The hydrodynamic characteristics of the canvas kite - 2. The characteristics of the triangular canvas kite - (캔버스 카이트의 유체역학적 특성에 관한 연구 - 2. 삼각형 캔버스 카이트의 특성 -)

  • As far as an opening device of fishing gears is concerned, applications of a kite are under development around the world. The typical examples are found in the opening device of the stow net on anchor and the buoyancy material of the trawl. While the stow net on anchor has proved its capability for the past 20 years, the trawl has not been wildly used since it has been first introduced for the commercial use only without sufficient studies and thus has revealed many drawbacks. Therefore, the fundamental hydrodynamics of the kite itself need to ne studied further. Models of plate and canvas kite were deployed in the circulating water tank for the mechanical test. For this situation lift and drag tests were performed considering a change in the shape of objects, which resulted in a different aspect ratio of rectangle and trapezoid. The results obtained from the above approaches are summarized as follows, where aspect ratio, attack angle, lift coefficient and maximum lift coefficient are denoted as A, B, $C_L$ and $C_{Lmax}$ respectively : 1. Given the triangular plate, $C_{Lmax}$ was produced as 1.26${\sim}$1.32 with A${\leq}$1 and 38$^{\circ}$B${\leq}$42$^{\circ}$. And when A${\geq}$1.5 and 20$^{\circ}$${\leq}$B${\leq}$50$^{\circ}$, $C_L$ was around 0.85. Given the inverted triangular plate, $C_{Lmax}$ was 1.46${\sim}$1.56 with A${\leq}$1 and 36$^{\circ}$B${\leq}$38$^{\circ}$. And When A${\geq}$1.5 and 22$^{\circ}$B${\leq}$26$^{\circ}$, $C_{Lmax}$ was 1.05${\sim}$1.21. Given the triangular kite, $C_{Lmax}$ was produced as 1.67${\sim}$1.77 with A${\leq}$1 and 46$^{\circ}$B${\leq}$48$^{\circ}$. And when A${\geq}$1.5 and 20$^{\circ}$B${\leq}$50$^{\circ}$, $C_L$ was around 1.10. Given the inverted triangular kite, $C_{Lmax}$ was 1.44${\sim}$1.68 with A${\leq}$1 and 28$^{\circ}$B${\leq}$32$^{\circ}$. And when A${\geq}$1.5 and 18$^{\circ}$B${\leq}$24$^{\circ}$, $C_{Lmax}$ was 1.03${\sim}$1.18. 2. For a model with A=1/2, an increase in B caused an increase in $C_L$ until $C_L$ has reached the maximum. Then there was a tendency of a very gradual decrease or no change in the value of $C_L$. For a model with A=2/3, the tendency of $C_L$ was similar to the case of a model with A=1/2. For a model with A=1, an increase in B caused an increase in $C_L$ until $C_L$ has reached the maximum. And the tendency of $C_L$ didn't change dramatically. For a model with A=1.5, the tendency of $C_L$ as a function of B was changed very small as 0.75${\sim}$1.22 with 20$^{\circ}$B${\leq}$50$^{\circ}$. For a model with A=2, the tendency of $C_L$ as a function of B was almost the same in the triangular model. There was no considerable change in the models with 20$^{\circ}$B${\leq}$50$^{\circ}$. 3. The inverted model's $C_L$ as a function of increase of B reached the maximum rapidly, then decreased gradually compared to the non-inverted models. Others were decreased dramatically. 4. The action point of dynamic pressure in accordance with the attack angle was close to the rear area of the model with small attack angle, and with large attack angle, the action point was close to the front part of the model. 5. There was camber vertex in the position in which the fluid pressure was generated, and the triangular canvas had large value of camber vertex when the aspect ratio was high, while the inverted triangular canvas was versa. 6. All canvas kite had larger camber ratio when the aspect ratio was high, and the triangular canvas had larger one when the attack angle was high, while the inverted triangluar canvas was versa.

