단백질 2-DE 이미지 분석에서 정확한 스팟 매칭 패턴 검색을 위한 효과적인 방법 (An Efficient Method to Find Accurate Spot-matching Patterns in Protein 2-DE Image Analysis)

  • 단백질 2-DE 이미지 분석에서 단백질 자체가 가지고 있는 불안정성과 2-DE 실험이 가지고 있는 근본적인 문제점으로 인하여 이미지 스팟 매칭 분석의 정확도가 낮아지게 된다. 이 논문에서는 다중 참조이미지를 사용하여, 스팟 매칭 패턴의 정확도에 큰 영향을 주는 이미지 찌그러짐을 보완하고, 그에 따른 노이즈 스팟 제거와 참조 이미지 품질에 의한 정확도 저하를 최소화하는 방법을 제안하였다. 또한 2-DE 이미지의 데이터 특성에 의하여 이미지 수가 증가할 때 성능이 급격히 떨어지는 문제를 해결하기 위하여, 다중 참조이미지를 사용하여 구축한 스팟 매칭 데이터베이스를 이미지의 생물학적 특성에 의하여 "분할 및 확장" 방법을 사용하여, 정확도를 향상시키는 동시에 패턴 길이를 보장하는 스팟 매칭 패턴을 효과적으로 생성하였다. 실험에서는 실제 인간 2-DE 이미지 데이터를 사용하여 제안한 방법의 타당성을 보여준다.

Application of Gel-based Proteome Analysis Techniques to Studying Post-mortem Proteolysis in Meat

  • This study was conducted to evaluate the possible application of 2 D-SDS-PAGE (2 DE)-based proteome analysis techniques to the assessment of extreme proteolysis in postmortem skeletal muscle. Eight Hanwoo longissimus muscles were incubated immediately after slaughter for 24 h at 5$^{\circ}C$, 15$^{\circ}C$ or 36$^{\circ}C$. Warner Bratzler (WB)-shear force and ultrastructural configuration were determined at 24 h, and rate of proteolysis to 24 h was determined by 1 D-SDS-PAGE (1 DE) and 2 DE. In addition, tentative protein identification was performed from peptide mass fingerprints of MALDI-ToF analysis of major protein groups on 2 DE profiles. The result showed that although ultrastructural configuration was similar between the 5$^{\circ}C$ and 36$^{\circ}C$ treatments, meat at 5$^{\circ}C$ had higher WBshear force (approximately 5 kg greater). A higher rate of protein degradation at 36$^{\circ}C$ was observed based on Troponin-T degradation, 1 DE, and 2 DE analysis. This indicates that proteolysis during the early postmortem period was a significant determinant of shear force at 24 h. Little difference in proteolysis between 5$^{\circ}C$ and 15$^{\circ}C$ treatments was found based on classic 1 DE profile assessment. Meanwhile, considerable differences in the 2 DE profiles between the two treatments were revealed, with substantially higher rate of proteolysis at 15$^{\circ}C$ compared to 5$^{\circ}C$. Nuclease treatment improved 2 DE profile resolution. 400 ${\mu}$g and 600 ${\mu}$g of sample loading appeared to be appropriate for 24 cm pH 3-10 and pH 5-7 IPG strips, respectively. Protein detection and quantification of the 5$^{\circ}C$, 15$^{\circ}C$ and 36$^{\circ}C$ 2 DE profiles revealed 78, 163 and 232 protein spots respectively that were differentially modified in terms of their electrophoretic properties between approximately pI 5.3-7.7 with the molecular weight range of approximately 71-12 kDa. The current results demonstrated that 2 DE was a superior tool to 1 DE for characterising proteolysis in postmortem skeletal muscle.

  • The maximum along-wind displacement of a considerable amount of building under simulated wind loads is computed with the aim to produce a simple prediction model using multiple regression analysis with variables transformation. The Shinozuka and Newmark methods are used to simulate the turbulent wind and to calculate the dynamic response, respectively. In order to evaluate the prediction performance of the regression model with longer degree of determination, two complex structural models were analyzed dynamically. In addition, the prediction model proposed is used to estimate and compare the maximum response of two test buildings studied with wind loads by other authors. Finally, it was proved that the prediction model is reliable to estimate the maximum displacements of structures subjected to the wind loads.

A surface chemical analysis strategy for the microstructural changes in a CuAgZrCr alloy cast under oxidation conditions

  • The aim of this work was to determine the behavior of alloy elements and compounds formed during solidification in the manufacturing process of the CuAgZrCr alloy under an oxidizing environment. Bulk and surface analysis techniques, such as Scanning Electron Microscopy (SEM), X-ray Photoelectron Spectroscopy (XPS), Raman and X-ray diffraction (XRD) were used to characterize the phases obtained in the solidification process. In order to focus the analysis on the on grain boundary interface, partial removal of the matrix phase by acid attack was performed. The compositional differences obtained by SEM-EDX, Raman and XPS on post-manufacturing materials allowed us to conclude that the composition of grain boundaries of the alloy is directly influenced by the oxidizing environment of alloy manufacturing.

