HPV-Associated p16INK4A Expression and Response to Therapy and Survival in Selected Head and Neck Cancers

  • Background: Development of squamous cell cancer of head and neck (SCCHN) is associated with human papillomavirus (HPV) infection, which in turn is closely related with expression of $p16^{INK4A}$. Loss of $p16^{INK4A}$ expression by deletion, mutation, or hypermethylation is common in SCCHN. We here evaluated $p16^{INK4A}$ as a prognostic marker of treatment response and survival in our SCCHN patients with laryngeal, hypopharyngeal or nasopharyngeal cancers. Materials and Methods: 131 patients diagnosed with SCCHN between January 2,2006 and July 17, 2010 were examined for $p16^{INK4A}$. The median age was 60 years (15-82 years). Fifty one patients were stage I-II and 80 were stage III-IV. Immunohistochemical expression of $p16^{INK4A}$ was analyzed in pretreatment paraffin-embedded tumor blocks. The influence of $p16^{INK4A}$ status on disease-free survival, and overall survival after treatment was evaluated. Results: $p16^{INK4A}$ positivity was found in 58 patients (44%). Tumor-positivity for$ p16^{INK4A}$ was correlated with improved disease free survival (70.1 months vs 59 months) and improved overall survival (2, 3 and 5-year values; 77% vs 72%, 70% vs 63% and, 63% vs 55%; respectively). On multivariate analysis, stage was determined as independent prognostic factor for disease-free survival. Conclusions: Stage was the major prognostic factor on treatment response and survival in our patients. $p16^{INK4A}$ status predicts better outcome in laryngeal, hypopharyngeal or nasopharyngeal cancer cases treated with surgery plus adjuvant radiochemotherapy as well as with definitive radiation therapy and/or chemotherapy.

Sonographic Findings of Second Branchial Cleft Cysts; Variable Appearances and Helpful Diagnostic Findings (제2새열 낭종의 초음파 소견;다양한 소견들과 진단에 도움이 될 만한 소견들)

  • Objectives:The purpose of this study was to evaluate the variable sonographic features in addition to classic findings and to find the helpful characteristic findings for diagnosis of branchial cleft cysts. Subjects and Methods:We retrospectively analyzed the sonographic finding of 16 histopathologically proven branchial cleft cysts. Assessment involved the following variables : location related to carotid artery, size, cyst wall, border, margin, shape, internal echotexture, post echoic enhancement, orientation of longitudinal axis relative to the long axis of the neck, and tapering edge. Results:Most branchial cleft cysts were seen as well defined(15/16), oval shape(13/16), smooth inner (12/16) and outer margin(13/16), and post echoic enhancement(16/16). Their diameter varied from 1 to 6cm (mean 3.3cm). The branchial cleft cysts showed variable internal echopatterns;hyperechogenicity or pseudosolid appearance(1/16), heterogeneous echogenicity(4/16) in addition to classic form of anechoic(5/16) or hypoechoic internal echogenicity(6/16). Longitudinal axis of most branchial cleft cysts were arranged in the direction of the long axis of the neck(13/16) and some branchial cleft cysts had tapering edge(6/16). Conclusion:If both atypical sonographic findings and location are showed, longitudinal arrangement of long axis and tapering edge of branchial cleft cysts are helpful findings for differential diagnosis.

Association between p16 Promoter Methylation and Thyroid Cancer Risk: A Meta-analysis

  • Background: The aim of the meta-analysis was to derive a more precise assessment of the association between p16 promoter methylation and thyroid cancer risk. Materials and Methods: The PubMed, Web of Science databases and Chinese CNKI were searched for relevant articles. Ultimately, seventeen case-control studies were included with a total of 804 thyroid cancer cases and 487 controls analysis by R Software (R version 3.1.2) including meta. Crude odds ratios with 95% confidence intervals were calculated using the random-effects model which were used to assess the strength of relationship between p16 methylation and lung carcinogenesis. Funnel plots were carried out to evaluate publication bias. Results: The meta-analysis results showed that the frequency of p16 promoter methylation in cancer tissue/blood was significantly higher than that normal tissue/blood (OR=5.46, 95%CI 3.12-9.55, P<0.0001) by random effects model with small heterogeneity. Conclusions: Thus, p16 promoter methylation may be associated with thyroid cancer risk.

  • Background: The normal functions of the cell cycle inhibitor p16INK4a are frequently inactivated in many human cancers. Over 80% of hepatocellular carcinoma (HCC) cases lack a functional p16/Rb pathway. p16/Rb pathway, as well as p53 pathway, is considered as one of key components of tumor suppression. Methods: To study the roles of p16INK4a in HCC, a stable cell line expressing exogenous p16 was generated from SNU-449 hepatocellular carcinoma cells lacking endogenous p16, and suppression subtractive hybridization (SSH) was performed in parallel with the control cells. Results: 1) SSH identifies fibronectin (FN1), crystallin ${\alpha}B$ (CRYAB), Rac1, WASP, RhoGEF, and CCT3 as differentially-expressed genes. 2) Among the selected genes, the up-regulation of FN1 and CRYAB was confirmed by Northern blot, RT-PCR and by proteomic methods. Conclusion: These genes are likely to be associated with the induction of stress fiber and stabilization of cytoskeleton. Further studies are required to clarify the possible role of p16 in the signal transduction pathway.

