Effect of Injin Butanol Fraction with Thin Layer Chromatography on Fas-mediated Apoptosis (인진butanol 분획의 TLC추출성분이 Fas-mediated Apoptosis에 미치는 영향)

  • Objective: The aim of this study is to investigate the effect of Injin butanol fractions with Thin Layer Chromatography on Fas-mediated Apoptosis. Method: Injin-butanol fraction separated by TLC. MIT assay, cell cycle analysis, Caspase-3 protease assay, DNA fragmentation assay and quantitative RT-PCR were performed to evaluate the effects of TLC extraction of lnjin-butanol fraction on cell viability, cell cycle progression and apoptosis. Results: Scopoletin, luteolin, apigenin and unknown powder was isolated by TLC. Fas-mediated apoptosis analysis shows that scopoletin has inhibiting function on apoptosis. Caspase- 3 protease assay analysis shows that scopoletin inhibits activity of caspase-3. Quantitative RT-PCR analysis shows that no activity on caspase-3, but apoptosis inhibition cytokine -Bcl-2- is activated, and apoptosis activating cytokine -Bax- is unactivated. Conclusion: These results show that each fraction of Injin-butanol TLC extraction, especially scopoletin, acts as a protective function on liver cell viability, and inhibitory function on apoptosis.   • This study measured the AITs of n-butanol + n-decane system from ignition delay time (time lag) by using ASTM E659 apparatus. The AITs of n-butanol and n-decane which constituted binary system were $340^{\circ}C$ and $212^{\circ}C$, respectively. The experimental AITs of n-butanol + n-decane system were a good agreement with the calculated AITs by the proposed equations with a few A.A.D. (average absolute deviation).

  • This study was carried out to develop antitumor effect of the n-butanol soluble fraction of Perilla frutescens on (KB cells) human oral epitheloid carcinoma cells. The cytotoxictty of methanollc extract of Perilla frutescens on KB cells was evaluated by 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide(MTT) assay. The antitumor activity of various fractions obtained from n-butanol soluble fraction of Perilla frutescens was evaluated in human oral epithelold carcinoma cells. The antitumor acavity of the n-butanol soluble fraction on human oral epitheloid carcinoma cells was evaluated by MTT assay of colorimetric method. The light microscopic study was carried out to observe morphological changes of cultured human oral epitheloid carcinoma cells. These results were obtained as follows; 1. The fractions 1,2 and 3 of the n-butanol soluble fraction of Perilla frutescens were shown significant antitumor activities. 2. The number of human oral epitheloid carcinoma cells were decreased and tend to form cell cluster by treatment with fractions 1,2,3 and 4 of the n-butanol soluble fraction of Perilla frutescens. 3. The fraction 1 of the n-butanol soluble fraction of Perllla frutescens showed the highest antitumor activity on Perilla frutescens. It has been selected as a lead fraction for further examinations.

Autoignition Phenomena of a Single Diesel/1-Butanol Mixture Droplet (디젤/1-부탄올 혼합연료 단일액적의 자발화 현상)

  • The goal of this study is to experimentally observe the autoignition phenomena of a diesel/1-butanol mixture droplet in ambient pressure and $700^{\circ}C$ condition. A volume ratio of 1-butanol in the fuel was set to 25, 50 and 75%. A single droplet was installed at the tip of fine thermocouple, and the electric furnace dropped down to make elevated temperature condition. Droplet behavior during the experiment could be divided into 3 stages including droplet heating, puffing and autoignition/combustion. Puffing process intensively observed for the case of 1-butanol volume ratio of 25 and 50%, but did not occur at 75%. Increase of 1-butanol volume ratio hindered rise of the droplet temperature and delayed ignition. In addition, puffing process also affected on autoignition, so the ignition delay of 1-butanol volume ratio of 50% was became longer than that of 75% case.

Studies of the actions of Aconiti tuber butanol fraction on the mechanical and electrical properties of isolated rabbit atrium (부자(附子) Butanol fraction의 강심작용(强心作用)에 관한 연구(硏究))

  • In Chinese medicine, it is said that Aconiti tuber has cardiotonic, diuretic and analgesic effects. Kim et al reported that alkaloid free part of Aconiti tuber, $CHCI_3$ insoluble fraction, showed inotropic effect on isolated frog heart and inotropic effect is potenciated by n-butanol fractionation. To investigate the effect of Aconiti tuber butanol fraction on the mechanical and electrical properties of heart, change of active tension, excitability and refractory period of isolated rabbit atrium in the presence of butanol fraction were measured and the comparison with that of ouabain and quinidine was done. The observed results are as follows. 1. $5{\times}10^{-4}g/ml$ concentration of Aconiti tuber butanol fraction showed approximately same effect with therapeutic concentration of ouabain on the increment of contractile force, and the effect of $2{\times}10^{-3}g/ml$ was greater than that of $1{\times}10^{-5}g/ml$ of ouabain. 2. Acceleration of rate of contractile force increment in the presence of Aconiti tuber butanol fraction was greater than in ouabain, and the time to maximum tension was shorter in Aconiti tuber butanol fraction than in ouabain. 3. The excitability of isolated atrium was slightly increased at low concentration of Aconiti tuber butanol fraction, while decreased at higher concentration. 4. Aconiti tuber butanol fraction slightly prolonged refractory period of isolated right atrium at the concentration of $2{\times}10^{-3}g/ml$.

