Relationship Among Nutritional Intake, Duration of Outdoor Activities, Vitamin D Status and Bone Health in High School Girls

  • This study investigated the interactions of bone health with several variables such as outdoor activity hours, nutritional status including habitual intake of calcium andvitamin D status in 72 high school girls aged 16-17 yearsattending day classes or night classes. The subjects consisted of 39 day-class students and 33 night-class students. Dietaty nutrient intakes were estimated using the 24-hour recall method. The daily activities of each subject were assessed using an activity questionnaire. Urinary calcium and creatinine excretion were assayed from subjects' 24-hoururine, while 25-OH-vitamin D[25-(OH)-D] and osteocalcin were measured from the subjects' fasting blood. Intake of energy, iron, vitamin A and vitamin C were worse in the night-class students. There was no significant difference in dietary calcium between the subjects in the different class types. Time spent on outdoor activities was significantly less in subjects attending night classes. Urinary calcium excretion of the night-class subjects was significantly higher than that of the day-class subjects (p<0.05). There was no significant difference in serum 25-(OH)-D level according to class type. Serum osteocalcin for night-class subjects was significantly higher than that for day-class subjects (p<0.01). It appeared that the night-class students had poorer dietary habits as well as fewer outdoor activities. Even though the estimated bone health of both groups of subjects appeared to be normal, the overall nutritional intake and duration of outdoor activities appeared to be important for maintaining bone health and lowering the future risk of osteoporosis.

Muscle Mass Changes After Daily Consumption of Protein Mix Supplemented With Vitamin D in Adults Over 50 Years of Age: Subgroup Analysis According to the Serum 25(OH)D Levels of a Randomized Controlled Trial

  • Early prevention of sarcopenia can be an important strategy for muscle maintenance, but most studies target subjects at slightly pre-sarcopenic state. Our previous paper describes the effect of protein supplements rich in leucine and vitamin D on muscle condition, and in this paper, we performed a sub-analysis to evaluate who benefitted the most in terms of improvement in muscle health. A 12-week randomized clinical trial of 120 healthy adults (aged 50 to 80) assigned to an intervention group (n = 60) or control group (n = 60) were analyzed. Subjects in the intervention group received, twice per day, a protein supplement containing (per serving) 800 IU of vitamin D, 20 g of protein (3 g of total leucine), 300 mg of calcium, 1.1 g of fat, and 2.5 g of carbohydrate. The subjects were classified into 'insufficient' and 'sufficient' groups at 25-hydroxyvitamin D (25[OH]D) value of 30 ng/mL. The skeletal muscle mass index normalized to the square of the skeletal muscle mass (SMM) height (kg/m2) increased significantly in the 'insufficient group' difference value of change between weeks 0 and 12 (Δ1.07 ± 2.20; p = 0.037). The SMM normalized by body weight (kg/kg, %) was higher, but not significantly, in the insufficient group (Δ0.38 ± 0.69; p = 0.050). For people with insufficient (serum 25[OH]D), supplemental intake of protein and vitamin D, calcium, and leucine and adequate energy intake increases muscle mass in middle-aged and older adults and would be likely to exert a beneficial effect on muscle health.

Effect of UV-B irradiated vitamin D enriched yeast supplementation on milk performance and blood chemical profiles in dairy cows

