옥수수 수염의 휘발성 성분 (Volatile Components of Cornsilk(Zea mays L.))

  • Volatile components of Cornsilk(Zea mays L.) were isolated by purge and trap headspace method and were analyzed by GC and GC/MSD. A total of 44 components were identified in the cornsilk volatile coponents including 9 alcohols 7 aldehydes and ketones 14 terpenes and terpene alcohols 3 pyrazines 5 hydrocarbons and 6 miscellaneous components. The major components were 2-propanol(8.08%) pen-tanol(1.82%) hexanol(2.86%) hexanal(3.68%) heptanal(7.40%) nonanal(7.93%) decanal (2.04%) $\alpha$-copaene(2.20%) limonene(1.68%) $\alpha$-selinene(1.03%) $\beta$-selinene(1.03%)

치주적으로 건강한 한국인에서 IL-1 ${\beta}$ 유전자의 유전자 다형성 발생빈도에 관한 연구 (Genetic polymorphisms of the IL-1 ${\beta}$ genes in periodontally healthy Korean population)

  • Interleukine-1(IL-1)은 여러 가지 기능을 가진 싸이토카인으로써 미생물에 대한 면역반응을 일으킨다. IL-1의 유전자 다형성과 치주질환과의 관련성에 대한 많은 연구가 있어왔지만, 대부분이 백인을 대상으로 한 연구였다. 이후 중국인과 일본인을 대상으로 한 연구에서 IL-1의 유전자 다형성의 분포가 인종간에 차이를 보인다는 점이 발견되었다. 이번 연구에서는 치주적으로 건강한 한국인에서 IL-1${\beta}$-511, IL-1${\beta}$+3954, IL-1RN에 대한 유전자형의 분포를 조사하고자 하였다. 서울대학교 치과병원에 근무하는 치과의사, 치과위생사, 간호조무사 및 서울대학교 치과대학 4학년 학생 중 치주낭 깊이와 부착소실이 4mm 이하인 치주적으로 건강한 한국인 65명을 대상으로 하였다. IL-1${\beta}$-511, IL-1${\beta}$+3954, IL-1RN의 유전자 다형성은 분리한 DNA에 각 대립유전자에 특이성을 지닌 primer를 넣고 PCR(Polymerase Chain Reaction)법을 이용하여 증폭시킨후 전기영동법을 이용하여 각 대립유전자의 존재를 확인함으로써 결정하였다. IL-1${\beta}$-511 대립유전자 11, 대립유전자 12, 대립유전자 22의 유전자형에 대하여 각각 23.1%, 49.2%, 26.2%의 분포를 보였다. IL-1${\beta}$+3954의 유전자 다형성은 대립유전자 11, 대립유전자 12의 유전자형에 대하여 각각 89.2%, 10.8%의 분포를 보였으며, 대립유전자 22의 유전자형을 갖는 사람은 한명도 발견되지 않았다. IL-1RN의 유전자형은 5가지의 대립유전자 중에서 1, 대립유전자 2, 대립유전자 4만일 발견되었으며, 대립유전자 11, 대립유전자 12, 대립유전자 14의 유전자형이 86.2%, 12.3%, 1.5%로 분포하였다. 이를 바탕으로 각 대립유전자의 발생빈도 계산한 결과 IL-1${\beta}$-511에서는 대립유전자 1과 2의 비율이 거의 유사하였으나 (47.7%, 52.3%), IL-1${\beta}$+3954, IL-1RN에서는 대립유전자 1이 90%이상 발견되었으며, 또한 대립유전자 1외의 다른 대립유전자가 발견된 경우, 모두 이형접합체였다. 이 연구는 IL-1${\beta}$-511, IL-1${\beta}$+3954, IL-1RN에 대한 유전자형의 분포를 조사한 것으로 한국인에서 이들 유전자의 유전자형의 분포는 백인에서의 분포와 차이를 보이고 있었다. 이후 치주질환자의 유전자형 분포와의 비교로 치주질환과 IL-1${\beta}$-511, IL-1${\beta}$+3954, IL-1RN의 유전자다형성과의 관련성에 관한 추가적인 연구가 필요할 것으로 여겨진다.

