  • Peroxynitrite and 4-hydroxynonenal (4-HNE) are highly reactive molecules which are generated under oxidative stress condition and during aging. Many proteins in living organism are modified by them and consequently associated with various diseases including cardiovascular and neurodegenerative diseases. We hypothesize that peroxynitrite and 4-HNE modified serum proteins are also associated with aging process. To establish information on peroxynitrite and 4-HNE adducted proteins for aging study, we used proteins methods, 2D-PAGE and MALD-TOF MS, to identify modified proteins from young (70month) and old (25-month) rat serum. 전통 청국장의 발효 기간 동안 변화하는 수용성 단백질 개요 (Changes of Protein Profiles in Cheonggukjang during the Fermentation Period)

  • 콩 발효식품인 청국장은 어느 정도는 그 기능성 때문에 많은 사람들이 선호하고 있다. 청국장의 발효 과정은 발효미생물이 분비하는 단백질 분해 효소를 포함하는 여러 효소들에 의해 이루어지기 때문에 발효기간 동안 청국장의 단백질체 분석은 발효의 최적화를 이루기 위해서 그리고 청국장에 생성되는 기능성물질 생성 과정을 이해하는 데 도움이 된다. 청국장의 수용성 단백질을 phenol/chloroform 추출 방법으로 분리하여 2-D gel 분석에 방해되는 물질들을 제거하였다. 각 발효 단계에서 단백질 분석을 하였을 때 20시간 안에 대부분의 단백질들이 작은 분자로 분해되었고 수용성 단백질에는 미생물 유래 단백질들이 점차 증가하였다. 청국장의 단백질 프로파일은 natto의 것과는 매우 다른 양상을 2-D gel 상에서 보였다. 각 gel에서 50개의 단백질을 임의로 선발하여 MALDI-TOF MS 분석을 하고 PMF로 단백질을 동정하였다. 결과는 콩이나 발효 미생물들의 유전체 정보가 부족하여 청국장에서 9종 natto에서 15종의 단백질만을 동정할 수 있었다. MS/MS 분석을 통한 아미노산 서열 분석을 통해 얻은 정보를 가지고 BLASTP 검색엔진으로 database를 검색한 결과 제한된 수의 단백질이 낮은 신뢰도 범위에서 동정되었다. 그렇지만 청국장과 같이 복잡한 단백질체를 분석하기에는 본 연구에서 고안한 전체 단백질 분리 기술과 2-D gel 분석적 접근은 단백질을 전체적으로 분석함에 있어 훌륭한 방법이다. 앞으로 청국장의 단백질 변화에 대한 연구는 의미 있는 변화를 보이는 소수의 단백질을 선발하고 이들에 대해 집중적으로 질량분석 하여 단백질을 동정하는 것이 필요하다.

  • Dung beetle larvae at the $3^{rd}$ instar were injected with lipopolysaccaride and inducible proteins were examined within a pI level of 3-10 and a size level by proteomics, including 1-D SDS PAGE analysis and antibacterial assay. The immune infected larvae extracts provided seven protein bands in one-dimensional electrophoresis and its antibacterial activity also checked. Hemolymph protein from immune infected larvae of the dung beetle were separated by twodimensional gel electrophoresis and compared with those from native larvae. In 2-D gel electrophoresis, we detected 63 immune infected unique and 32 up-regulated proteins, and 36 proteins that were down-regulated or not present in treated gel. Ten protein spots from unique proteins and those presented as different level of abundance in infected and native larvae were specially expressed. These differentially expressed proteins were proposed to be involved in the defense mechanism against microorganism.

Proteomic Analysis of Fructophilic Properties of Osmotolerant Candida magnoliae

  • Candida magnoliae, an osmotolerant and erythritol producing yeast, prefers D-fructose to D-glucose as carbon sources. For the investigation of the fructophilic characteristics with respect to sugar transportation, a sequential extraction method using various detergents and ultracentrifugation was developed to isolate cellular membrane proteins in C. magnoliae. Immunoblot analysis with the Pma1 antibody and two-dimensional electrophoresis analysis coupled with MS showed that the fraction II was enriched with membrane proteins. Eighteen proteins out of 36 spots were identified as membrane or membrane-associated proteins involved in sugar uptake, stress response, carbon metabolism, and so on. Among them, three proteins were significantly upregulated under the fructose supplying conditions. The hexose transporter was highly homologous to Ght6p in Schizosaccharomyces pombe, which was known as a predominant transporter for the fructose uptake of S. pombe because it exhibited higher affinity to D-fructose than D-glucose. The physicochemical properties of the ATP-binding cassette transporter and inorganic transporter explained their direct or indirect associations with the fructophilic behavior of C. magnoliae. The identification and characterization of membrane proteins involved in sugar uptake might contribute to the elucidation of the selective utilization of fructose to glucose by C. magnoliae at a molecular level.