  • Objective: The objective was to evaluate the effects of UV-B irradiated vitamin D-enriched yeast supplementation on milk yield, milk composition, vitamin D in milk, milk fatty acids, blood chemistry, and 25(OH)D status in dairy cows. Methods: Six Thai Friesian cows (milk production, 11.2±2.0 kg/d; body weight, 415.0±20.0 kg; and days in milk, 90.0±6.0) were allocated to each treatment in a 3×3 Latin square design, with three treatments and three periods. Each period of the Latin square lasted 49 days consisting of 14 days for diet adaptation and 35 days for sample collection. Dairy cows were randomly assigned to one of three treatments: i) feeding a basal diet without yeast (CON); ii) basal diet + 5 g of live yeast (75 IU/head/d of vitamin D2; LY); and iii) basal diet + 5 g of UV-B irradiated vitamin D enriched yeast (150,000 IU/head/d of vitamin D2; VDY). Feed intake and milk production were recorded daily, milk sample collection occurred on days 14 and 35 of each collection period, and blood plasma was collected on days 0, 7, 14, 21, 28, and 35 of each collection period. Results: The results show that after a trial period of 14 and 35 days, the VDY group had significantly higher vitamin D content in milk than the LY and CON groups (376.41 vs 305.15, 302.14 ng/L and 413.46 vs 306.76, 301.12 ng/L, respectively). At days 7, 14, 21, 28, and 35 of the experiment, cows fed the VDY group had significantly higher 25(OH)D2 status in blood than the CON and LY groups (51.07 vs 47.16, 48.05 ng/mL; 54.96 vs 45.43, 46.91 ng/mL; 56.16 vs 46.87, 47.16 ng/mL; 60.67 vs 44.39, 46.17 ng/mL and 63.91 vs 45.88, 46.88 ng/mL), respectively. Conclusion: In conclusion, UV-B irradiated vitamin D-enriched yeast supplementation could improve vitamin D content in the milk and 25(OH)D status in dairy cows during the lactation period.

대학 연구활동종사자의 식생활실태, 비타민 D 영양상태 및 혈액 임상지표 분석 (Dietary Life, Vitamin D Status and Blood Clinical Indices of University Laboratory Workers)

  • Objectives: Although the number of laboratory workers is constantly increasing every year, few studies have been conducted on the health and nutritional status of these research workers. This study determined the health status of laboratory workers by analyzing their anthropometric indices, dietary life, vitamin D status and blood clinical indices. Methods: The subjects consisted of 100 female laboratory workers. This study investigated their diet, anthropometric indices, vitamin D status and blood clinical indices. The subjects were divided into two groups according to their duration of working in a laboratory (<1 year, $${\geq}_-1year$$). Results: The average age and body mass index (BMI) of subjects were 23.18 years and $21.51kg/m^2$, respectively Those subjects with over 1 year employment ($${\geq}_-1year$$) had a significantly higher waist-hip ratio than that of the subjects with the less than 1 year employment (<1 year). The mean serum vitamin D level of all the subjects was 10.04 ng/mL, which is close to a level of vitamin D deficiency. There was a significantly higher average intake of calories in the over 1 year employment group as compared to that of the less than 1 year employment group. The frequency of eating sweet snacks was significantly higher for the over 1 year employment group. The correlation analysis showed a significant positive correlation between the serum 25-(OH)-vitamin D level and the time of exposure to sunlight, while dietary intake of vitamin D did not show correlation with the serum 25-(OH)-vitamin D level. However, the serum 25-(OH)-vitamin D level was also negatively correlated with both the percentage of body fat and visceral fat. Conclusions: Laboratory workers are a very high risk group in terms of their nutritional status of vitamin D. Therefore, they need greater time of exposure to sunlight as well as increasing their dietary consumption of vitamin D. In addition, it is important for laboratory worker to practice regular and balanced dietary habits in order to maintain a healthy life.

Inhibition of Vitamin D Receptor Translocation by Cigarette Smoking Extracts

  • Background: Vitamin D can translocate a vitamin D receptor (VDR) from the nucleus to the cell membranes. The meaning of this translocation is not elucidated in terms of a role in pathogenesis of chronic obstructive pulmonary disease (COPD) till now. VDR deficient mice are prone to develop emphysema, suggesting that abnormal function of VDR might influence a generation of COPD. The blood levels of vitamin D have known to be well correlated with that of lung function in patients with COPD, and smoking is the most important risk factor in development of COPD. This study was performed to investigate whether cigarette smoke extracts (CSE) can inhibit the translocation of VDR and whether mitogen activated protein kinases (MAPKs) are involved in this inhibition. Methods: Human alveolar basal epithelial cell line (A549) was used in this study. 1,25-$(OH_2)D_3$ and/or MAPKs inhibitors and antioxidants were pre-incubated before stimulation with 10% CSE, and then nucleus and microsomal proteins were extracted for a Western blot of VDR. Results: Five minutes treatment of 1,25-(OH2)D3 induced translocation of VDR from nucleus to microsomes by a dose-dependent manner. CSE inhibited 1,25-$(OH_2)D_3$-induced translocation of VDR in both concentrations of 10% and 20%. All MAPKs inhibitors did not suppress the inhibitory effects of CSE on the 1,25-$(OH_2)D_3$-induced translocation of VDR. Quercetin suppressed the inhibitory effects of CSE on the 1,25-$(OH_2)D_3$-induced translocation of VDR, but not in n-acetylcysteine. Conclusion: CSE has an ability to inhibit vitamin D-induced VDR translocation, but MAPKs are not involved in this inhibition.

