DNA Probes에 의한 토양의 이사디 (2,4-D) 분해세균의 검출 (Application of DNA Probe Method for Detection of 2,4-Dichlorophenoxyacetic Acid Degrading Bacteria in Soil)

  • 토양에서 세균군집의 DNA를 추출하여 이사디 분해세균의 밀도와 군집변화를 tdfA 유전자와 Spa Probe를 이용하여 조사하였다. 이사디 분해균주인 Pseudomonas cepacia/pJP4을 토양에 여러 가지 밀도로 접종한 후 추출된 토양세균군집의 DNA를 Southern blot에서 분석한 결과, 본 실험에 사용된 DNA probe method에 의해 이 세균을 $10^5\;cells/g$ soil 수준까지 검출할 수 있는 것으로 나타났다. 이사디를 가해준 microcosm 토양에서 추출된 세균군집의 DNA를 분석한 실험에서는 Pseudemonas pickettii와 Sphingomonas Paucimobilis가 우점종으로 검출되었고, 사용된 두 가지의 DNA probes는 토양의 이사디 분해미생물에 대해 매우 높은 특이성을 가지고 있는 것으로 나타났다. 밭에 이사디를 장기 적으로 가해준 후 추출된 토양세균군집의 DNA를 분석 한 실험에서는 이사디를 최소한 10 ppm 이상 가해주어야 토양의 이사디 분해세균을 DNA probe method에 의해 검출할 수 있었고, tfdA 유전자는 실제의 밭토양에서도 높은 특이성을 나타냈으나 Spa probe는 일부의 토착세균에 비특이적으로 반응하는 것으로 나타났다. 토양에서 추출된 세균군집의 DNA를 분석하는 DNA probe method는 Southern blot과 함께 사용되었을 때 토양에 존재하는 이사디 분해미생물을 실험실 배지에 배양하지 않고 검출할 수 있었고,이 미생물들의 밀도, 군집변화, 유전적 변화 등을 효과적으로 분석할 수 있는 것으로 나타났다.

전기비저항을 이용한 금속합금 열화도 평가기술 (Evaluation Technology of Degradation of Metallic Alloy using Electrical Resistivity)

  • 재료의 노화에 의한 강도 및 인성 감소치를 정량적으로 평가하는 비파괴적 기법의 개발이 시도되고 있으며 전기비저항법도 그 중의 하나이다. 본 연구에서는 전기비저항법의 열화도 평가에의 적용성을 살펴보기 위해 먼저 6종 10 가지의 비자성 금속을 측정시료로 선택하여 직류 two-point probe법(혹은 2탐침법)으로 전기비저항을 계산한 결과와 비접촉식 와전류 방법으로 전기비저항을 측정한 결과를 서로 비교하였다. 또한, 1Cr-1Mo-0.25V강에 대하여 비파괴적 측정 방법인 four-point probe 방법(혹은 4탐침법)을 사용하여 얻은 결과와 기존의 2탐침법을 사용하여 얻은 결과를 서로 비교하여 현장에서의 4탐침 기술의 적용 가능성을 살펴보았다. 1Cr-1Mo-0.25V강에 대하여 2탐침법으로 구한 비저항 값과 4탐침법으로 구한 비저항 측정값의 차이는 0.6%였다. 따라서 4탐침법을 사용하여 현장에서 금속소재의 열화도의 비파괴적 평가가 가능하다고 사료된다.

  • Portable sheet resistance-measuring instrument using the dual-configuration Four-Point Probe method is developed for the purpose of precisely measuring the sheet resistance of conducting thin films. While single-configuration Four-Point Probe method has disadvantages of applying sample size, shape and thickness corrections for a probe spacing, the developed instrument has advantages of no such corrections, little edge effects and measuring simply and accurately the sheet resistance between $0.2\Omega/sq$ and $2k\Omega/sq$.

  • The theoretical model for the transient probe method is the modified Jaeger model which is used perfect line source theory. The transient probe technique has been developed for the simultaneous determination of thermal conductivity, diffusivity and volumetric heat capacity of liquids. The Levenberg-Marquardt iteration method is adapted to obtain thermal property within nonlinear range. Experimental results of liquids were found to agree well with recommended thermal property data.

  • We estimated the critical current distribution of the long length CCs using hall probe method, and checked the reliability by comparing with the four-probe dc transport method. The deviation ratio of $I_{c-T}$ (a measured critical current by four probe dc transport method) and $I_{c-H}$ (calculated critical current by formula (3).) was nearly smaller than 12%. The Hall probe method appeared powerful to evaluate the critical current of CCs in a reel to reel system.

3차원 음향 인텐시티 프로브의 고주파 영역 감도 보상 연구 (A Study on the Sensitivity Compensation of Three-dimensional Acoustic Intensity Probe in the Higher Frequency Range)

  • 본 논문에서는 3차원 음향 인텐시티 프로브의 고주파 영역에서 발생하는 감도 저하 현상을 보정하는 방법에 대해 연구하였다. 고주파 영역에서 측정오차는 음향 인텐시티 프로브를 구성하는 마이크로폰으로 수음한 신호들의 위상차로 인하여 발생한다. 이 오차는 마이크로폰 사이의 간격보다 측정대상 음향신호의 파장일 작을 경우, 즉 신호의 주파수가 높을수록 크게 발생한다. 이 연구에서는 두개의 마이크로폰으로 구성된 1차원 프로브의 보정법들을 제안하여 그 유효성을 컴퓨터 수치계산으로 검토하였다. 음향 인텐시티 프로브를 구성하는 마이크로폰 사이의 위상차를 음원의 추정된 방향으로 보정하는 방법이 가장 유용하였으며, 이를 3차원 프로브에 적용하여 임의의 방향으로 전파하는 음원에 대해 컴퓨터 수치계산으로 검증하였다. 그 결과, 4개의 마이크로폰을 60mm 간격으로 구성한 3차원 프로브로 1dB 이하의 정밀한 측정이 가능한 주파수 범위가 약 1.2kHz 이였던 것을 제안한 보정방법을 적용한 후, 약 2.8kHz까지 감도가 향상됨이 확인되어 제안한 보정법의 유효성이 증명되었다.