Critical Discourse Analysis of Deinstitutionalization News Articles for the Disabled: Focusing on Fairclough's critical discourse analysis

  • This study aims to derive discourse's linguistic meaning, production method, and social practice implications by analyzing news reports on de-facility for people with disabilities. To this end, the discourse was analyzed by applying Fairclough's framework of critical discourse analysis. The subject of analysis is a news article on the de-facility of the disabled on the N portal site, and the analysis period is one year, from January 1 to December 31, 2022. First, as a result of the study, the surface meaning of the news discourse on the de-facility for disabled people was ideological through the seriousness of the problem for disabled people, the poor environment, and the policy of de-facility for disabled people separated from reality. Second, the social meaning of the de-facility news discourse for disabled people appeared from a realistic perspective, such as the structural cause of the problem for disabled people and the need for sensible government policies and measures to practice de-facility for disabled people. Finally, the socio-cultural practical implications of the de-facility news discourse for people with disabilities proposed the development of a systematic and realistic de-facility management manual for the disabled, practical government policy support, and changes in self-support perception for disabled people. The results of this study are expected to help find an alternative direction to reduce the gap between actual policies for de-facility for disabled people and practice in the field in the future.

Nonlinear earthquake capacity of slender old masonry structures prestressed with steel, FRP and NiTi SMA tendons

  • This paper focuses on the seismic protection of slender old masonry structures by the implementation of prestressing devices at key locations. The devices are vertically and externally located inside the towers in order to be reversible and calibrated. An extensive parametric study on a selected slender tower is carried out based on more than 100 nonlinear static simulations aimed at investigating the impact of different parameters on the seismic performance: (i) different prestressing levels; (ii) shape memory alloy superelasticity and (iii) changes in prestressing-forces in all the stages of the analysis until failure and masonry toe crushing. The tendon materials under analysis are conventional prestressing steel, fiber-reinforced polymers of different fibers and shape memory alloys. The parametric study serves to select the most suitable prestressing device and optimal prestressing level able to dissipate more earthquake energy. The seismic energy dissipation is evaluated by comparing the structural capacity curves in original state and retrofitted.

Nonlocal strain gradient theory for bending analysis of 2D functionally graded nanobeams

  • This article presents an analytical approach to explore the bending behaviour of of two-dimensional (2D) functionally graded (FG) nanobeams based on a two-variable higher-order shear deformation theory and nonlocal strain gradient theory. The kinematic relations are proposed according to novel trigonometric functions. The material gradation and material properties are varied along the longitudinal and the transversal directions. The equilibrium equations are obtained by using the virtual work principle and solved by applying Navier's technique. A comparative evaluation of results against predictions from literature demonstrates the accuracy of the proposed analytical model. Moreover, a detailed parametric analysis checks for the sensitivity of the bending and stresses response of (2D) FG nanobeams to nonlocal length scale, strain gradient microstructure scale, material distribution and geometry.

Mercury recovery from aqueous solutions by polymer-enhanced ultrafiltration using a sulfate derivative of chitosan

  • The sulfatation of chitosan, by reaction with chlorosulfonic acid under controlled conditions, allowed increasing the pH range of chitosan solubility. The biopolymer was characterized using FTIR and $^{13}C$-NMR spectroscopy, elemental analysis and titration analysis and it was tested for mercury recovery by polymer enhanced ultrafiltration (PEUF). In slightly alkaline conditions (i.e., pH 8) mercury recovery was possible and at saturation of the polymer the molar ratio $-NH_2$/Hg(II) tended to 2.6. Polymer recycling was possible changing the pH to 2 and the polymer was reused for 3 cycles maintaining high metal recovery. The presence of chloride ions influences metal speciation and affinity for the polymer and "playing" with metal speciation allowed using the PEUF process for mercury separation from cadmium; at pH 11 the formation of hydroxo-complexes of Hg(II) limits it retention. Cake formation reveals the predominant controlling step for permeation flux.

Molecular Characterization of Trypanosoma cruzi Tc8.2 Gene Indicates Two Differential Locations for the Encoded Protein in Epimastigote and Trypomastigote Forms

  • This report describes the molecular characterization of the Tc8.2 gene of Trypanosoma cruzi. Both the Tc8.2 gene and its encoded protein were analyzed by bioinformatics, while Northern blot and RT-PCR were used for the transcripts. Besides, immunolocalization of recombinant protein was done by immunofluorescence and electron microscopy. Analysis indicated the presence of a single copy of Tc8.2 in the T. cruzi genome and 2-different sized transcripts in epimastigotes/amastigotes and trypomastigotes. Immunoblotting showed 70 and 80 kDa polypeptides in epimastigotes and trypomastigotes, respectively, and a differential pattern of immunolocalization. Overall, the results suggest that Tc8.2 is differentially expressed during the T. cruzi life cycle.

Replication origins oriGNAI3 and oriB of the mammalian AMPD2 locus nested in a region of straight DNA flanked by intrinsically bent DNA sites

  • The aim of this work was to determine whether intrinsically bent DNA sites are present at, or close to, the mammalian replication origins oriGNAI3 and oriB in the Chinese hamster AMPD2 locus. Using an electrophoretic mobility shift assay and in silico analysis, we located four intrinsically bent DNA sites (b1 to b4) in a fragment that contains the oriGNAI3 and one site (b5) proximal to oriB. The helical parameters show that each bent DNA site is curved in a left-handed superhelical writhe. A 2D projection of 3D fragment trajectories revealed that oriGNAI3 is located in a relatively straight segment flanked by bent sites b1 and b2, which map in previously identified Scaffold/Matrix Attachment Region. Sites b3 and b4 are located approximately 2 kb downstream and force the fragment into a strong closed loop structure. The b5 site is also located in an S/MAR that is found just downstream of oriB.
