Isolation and Degradation Characteristics of 2,4,4l-Trichloro-2l-Hydroxydiphenyl Ether Degrading Bacterium (2,4,4l-Trichloro-2l-Hydroxydiphenyl Ether 분해균의 분리 및 분해특성)

  • The bacterial strains, which utilizes 2,4,4'-trichloro-2'-hydroxydiphenyl ether(TCHDPE) as a sole carbon source, were isolated by selective enrichment culture from soil samples of industrial waste deposits. The bacterium that showed the highestt biodegradation activity was designated as EL-O47R The isolated strain EL-O47R was Identified as the genus Pseudomonas from the results of morphological, cultural, and biochemical tests. The optimum conditions of medium for the growth and the degradation of TCHDPE were TCHDPE 500 ppm, (NH4)2SO4 0.1% as the nitrogen source, initial pH 7.0±0.1, and 37℃, respectively. In this conditions, the regradation rate of TCHDPE was about 97%. Pseudomonas sp. EL-O47R was tested for resistance to several metal compounds and antibiotics. Pseudomonas sp. EL-O47R was moderately grown to Cd(NO3)2, ZnCl2, AgSO4, CuSO4 and HgCl2. This strain was sensitive to rifampicin and kanamycln but resistant to ampicillin, penicillin, tetracyclin and chloramphenlcol. Pseudomonas sp. EL-O47R was grown structurally related com- pounds and potential metabolites of TCHDPE, and has the stability on TCHDPE biodegradation.

  • Microcapsules containing 2,4,4'-trichloro-2-hydroxydiphenyl ether (DP) and perfumes were prepared by the coacervation using poly (vinyl alcohol) and crosslinking agents. Internal phase content, percent releasing of core materials, antimicrobial activities of microcapsules and fabrics treated with them were investigated. The internal content of microcapsules containing perfumes and DP are increased with increasing perfumes. The internal content of cacharia in the microcapsue was higher than that of lemon and its maimum value was 93%. The microcapsules containing perfumes and DP did not release cacharia and lemon at roonl temperature. But the amount of perfumes released from microcapsules were increased with temperature and time of microcapsules containing DP and perfumes showed 100% reduction percentage of bacteria. The size of obstruction of fabrics treated with microcapsule containing DP and perfumes were increased with DP content in microcapsules.

  • AcDP and AcAP were prepared by the reaction of acryloyl chloride (Ac) with 2,4,4' -trichloro-2' -hydroxydiphenyl ether (DP) and p-hydroxyacetophenone respectively in presence of triethylamine (TEA) in MEK at $0^{\circ}C$. The reaction was monitored by TLC and the prepared monomer was characterized by UV, IR, $1^H-NMR$ and GC-MS. Further, copolymers poly (AcDP-MMA-AcAP) were prepared in different feed ratio of monomers by free radical polymerization at $70^{\circ}C$, in which BPO as initiator and their molecular weight was determined by GPC. The AF activity of prepared polymers was investigated against representatives of marine microfoulers, shipfouling bacteria (B. macroides & P. aeruginosa) and microalgae (A.coffeaeformis & N. incerta). The antibacterial activity and diatom attachment assays showed significant AF potential of these polymers.

Oxidative Transformation of Triclosan by Laccase (Laccase를 이용한 Triclosan의 처리)

  • The oxidative transformation of triclosan with laccase from Trametes versicolor was conducted in a closed, temperature controlled system containing phosphate buffer for pH control. The optimum pH for triclosan transformation showed about 5. Despite the observation that elevated temperatures tended to inactivate the enzyme, increased transformation of triclosan was observed up to $50^{\circ}C$. Of the mediators studied, ABTS was most successful at enhancing triclosan transformation. About 80% of the toxicity of the initial mixture was reduced after the enzymatic treatment. In the presence of 1.0 mM of anions such as sulfite, sulfide, and cyanide, triclosan transformation was greatly inhibited. Chloride and fluoride ions exhibited inhibition of triclosan transformation at 25 mM. Ferric ion substantially inhibited triclosan transformation at 1.0 mM.

  • Microcapsules containing 2, 4, 4'-trichloro-2-hydroxydiphenyl ether and perfumes were prepared by the coacervation using poly (vinyl alcohol) and crosslinking agents. Effects of dispersing agents, core materials, agitating speed and crosslinking agents on microcapsule size were investigated. The mean and deviation of microcapsule diameters decreased with increasing agitation speed. The diameters of m;crocapsules decreased with increasing dispering agent concentration at 6, 000 rpm of agitation speed, but it was not changed at 10, 000 rpm. The dispering effect of PVA is better than that of gum arabic. The slight increase in the diameter of microcapsule was observed when the amount of core material was increased. As the amount of crosslinking agent was increased, the diameter of microcapsule was decreased.