Microorganism Contaminants of Dutch Coffee and Change according to the Storage Period (시판 더치커피의 미생물 오염도와 저장 기간에 따른 미생물 변화)

  • This study was carried out to investigate the microbiological contamination levels of Dutch coffee products marketed in Korea. The temperature conditions during distribution and storage were also considered in this experiment. Retailed Dutch coffee were purchased from regional cafes, that is, these were self-blended by the cafes, and the marketed products were purchased from department stores and from Internet sites. The 21 samples were blended in a coffee house and 9 were obtained from department stores or were delivered from internet sites. House blended Dutch coffee contained $35.2{\pm}15.8CFU/mL$ of general bacteria, and this increased to $78.4{\pm}29.7CFU/mL$ at room temperature or $51.2{\pm}32.1CFU/mL$ after refrigeration for 5 days. These almost reached the highest criteria level for the Korea Food Sanitation Law. After 10 days, the count increased to $98.5{\pm}58.4CFU/mL$ at room temperature and $86.7{\pm}44.2CFU/mL$ at refrigeration temperature. In the Dutch coffee for distribution, $39.6{\pm}20.1CFU/mL$ of general bacteria were detected, but these did not increase after 5 days or 10 days both for room temperature and under refrigeration. The Coliform group was not found in any kind of Dutch coffee, and Fungi was founded in 60% of the Dutch samples purchased in coffee houses, department stores, and shopping sites mall. On day 0 day, $2.6{\pm}1.7CFU/mL$ of fungi were detected in the coffee house Dutch, and it did not increase significantly during the storage period at room and in a cold temperature. $3.5{\pm}3.4CFU/mL$ of fungi were detected in the Dutch coffee for distribution, and it didn't increase during further storage under any temperature.

Acute Toxicity of TBT Influencing on the Production of Coastal Olive Flounder (연안 어중의 넙치 생산성에 영향을 미치는 TBT의 급성 독성)

  • The toxic effect of TBTO on Chlorella and Rotifer was observed. The value of 48hr-LC50 for Chlorella (3.3$\mu\textrm{g}$/L) estimated to be almost 500 times as high as that for Rotifer (6.7ng/L). A fertilized egg of olive flounder exposed in an embryo-formation stage was mostly influenced by TBTO toxicity when the fertilized egg at each stage until hatching was exposed to TBTO at the concentrations of 5 to 200ng/L. The values of LT50 were estimated to be 68.0, 41.0, 21.0, 13.0, 7.7 and 4.7 hours at 5, 10, 25, 50, 100 and 200ng/L of TBTO, respectively when the fertilized egg in a morula stage was exposed to TBTO, and the 48hr-LC50 was 8 ng/L. In case of TBTO treatment in an embryo-formation stage, the values of LT50 were 33.0, 12.5, 3.5, 1.3, 0.5 and 0.2 hours at 5, 10, 25, 50, 100 and 200ng/L of TBTO, respectively, and the value of 48hr-LC50 was 4ng/L. The values of LT50 were estimated to be 17.0, 11.0, 6.2, 4.0, 2.6 and 1.7 hours at 5, 10, 25, 50, 100 and 200ng/L of TBTO, respectively when the fertilized egg in a stage just before hatching was exposed to TBTO, and the 48hr-LC50 was below 1ng/L. The percentages of hatching were 46.2, 20.6, 21.9, 20.6 and 13.2% at 25, 50, 100, 250 and 500ng/L of TBTO, respectively when the fertilized egg in the stage just before hatching was exposed to TBTO and the measurement was done at second day after the completion of hatching. However no survival after the completion of hatching was found in all cases. With the treatment of 1, 5 and 10ng/L of TBTO, the percentages of hatching were 80.5, 70.0 and 44.1%, respectively. The percentages of survival until second day after the completion of hatching were 80.0, 63.3 and 9.1%, respectively. The percentages of hatching and survivability after the completion of hatching for the control were 84.5 and 82.5%, respectively.

  • Hydrocotyle maritima Honda used as medicinal plants for a hemostatic agent was investigated for in vitro regeneration. The petiole explants of H. maritima were cultured on callus induction medium containing growth regulators ($0{\sim}5\;mg/l$, NAA and $0{\sim}5\;mg/l$ 2,4-D) either in single or in combination with $0.1{\sim}2\;mg/l$ BA for 6 weeks. Although single treatments of 2,4-D or NAA resulted in callus formation, the best results were combination of $0.5\;mg/l$, 2,4-D and $0.5\;mg/l$, BA. $5\;mg/l$, NAA and $0.5\;mg/l$, BA, respectively. The highest number of shoot (12 shoots per callus) was achieved with $3\;mg/l$, Kinetin. Also, when pieces of embryogenic callus induced on the medium supplemented with $0.5\;mg/l$, 2,4-D and $0.5\;mg/l$, BA were subcultured on hormone-free medium, somatic embryos were differentiated and developed further into welldeveloped plants.