Cloning and Expression of Thermostable $\beta$-Glycosidase Gene from Thermus filiformis Wai33 A1 in Escherichia coli and Enzyme Characterization

  • A thermostable $\beta$-glycosidase gene, tfi $\beta$-gly, was cloned from the genomic library of Thermus filiformis Wai33 A1. ifi $\beta$-gly consists of 1,296 bp nucleotide sequence and encodes a polypeptide of 431 amino acids. It shares a strong amino acid sequence similarity with the $\beta$-glycosidases from other Thermus spp. belonging to the glycosyl hydrolase family 1. In the present study, the enzyme was overexpressed in Escherichia coli BL21 (DE3) using the pET21b(+) vector system. The recombinant enzyme was purified to homogeneity by heat treatment and a $Ni^{2+}$-affinity chromatography. Polyacrylamide gel electrophoresis (PAGE) showed that the recombinant Tfi $\beta$-glycosidase was a monomeric form with molecular mass of 49 kDa. The temperature and pH range for optimal activity of the purified enzyme were 80- $90^{\circ}C$ and 5.0-6.0, respectively. Ninety-three percent of the enzyme activity was remained at $70^{\circ}C$ after 12 h, and its half-life at $80^{\circ}C$ was 6 h, indicating that Tfi $\beta$-glycosidase is highly thermostable. Based on its K_m$, or $K_{cat}K_m$, ratio, Tfi $\beta$-glycosidase appeared to have higher affinity for $\beta$-D-glucoside than for $\beta$-D-galactoside, however, $K_{cat} for \beta$-D-galactoside was much higher than that for $\beta$-D-glucoside. The activity for lactose hydrolysis was proportionally increased at $70^{\circ}C$ and pH 7.0 without substrate inhibition until reaching 250 mM lactose concentration. The specific activity of Tfi TEX>$\beta$-glycosidase on 138 mM lactose at $70{^\circ}C$ and pH 7.0 was 134.9 U/mg. Consequently, this newly cloned enzyme appears to have a valuable advantage of conducting biotechnological processes at elevated temperature during milk pasteurization in the production of low-lactose milk.

삽주의 휘발성 향기성분 (Volatile Flavor of Atractylodes japonica koidzumi)

  • 삽주(Atractylodes japonica Koidzumi)의 약명인 창출(蒼朮)과 백출(白朮)로 휘발성 향기성분을 조사하기 위하여 n-pentane/diethyl ether용매로 사용하여 SDE방법으로 정유성분을 분리한 다음 GC/MS를 이용하여 성분을 확인하였다. 백출에서 30종 그리고 창출에서 28종류의 휘발성 향기성분을 확인하였으며, 백출의 향기성분을 관능기별로 보면 hydrocarbons 18종, carbonyls 2종, alcohols 5종, esters 5종이고, 창출은 hydrocarbons 14종, carbonyls 6종, alcohols 4종, esters 3종, acids 3종으로 각각 조사되었고, 백출 및 창출의 주요 향기성분으로서는 furanodiene 성분이 각각 27.9%, 15.7%, $\alpha$-cyperone 성분이 8.1%, 22.5%, alloaromadendrene 성분이 2.9%, 4.7% (1,1-biphenyl)-4-carbon aldehyde 성분은 창출에서만 8.7%로 확인되었다. 백출에서 limonene, p-cymene, p-cymen-8-ol, (1,1-biphenyl)-4-carbox aldehyde 등을 포함한 10종의 성분이 확인된 반면에 창출에서는 확인되지 않았고, 반면에 창출에서 $\alpha$-copaene; isocaryophyllene, $\beta$ -himachalene, germacrene B 등을 포함한 8종의 성분이 확인된 반면에 백출에서는 확인되지 않았다.

The Reaction of Superoxide with Carbohydrate Sulphonates

  • The reaction between methyl 2,3-di-O-benzyl-4,6-di-O-mesyl-${\alpha}$-D-glucopyranoside (1b) and potassium superoxide resulted in hydrolysis, and gave methyl 2,3-di-O-benzyl-${\alpha}$-D-glucopyranoside (1) as a sole product. When the reaction was performed with a vicinal dimesylate, methyl 4,6-O-benzylidene-2,3-di-O-mesyl-${\alpha}$-D-altropyranoside (4b), again the hydrolysis product, methyl 4,6-O-benzylidene-${\alpha}$-D-altropyranoside (4) was obtained. However, the reaction of potassium superoxide with another vicinal dimesylate, methyl 4,6-O-benzylidene-2,3-di-O-mesyl-${\alpha}$-D-glucopyranoside (3b), nucleophilic displacement took place to afford methyl 4,6-O-benzylidene-${\alpha}$-D-altropyranoside (4). Apparently different results from two trans vicinal dimesylates, 3b and 4b are explained by the transient formation of epoxides, methyl 2,3-anhydro-4,6-O-benzylidene-${\alpha}$-D-allopyranoside (8) and methyl 2,3-anhydro-4,6-O-benzylidene-${\alpha}$-D-mannopyranoside (9) by $KO_2$. The reaction between the allo epoxide 8 and $KO_2$ gave altro 4. The manno epoxide 9 also afforded altro 4 as the major product. Facile epoxide formation by the reaction of a vicinal dimesylate and superoxide was also observed with 3-O-benzyl-1,2-O-isopropylidene-5,6-di-O-mesyl-${\alpha}$-D-glucofuranose: 5,6-anhydro-3-O-benzyl-1,2-O-isopropylidene-${\beta}$-L-idofuranose was obtained.