  • This paper deals with an inexpensive, simple and fast Langmuir probe sweeping circuit and its application. This sweeper completes a probe trace in a 1 ms order. Futhermore, the circuit drives a maximum probe voltage of $\pm$30V and has a maximum probe current capability of a few amperes. The plasma parameters are successfully determined using the fast Langmuir probe method.

  • We developed a 2-channel fiber-optic temperature sensor (FOTS) using a temperature sensing probe, a fiber-optic coupler, transmitting optical fiber, and an optical time domain reflectometer (OTDR). The temperature sensing probe is divided into a sensing probe and a reference probe for accurate thermometry. A sensing probe is composed of a silicon oil, a FC terminator, a brass pipe, and a singlemode optical fiber and the structure of a reference probe is identical with that of the sensing probe excluding a silicon oil. In this study, we measured the modified optical powers of the light signals reflected from the temperature sensing probe placed inside of the water with a thermal variation from 5 to $70^{\circ}C$. Although the optical power of the reference probe was constant regardless of the temperature change, the optical power of the sensing probe decreased linearly as the temperature increased. As experimental results, the FOTS using a subtraction method showed a small difference (i.e., hysteresis) in its response due to heating and cooling. The reversibility and reproducibility of the FOTS were also evaluated.

Machining Center의 2차원 원호보간정밀도 진단 System의 개발 (A development of accuracy diagnostic system 2-dimensional circular interpolation of machining centers)

  • The paper describes and alternative method based on a new idea to measure the circular movement of machining centers. ISO has employed three testing methods for the acceptance tests of machine tools; the first is a rotating one-dimensional probe method, the second is a two-dimensional probe and a master circular ring, and the third is a kinematic ball bar. The last two methods were proposed and introduced by W. Knapp and J. B. Bryan, respectively. The newly developed method is superior to above two methods; the rotating angle can be detected and the rotating radius is variable. Circular movement errors of machining centers were investigated by the analysis of data measured by R- .THETA. method. Followint observations are obtained 1) The errors which depend on positions, i.e., periodical errors by the pitch of ball screws, errors by compensation of backlash and errors by perpendicularity of X and Y-axis, were analyzed. 2) The errors which depend on NC control system, i.e., errors by the unbalance of position-loop-gaians, errors by velocity-loop-gains and errors by feed speeds, were quantiatively analyzed. 3) The method of extracting error information, which uses moving technique of averaging angle and fourier's analysis data mesured by the R- .THETA. method, was proposed.

감염 근관에서 혐기성 배양법과 간접 면역 형광법 및 DNA 프로브법에 의한 Porphyromonas endodontalis의 검출에 관한 비교 연구 (COMPARATIVE STUDY ON THE DETECTION OF PORPHYROMONAS ENDODONTALIS BY ANAEROBIC CULTURE, IIF AND DNA PROBE METHOD IN INFECTED ROOT CANALS)

  • There are many advantages when using IIF and DNA probe methods over anaerobic culture method in that they are time-and effort-saving, more precise and more sensitive. Furthermore, in IIF and DNA probe methods, the detection is possible only with small amount of bacteria, the quantitative analysis is possible, and the cell viability is not necessary. The purpose of this study is to observe the incidence of P.endodontalis by carrying out anaerobic culture, IIF and colony lift using DNA probe method respectively, and to compare these 3 methods in terms of effectiveness and sensitivity in order to identify the most effective detection method. 30 teeth with at least one clinical symptoms, with single canal, and with pulp necrosis were sampled. For sampling bacteria, access cavity was prepared after disinfecting tooth and its surroundings. Then the paper point was inserted up to the periapical area, leave there for a while, and finally it was placed into PRAS Ringer's sol. and PBS sol. In anaerobic culture method, P.endodontalis was identified by biochemical tests after subculturing black and brown colonies which were produced after 7 days of incubation on BAP and Brucella BAP in anaerobic chamber. To identify P.endodontalis in IIF method, species-specific polyclonal rabbit-antisera of P.endodontalis(ATCC 35406) was reacted with sampled PBS sol. dispensed onto glass slide, and then P.endodontalis was examined by phase contrast microscopy after incubating with Goat anti-rabbit lgG conjugated to Fluorescein isothiocyanate. For colony lift using DNA probe method, membranes were laid over colonies on the surface of BAP and were hybridized with cloned DNA probe of P.endodontalis. The existence of P.endodontalis was then identified by the methods of chemiluminescent detection and color metric detection. Black colony was found in 11 teeth out of 30 teeth and P.endodontalis was detected in 6 teeth (20 %) by anaerobic culture method, 16 teeth (53 %) by IIF method, and 7 teeth (23 %) by DNA probe method. IIF method is significantly better in detecting P.endodontalis than DNA probe method and anaerobic culture method. There was no significant differences between DNA probe method and anaerobic culture method. There was significant correlation between the formation of black colony and the existence of P.endodontalis. The probability of detecting P.endodontalis when black colony being present is 2.89 times higher than when not being present. There was significant relationship between the foul odor of clinical symptoms and P.endodontalis. The sensitivity of existing P.endodontalis when foul odor being present was 93.75 %, while the specificity of not existing P.endodontalis when foul odor not being present was 28.57 %. These results suggested that the probes of P.endodontalis will be used to decide the method and prognosis in endodontic treatments.
