affect a consumer's ethical consciousness, brand loyalty and purchase intention. The results of this study show that firstly, all sustainable activities have positive effects on the ethical consciousness of Russian and, in particular, Korean consumers. Secondly, ethical consciousness has a positive effect on brand loyalty, which, in turn, has a positive effect on purchase intention. Consequently, by adding to the existing sustainable and cultural activities, we can propose new perspectives for future research on sustainability. Due to the effect on a consumer's ethical consciousness, future research might consider ethical consciousness as the main factor. By focusing on H&M as a representative of SPA brands with Russian and Korean consumers as target audiences, we can advise global SPA brands on the direction of their sustainable activities and exporting of their products to foreign markets.

The influence of sustainability management fit of SPA brand on consumer purchase intention (SPA 브랜드의 지속가능경영 활동 적합성이 구매의도에 미치는 영향)

  • The purpose of this study is to examine the impact of perceived sustainability management fit of SPA brands on consumer purchase intention. A survey was conducted targeting women in their 20s and 30s from April 27th to May 6th 2013, and a total of 350 questionnaires were used in the final analysis. Frequency analysis, reliability analysis, factor analysis, ANOVA, simple regression analysis and multiple regression analysis were carried out using SPSS ver 21.0. The results of this study are as follows. First, fit of all three areas- economic, ecological and social-of sustainability management activities have a significant impact on consumer perception on sustainability activities. Second, perceived sustainability activities by SPA brands are found to have a positive effect on brand attitude, brand trust and consumer satisfaction. Third, brand attitude and consumer satisfaction forged by perceived sustainability activities by SPA brands have a positive impact on consumers' purchase intention. Based on the result of this study, the following marketing implications can be suggested. First, SPA brands need to be more active in all three area of sustainability activities while coming up with measures for differentiated social sustainability activities. Second, SPA brands need to meet their primary obligation by providing consumers with quality products at a reasonable price. Third, SPA brands need to recognize the potential value of sustainability activities as a driver of a longstanding relationship with consumers as well as long-term profit and value creation, leading to a great financial performance.

The effect of aroma foot spa on stress (아로마풋스파가 스트레스에 미치는 영향)

  • To identify the effect of the aroma foot spa on the stress, 12 university students with stress were divided into control group (6 subjects) and aroma foot spa group (6 subjects). Control group was not subjected to any kind of intervention while aroma foot spa group was subjected to 10 minutes of foot bath using 5% Juniper, lavender and lemon essential oil and 10 minutes of foot massage using massage cream during four weeks, two times per week. As for the evaluation method, brain wave was studied to measure the amount of change in stress. After measuring the change in the brain wave before and after the experiment, it was proven that the aroma foot spa group s Alpha wave (Z=-2.364, p<.05) and SMR wave (Z=-1.981, p<.05) were higher than those of the control group. Moreover, when the pre and post experiment results of the aroma foot spa group were measured, it was proven that the Theta wave (Z=-2.366, p<.05) decreased while Alpha wave (Z=-2.371, p<.05) increased. In other words, aroma foot spa that included foot bath using essential oil and foot massage increased Alpha wave which in turn influenced the brain wave due to increased blood circulation resulting from muscle relaxation. Moreover, SMR wave increase was closely related to the change in Alpha wave, which demonstrates that SMR wave increased due to stress alleviation. Accordingly, it may be possible to assume that aroma foot spa is effective in relieving stress.

Clinical experience with single-port access laparoscopic cystectomy and myomectomy

  • Objective: This study was performed to assess our clinical experience with single-port access (SPA) laparoscopic cystectomy and myomectomy and the surgical outcomes of those procedures at our institution. Methods: The authors evaluated the surgical outcomes of SPA laparoscopic cystectomy in 293 patients and SPA laparoscopic myomectomy in 246 patients. The surgical outcomes comprised operation time, the amount of blood loss during the operation, the change in hemoglobin (before and after the operation), the change in hematocrit (before and after the operation), switching to the multi-port access method, complications, transfusions, and the duration of the postoperative hospital stay. Results: The Pearson correlation coefficient and the Spearman correlation coefficient between the operation time and the amount of blood loss were 0.312 and 0.321 for SPA laparoscopic cystectomy, respectively, and 0.706 and 0.674 for SPA laparoscopic myomectomy, respectively. The drops in hemoglobin and hematocrit were $1.33{\pm}0.78g/dL$ and $4.14%{\pm}2.45%$, respectively, in SPA laparoscopic cystectomy, while the corresponding figures were $1.34{\pm}1.13g/dL$ and $4.17%{\pm}3.24%$ in SPA laparoscopic myomectomy, respectively. Conclusion: This study reported the surgical outcomes of SPA laparoscopic cystectomy and myomectomy and compared them to previously published findings on traditional laparoscopic cystectomy and myomectomy. No significant differences were found in the surgical outcomes between SPA and traditional laparoscopic cystectomy and myomectomy.
