맥문동 열수추출액을 이용한 인삼정과의 품질 특성 (Physicochemical Characteristics of Ginseng JungKwa Produced with Hot-Water Extract from Maegmundong (Liriope platyphylla tubers))

  • 인삼정과 제조 시 이용되는 당 용액 대신에 올리고당 함량이 높은 맥문동의 열수추출액을 이용하여 인삼정과를 제조한 후 품질 특성을 조사하였다. 맥문동 추출액으로 제조한 인삼정과는 가용성 무질소물인 탄수화물의 함량이 76.40%로 설탕으로 제조한 인삼정과의 80.01%에 비하여 약 4% 정도 낮았고, 조단백질 및 조회분 함량은 각각 1.46% 및 3.49%로 일반인삼정과의 0.59% 및 0.96%에 비하여 약 3배 이상 높았다. 맥문동 열수추출액을 이용한 인삼정과에서 유리당 조성 및 함량은 fructose가 32.42%로 가장 많았고, 그 외 올리고당이 16.91%, 설탕 및 이당류가 13.91%, glucose가 13.16%이었으며, Rh2를 비롯한 진세노사이드 11종이 검출되어 총 함량이 740.1 mg%로 일반 인삼정과(675.6 mg%)보다 더 높았다. 총 페놀화합물과 플라보노이드 함량은 각각 5.46과 0.016%로 일반인삼정과의 5.02와 0.014%보다 높았다. 맥문동 열수추출액으로 제조한 인삼정과의 항산화활성은 DPPH $IC_{50}$값이 인삼정과 중량기준으로 약 34.5 mg/mL 이었으며, hydrogen peroxide 소거능은 $50{\mu}g$/mL의 농도에서 92%의 소거능을 보여주었다.

QTL Scan for Meat Quality Traits Using High-density SNP Chip Analysis in Cross between Korean Native Pig and Yorkshire

  • We attempted to generate a linkage map using Illumina Porcine 60K SNP Beadchip genotypes of the $F_2$ offspring from Korean native pig (KNP) crossed with Yorkshire (YS) pig, and to identify quantitative trait loci (QTL) using the line-cross model. Among the genotype information of the 62,136 SNPs obtained from the high-density SNP analysis, 45,308 SNPs were used to select informative markers with allelic frequencies >0.7 between the KNP (n = 16) and YS (n = 8) F0 animals. Of the selected SNP markers, a final set of 500 SNPs with polymorphic information contents (PIC) values of >0.300 in the $F_2$ groups (n = 252) was used for detection of thirty meat quality-related QTL on chromosomes at the 5% significance level and 10 QTL at the 1% significance level. The QTL for crude protein were detected on SSC2, SSC3, SSC6, SSC9 and SSC12; for intramuscular fat and marbling on SSC2, SSC8, SSC12, SSC14 and SSC18; meat color measurements on SSC1, SSC3, SSC4, SSC5, SSC6, SSC10, SSC11, SSC12, SSC16 and SSC18; water content related measurements in pork were detected on SSC4, SSC6, SSC7, SSC10, SSC12 and SSC14. Additional QTL of pork quality traits such as texture, tenderness and pH were detected on SSC6, SSC12, SSC13 and SSC16. The most important chromosomal region of superior pork quality in KNP compared to YS was identified on SSC12. Our results demonstrated that a QTL linkage map of the $F_2$ design in the pig breed can be generated with a selected data set of high density SNP genotypes. The QTL regions detected in this study will provide useful information for identifying genetic factors related to better pork quality in KNP.