Isolation and Characterization of Antioxidative Compounds from the Aerial Parts of Angelica keiskei

  • Ethyl acetate-soluble neutral fraction of hot water extracts from the aerial parts of Angelica keiskei showed a 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity. Six antioxidative compounds were purified and isolated by various chromatographic procedures. Based on the analyses of FAB-MS and NMR, the isolated compounds were structurally elucidated as luteolin 7-O-${\beta}$-D-glucopyranoside (1), quercetin 3-O-${\beta}$-D-galactopyranoside (2), quercetin 3-O-${\beta}$-D-glucopyranoside (3), quercetin 3-O-${\alpha}$-D-arabinopyranoside (4), kaempferol 3-O-${\alpha}$-D-arabinopyranoside (5), and luteolin 7-O-rutinoside (6). The glycosides of flavonols and luteolin showed DPPH radical-scavenging activity. One molecule of 2, 3, 4, 6, 1, and 5 scavenged 4.2, 4.2, 4.1, 2.5, 2.2, and 1.4 molecules of DPPH radical, respectively.

간호대학생의 자아탄력성과 자기효능감이 전공만족도에 미치는 영향 (Influence of Ego-Resilience and Self-Efficacy on Satisfaction in major of Nursing Student)

  • Purpose: The purpose of this study was to investigate the influence of ego-resilience and self-efficacy on satisfaction in major of nursing student. Method: Data were collected from 275 nursing students by means of self reported questionnaires about ego-resilience, self-efficacy and satisfaction in major from December 3 to 20, 2012. Results: There were positive correlations between ego-resilience, self-efficacy and satisfaction in major. The optimistic attitude in ego-resilience (${\beta}$=.56, p<.001), an associate's degree (${\beta}$=.38, p<.001), suitable to aptitude (${\beta}$=.17, p<.001), social self-efficacy (${\beta}$=.14, p=.039) and high employment rate (${\beta}$=.12, p=.033) were significant predictors of satisfaction in major. Conclusion: It is necessary to develop an effective intervention program to improve nursing student's ego-resilience, self-efficacy and satisfaction in major of nursing student.

Norfloxacin과 ${\beta}-Cyclodextrin$간의 Inclusion Complex에 관한 약제학적 연구 (Pharmaceutical Studies on Inclusion Complex of Norfloxacin with ${\beta}-Cyclodextrin$)

  • To increase the bioavailability of norfloxacin, inclusion complex of antimicrobial agent norfloxacin with ${\beta}-Cyclodextrin$ was prepared and studied by the solubility method, spectrophotometric methods(UV, IR, $^1H-NMR$), differential thermal analysis, powder X-ray diffractometry, the physical properties, the antimicrobial activity, DNA binding and in situ recirculation technique. The conclusions are summerized as following; 1) The inclusion complexation was identified by means of solubility, spectrophotometry(UV, IR, NMR), DTA and X-ray diffraction. 2) The molar ratio of $norfloxacin-{\beta}-cyclodextrin$ complex was 1 : 1. 3) The stability constant of $norfloxacin-{\beta}-cyclodextrin$ complex was $21.5\;M^{-1}$, and both true and apparent partition coefficients of the inclusion complex were larger than those of norfloxacin. 4) The time required to dissolve 60% $(T_{60}%)$ of the inclusion complex was 120 min. in distilled water and in the artificial intestinal juice, while norfloxacin did not reach to 60% dissolution within 120 min. 5) The antimicrobial activity of the inclusion complex against Pseudomonas aeruginosa, Klebsiella pneumoniae and Staphylococcus aureus showed no significant difference compared to that of norfloxacin alone. 6) Studies on binding properties between the inclusion complex and norfloxacin alone to DNA according to equilibrium dialysis showed no significant differency. 7) In situ absorption rates (Ka) of inclusion complex and norfloxacin alone were 0.229 and $0.102hr^{-1}$, respectively.