  • A thermotropic liquid crystalline polyester (TLCP) containing a side group was, synthesized from ethoxyhydroquinone and bromoterephthalic acid. Intercalation of TLCP in layered clays is accomplished by heating the polymer with hexadecyl ammonium-montmorillonite ($C_{16}$) above melting transition temperature ($T_m$). Liquid crystallinity of the TLCP/$C_{16}$-hybrid was observed up to 6 wt% $C_{16}$-MMT. Some of the $C_{16}$-MMTs in TLCP were highly dispersed in a nanometer scale, but some of them were agglomerated. Thermal and morphological properties of the nanocomposites were examined by differential scanning calorimetry (DSC), thermogravimetric analyzer (TGA), polarized optical microscope, and electron microscopes (SEM and TEM).

  • In this paper, we investigate the most commonly adopted networking technology for Network Centric Warfar (NCW), namely Link-16 in the perspectives of physical, data link, and network layers (specifically, strategic message exchanges) and derive its merits with some constraints therein. Furthermore, we perform a quantitative evaluation on its communication capacity, focusing on whether Link-16 can indeed meet the capacity requirements for major operations, based on the time slot structure, NPG, and multi-netting concepts.

  • Purpose: This study was performed to determine the inhibitory effect of Polygonum multiflorum(PM) on melanin synthesis in B16F10. Methods: The Inhibitory effects of Polygonum multiflorum(PM) on melanin synthesis were determined by in-vitro assay. To elucidate inhibitory effects of Polygonum multiflorum on melanin synthesis, we determined the melanin release and melanin production in B16F10. And to investigate the action mechanism, we assessed the gene expression of tyrosinase, TRP-1, TRP-2, MMP-2, PKA, PKC, ERK-1 ERK-2, AKT-1, MITF in B16F10. Results: 1. PM inhibited melanin-release, melanin production in B16F10. 2. PM inhibited tyrosinase activity in vitro and in B16F10. 3. PM suppressed the expression of tyrosinase, TRP-1 in B16F10. 4. PM suppressed the expression of PKA in B16F10. 5. PM suppressed the expression of ERK-1, ERK-2, AKT-1 in B16F10. 6. PM suppressed the expression of MITF in B16F10. Conclusion: From these results, it may be concluded that PM possesses the antimelanogenetic effects.

A Study on the Melanin Synthesis Inhibition and Whitening Effect of Bombysis Corpus (백강잠의 멜라닌 생성 억제와 미백효과에 관한 연구)

  • Objective : This study was performed to assess the whitening effect of Bombysis Corpus on melanin synthesis. Methods : The whitening effects of Bombysis Corpus were examined by in vitro melanin production assay. We assessed inhibitory effects of Bombysis Corpus on melanin-release from B16F10, on melanin production in B16F10, on mushroom tyrosinase activity in vitro, on tyrosinase activity in B16F10, effect of Bombysis Corpus on the expression tyrosinase, TRP-1, PKA, ERK-1 ERK-2, AKT-1, MITF in B16F10. Results : 1. Bombysis Corpus inhibited melanin-release, melanin production in B16F10. 2. Bombysis Corpus inhibited tyrosinase activity in vitro and in B16F10. 3. Bombysis Corpus suppressed the expression of tyrosinase, TRP-1 in B16F10. 4. Bombysis Corpus suppressed the expression of PKA in B16F10. 5. Bombysis Corpus suppressed the expression of ERK-1, ERK-2, AKT-1 in B16F10. 6. Bombysis Corpus suppressed the expression of MITF in B16F10. Conclusion : The study shows that Bombysis Corpus inhibited melanin production on the melanogenesis.

  • Link-16 is a up-to-date tactical data link used in the armed forces of U.S. and NATO. In order to guarantee the required capabilities of anti-jamming and low probability of detection, Link-16 uses the techniques of frequency hopping and spreading code etc. In this paper, under Nakagami fading channels, we ananlyze the symbol error rates (SERs) of JTIDS/Link-16-type waveform based on both the single- and double-pulse schemes by considering the partial band noise jamming and the pulse jamming models, respectively.

Flutter Analysis of F-16 Aircraft Using Test Modal Data (시험 모달 데이터를 이용한 F-16 항공기의 플러터 해석)

  • Flutter analyses are performed for the KF-16D aircraft with brand new ALQ-X ECM pod. A flutter analysis method using test modal data is proposed and validated using published F-16 modal data and flutter analysis results. Ground vibration test is performed for KF-16D stands on its landing gears. Attained modal data are transformed to free-free condition of KF-16D aircraft with ALQ-X pod and ALQ-119 pod, respectively. As the results of comparison of flutter analyses, ALQ-X is cleared to be operated in the flight envelope authorized for existing ECM pods.