  • This study describes the effects of novel1, 25-dihydroxyvitamin D$_3$ analong[1,25(OH)$_2$-16ene-23yne-26, 27-F6-D$_3$] on proliferation of the human histiocytic lymphoma cell line U937 in vitro. We also examined the expression of c-myc oncogene in U937 cells was apparently inhibited to 62% and 87% of the control level after 4 days in the presence of 10-8M and 10-7 M of this analog, respectively. This compound morpholgically and functionally differentiated U937 cells to nonocyte-macrophage phenotype showing the increase of adherence ability to surface and a decrease of N/C ratio in Giemsa staining . Especially, nonspecific esterase activity which is a marker of cell differentiation to monocyte-macrophage was positive, and production of the positive stained cells increased in a dose dependent fashion . The expression of c-myc oncogene by 1, 25(OH)$_2$D$_3$ analog(10-7 M) was reduced by 60% at the mRNA level as determined by Northern blotting. The effects of this novel analog on cell proliferation and cell differentiation may open op new therapeutic strategies for human disorders such as psoriassis and may provide a tool to understand the mechanism of action of vitamin D$_3$ seco-steroids in malignancy.

Levels of Serum 25-Hydroxy-Vitamin D in Benign and Malignant Breast Masses

  • Background: The true association between breast cancer and vitamin D is currently under investigation. We compared serum 25-hydroxy-vitamin D levels in women with benign and malignant breast masses and controls. Materials and Methods: Levels of vitamin D were measured by electrochemiluminescense. Serum levels >35 ng/ml, 25-35 ng/ml, 12.5-25 ng/ml and <12.5 ng/ml were considered as normal, mild, moderate and severe vitamin D deficiency, respectively. Results: Overall, 364 women were included in the control, 172 in the benign and 136 in the malignant groups. The median serum vitamin D level was significantly lower in breast cancers than controls. Levels were also lower in malignant than benign cases and in benign cases than controls although statistically non-significant. Conclusions: Multinomial logistic regression analysis showed that severe vitamin D deficiency causes a three-fold increase in the risk of breast cancer while this was not the case for moderate and mild deficiency.

  • 비타민은 우리 몸에서 많은 양을 필요로 하지는 않지만 없어서는 안 될 필수 영양성분으로 음식물과 태양광등을 통하여 획득하고 그 농도를 유지하고 있다. 그러나 때로는 어떤 요인에 의하여 농도가 변하는 경우가 있는데 그중 간기능 인자들과 $25OH-VIT.D_3$ 농도값의 영향요인을 분석하고자 실시하였다. 연구 대상은 2012년 8월부터 2012년 12월까지 모대학병원 검진센타에 건강검진을 받은 검체를 대상으로 하여 실험군 검체와 대조군 정상검체 두 군으로 분류한 후 성별, 연령, 비만도, AST, ALT, rGTP와 $25OH-VIT.D_3$의 상관성을 조사하였다. 분석결과로 $25OH-VIT.D_3$농도에 영향을 미치는 요인으로 선정된 변수는 연령과 B형간염역가로 나타났으며 특히 B형간염역가는 역 상관관계를 나타내었다($R^2$=0.40). 이는 간염바이러스에 의한 간기능에 의해 그 농도값에 영향을 줄 수 있으므로 간염 보균자인 경우 간기능 검사와 더불어 $25OH-VIT.D_3$의 영향인자로 관리 되어져야 한다고 사려 된다.