  • Soybean mosaic virus(SMV)는 potyvirus 속에 속하며, 모자이크, 괴사, 기형 등의 병징을 야기하고 국내에서는 11개 계통(G1 to G7, G5H, G6H, G7H, G7a)이 보고되어있다. Reverse transcription loop-mediated isothermal amplification(RT-LAMP) 방법은 등온에서 유전자 증폭이 가능하게 하며, 이 방법은 PCR 과정이나 전기영동 없이도 바이러스에 감염된 식물을 검출할 수 있는 이점이 있다. RT-LAMP의 최적반응 조건은 $58^{\circ}C$, 60분으로 확인되었다. 특이성 검정을 위해 콩에서 발생하는 여러 바이러스들과 보유중인 SMV의 9 계통에서 그 특이성을 확인하였다. 그 결과 SMV에 대한 RT-LAMP primer들의 종 특이성이 확인되었으며, SMV의 계통들에 대해서도 적용이 가능한 것으로 확인되었다. 항온수조와 heating block과 같은 간편한 등온 장치에서 재현성을 확인하기 위해 Thermocycler 기기와 비교하여 증폭 여부를 확인한 결과 반응의 차이는 나타나지 않았다. RTLAMP 반응 이후, 반응물을 전기영동과 SYBR Green I을 이용하여 자연광과 UV광에서 증폭 여부를 확인하였다. 그 결과 전기 영동, 자연광, portable UV light와 UV transilluminator에서 모두 반응이 확인되었다.

Effects of Different Sources of Dietary Chromium on Growth, Blood Profiles and Carcass Traits in Growing-finishing Pigs

  • This study was carried out to evaluate the effects of dietary supplementation of different sources of chromium on growth performance, blood profile and carcass trait in growing-finishing pigs. A total of 200 growing pigs (Landrace${\times}$Yorkshire)${\times}$Duroc, average initial weight 8.5 kg) were allotted to 5 treatments with 4 replicates per treatment and 10 pigs per replicate. Five treatments were designated as follows according to the source of chromium. i) Control (No chromium): corn-soybean meal based basal diet, ii) $CrCl_{3}$: control diet+200 ppb Cr as $CrCl_{3}$, iii) CrPic: control diet+200 ppb Cr as Cr picolinate, iv) CrMet-1: control diet+100 ppb Cr as Cr methionine, and v) CrMet-2: control diet+200 ppb Cr as Cr methionine. After the feeding trial, three pigs per replicate (12 pigs per treatment) were slaughtered for the evaluation of carcass traits. Average daily gain (ADG), average daily feed intake (ADFI), and feed: gain ratio (F/G) were not different (p>0.05) among dietary Cr sources. However, whole-period ADG of pigs fed CrPic, CrMet-1 and CrMet-2 diets was higher (p<0.05) than for the control diet. Nutrient digestibility was not different (p>0.05) among dietary Cr sources, but the nutrient digestibility of pigs fed CrPic, CrMet-1 and CrMet-2 diets was higher (p<0.05) than for the control diet. BUN level decreased with more magnitude (p<0.05) in pigs fed Cr during the 20 to 50 kg period. Although both serum cholesterol and triglyceride were different (p<0.05) among treatments, there was no consistent response that could be related to the dietary Cr sources regardless of growth phase. However, the overall data suggested that serum cholesterol level increased as BW of pigs increased. Blood total protein (TP) increased (p<0.05) in pigs fed Cr only during the 90-110 kg phase, and blood creatinine (Creat) level was higher in $CrCl_{3}$ and CrPic treatments than in the control only during the 90-110 kg phase. Backfat thickness was thinner (p<0.05) in pigs fed CrMet-2 than in the control treatment. Therefore, lean percentage was higher (p<0.05) in CrMet-2 than in control pigs. However, dressing percentage and Longissimus muscle area (LMA) were not different (p>0.05) among treatments. In conclusion, dietary supplementation of 200 ppb Cr, via either CrPic or CrMet, improved pig growth performance and nutrient digestibility. Moreover, dietary CrMet supplementation for the growing-finishing pig is evidently remarkable for improving both lean percentage of the carcass and backfat thickness.