  • Objective : The purpose of this study was to investigate the effect of Bee Venom on the lipopolysaccharide, sodium nitroprusside and hydrogen peroxide induced expression phospholipase $A_2$ and calcium concentration in RAW 264.7 cells, a murine macrophage cell line. Method : The expression of phospholipase $A_2$ was determined by western blotting with corresponding antibodies, and the generation of intracellular calcium concentration was investigated by delta scan system in RAW 264.7 cells. Results : 1. Compared with control, expressions of lipopolysaccharide-induced phospholipase $A_2$ were decreased significantly by $1\;{\mu}g/{\mu}l$ of bee venom and decreased by 0.5, $5\;{\mu}g/{\mu}l$ of bee venom. 2. Compared with control, expressions of sodium nitroprusside-induced phospholipase $A_2$ were decreased significantly by $5\;{\mu}g/{\mu}l$ of bee venom but increased by 0.5, $5\;{\mu}g/{\mu}l$ of bee venom. 3. Compared with control, expressions of hydrogen peroxide-induced phospholipase $A_2$ were decreased significaltly by $1{\mu}g/{\mu}l$ of bee venom and decreased by $0.5\;{\mu}g/{\mu}l$ of bee venom but increased by $5\;{\mu}g/{\mu}l$ of bee venom. 4.   • The taxonomical properties of strain J-275L isolated from soil as a microorganism which produces extracellular adenine deaminase and cultural conditions for the enxyme production were studied. The hyphae of strain J-275L is fragmented into rod-or coccus-like elements. The elements of fragmented aerkal hyphae has smooth surfaces. The cell wall of the organism contains LL-diaminopimelic acid. Mycolic acid are not produced. As a result of taxonomical studies, strain J-275L is designated as Nocardioides sp. J-275L. The optimum medium for the enzyme production from Nocardioides sp.J-275L wascomposed of 0.5% peptone, 0.5% dextrin, 1% yeast extract, and 0.2% $K_{2}HPO_{4}$. The optimum initial pH of the medium was pH 7.5.

Population Pharmacokinetics for Gentamicin in Korean and Caucasian Appendicitis Patients Using Nonparametric Expected Maximum (NPEM) Algorithm (한국인과 코카시안 충수돌기염 환자에서 비모수적 기대최대치(NPEM) 연산방법에 의한 겐타마이신의 모집단 약물동태학)

  • Population pharmacokinetics for gentamicin were compared with 20 Korean patients (14 male and 6 female) and 25 Caucasian appendicitis patients (16 male and 9 female). Two to six blood specimens were collected from all patients at the following times : just before a regularly scheduled infusion and at 0.5 hour after the end of a 0.5 hour infusion. Nonparametric expected maximum(NPEM) algorithm for population modeling was used. The estimated parameters were the elimination rate constant(K), the slope(KS) of the relationship between K versus creatinine clearance(Ccr), the apparent volume of distribution (V), the slope(VS) of the relationship between V versus weight, gentamicin clearance(CL) and the slope(CS) of the relationship between CL versus Ccr and the V. The output includes two marginal probability density function(PDF), means, medians, modes, variance, skewness, kurtosis, and CV%. The mean K(KS) were$0.402{\pm}0.129hr^{-1}$ ($0.00486{\pm}0.00197[hr{\cdot}mL/min/1.73m^2]^{-1}$) and $0.425{\pm}0.137hr^{-1}$($0.00432{\pm}0.00168[hr{\cdot}mL/min/1.73m^2]^{-1}$) for Korean and Caucasian populations, respectively. The mean V(VS) were not different at $14.3{\pm}3.69L$($0.241{\pm}0.0511L/kg$) and $15.8{\pm}4.81L$($0.236{\pm}0.0531L/kg$) for Korean and Caucasian populations, respectively (P>0.2). The mean CL(CS) were $5.68{\pm}1.69L/hr$ ($0.0714{\pm}0.0222L/kg[hr{\cdot}mL/min/1.73m^2]$) and $6.29{\pm}1.84L/hr$ ($0.0629{\pm}0.0189L/kg[hr{\cdot}mL/min/1.73m^2]$) for Korean and Caucasian populations, respectively. There are no differences in gentamicin pharmacokinetics between Korean and Caucasian appendicitis patients.

