Antioxidant and Anti-wrinkling Effects of Extracts from Vitex trifolia L. (만형자 추출물의 항산화 및 주름개선 효과)

  • In this study, the antioxidant and anti-wrinkling effects of fraction from Vitex trifolia L. were investigated. Among the fractions, ethyl acetate fraction showed the highest antioxidant activities in 1-1-diphenyl-2-picryl-hydrazyl, 2,2'-azino-bis(3-ethylbenzothiazoine-6-sulfonic acid) radical scavenging and elastase inhibition with 76, 89, and 74%, respectively, at a concentration of $1,000{\mu}g/mL$. This fraction, at the concentration of $25{\mu}g/mL$, inhibited 70% fibroblast cell viability and 86% the matrix metalloprotease (MMP)-1. In addition, the results from Western blot assay showed that this fraction ($25{\mu}g/mL$) expressed the MMP-1 protein level by decreasing 50%. The findings suggest that the ethyl acetate fraction from V. trifolia has great potential as a cosmeceutical ingredient with antioxidant and anti-wrinkling effects.

Complete $^1H$-NMR and $^{13}C$-NMR spectral analysis of the pairs of 20(S) and 20(R) ginsenosides

  • Background: Ginsenosides, the major ingredients of Panax ginseng, have been studied for many decades in Asian countries as a result of their wide range of pharmacological properties. The less polar ginsenosides, with one or two sugar residues, are not present in nature and are produced during manufacturing processes by methods such as heating, steaming, acid hydrolysis, and enzyme reactions. $^1H$-NMR and $^{13}C$-NMR spectroscopic data for the identification of the less polar ginsenosides are often unavailable or incomplete. Methods: We isolated 21 compounds, including 10 pairs of 20(S) and 20(R) less polar ginsenosides (1-20), and an oleanane-type triterpene (21) from a processed ginseng preparation and obtained complete $^1H$-NMR and $^{13}C$-NMR spectroscopic data for the following compounds, referred to as compounds 1-21 for rapid identification: 20(S)-ginsenosides Rh2 (1), 20(R)-Rh2 (2), 20(S)-Rg3 (3), 20(R)-Rg3 (4), 6'-O-acetyl-20(S)-Rh2 [20(S)-AcetylRh2] (5), 20(R)-AcetylRh2 (6), 25-hydroxy-20(S)-Rh2 (7), 25-hydroxy-20(S)-Rh2 (8), 20(S)-Rh1 (9), 20(R)-Rh1 (10), 20(S)-Rg2 (11), 20(R)-Rg2 (12), 25-hydroxy-20(S)-Rh1 (13), 25-hydroxy-20(R)-Rh1 (14), 20(S)-AcetylRg2 (15), 20(R)-AcetylRg2 (16), Rh4 (17), Rg5 (18), Rk1 (19), 25-hydroxy-Rh4 (20), and oleanolic acid 28-O-b-D-glucopyranoside (21).

  • Morphological characteristics, developmental period, and seasonal occurrence of Adeia leucomelas (L.) were investigated from 1999 to 2000. In addition, consumption of sweetpotato as food was also examined. Adults of A. leucomelas were dark-brown and body lengths of females and males were 20.2 mm and 18.9 mm, respectively, Wing expanse of female and male was 33.7 mm and 29.4mm, respectively. Egg was flat round-shape. Larva was light yellow-green to dark-brown with 3.3-53.5 mm. Pupa was deep-brown and 15.1 mm in length. Developmental periods of A. leucomelas from egg to adult emergence at different temperatures of 15, 20, 25, and 30$^{\circ}$C were 108.5, 70.7, 40.2, and 29.1 days, respectively, Developmental threshold (DT) and effective accumulative temperatures were estimated as 10.7$^{\circ}$C and 67.5 DD in egg stage, 11.0$^{\circ}$C and 275.1 DD in larval stage and 9.3$^{\circ}$C and 244.6 DD in pupal stage, respectively. The longevity of adult female was shortened with increment of temperature, whereas the total numbers of eggs laid by a female were increased. The larvae of A.leucomelas occurred from mid-June to early October, and population reached its peak during early to mid-September in Jeonbuk province. Food consumption of A. leucomelas was highest at 20-25$^{\circ}$C. Food consumption of 1 st, 2nd, 3rd, 4th, and 5th larvae of A. leucomelas per day at 25$^{\circ}$C was 0.4, 3.6, 19.6, 40.7, and 78.9 $\textrm{cm}^2$, respectively.