Melanocyte-stimulating Hormone Receptor (MC1R) Genotype and Its Effects on Coat Color in Korean Jindo Dogs

  • The Jindo dog is a Korean natural monument and is recognized by the Fédération Cynologique Internationale. A prominent feature is the diverse coat color within the breed. To analyze the genetic basis of variation in the Jindo coat color, we sequenced the protein-coding regions of the melanocortin 1 receptor gene (MC1R). The MC1R coding sequence was determined from 154 dogs in five breeds (Jindo, Labrador Retriever, English Springer Spaniel, Belgian Malinois, and German Shepherd). To confirm the genetic structure of sampled populations, we tested for Hardy-Weinberg equilibrium (HWE) and computed $F_{st}$ The sample populations did not significantly deviate from HWE. $F_{st}$ was 0.02 between white and fawn Jindo dogs; this was lower than $F_{st}$ between breeds. Six single nucleotide polymorphisms (SNPs) were detected in the MC1R coding region. Among the six SNPs, five were non-synonymous (S90G, T105A, Q159P, M264V, and R306ter) and one was synonymous SNP (Y298Y). From the SNPs, we predicted four haplotypes (H1, H2, H3, and H4) for Jindo MC1R. Jindo dogs had different haplotypes corresponding to different coat colors. H1 was frequently observed in white Jindo dogs with an odds ratio of 5.03 (95% CI: 2.27-11.18, p<0.0001), whereas H2 and H4 were observed only in fawn Jindo dogs. Our findings indicate that SNP haplotype can influence coat color. Knowledge of MC1R haplotypes can help discriminate white and fawn coats in Jindo dogs. We hope this report will trigger more research into the genetics of this traditional Korean dog and will be a reference for dogs of Asian origin. Also, our results will provide a useful genetic marker for Jindo dog breeders who have selected for specific colors.

증포 처리한 삼채 뿌리의 이화학적 특성 및 항산화 활성 (Physicochemical Properties and Antioxidant Activities of Steam-Dried Allium hookeri Root)

  • 증포 처리가 삼채 뿌리의 품질 특성 및 항산화 활성에 미치는 영향을 조사하였다. 증포 처리한 삼채 뿌리는 생 삼채 뿌리보다 황 함량이 각각 2.0배와 2.1배 감소하였다. pH와 총당은 증포 처리에 의해 감소하였으나 적정산도와 갈변도는 증가하였다. 색도의 경우 L과 b 값은 감소하였으나 a 값은 증가하였다. 열수 추출물의 항산화 활성은 증포 4회가 DPPH radical assay, ABTS radical assay 및 reducing power에서 $EC_{50}$ 값이 각각 0.44 mg/mL, 9.01 mg/mL 및 0.48 mg/mL로 가장 낮게 나타났으나 TPC($34.47{\mu}g/mg$)와 갈변도(2.05 at 280 nm와 0.20 at 420 nm)는 가장 높게 나타났다. 이들 성분과 각 항산화 활성에 대한 $EC_{50}$ 값의 $R^2$는 0.87 이상의 높은 값을 나타내었다. 결과적으로 증포 처리는 삼채 뿌리의 항산화 활성을 증가시키는 데 효과적인 가공 방법으로 판단된다.

흑미 미강 쿠키의 항산화활성 및 품질특성 (Antioxidant Activity and Quality Characteristics of Black Rice Bran Cookies)