  • Synthesis of butenes through dehydration of 2-butanol was investigated over HY zeolite catalysts. 2-Butanol dehydration reaction was carried out in a fixed bed catalytic reactor. 2-Butanol conversion was increased with increase of $Si/Al_2$ ratio of HY zeolite catalysts, which can be ascribed to increase of acid strength with increase of $Si/Al_2$ ratio. Selectivities to 1-butene, trans-2-butene, and cis-2-butene were not greatly influenced by the change of the $Si/Al_2$ ratio of HY zeolite. As a result, it was advantageous to use a HY zeolite catalyst with 60 $Si/Al_2$ ratio for maximizing the yield of 1-butene in the dehydration of 2-butanol. The optimal reaction temperature for maximizing the yield of 1-butene was $250^{\circ}C$ over HY (60) catalyst.

Effects of Chenopodium album Linne on Gastritis and Gastric Cancer Cell Growth

  • In our previous study, we investigated Chenopodium album Linne (CAL) ethanol extract and its fractions on anti-gastritic actions using the HCl/ethanol and indomethacin induced gastric lesion model and Helicobacter pylori (H. pylori). Based on the results, butanol fraction was most effective among fractions obtained from CAL. This study aims to elucidate the mechanisms of butanol fraction, and betaine as a constituent of the butanol fraction, on gastritis and anti-gastric cancer cell growth. First, we examined antioxidant properties using hydrogen peroxide and superoxide radical, and we found that butanol fraction and betaine may be good antioxidants. Second, cytotoxicity was assessed by measuring cell viability and 4,6-diamidino-2-phenylinodole dihydrochloride (DAPI) staining of human gastric cancer cells (AGS cells). We also examined the relationship between the cytotoxicity and intracellular $Ca^{2+}$ signaling mechanism. The butanol fraction demonstrated cell viability 71.49% at the concentration of 100 ${\mu}g/ml$ and increased intracellular $Ca^{2+}$ concentration in a dose dependent manner. Finally, we observed the mucus content as a defensive factor and gastric secretion as an aggressive factor, and found that the mucus content noticeably increased when treated with butanol fraction and betaine and gastric secretion decreased when treated with betaine in vivo study. From these results, we suggest that CAL butanol fraction and betaine may have protective effects on gastritis.

Bubble Point Calculation using Experimental Flash Points of Binary Solutions (이성분계 용액의 인화점 실험값을 이용한 기포점 계산)

  • Suitable design and operation of distillation process is very dependent on vapor-liquid equilibrium calculation. The usual calculation method is use binary interaction parameter. Flash points of n-propanol+n-butanol and 2-butanol+n-butanol were measured by Seta-flash closed cup tester. Experimental Flash points were compared with those calculated by the method based on Raoult's law and the optimization method using Wilson equation. The binary interaction parameters obtained by the optimization method are then used to calculate the bubble points of n-propanol+n-butanol and 2-butanol+n-butanol.

  • Objectives: This study was camed out to examine the effect of five fractions of aqueous extract from Artemisia capillaris THUNB(ACT), on TGF, 1-induced apoptosis, cell viability, cell cycle progression and mRNA expression of apoptosis-related genes in human hepatocyte cell line HepG2. Methods: This study employed Tryphan blue exclusion assay, DNA fragmentation assay, Cpp32 protease activity assay and Quantitative RT-PCR analysis. Results: In the Tryphan blue exclusion assay, the butanol fraction of ACT with $TGF{\beta}$, l showed magnificent (Nice word, ut is it appropriate in a medical abstract\ulcorner) viability and the H2O fraction of ACT with $TGF{\beta}$, l also showed higher viability than only $TGF{\beta}$, l-treated group. DNA fragmentation assay showed that the butanol fraction and the H2O fraction carried inhibitory effects on apoptosis induction, with the butanol fraction displaying greater effects. The Cpp32 protease activity assay showed that the butanol fraction decreased Cpp32 protease activity. The H2O fraction of ACT had no significant effect on the Cpp32 protease activity. Quantitative RT-PCR showed that the butanol fraction suppressed Bax, p 15/INK4B, p21/Waf1, PAI-1 and increased Bcl-2 gene. Conclusions: The data shows that butanol fraction of ACT increases the hepatocyte viability and has the hepatocellular protective effect by the suppression of $TGF{\beta}$, l induced-apoptosis through gene regulation.

Functional Properties of Korean Atractylodes japonica Koidz (한국산 삽주의 기능성)

  • To study the availability of the Korean Atractylodes japonica Koidz. as ingredients for functional flood, functional properties of solvent extracts were investigated and the results were followed. Yield was 14.8% by ethanol extraction of fresh Korean A. Japonica and 17.7% by water fraction. Acetone extract and butanol fraction showed stronger activity of the hydrogen donating activities, each of 72.9% and 74.2%, respectively, in fresh Korean A. japonica and methanol extract and butanol fraction showed stronger activity of the nitrite scavenging effects, each of 95.0% and 79.2%. in fresh Korean A. japonica. Among the solvent extacts from fresh Korean a. japonica, extract by methanol showed strong antimicrobial activity in which clear zone showed 20 mm for Bacillus subtilis and 19 mm for Pseudomonas aeruginosa. Butanol fraction derived from methanol extract showed moderate antimicrobial activity : 18 mm clear zone for Bacillus subtilis and Vibrio parahaemolyticus. Minimum inhibitory concentrations of methanol extract and butanol fraction were about 2 mg/disc and 4 mg/disc against gram(+) bacteria and 6 mg/disc against gram(-) bacteria, respectively.