Proteome Analysis of Bovine Longissimus dorsi Muscle Associated with the Marbling Score

  • The breeding value of marbling score in skeletal muscle is an important factor for evaluating beef quality. In the present study, we investigated proteins associated with the breeding value of the marbling score for bovine sirloin to select potential biomarkers to improve meat quality through comparative proteomic analysis. Proteins isolated from muscle were separated by two-dimensional gel electrophoresis. After analyzing images of the stained gel, seven protein spots for the high marbling score group were identified corresponding to changes in expression that were at least two-fold compared to the low marbling score group. Four spots with increased intensities in the high marbling score group were identified as phosphoglycerate kinase 1, triosephophate isomerase, acidic ribosomal phosphoprotein PO, and capping protein (actin filament) Z-line alpha 2. Spots with decreased intensities in the high marbling score group compared to the low score group were identified as 14-3-3 epsilon, carbonic anhydrase II, and myosin light chain 1. Expression of myosin light chain 1 and carbonic anhydrase 2 was confirmed by Western blotting. Taken together, these data could help improve the economic performance of cattle and provide useful information about the underlying the function of bovine skeletal muscle.

  • In Vietnam, Celastrus hindsii Benth, a medicinal plant rich in secondary metabolites, has been used to alleviate distress caused by ulcers, tumors, and inflammation for generations. The occurrence of two phenotypes, Broad Leaf (BL) and Narrow Leaf (NL), has raised questions about the selection of appropriate varieties for conservation and crop improvement to enhance medicinal properties. This study examined molecular differences in C. hindsii by comparing protein profiles between the NL and BL types using 2D-PAGE and MS. Peptide sequences and proteins were identified by matching MS data against the MSPnr100 databases and verified using the MultiIdent tool on ExPASy and the Blast2GO software. Our results revealed notable variations in protein abundance between the NL and BL proteomes. Selected proteins were confidently identified from 12 protein spots, thereby highlighting the molecular variation between NL and BL proteomes. Upregulated proteins in BL were found to be associated with flavonoid and amino acid biosynthesis as well as nuclease metabolism, which probably attributed to the intraspecific variations. Several bioactive proteins identified in this study can have applications in cancer therapeutics. Therefore, the BL phenotype characterized by healthier external morphological features has higher levels of bioactive compounds and could be better suited for medicinal use.

무기비소에 의한 마우스 간의 단백질 발현 조절 : 단백체 분석 (Regulation of Protein Expression in Mouse Liver by Inorganic Arsenic: Proteomic Analysis)

  • Background: Inorganic arsenic is a human carcinogen that can target the liver, but its carcinogenic mechanisms are still unknown. Inorganic arsenic induces a spectrum of tumors including hepatocellular carcinoma in mice. Methods: Pregnant C3H mice were supplied with drinking water containing 50 ppm sodium arsenite during their pregnancy. The protein expression profile in the liver of 0.5-day-old. male offsprings exposed transplacentally to sodium arsenite was analyzed using protein 2D gel electrophoresis followed by mass spectrometry (MALDI-TOF). Results: Expression of proteins such as hydroxymethylglutaryl-CoA synthase mitochondrial precursor (HMG-CoA synthase), ${\beta}$-actin (cytoplasmic 1) and apolipoprotein A-IV precursor (Apo-AIV) were induced in mouse liver by sodium arsenite, while uricase (urate oxidase), guanine nucleotidebinding protein beta subunit 2-like 1 (RACK1) and fructose-bisphosphate aldolase B (Aldolase 2) were down-regulated. Summary: Expression of proteins that have been implicated in carcinogenesis, such as HMG-CoA, ${\beta}$-actin, and RACK1, was regulated in the liver of mice transplacentally exposed to inorganic arsenic.

Analysis of Differentially Expressed Proteins in Bovine Longissimus Dorsi and Biceps Femoris Muscles

  • Skeletal muscle contains slow and fast twitch fibers. These skeletal muscle fibers express type I and type II myosin, respectively, and these myosin isoenzymes have different ATPase activity. The aim of this study was to investigate protein profiles of bovine skeletal muscles by proteomic analysis. Fifty seven spots of distinct proteins were excised and characterized. The expression of sixteen spots was differed in longissimus dorsi muscle with a minimal 2-fold change compared to biceps femoris muscle. The majority of differentially expressed proteins belonged to metabolic regulation-related proteins such as glyceraldehyde 3-phosphate dehydrogenase, triosephosphate isomerase and carbonic anhydrase 3. The real time-PCR assay confirmed an increase or induction of specific genes: RGS12TS isoform, GAPDH, triosephosphate isomerase and carbonic anhydrase. These results suggest that the expression of metabolic proteins is under a specific control system in different bovine skeletal muscle. These observations could have significant implications for understanding the physiological regulation of bovine skeletal muscles.