  • 본 연구에서는 흑미 미강 분말을 첨가(0, 1, 3, 5, 7, 9%)한 쿠키를 제조하여 쿠키의 항산화 활성을 입증하고 품질특성을 측정하였다. 흑미 미강 분말의 총 페놀화합물 함량은 $11.81{\pm}0.03$ mg GAE/g이며, 이를 쿠키에 첨가한 경우 시료의 첨가량에 비례하여 증가하였다. 제조된 쿠키의 DPPH radical 소거능을 측정한 결과 흑미 미강 분말의 첨가량에 따라 항산화 활성도 유의적으로 증가하는 결과를 나타내었다. 항산화 활성의 기능성과 함께 제품의 적합성 판단을 위해 흑미 미강 분말과 쿠키의 품질평가를 실시하였다. 일반성분 분석의 결과 흑미 미강에는 섬유소와 지방의 성분이 많은 부분을 차지하고 있음을 알 수 있었다. 지방산의 분석에서 흑미 미강은 약 80%의 불포화지방산을 함유하고 있었으며 그 성분은 주로 oleic acid와 linoleic acid로 나타났다. 또한 아미노산의 분석에서 대조군에 비해 7% 첨가 쿠키가 높은 아미노산 함량을 나타내었다. 반죽의 밀도는 흑미 미강 첨가량이 증가할수록 높게 나타났고 pH는 감소하였다. 쿠키의 수분함량에는 유의적 차이가 나지 않았으며, 퍼짐성 지수는 흑미 미강 분말의 첨가량이 많아질수록 증가하였고, 손실율과 팽창률은 감소하였다. 쿠키의 색도는 흑미 미강 분말의 첨가량이 증가할수록 L값과 b값이 낮아지고, a값이 높아졌으며, 경도는 감소하는 경향을 나타내었다. 쿠키의 기호도 검사(전반적인 기호도, 외관, 향, 맛, 조직감)에서 5%와 7% 흑미 미강 쿠키가 유의적으로 높게 나타났다. 특성 강도 검사(흑미 미강 향, 고소한 맛, 부드러운 정도, 삼킨 후의 느낌)에서는 흑미 미강을 첨가한 쿠키에서 유의적으로 높은 강도를 보였으며, 느끼한 향은 대조군이 유의적으로 가장 강하다는 결과를 얻었다. 이러한 결과로 보아 쿠키에 흑미 미강분말을 첨가하는 것은 쿠키의 기호도를 증가시켜주고 동시에 총 페놀함량과 DPPH 라디칼 소거능을 높여주어 쿠키의 가치를 높일 수 있어 바람직할 것으로 사료되며, 쿠키 제조시 기호도가 가장 높고 항산화능이 좋았던 7% 흑미 미강 분말을 첨가하는 것이 적합할 것으로 생각된다.

Identification of Genes Involved in Primordial-primary Follicle Transition by Suppression Subtractive Hybridization

  • Recruitment of primordial follicles(PMF) is crucial for female fertility. however, factors and mechanisms that regulate this process is poorly understood. The present study was conducted to obtain an inclusive view of the gene expression and to identify novel factors and their pathways of regulating PMF arrest and/or growth initiation. Ovaries from one-day neonatal(consists of oocyte and PMF) and five-day old(consists of PMF and primary follicles, PRIF) mice were collected, either total RNA or mRNA was isolated, and suppression subtractive hybridization(SSH) was used to isolate and clone genes that differentially expressed in day 1 and day 5 ovaries. Confirmation that some of these genes are differentially expressed in PMF and/or in PRIF was accomplished by using laser captured microdissection(LCM), RT-PCR. in situ hybridization(ISH) and/or immunohistochemistry(IHC). In toto, 357 clones were sequenced and analyzed by BLAST and RIKEN program. Sequences of 330 clones significantly matched database entries while 27 clones were novel. Forty-two and 47 different genes were identified as differentially expressed in day 1 and day 5 ovaries, respectively, while 7 genes were expressed in both stages of ovaries. Day 5-subtracted library included several genes known as markers far growing follicles, such as ZP2, MATER, and fetuin. Among the genes with assigned functions, 23.8% was associated with cell cycle/apoptosis regulation, 7.1% with cellular structure, 11.9% with metabolism, 26.2% with signal transduction, and 31.0% with gene/protein expression in day 1; while 10.6%, 17.0%, 23.5%, 25.5%, and 23.4% in day 5, respectively. Genes such as GDF-8, Lats2, Septin2, and Weel were the highly expressed genes in PMF, while HSP84, Laminin2, MATER, MTi7, PTP, and Wrn were highly expressed genes in PRIF. We have successfully discovered list of genes expressed in day 1 and day 5 ovaries and confirmed that some of them are differentially expressed in PMF and/or PRIF. Gene expression profile from the present study would provide insight for the future study on the mechanism(s) involved in primordial-primary follicular transition. The Respective Effects of Shoot Height and Conservation Method on the Yield and Nutritive Value, and Essential Oils of Wormwood (Artemisia montana Pampan)

