Culture study on the hybrid by interspecific crossing between Porphyra pseudolinearis and P. dentata (Bangiales, Rhodophyta), two dioecious species in culture

  • Physiological studies on the hybrid by crossing between two dioecious species, Porphyra pseudolinearis and P. dentata from Korea were conducted at constant temperatures (5, 10, 15, 20, and $25^{\circ}C$), at photon flux densities (10, 20, 40, and $80\;{\mu}mol\;m^{-2}s^{-1}$) under photoperiods (14 L : 10 D and 10 L : 14 D). In the hybrid, higher growth of conchocelis was observed at 20 and $40\;{\mu}mol\;m^{-2}s^{-1}$ under 14 L : 10 D. Conchosporangial branches were produced under $10-80\;{\mu}mol\;m^{-2}s^{-1}$ at only $25^{\circ}C$, and were abundant when the conchocelis was cultured under 10 L : 14 D. Foliose thalli of the hybrid grew well at the conditions of $10-20^{\circ}C$, 10 L : 14 D and $15-20^{\circ}C$, 14 L : 10 D. The foliose thalli grew very slowly at $5^{\circ}C$ and $30^{\circ}C$, respectively. No archeospores were observed at any culture conditions. Spermatangial and zygotosporangial sori were formed at the marginal portion of mature thallus. Zygotospores from the hybrid were released at $10-2^{\circ}C$ under both photoperiods, and gave rise to form conchocelis filament. Monoecious thalli were observed at $10^{\circ}C$ under 14 L : 10 D. Neither monospores nor protothalli were produced from the conchocelis in culture.

  • This study pointed at isolation of bunker-C oil degrading bacteria and then estimation of it's degrading capability in environmental conditions. Degradation ratio of the excellent isolate was appeared to 40.5% and 44.7% when the oil was treated to 1% and 5%, respectively. The isolate was identified to Acinetobater calcoaceticus SEBCM. In pH test, high degrading effect was appeared to about 73% at pH 6 and pH 7, and low degrading ratio was 37% at pH 4. Its growth condition at temperature has not large variation in $15^{\circ}C\;{\sim}30^{\circ}C$, Quantity of nitrogen for it's good growth was ranged of $0.5\;g/L{\sim}2\;g/L$. As these results, we realized that this isolate have good activity when treated to $15\;{\sim}30^{\circ}C$ of temperature and $6{\sim}7$ of pH.

  • In order to design a wave energy generating system, a 6-DOF analysis technique is applied to the three-Dimensional CFD analysis on of a floating body and the behavior is interpreted according to the nature of the incoming wave. A wave period of 5.5s & amplitude of 0.57m from Marado is chosen. 12 case of natural pitching period from 1.25 to 2.8s has been modeled. The relation between tuning factor & pitch angle for the waves generated is compared to analyze the effects of energy absorption variables, namely mass moment of inertia, angular velocity and angular acceleration. From the results obtained, we conclude that model L is the maximum power absorbed, 6kW approximately. A maximum pitch angle of 1.91 degree was attained by Model F, and the maximum displacement of nearly 0.7m was attained by Model L among models D, F and L.

  • A new potassium sensitive field effect transistor modified with 4'-aminobenzo-15-crown-5 was prepared and its response characteristics were evaluated. The response slope of $K^{+}-ISFET$ for pH was 30.0 mV/decade and the response time was mere than 3 minutes. And the response slope and time of the $K^{+}-ISFET$ for potassium ion as $19.5{\pm}0.2{\;}mV/decade$ and about 3 minutes, respectively. The linear response range of the sensor for potassium ion was $2.0{\times}10^{-4}{\sim}1.0{\times}10^{-2}M$. The selectivity coefficients of the $K^{+}-ISFET$ for the alkali and alkaline earth metal ions were also evaluated. Sodium, ammonium and calcium ions exhibited relatively significant interference. The long term stability of the sensor was remarkably improved and it could be used for more than 50 days.

The Role of Computed Tomography in the Presurgical Diagnosis of Foraminal Entrapment of Lumbosacral Junction

  • Objective: On the basis of preoperative computed tomography (CT) scans, we studied the change of the size of anterior primary division (APD) of the L5 spinal root in the presence of foraminal/extraforaminal entrapment of the L5 spinal root. Methods: Two independent radiologists retrospectively reviewed the preoperative CT scans of 27 patients treated surgically and compared the sizes of the APDs on bilateral L5 spinal roots. If one side APO size was larger than the other side APD size, it was described as left or right "dominancy" and regarded this as "consensus (C)" in case that there was a consensus between the larger APD and the location of sciatica, and regarded as "non-consensus (NC)" in case that there was not a consensus. Oswestry Disability Index (ODI) scores were used for preoperative and postoperative evaluation. Results: On CT scans, twenty-one (77%) of 27 patients were the consensus group (APD swelling) and 6 (22%) were a non-consensus group (APD no swelling). In 9 patients with acute foraminal disc herniations, asymmetric enlargement of the APD on L5 spinal root was detected in all cases (100%) and detected in 11 (64%) of 17 patients with stenosis. Preoperative ODI score was 75-93 (mean 83) and postoperative 001 scores were improved to 13-36 (mean 21). The mean follow-up period was 6 months (range, 3-11 months). Conclusion: An asymmetric enlargement of the APD on L5 spinal root on CT scans is meaningfully associated with a foraminal or extraforaminal entrapment of the L5 spinal root on the lumbosacral junction.