Effects of an in vitro vitamin D treatment on the inflammatory responses in visceral adipose tissue from Ldlr-/- mice

  • BACKGROUND/OBJECTIVES: Atherosclerosis is associated with increased inflammation in the visceral adipose tissue (VAT). Vitamin D has been reported to modulate the inflammatory responses of stromal vascular cells (SVCs) and adipocytes in adipose tissue, but the role of vitamin D in atherosclerosis biology is unclear. This study examined the effects of in vitro 1,25-dihydroxyvitamin D3 (1,25[OH]2D3) treatment on the inflammatory responses of SVCs and adipocytes from atherosclerotic mice. MATERIALS/METHODS: C57BL/6J (B6) mice were divided randomly into 2 groups and fed a 10% kcal fat control diet (control group, CON) or 41% kcal fat, 0.21% cholesterol (high fat + cholesterol, HFC) diet (obese group, OB), and B6.129S7-Ldlrtm1Her/J (Ldlr-/-) mice were fed a HFC diet (obese with atherosclerosis group, OBA) for 16 weeks. SVCs and adipocytes isolated from VAT were pre-incubated with 1,25(OH)2D3 for 24 h and stimulated with lipopolysaccarides for the next 24 h. Proinflammatory cytokine production by adipocytes and SVCs, the immune cell population in SVCs, and the expression of the genes involved in the inflammatory signaling pathway in SVCs were determined. RESULTS: The numbers of total macrophages and SVCs per mouse were higher in OB and OBA groups than the CON group. The in vitro 1,25(OH)2D3 treatment significantly reduced macrophages/SVCs (%) in the OBA group. Consistent with this change, the production of interleukin-6 and monocyte chemoattractant protein 1 (MCP-1) by SVCs from the OBA group was decreased by 1,25(OH)2D3 treatment. The 1,25(OH)2D3 treatment significantly reduced the toll-like receptor 4 and dual-specificity protein phosphatase 1 (also known as mitogen-activated protein kinase phosphatase 1) mRNA levels in SVCs and MCP-1 production by adipocytes from all 3 groups. CONCLUSIONS: These findings suggest that vitamin D can attribute to the inhibition of the inflammatory response in VAT from atherosclerotic mice by reducing proinflammatory cytokine production.

치주인대세포의 증식 및 세포활성에 미치는 $1,25-(OH)_2D_3$의 영향에 관한 연구 (THE EFFECT OF $1,25-(OH)_2D_3$ ON THE PROLIFERATION AND ALKALINE PHOSPHATASE ACTIVITY OF HUMAN PERIODONTAL LIGAMENT CELLS)

  • [ $1,25-(OH)_2D_3$ ]가 치주인대세포의 증식에 미치는 영향과 조골세포 기능을 나타내는 지표이며 골의 석회화 과정에 관여하는 alkaline phosphatase의 활성도에 미치는 영향을 관찰하여 아직 확실히 밝혀져 있지 않은 치주인대조직에 대한 $1,25-(OH)_2D_3$의 생물학적 기능을 알아보고자 교정 치료 목적으로 발거된 치아로부터 치주인대세포를 배양하였다. $1,25-(OH)_2D_3$를 첨가하여 24시간 후 [${^3}H$]thymidine으로 DNA를 표지하여 세포의 증식을 관찰하였으며 $1,25-(OH)_2D_3$가 치주인대세포에서 조골세포의 표지효소인 alkaline phosphatase의 활성도에 미치는 영향을 24시간 또는 6일간 $1,25-(OH)_2D_3$ 처리후 측정한 결과 100nM농도의 $1,25-(OH)_2D_3$은 치주인대세포의 증식을 유의하게 증가시켰으며 10nM에서는 차이를 보이지 않았다. $1,25-(OH)_2D_3$을 24시간 첨가시 10nM에서는 ALP활성도는 $80.8\pm31.4$nmol로 서 대조군 $38.5\pm5.3$nmol에 비해 유의하게 증가하였으며 또한 $1,25-(OH)_2D_3$을 6일동안 첨가하였을때에도 10nM에서 $106.7\pm23.0$nmol로 대조군 $29.3\pm1.0$nmol에 비해 유의하게 증가되었다. 이상의 결과를 종합하면 $1,25-(OH)_2D_3$이 치주인대세포의 증식 및 세포기능을 시사하는 결과로 사료된다.