  • This study was conducted to evaluate the shoot height at which the yield and nutritive value of wormwood (Artemisia montana) is optimized in order to provide information on its potential to support animal production (Experiment 1). A second objective was to determine how the essential oil (EO) concentration in wormwood hay and silage differ (Experiment 2). In Experiment 1, Artemisia montana was harvested at five different shoot heights (20, 40, 60, 80 and 100 cm) from triplicate $1.8{\times}1.8m$ plots. Dry matter (DM) yield was measured at each harvest date and the harvested wormwood was botanically separated into leaf, stalk and whole plant fractions and analyzed for chemical composition and in vitro dry matter digestibility (DMD). Values for total digestible nutrients (TDN), digestible energy (DE) and metabolizable energy (ME) were subsequently calculated using prediction equations. Dry matter yields of stalk and whole plant increased linearly (p<0.001) and leaf yield increased quadratically (p<0.01) with shoot height, whereas the leaf/stalk ratio decreased linearly (p<0.001). As shoot height increased, there was a linear increase (p<0.001) in leaf DM, ether extract (EE) and neutral detergent fiber (NDF) contents and a quadratic increase (p<0.05) in leaf acid detergent fiber (ADF) and nitrogen free extract (NFE) contents, and stalk and whole plant DM (p<0.001), organic matter (OM, p<0.01 and p<0.05), NDF (p<0.001 and 0.05) and NFE (p<0.05) contents. However, there were decreases in leaf crude protein content (CP, quadratic, p<0.001) and stalk and whole plant EE content (linear, p<0.001), CP (quadratic, p<0.05) and ash (quadratic, p<0.05) contents. Digestibility of DM and TDN, and DE and ME value in leaves were not affected by increasing shoot height, but these measures linearly decreased (p<0.001) in stalk and whole plant. In Experiment 2, the hay had higher DM and CP concentrations, but lower EE concentration than the silage. Essential oil (EO) content in wormwood silage (0.49 g/100 g DM) was higher (p<0.05) than that in wormwood hay (0.32 g/100 g DM). Wormwood hay contained 25 essentail oils (EO) including camphor (10.4 g/100 g), 1-borneol (11.6 g/100 g) and caryophyllene oxide (27.7 g/100 g), and wormwood silage had 26 EO constituents including 3-cyclohexen-1-ol (8.1 g/100 g), trans-caryophyllene (8.6 g/100 g) and ${\gamma}$-selinene (16.8 g/100 g). It is concluded that the most ideal shoot height for harvesting wormwood is 60 cm based on the optimization of DM yield and nutritive value. Wormwood silage had a greater quantity and array of EO than wormwood hay.

Aberrant Expression of Markers of Cancer Stem Cells in Gastric Adenocarcinoma and their Relationship to Vasculogenic Mimicry

  • Background: Gastric cancer is the second leading cause of cancer-related death in Asia, and the majority type is gastric adenocarcinoma (GAC). Most GAC patients die of recurrence and metastasis. Cancer stem cells (CSCs) have been thought to be responsible for the initiation, development, metastasis, and ultimately recurrence of cancer. In this study, we aimed to investigate expression and clinical significance of CSCs markers, CD133 and Lgr5, and vasculogenic mimicry (VM) in primary GAC. Materials and Methods: Specimens from 261 Chinese patients with follow-up were analyzed for CD133, Lgr5 protein expression and VM by immunohistochemical and histochemical staining. The Pearson Chi's square test was used to assess the associations among the positive staining of these markers and clinicopathological characteristics. Postoperative overall survival time was were studied by univariate and multivariate analyses. Results: In GAC tissues, positive rates of 49.0%, 38.7%, and 26.8% were obtained for CD133, Lgr5, and VM, respectively. The mean score of microvessel density (MVD) was $21.7{\pm}11.1$ in GAC tissues. There was a significantly difference between the positive and negative groups. There was a positive relationship between the VM, the expression of CD133 and Lgr5, and the score of MVD and the grades of tumor, lymph node metastasis, TNM stages (all p<0.05). The overall mean survival time of the patients with CD133, Lgr5, VM, and MVD (${\geq}22$) positive expression was lower than that of patients with negative expression. The score of MVD, positive expression of CD133 and VM were independent prognostic factors of GAC (p<0.05). Conclusions: VM, and expression of CD133, Lgr5, and the score of MVD are related to grades of tumor, lymph node metastasis, TNM stages, and overall mean survival time. It is suggested that CSCs and VM could play an important role in the evolution of GAC.
