Synthesis and Preliminary Antimicrobial Screening of New Benzimidazole Heterocycle

  • A series of 2-methylbenzimidazole incorporated to different heterocycles through ethyl or carbamoylethyl groups at position 1 of benzimidazole were synthesized. Also 3-(2-methylbenzimidazol-1-yl)propanoic acid hydrazide incorporated with semicarbazides and thiosemicarbazides were prepared. Moreover, the triazole 5e underwent Michael addition and alkylation reaction. Some of the newly synthesized compounds showed considerable antimicrobial activity against gram positive, negative bacteria and yeast.

Synthesis of New Heterocycles Derived from 3-(3-Methyl-1H-indol-2-yl)-3-oxopropanenitrile as Potent Antifungal Agents

  • New thiazoline derivatives 7a-c, and thiophenes 9a-c linked to indole moiety were easily prepared via the reaction of the acrylamide derivative 3 with phenacyl bromides 4a-c, depending on the reaction conditions. In addition, the reaction of compound 3 with hydrazonoyl chlorides 11a-f afforded a series of 1,3,4-thiadiazole derivatives 13a-f. Moreover, coupling of 3-(3-methyl-1H-indol-2-yl)-3-oxopropanenitrile (2) with the diazonium salts of 3-phenyl-5-aminopyrazole 16 or 3-amino-1,2,4-triazole 17 gave the corresponding hydrazones 18 and 19, respectively. Cyclization of the latter hydrazones yielded the corresponding pyrazolo[5,1-c]-1,2,4-triazine and 1,2,4-triazolo[5,1-c]-1,2,4-triazine derivatives 20 and 21, respectively. The structures of the synthesized compounds were assigned on the basis of elemental analysis, IR, $^1H$ NMR and mass spectral data. All the synthesized compounds were tested for in vitro activities against certain strains of fungi such as Aspergillus niger, Aspergillus nodulans, Alternaria alternate. Compounds showed marked inhibition of fungal growth nearly equal to the standards.

오만둥이 된장찌개의 Aroma-active 화합물 (Aroma-Active Compounds in Omandungi (Styela plicata)-Doenjang (Soybean Paste) Stew)

  • Volatile compounds in Omangdungi (Styela plicata)-Doenjang (soybean paste) stew were analyzed using solvent-assisted flavor evaporation/gas chromatography/mass-selective detection/olfactometry (SAFE/GC/MSD/O) and aroma extract dilution analysis (AEDA). The GC/O analysis detected 37 volatile compounds, of which 32 were positively identified, and included 9 aldehydes, 5 alcohols, 4 aromatic hydrocarbons, 4 ketones, 3 esters, 3 N-containing compounds, 2 acids, 1 S-containing compound, and 1 furan. Nine aroma-active odorants ($\log_3FD{\geq}3.0$) in the sample included six compounds derived from Doenjang (3-methyl(thio)propanal, tetramethylpyrazine, 4-vinyl-2-methoxyphenol, 2-acetylpyrrole, butyric acid, and 2-methoxyphenol) and three compounds from Omangdungi (2-acetyl-2-thiazoline, 9-decanol, and 6-decenol). Three compounds derived from Omangdungi (9-decanol, 6-decenol, and 6-nonenol) were thought to enhance the seafood-like flavor of Omangdungi-Doenjang stew.

Antioxidative Properties and Flavonoids Contents of Matured Citrus Peel Extracts

  • We assessed various antioxidant activities, such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, 2,2'-azino-bis 3-ethylbenz-thiazoline-6-sulfonic acid (ABTS) radical, and reactive oxygen species (ROS) radical scavenging effect, along with antioxidant capacity, of soybean oil with added Citrus species peel extracts (CPEs). These extract oils showed higher radical scavenging effects than grape fruit seed extract, the natural antioxidant agent, did. When CPEs were added to soybean oil, they showed peroxide value (POV) and acid value (AV) increasing inhibition effects. Furthermore, none of the CPEs showed any cytotoxicity over the tested concentration range of 0.01-100 ppm. The major flavonoid contents of Citrus junos, as determined by HPLC, were naringin ($7.5\;{\mu}g/mg$) and neohesperidin ($7.5\;{\mu}g/mg$), and those of Citrus unshiu were narirutin ($3.13\;{\mu}g/mg$) and hesperidin ($1.97\;{\mu}g/mg$). However, the aglycone form was not found. This study showed that CPEs might be a potent source of natural antioxidant, without any toxic effects.

Anti-Inflammatory Effects of Fermented Products with Avena sativa on RAW264.7 and HT-29 Cells via Inhibition of Inflammatory Mediators

  • This study investigated therapeutic candidates with anti-inflammatory potential among traditional dietary ingredients targeting inflammatory bowel disease (IBD). Both Avena sativa and traditional fermented products, such as Korean soy paste, are popular health foods. We investigated the anti-inflammatory effects of soy paste combined with A. sativa (KDA), compared with soy paste without A. sativa (KD) by evaluating the expression of pro-inflammatory cytokines in lipopolysaccharide-stimulated RAW264.7 mouse macrophages and HT-29 human colon epithelial cells. KDA significantly inhibited the production of nitric oxide (NO) and downregulated the pro-inflammatory cytokines, such as interleukin (IL)-1β, IL-6, and tumor necrosis factor-α in lipopolysaccharide (LPS)-induced RAW264.7 cells. In another in vitro experiment involving LPS-stimulated HT-29 cells, KDA suppressed the levels of IL-8, which is the chemokine elevated in IBD. In addition, KDA exhibited anti-oxidative properties, such as 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and 2,2'-azino-bis(3-ethylbenz-thiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. Our findings revealed that A. sativa combined with soy paste exhibits a synergistic anti-inflammatory and anti-oxidant effect following fermentation. These results suggest that KDA may be used as a potential anti-inflammatory therapy against IBD.