Construction of a Genetic Linkage Map of Shiitake Mushroom Lentinula Edodes Strain L-54

  • From fruiting bodies of L. edodes strain L-54, single-spore isolates (SSIs) were collected. Two parental types of L-54 were regenerated via monokaryotization. By means of random-amplified polymorphic DNA (RAPD), DNA samples from L-54, its two parental types, and 32 SSIs were amplified with arbitrary primers. Dedikaryotization was demonstrated, and 91 RAPD-based molecular markers were generated. RAPD markers that were segregated at a 1:1 ratio were used to construct a linkage map of L. edodes. This RAPD-linkage map greatly enhanced the mapping of other inheritable and stable markers [such as those that are linked to a phenotype (the mating type), a known gene (priA) and a sequenced DNA fragment (MAT)] with the aid of mating tests, bulked-segregant analysis, and PCR-single-strand conformational polymorphism. These markers comprised a genetic map of L. edodes with 14 linkage groups and a total length of 622.4 cM.

오미자추출물의 Listeria Monocytogenes에 대한 항균효과

  • To development food preservative, antimicrobial activities of Schizandra chinensis (SC) against Listeria monocytogenes Scott A, L. monocytogenes Brie I and L. monocytogenes ATCC 19111 were investigated. The growth of L. monocytogenes Scott A, L. monocytogenes Brie I and L. monocytogenes ATCC 19111 was inhibited apparently in Tryptic Soy Broth (TSB) containing 1% SC at 35$\circ$C and it was found that these had antibacterial effects against a broad spectrum of pathogenic bacteria such as S. aureus ATCC 29737, B. subtilis KCTC 1021, E. coli ATCC 11775. The growth of L. monocytogenes was also inhibited about 3 to 5 log$_{10}$ cycle by 0.1% of three fractions of the alcohol extract such as ether, ethyl acetate and butanol. Acidic, weakly acid and neutral fraction of ether fraction showed inhibitory properties against L. monocytogenes.

  • 5-Aminosalicyl-L-aspartic acid (5-ASA-Asp) and 5-aminosalicyl-L-glutamic acid (5-ASA-Glu) were synthesized as new colon-specific prodrugs of 5-aminosalicylic acid (5-ASA), their apparent partition coefficients, and the extent of conversion in the homogenates of tissue and contents of various G.I. Tract segments of rats were evaluated. These prodrugs were stable in the homogenate of tissue and contents of stomach, proximal small intestine (PSI) or distal small intestine (DSI). Release of 5-ASA from 5-ASA-Asp after incubation with the cecal and colonic contents for 8hrs at $37^{\circ}C$ was 18%, and 8%, respectively. No significant conversion of prodrug was observed in the cecal and colonic contents of rats pretreated with kanamycin sulfate, which indicated that microbial enzymes were responsible for the cleavage of these prodrugs.

  • This experiment was conducted to investigate growth and flowering characteristics of Celosia argentea native to Korea treated by pinching and growth retardants, diniconazole, hexaconazole and daminozide. The more increased from one to three times of pinching, the more decreased in plant height and internode length but increased number of spadix and lateral branch. Spray application of diniconazole $50mg{\cdot}L^{-1}$ and daminozide $6,800mg{\cdot}L^{-1}$ decreased plant height, internode length and leaflength, but increased number of spadix, number of leaves and number of lateral branch.

In vitro Screening of Jeju Medicinal Plants for Cosmeceutical Materials

  • One of the important functions of skin is protection from harmful environments. Many studies have explored how to prevent skin from wrinkling and the occurrence of pigmentation changes. Skin wrinkling and pigmentation changes could be caused by unusual disruption of connective tissue, the formation of free radicals and ultraviolet radiation. In this study, extracts obtained from 254 different kinds of Jeju medicinal plants were screened for inhibitory effects on tyrosinase and elastase, and for free radical scavenging effects. Four herbs, Phormium tenax, Morus bombycis, Morus alba, and Cudrania tricuspidata, were potent inhibitors of tyrosinase ($IC_{50}$ values 4.62, 5.46, 8.17, and 64.17 ${\mu}g$/mL, respectively). Aleurites fordii [$IC_{50}$: 5.29 ${\mu}g$/mL, 1,1-diphenyl-2-picrylhydrazyl (DPPH)], Distylium racemosum ($IC_{50}$: 6.14 ${\mu}g$/mL), Acer palmatum ($IC_{50}$: 5.44 ${\mu}g$/mL), and Spiraea salicifolia ($IC_{50}$: 5.25 ${\mu}g$/mL) showed good antioxidative effects. Furthermore, Distylium racemosum ($IC_{50}$: 7.51 ${\mu}g$/mL), Diospyros kaki ($IC_{50}$: 15.1 ${\mu}g$/mL), Cornus macrophylla ($IC_{50}:$ 16.59 ${\mu}g$/mL), and Psidium guajava ($IC_{50}$: 40.25 ${\mu}g$/mL) exhibited potent inhibitory effects on elastase. These results suggest that medicinal plants possessing several biological activities may be potent inhibitors of the processes involved in pigmentation increases and aging. Further investigations will focus on in vivo assays and on the chemical identification of the major active components responsible for whitening and anti-aging activity in the screened efficacious extracts.