  • Thermal energy transport in a two-dimensional horizontal and vertical channel with an isothermal rectangular beam attached to one adiabatic wall is investigated from the numerical solution of Navier-Stokes and energy equations. The solutions have been obtained for dimensionless aspect equations. The solutions have been obtained for dimensionless aspect ratios of beam, H/B=$0.25{sim}4$, Reynolds numbers, Re=$50{\sim}500$ and Grashof numbers, Gr=$0{\sim}5{\times}10^4$. The mean Nusselt number, $\overline{Nu}$ for horizontal and vertical channels shows same value at Gr=0 and increases as Gr increases and decreases as H/B increases at Re=100. $\overline{Nu}$ of vertical channel shows higher in $0.25{\leq}H/B<1.1$ and lower in $1.1{\leq}H/B{\leq}4.0$ than that of horizontal channel at $Gr=10^4$, Re=100. $\overline{Nu}$ of vertical channel shows higher in $0.25{\leq}H/B<1.1$ and lower in $1.1{\leq}H/B=1.0$ than that of horizontal channel at Re=100, $0<Gr{\leq}5{\times}10^4$. A comparison between the experimental and numerical results shows good agreement.

Distance Identification for Maximum Change in Ship Collision Risk through a Coast Guard Patrol Ship Experiment (해양경찰 함정 실험을 통한 선박충돌 위험도의 변화가 최대인 거리 식별에 관한 연구)

  • Using two large coast guard ships at sea, we created four encounter situations ($000^{\circ}$, $045^{\circ}$, $090^{\circ}$, $135^{\circ}$) with high possibility of collision, from 3 NM up to 0.25 NM. As relative distance was gradually decreased, the subjects were measured at 0.25 NM intervals and perceived ship collision risk (PSCR) was determined by looking at the opponent ship. Characteristics were statistically analyzed using the obtained data. The purpose of this study was to analyze the characteristics of collision risk values obtained from twelve intervals, from 3 NM to 0.25 NM relative to encounter situations by curve fitting with appropriate polynomials, to determine the distance from which the change in perceived collision risk is greatest. As a result, an optimal regression equation for each distance interval was derived from each analysis direction. The greatest variation in average collision risk value was over the range 1.25 ~ 1 NM, and the collision risk value was largest at 1 NM. The maximum change in perceived collision risk was at 1 NM. These results can contribute to preventive guidelines to minimize human error in close proximity situations with a high probability of ship collision.

Buckling Behavior of Sandwich Composite Columns by Varying Hole Size and Hole Position (원공 크기 및 원공 위치에 따른 샌드위치 복합재 기둥의 좌굴 거동)

  • The study investigated the buckling behavior of sandwich composite columns with different hole sizes and hole positions when they were applied to a compressive load. The columns consisted of 1.7mm thick faces of glass fabric/epoxy and 23mm, 37mm, 48mm, and 61mm thick cores of urethane-foam. Different hole sizes with the diameter of 25mm and 38mm were considered in this experiment. To evaluate the effect of hole position on the buckling behavior, we considered three types of hole position: 25mm diameter hole located at the center, 25mm diameter hole at 1/4 position from the center to the end of the column, and 25mm diameter hole at 1/2 position from the center to the end of the column. According to the results, buckling and maximum loads of the column having 25mm diameter hole were lower by 10% compared to those of the column without hole, whereas the loads for the column having 38mm diameter hole were 30% less than those of the column without hole. Hole position appeared to have no effect on buckling and maximum loads. Major failure modes were observed as follows: the core shear failure for the thin columns having 23mm and 37mm thick cores, and the face-core debonding for the thick columns having 48mm and 61mm thick cores.

Antimicrobial Activity of Niaouli (Melaleuca quinquenervia) Leaf Extracts against Skin Flora (피부 상재균에 대한 니아울리 잎 추출물의 항균활성)