Cloning and Sequencing Analysis of the Repressor Gene of Temperate Mycobacteriophage L1

  • The wild-type and temperature-sensitive (ts) repressor genes were cloned from the temperate mycobacteriophage L1 and its mutant L1cIts391, respectively. A sequencing analysis revealed that the $131^{st}$ proline residue of the wild-type repressor was changed to leucine in the ts mutant repressor. The 100% identity that was discovered between the two DNA regions of phages L1 and L5, carrying the same sets of genes including their repressor genes, strengthened the speculation that L1 is a minor variant of phage L5 or vice versa. A comparative analysis of the repressor proteins of different mycobacteriophages suggests that the mycobacteriophage-specific repressor proteins constitute a new family of repressors, which were possibly evolved from a common ancestor. Alignment of the mycobacteriophage-specific repressor proteins showed at least 7 blocks (designated I-VII) that carried 3-8 identical amino acid residues. The amino acid residues of blocks V, VI, and some residues downstream to block VI are crucial for the function of the L1 (or L5) repressor. Blocks I and II possibly form the turn and helix 2 regions of the HTH motif of the repressor. Block IV in the L1 repressor is part of the most charged region encompassing amino acid residues 72-92, which flanks the putative N-terminal basic (residues 1-71) and C-terminal acidic (residues 93-183) domains of L1 repressor.

HPLC Assay and Renal Excretion Characteristics of Theophylline and Its Metabolites in Rat (테오필린과 그 대사체의 HPLC 동시 정량 및 신(腎) 배설 특성)

  • A high-performance liquid chromatographic (HPLC) method was developed for the simultaneous determination of theophylline(TP) and its metabolites, 1-methyluric acid (1-MU) and 1,3-dimethyluric acid (1,3-DMU), in rat plasma and urine. An $100\;{\mu}l$ aliquot of a plasma or urine sample was mixed with $250\;{\mu}l$ of acetonitrite and vortexed. After centrifugation, $200\;{\mu}l$ (plasma) or $20\;{\mu}l$ (urine) aliquot of the supernatant was dried by $N_2$ stream and redissolved in $100\;{\mu}l$ (plasma) or $200\;{\mu}l$ (urine) of the mobile phase. A $20\;{\mu}l$ of the mobile phase solution was injected onto a $C_{18}$ reversed-phase column. The column was maintained at $45^{\circ}C$ by the aid of electric heating jacket. The mobile phase was a 3%(v/v) methanol solution in deionized water which contains sodium acetate (100 mM) and tetrabutyl ammonium hydroxide (4 mM). pH of the mobile phase was adjusted 4.5 by the addition of acetic acid. Detection limits for TP, 1-MU, and 1,3-DMU in plasma were 0.2, 0.1 and $0.1\;{\mu}/ml$, respectively and the corresponding values in urine were all $5\;{\mu}g/ml$. Inter- and intra-day variability of the assay for all compounds in the plasma samples was less than 5.5 and 3.8%, respectively. The retention times for 1-MU, 1,3-DMU, and TP were approximately 7, 8.5 and 18 min, respectively. Sample preparation procedure used in this method was simple, rapid and reproducible. Renal clearance of TP and its metabolites in rats showed plasma concentration dependency indicating renal tubular secretion and reabsorption of them.

Effect of Plant Crowth Regulators on Production of Good Quality Plant for Korean Gerbera Lines (국내 육성 거베라의 우량묘 생산에 미치는 식물생장조절제의 영향)

  • This study was carried out to obtain information about the valuable combination and concentration to produce good plants of cut gerbera flowers of 19 domestically - bred gerbera cultivars. BA and kinetin combination is more effective than NAA and BA combination to produce good plants and mass propagation of domestically - bredgerbera cultivars. Normally almost of 19 cultivars, mass propagation was more effective on the medium containing 1.0 mg/L BA + 0.5, 1.0 mg/L kinetin. But some cultivars, 'Sunnyeo', 'Oksaem','Piny', and 'Pink Light', vitrified plants were induced on MS medium high level of BA concentration(BA 1.0 mg/L), in comparison with those on the medium with low level of BA(0.1 mg/L).Fresh and dry weight, more effective on the medium containing BA 0.5 mg/L . Kinetin 0.1, 0.5 mg/L. Anatomical investigation of vitrified leaf, stomata of vitrified leaves were circular and inflated, where-as those of normal leaves.