전자레인지 조리에 의한 적양배추의 항산화력 및 대장암세포 증식억제 (Comparison of Antioxidant and Anti-colon Cancer Activities of Red Cabbage (Brassica oleracea) by Microwave Cooking)

  • The present study was performed to investigate antioxidant and anti-colon cancer activities of red cabbage (Brassica oleracea L. var. capitata f. rubra DC) according to the cooking conditions (raw, microwave, blanching and steaming). The contents of red cabbage extracts were determined as follow: total phenolic contents, 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethy lbenzo-thiazoline-6-sulfonic acid) (ABTS), 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay, western blot analysis. The total contents of polyphenol and flavonoid of red cabbage were 20.27 mg GAE/g Dry weight ${\pm}0.03$ and $2.55{\pm}0.02mg$ RE/g Dry weight. In this study, the total contents of polyphenol were decreased to both microwave and steam cooking. Total antioxidant activity and growth inhibition of HCT116 human colon cancer cells were in the order of raw > microwaving > steaming cooking methods. These results indicate that red cabbage extracts might have antioxidant and anti-proliferative activity according to the cooking conditions.

Cytotoxic and antioxidant properties of four plants belonging to the genus Solanum

  • The aim of the study was to evaluate in vitro antioxidant and cytotoxic activities of the methanolic extracts of leaves of Solanum sisymbrifolium, Solanum anguivi multiflora, Solanum barbisetum and Solanum jasminoides. In the in vitro antioxidant screening using ABTS [2,2'-azino-bis (3-ethylbenzo-thiazoline-6-sulphonic acid) diammonium salt] method, the methanol extracts of Solanum jasminoides, Solanum barbisetum and Solanum anguivi multiflora exhibited potent antioxidant activity with $IC_{50}$ values 31.25 $\pm$ 0.35, 40.33 $\pm$ 0.57 and 54.33 $\pm$ 0.57 ${\mu}g$/ml, respectively. Solanum barbisetum also showed potent activity in DPPH [1,1-diphenyl-2-picryl hydrazyl] method with an $IC_{50}$ value of 55.33 $\pm$ 1.66 ${\mu}g/ml. In the cytotoxicity studies, the methanol extract of Solanum barbisetum exhibited moderate activity against Vero, HEp-2, HeLa and A-549 cell lines with $IC_{50}$ values in the range of 83.30 - 127.30 ${\mu}g$/ml. Solanum anguivi multiflora extract also showed moderate activity against Hep-2 cell line with $IC_{50}$ value of 80.13 ${\mu}g$/ml. Solanum barbisetum possessing both the activities requires further investigation.

새로운 2-이미노티아졸린 유도체의 합성과 항균 활성(I) (Synthesis of New 2-Iminothiazolines and Their Antifungal Activities)

  • 새로운 살균제를 개발할 목적으로 2-이미노티아졸린 유도체를 합성하여 그 생물활성을 시험하였다. 1차아민과 이소티오시아네이트를 결합시켜 티오우레아 유도체를 제조한 다음 이것을 클로로아세트알데히드 또는 알파-할로케토산과 반응시켜 새로운 36종의 2-이미노티아졸린 유도체를 합성하였으며 이들의 6가지 중요한 식물병원균에 대한 항균력을 조사한 결과 티아졸린의 C-2 위치에 페닐이미노기, C-3 위치에 메틸, C-4 위치에 페닐카바모일메틸기, 그리고 C-5위치에 수소원자가 있을 때 벼 도열병균(Pyriculraria oryzae, rice blast)에 대한 항균활성이 가장 높았다.

겉보리에서 배양한 영지버섯 추출물의 항산화 및 항염증 효능 평가 (Evaluation of Anti-Oxidant and Anti-Inflammatory Activities of Ganoderma lucidum Cultured on Hulled Barley)

  • Background: Ganoderma lucidum cultured on hulled barley was investigated as a potential natural source of antioxidants and anti-inflammatory agents. Methods and Results: The yields from Ganoderma lucidum cultured on hulled barley water and ethanol extract were 17.69% and 25.77%, respectively. The antioxidant activity of Ganoderma lucidum cultured on hulled barley extracts was confirmed by various methods including assayss of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethylbenzo thiazoline-6-sulfonic acid) (ABTS), nitrite radical scavenging, and $Fe^{3+}$ to $Fe^{2+}$ reducing power activity. The ethanol extract of Ganoderma lucidum cultured on hulled barley showed improved DPPH, ABTS and nitrite radical scavenging activity compared with the water extract. After treatment of RAW264.7 cells with Ganoderma lucidum cultured on hulled barley ethanol extracts, the cell viability compared with the control was 92.82%, even at a concentration of $3,000{\mu}g/m{\ell}$. The ethanol extract inhibited reactive oxygen species (ROS) generation in RAW264.7 cells stimulated with $H_2O_2$, even at low concentrations. In addition, the ethanol extract showed an inhibitory effects on the production of lipopolysaccharide-induced nitric oxide (NO) in RAW264.7 cells. Conclusions: This study suggests that the extract of Ganoderma lucidum cultured on hulled barley is a potential source of natural antioxidants and anti-inflammatory agents.