Proteomic Analysis of Protein Expression in Streptococcus pneumoniae in Response to Temperature Shift

  • From its initial colonization to causation of disease, Streptococcus pneumoniae has evolved strategies to cope with a number of stressful in vivo environmental conditions. In order to analyze a global view of this organism's response to heat shock, we established a 2-D electrophoresis proteome map of the S. pneumoniae D39 soluble proteins under in vitro culture conditions and performed the comparative proteome analysis to a 37 to $42^{\circ}C$ temperature up-shift in S. pneumoniae. When the temperature of an exponentially growing S. pneumoniae D39 culture was raised to $42^{\circ}C$, the expression level of 25 proteins showed changes when compared to the control. Among these 25 proteins, 12 were identified by MALDI-TOF and LC-coupled ESI MS/MS. The identified proteins were shown to be involved in the general stress response, energy metabolism, nucleotide biosynthesis pathways, and purine metabolism. These results provide clues for understanding the mechanism of adaptation to heat shock by S. pneumoniae and may facilitate the assessment of a possible role for these proteins in the physiology and pathogenesis of this pathogen.

Streptococcus pneumonia 감염으로 변화한 사람 폐 상피세포 단백질의 프로테오믹 분석 (Proteomic Analysis of Protein Changes in Human Lung Cancer Epithelial Cells Following Streptococcus pneumoniae Infection)

  • Streptococcus pneumoniae는 전 세계적으로 급성 호흡기 질환 높은 사망률 나타내며, 정상인의 비인후부에 존재하여 호흡기 감염을 통해 폐렴, 수막염, 중이염, 패혈증, 복막염, 골수염 등을 일으킨다. 그러나 S. pneumoniae가 폐 조직에 침입하는 분자적 메커니즘과 혈류를 통한 침입은 많은 연구에도 불구하고 아직 명확하게 알려지지 않았다. 그러므로 본 실험에서는 S. pneumoniae D39의 감염 및 침입에 대한 분자 메카니즘을 알고자 사람의 폐암상피 세포 유래 A549 세포를 이용하여 감염 후 시간의 경과에 따라 변화되는 A549 세포의 모양을 관찰하였으며, 또한 숙주세포의 단백질 패턴 변화를 조사하였다. 일부 A549 세포는 감염 후 2 시간부터 세포의 모양이 둥근형태로 변화된 것으로 관찰되었으며, 감염 3 시간째에는 세포의 모양이 둥글며 filopodia가 아주 잘 발달하였다. 감염 4 시간에 도달하게 되면 거의 모든 A549 세포가 둥글며 잘 발달된 filopodia를 형성하였다. 감염 후 각 시간 별 A549 세포의 총 단백질들을 추출하여 시간의 경과에 따라 특이적으로 양 적인 변화를 나타내는 단백질을 MALDI-TOF 분석법을 사용하여 동정하였다. Streptococcus pneumoniae D39 감염 후 시간에 따라 변화하는 단백질 중 대다수가 특이하게도 molecular chaperone에 속하는 단백질들이었다. 대표적인 cytosol chaperone인 Hsp90과 Hsp70의 경우 감소하는 패턴을 나타낸 반면에 endoplasmic reticulum (ER)에 존재하는 chaperone인 Grp94와 Grp78 (BiP)은 감염 후 점차 증가하는 패턴을 나타내었다. ER chaperone인 Grp94와 Grp78의 증가는 ER stress signaling pathway와 관련 있는 것으로 알려져 있어, S. pneumoniae D39의 감염에 의한 이들 단백질의 변화 패턴을 ER stress를 유발 시켰을 때와 비교하였다. Tunicamycin 또는 thapsigargin으로 처리하여 ER stress를 유발시킨 A549 세포의 형태는 변화하지 않았으며 흡착세포의 형태를 유지하였다. 그러나 Western blot을 통한 molecular chaperone의 분석 결과는 S. pneumoniae D39 감염의 경우와 일치하였다. 본 연구에서 얻은 결과는 S. pneumoniae D39의 감염은 A549 세포의 형태적 변화를 유발하며 또한 molecular chaperone 증가와 감소를 유발한다는 것을 보여주며, 특이적으로 Grp94와 Grp78이 증가되는 것으로 보아 S. pneumoniae D39 감염은 A549 세포 내 ER stress를 유발한다고 생각된다.