  • In this study, the antimicrobial activity of niaouli leaf extracts was evaluated against skin flora. The skin flora used for experiments were three gram-positive bacteria such as Bacillus subtilis (B. subtilis), Staphylococcus aureus (S. aureus), Propionibacterium acnes (P. acnes), and two gram-negative, Escherichia coli (E. coli), Pseudomonas aeruginosa( P. aeruginosa), and the yeast, Plasmodium ovale (P. ovale). The bioassay applied for determining the antimicrobial effects of niouli leaf extracts or fraction included the disc diffusion assay and broth dilution assay. Minimum inhibitory concentration (MIC) values of 50% ethanol extract on B. subtilis, S. aureus, P. acnes, E. coli and P. aeruginosa were 0.25%, 0.50%, 1.00%, 0.13% and 0.25% respectively and the MIC values of water fraction were 0.25%, 0.25%, 4,00%, 0.25% and 0.25%. P. ovale did not show antimicrobial activities. The MIC values of methyl paraben used as positive control indicated 0.25%, 0.25%, 0.25%, 0.13% and 0.50%. Also, Minimum bactericidal concentration (MBC) values of 50% ethanol extract were 2.00%, 2.00%, 1.00%, 0.50% and 2.00% individually and the MBC values of water fraction were 0.50%, 0.25%, 4.00%, 0.50% and 1.00%. The MBC values of methyl paraben indicated 1.00%, 0.500%, 0.50%, 0.50% and 1.00%. These results showed that water fraction was as good as methyl paraben except for P. acnes. The 50% ethanol extract also showed activity similar with it. Thus, it is concluded that the 50% ethanol extract/fraction of niaouli could be applicable to cosmetics as a natural preservatives effective in antimicrobial activity against skin flora.

  • A determination of proper maturity is a significant factor for a good quality and storage. This study is aimed at a determination of optimum harvest date related to a decisive measuring of maturity in Spur Golden Delicious. In this case, respiratory status and other characteristics are taken as a indicator for maturing determination. The results are as follows. 1. Respiratory rate has been decreased through a fruit development and come to its minimized point on Sep. 25. at last. On the contrary its tendency has been increased after Sep. 25. It is also estimated that the former is pre-climacteric, the latter climacteric rise based on dimacteric minimum Sep. 25. 2. A flesh hardness has maintained comparably high value until Sep. 18. But in the point of climacteric minimum, Sep. 25, curve of flesh firmness has rapidly been broken down, and after Sep. 25, tendency has slowly decreased at the sucessive date. 3. Pectin content, optimum acidity are remarkably decreased after Sep. 25, Oct. 2 respectively. 4. When water soluble sugar content shows to be swung from climacteric minimum, any resonable tendency are not found. 5. In this experiment, optimum harvest time is found by Oct. 2, in time of 145 days after flowering.

Usefulness of Single Voxel Proton MR Spectroscopy in the Evaluation of Hippocampal Sclerosis

  • Objective: The purpose of our study was to determine the ability of H-1 MR spectroscopy (MRS) to lateralize the lesion in patients with hippocampal sclerosis. Materials and Methods: Twenty healthy volunteers and 25 patients with intractable temporal lobe epilepsy whose MR imaging diagnosis was unilateral hippocampal sclerosis were included. This diagnosis was based on the presence of unilateral atrophy and/or high T2 signal intensity of the hippocampus. Single-voxel H-1 MRS was carried out on a 1.5-T unit using PRESS sequence (TE, 136 msec). Spectra were obtained from hippocampal areas bilaterally with volumes of interest (VOIs) of 6.0 cm3 and 2.25 cm3 in healthy volunteers, and of either 6.0 cm3 (n = 14) or 2.25 cm3 (n = 11) in patients. Metabolite ratios of NAA/Cho and NAA/Cr were calculated from relative peak height measurements. The capability of MRS to lateralize the lesion and to detect bilateral abnormalities was compared with MR imaging diagnosis as a standard of reference. Results: In healthy volunteers, NAA/Cho and NAA/Cr ratios were greater than 0.8 and 1.0, respectively. In patients, the mean values of these ratios were significantly lower on the lesion side than on the contralateral side, and lower than those of healthy volunteers (p < .05). The overall correct lateralization rate of MRS was 72% (18/25); this rate was lower with a VOI of 6.0 cm3 than of 2.25 cm3 (64% versus 82%, p < .05). Bilateral abnormalities on MRS were found in 24% (6/25) of cases. Conclusion: Although its rate of correct lateralization is low, single-voxel H-1 MRS is a useful and promising diagnostic tool in the evaluation of hippocampal sclerosis, particularly for the detection of bilateral abnormalities. To improve the diagnostic accuracy of H-1 MRS, further investigation, including the use of a smaller VOI and measurement of the absolute amount of metabolites, are needed.

  • Nonlinear acoustic effect has been considered as an effective tool for the evaluation of material degradation. In this paper, the applicability of nonlinear acoustic effect to the evaluation of degradation of 2.25Cr-1Mo steel is investigated. Firstly, a number of 2.25Cr-1Mo steel samples were heat-treated, and their damage mechanism was examined. Secondly, Ultrasonic nonlinear parameter was measured. Nonlinear acoustic parameter was found to be clearly sensitive to the material degradation.
