A Study on the Stratified Cluster Replicated Systematic Unrelated Question Model (층화 집락 반복계통 무관질문모형에 관한 연구)

  • We apply stratified cluster sampling to a replicated systematic unrelated question model for a large scale survey in which the population is comprised of several strata developed by several clusters and with sensitive parameters. We first present a replicated systematic unrelated question model using an unrelated question model to procure sensitive information from the population of clusters and then develop a suggested model to an unrelated question by a stratified cluster replicated systematic sampling that can be used in large population of strata. We cover the proportional and optimum allocation for the suggested model. Finally, we compare and analyze the efficiency of the suggested model with the replicated systematic unrelated question model.

  • In this paper we point out another advantage of systematic sampling over simple random sampling, which have not yet been spelled out in the literature. After a single sample is drawn by a sampling scheme, it is important to check whether the achived sample represents the population well or not. Therefore. a sampling scheme which avoids the possibility of selecting non-preferred samples is desirable. The simulation results are given to illustrate that, in the ordered population, the possibility of selecting non-preferred sample by systematic sampling is lower than that by simple random sampling.

  • In this paper, we investigate several variance estimators for pps systematic sampling. Unfortunately, there is no unbiased variance estimators for a systematic sample because systematic sampling can be regarded as a random selection of one cluster. This study provides guidance on which variance estimator may be more appropriate than others in several circumstances. We judge the efficiency of variance estimators for systematic sampling based on of their relative biases and relative mean square error. Also, we investigate variance estimation problems for two-stage systematic sampling applied for the Food Raw Material Consumption Survey and the Establishment Labor Force Survey simulation study, in order to consider the popular two-stage pps systematic sample design for establishment and household survey in Korea.

Efficient Estimation of the Mean for Populations with a Linear Trend : An Extension of Systematic Sampling (선형추세를 갖는 모집단에 대한 효율적인 모평균 추정 : 계통추출의 확장)

  • In this study, we have proposed a sampling method and an estimation method for efficiently estimating the mean of a population which has a linear trend. These methods involve drawing a sample by the so-called "centered balanced systematic sampling", which is an extension of systematic sampling, and then estimating the population mean with an adjusted estimator, not with the sample mean itself. We used the concept of interpolation in determining the adjusted estimator.\Ve compared the efficiency of the proposed estimator with those of the estimators from existing methods, under the expected mean square error criterion based on the infinite superpopulation model introduced by Cochran(1946). The proposed method is for use in the case when the sample size n(2 5) is an odd number and k(the reciprocal of the sampling fraction) is an even number. A good result was also obtained in an example using computer simulation. simulation.

Estimation of Population Mean Using Modified Systematic Sampling and Least Squares Method (변형된 계통추출과 최소제곱법을 이용한 모평균 추정)

  • In this paper, a new method is developed for estimating the mean of a population which has a linear trend. This method involves drawing a sample by the modified systematic sampling, and then estimating the population mean with an adjusted estimator, not with the sample mean itself. We use the method of least squares in determining the adjusted estimator. The proposed method is shown to be more and more efficient as the linear trend becomes stronger. It turns out to be relatively efficient as compared with the conventional methods if $\sigma$$^2$the variance of the random error term in the infinite superpopulation model, is not very large.

  • Recently, much attention has been focused on plant antioxidants, because they are expected to protect against oxidative damage, possibly preserving biological functions of cells. Antioxidant compounds were isolated from Angelica keiskei through extraction with 80% EtOH, and fractionations were carried out sequentially with n-hexane, chloroform, ethyl acetate, n-butanol, and water. Two active compounds were isolated from ethyl acetate fraction by silica gel column chromatography, and were identified as isoquercitrin ($quercetin-3-O-{\beta}-D-glucose$) and hyperoside ($quercetin-3-O-{\beta}-D-glucose$). Isoquercitrin and hyperoside showed strong antioxidative potency, as revealed by evaluation of their ABTS, DPPH, OH, and $H_{2}O_{2}$ radical-scavenging activities, and ex vivo DNA damage-protecting effects.

Exocyclic GpC DNA methyltransferase from Celeribacter marinus IMCC12053 (Celeribacter marinus IMCC12053의 외향고리 GpC DNA 메틸트랜스퍼라아제)

  • DNA methylation is involved in diverse processes in bacteria, including maintenance of genome integrity and regulation of gene expression. CcrM, the DNA methyltransferase conserved in Alphaproteobacterial species, carries out $N^6$-adenine or $N^4$-cytosine methyltransferase activities using S-adenosyl methionine as a co-substrate. Celeribacter marinus IMCC12053 from the Alphaproteobacterial group was isolated from a marine environment. Single molecule real-time sequencing method (SMRT) was used to detect the methylation patterns of C. marinus IMCC12053. Gibbs motif sampler program was used to observe the conversion of adenosine of 5'-GANTC-3' to $N^6$-methyladenosine and conversion of $N^4$-cytosine of 5'-GpC-3' to $N^4$-methylcytosine. Exocyclic DNA methyltransferase from the genome of strain IMCC12053 was chosen using phylogenetic analysis and $N^4$-cytosine methyltransferase was cloned. IPTG inducer was used to confirm the methylation activity of DNA methylase, and cloned into a pQE30 vector using dam-/dcm- E. coli as the expression host. The genomic DNA and the plasmid carrying methylase-encoding sequences were extracted and cleaved with restriction enzymes that were sensitive to methylation, to confirm the methylation activity. These methylases protected the restriction enzyme site once IPTG-induced methylases methylated the chromosome and plasmid, harboring the DNA methylase. In this study, cloned exocyclic DNA methylases were investigated for potential use as a novel type of GpC methylase for molecular biology and epigenetics.

Characterization and Identification of an Agar-Degrading Motile Bacteria Strain (Agar를 분해하는 swarming 박테리아 균주의 특성과 동정)

  • A bacterial strain, CK214, exhibiting high motility on an LB agar (1.5%, w/v) surface was isolated from the environment. The formation of unusual agar shrinking around colonies on agar plates was observed. The strain grew on minimal media containing pure agar as a sole carbon source. The cell-free culture supernatant of CK214 generated a reduced form of sugar in the in vitro reaction with the use of pure agar as a substrate, suggesting the secretion of an agar-degrading enzyme. The CK214 strain showed swarming motility on the solid media containing a wide range of concentrations of agar (0.5, 1.0, 1.5, 2.0% w/v). Various tests, including Gram staining, API analysis, and phylogenetic analysis based on 16S rDNA sequences identified that the CK214 strain was a G(+) rod-shaped bacterium grouped in genus Paenibacillus. Electron microscopic analysis demonstrated that the P. CK214 strain is peritrichously flagellated. Through transposon random mutagenesis, several agar-degrading activity defective mutants (ADMs) were generated. These mutants will be used in the future experimentation for the study of the correlation between agar-degrading activity and motility.