The Crystal Structure of Tolfenamic Acid $(C_{14}H_{12}ClNO_2)$, an Antiinflammatory Fenamate

  • The structural analysis of tolfenamic acid, 2-[(3-chloro-2-methylphenyl)-amino]benzoic acid, was performed by single crystal X-ray diffraction technique. The compound was recrystallized from a mixture of ether and toluene in triclinic, space group $P2_1/c, \;with\; \partial=3.914(1), \; b=22.\; 020(2), \; c=14.271(1)\;{\AA}, \beta.=94.68(1)^{\circ}, $ and Z=4. The calculated density is $1.418 g/cm^3$. The structure was solved by the direct method and refined by full matrix least-squares procedure to the final R value of 0.039 for 1773 independent reflections. In the molecule, carboxyl group at the anthranilic acid is coplanar to the phenyl ring. The dihedral angle between the two aromatic rings of the molecule is $44.2^{\circ}$ The molecules are dirnerized through the intermolecular hydrogen bonds at the carboxyl group in the crystal.

Cinnamonitrile의 알카리 가수분해(加水分解) 반응(反應)메카니즘 (II) (Kinetics and Mechanism of Alkalie Hydrolysis of Cinnamonitrile (II))

  • pH 7.0~14.0의 중성에서 강염기성 용액에 이르는 넓은 pH 범위에서 Cinnamonitrile의 알카리 가수분해 반응속도 상수를 $25^{\circ}C$의 50% methanol 물 혼합용매로 부터 측정하여 실험사실을 잘 설명할 수 있는 반응 속도식을 유도하여 가수분해 반응 메카니즘을 제안하고, 반응 속도론적으로 고찰 검토하였으며 (Z)형의 형태에 관하여 분자의 안정성과 반응성을 알아보기 위하여 cinnamonitrile의 분자궤도 함수를 EHT와 CNDO/2방법으로 계산하였다. (1) Cinnamonitrile의 전체 에너지 계산 결과로부터 ${\theta}_1$ 회전에 따르는 형태의 안정도는 (E)형을 포함하여 (E)-planar > (E)-gauche > (Z)-gauche > (Z)-planar이였고 (E)-gauche와 (Z)-gauche의 에너지 차는 1.94Kcal/mole이였다. 따라서 반응은 가장 안정한 (E)-planar의 $C_7({\alpha})$ 원자에 chydroxide 이온이나 물분자의 첨가로 ${\alpha}C-{\beta}C$ 결합이 분열하게 된다. (2) pH7.0~14.0 범위에서 측정한 반응속도상수로 부터 유도된 전체 반응속도식은 다음과 같다. $$k_t=k_o+k^{\prime}[OH^-]=({\frac{1.41{\times}10^{-14}+1.21{\times}10^{-7}/[H_3O^+]}{2.65{\times}10^{-7}+1.64/[H_3O^+]})+9.14{\times}10^9/[H_3O^+]$$ (3) 분자 궤도함수의 계산 결과와 반응 속도식을 토대로 하여 pH10.0~14.0에서는 hydroxide 이온의 첨가로 가수분해가 진행되는 Scheme(I)과 같은 Michael형의 친핵성 첨가반응 메카니즘을 그리고 pH7.0~10.0사이의 중성 용액중에서는 물분자의 첨가로 반응이 진행되는 Scheme(II)와 같은 가수분해 반응메카니즘을 제안하였다. (4) pH10.0~12.0사이의 약 알칼리성 용액중에서는 Scheme(I)과 Scheme(II)의 두 반응이 서로 경쟁적으로 일어나는 대단히 복잡한 일련의 가수분해 반응이 진행됨을 알았으며 분해 생성물은 공히 benzaldehyde와 methylnitrile 그리고 acetic acid 이였다. 앞으로의 과제는 cinnamonitrile 의 ${\alpha}{\cdot}{\beta}$ 탄소불포화 이중결합에 미치는 치환기에 의한 자유에너지 직선성의 상관관계와 물 L-cysteine 및 hydrogene cyanide 등의 서로 다른 친핵체들과의 반응으로 부터 이들의 친핵 첨가반응성을 검토하고자 한다.
