Characterization and Epitope Mapping of KI-41, a Murine Monoclonal Antibody Specific for the gp41 Envelope Protein of the Human Immunodeficiency Virus-1

  • In this study, a mouse monoclonal antibody (mAb) against gp41(584-618), the immunodominant epitope protein, was generated. For this purpose, BALB/c mice were immunized with double branched multiple antigenic peptides derived from the HIV-1 gp41(584-618) sequence, and antibody-secreting hybridoma were produced by fusion of mice splenocytes with SP2/0 myeloma cells. One clone producing an antigen specific mAb, termed KI-41(isotype IgG1) was identified, whose specific reactivity against gp41(584-618) could be confirmed by ELISA and Western blot analysis. Epitope mapping revealed the recognition site of the mAb KI-41 to be located around the sequence RILAVERYLKDQQLLG, which comprises the N-terminal region within the immunized gp41(584-618) peptied. Since this mAb recognizes this specific epitope within the HIV-1 gp41 without any cross-reactivity to other immunodominant regions in the HIV-2 gp35, KI-41 will provide some alternative possibilities in further applications such as the development of indirect or competitive ELISA for specific antibody detection in HIV-1 infection or for other basic researches regarding the role and function of HIV-1 gp41.

환자간호실무 표준에 관한 연구 (A Study on the Patient Care Standard)

  • The purpose of this study is to develop a patient care standard which is the basis of unit based quality assurance. The subjects were 570 nurses of 6 hospitals is Seoul. Patient Care Standards were developed from 3 times of clinical Nurses Association's workshop & the joint meeting of Clinical Nurses Association & the Korean Nurses Academic Socity of Nursing Administration. Respondents were instructed to rate of the 2 types of 5 - point Likert type questionnaire(one is the level of perceived importance, the other is the level of actual performance) Findings of this study were as follows 1. As a results of reliability analysis, each questionnaire ranged from $\alpha$=0.79 to 0.94

    2. The Correlations between the levels of perceived importance & actual performances were ranged from r=.40 to 0.72(P=.00) 3.     • 본 논문에서는 소형화, 경량화 및 저전력화를 목표로 개발된 과학기술위성 2호 대용량 메모리 유닛(Mass Memory Unit, MMU)의 준비행모델(Proto Flight Model, PFM)을 통해서 과학기술위성 1호 대용량 메모리 유닛과의 성능을 비교 하였다. 탑재체로부터의 수신 속도와 XTX로의 전송 속도는 각각 200Kbps와 10Mbps이다. 실험실에서의 성능시험과 위성에 조립된 상태에서의 성능 및 환경 시험을 통해서 그 성능을 확인 하였다.

    낙동강유역 과수재배지의 단위면적당 비점오염부하량 산정에 관한 연구 (The Calculation of NPS Load per Unit Area in Orchard to the Nakdong River Basin)

    • In this study, Calculated the nonpoint sources(NPS) load per unit area about various rainy events in vineyard of Nakdong River basin. NPS monitoring and calculation for NPS load per unit area were estimated from 'Investigation method of precipitation discharge(National Institute of Environmental Research, 2007)'. The evaluation of applicability for NPS load per unit by compared with prior research data and Total Maximum Daily Load(TMDL) data. Five target areas were each $2000m^2$, $1800m^2$, $1943m^2$, $2484m^2$, $864m^2$ and located in Gyeongsangbukdo Gyeongju, Gyeongsangbukdo Sangju, Gyeongsangnamdo Hapcheon in Korea. Since fruits were the only crop on the target area, the characteristics of stormwater discharge at survey sites could be evaluated independently. A total of 115 rainfall events in the Orchard area during five years(2008-2012) was surveyed, and 38 of them became stormwater discharge. In the Nakdong River watershed, average of event mean concentrations(EMCs) in Orchard area for biochemical oxyzen demand(BOD), Chemical oxyzen demand(COD), total nitrogen(T-N), total phosphorus(T-P) were 2.0mg/L, 10.1mg/L, 3.195mg/L, 0.578mg/L, respectively. NPS load per unit area in Orchard area showed BOD : $2.0kg/km^2{\cdot}day$, COD : $10.2kg/km^2{\cdot}day$, T-N : $3.220kg/km^2{\cdot}day$, T-P : $0.606kg/km^2{\cdot}day$.

    소형 임대주택 가구특성 및 실내공간 이용실태 조사 연구 (A Study on the Residents Characteristic and the Inner Space Use of Small-Sized Rental Multi-family Housing)

    • As the provision of National Rental Housing has led the consciousness on rental housing changed, it becomes more necessary to improve the quality of rental housing. The purpose of this study is to analyze the inner space use of a unit and residents' consciousness on the results of surveying the residents in order to construct rental housing which provides comfort residental environment for the residents. Data was collected through questionnaire surveys(459 residents) from 5 estates including 3 estates of National Rental Housing and 2 estates of Public Rental Housing, which is $36m^{2}\;and\;45m^{2}$ of net area of a unit, for the actual use conditions and their requirements on the inner space of a unit. The results are as follows ; 1) The rental housing differentiated according to the needs from the residents' characteristics should be developed. 2) The main demand household for rental housing is established to that of 4 families including a 40s year-old couple and that of 3 or 4 families including a 30s year-old couple, and a double income family. But the type of household becomes more diverse than before. 3) It is necessary to plan the living room for the Multi-functional inner space emphasizing commom space 4) The various and efficient storage space and system for the residents' demands.

    • A crystallographic unit cell can be transformed into another one by a 3×3 transformantion matrix. If the determinant of the transformation matrix has a negative value, the newly transformed unit cell becomes a left-handed cell. The best way of transforming the left-handed cell to the right-handed one is to multiply each element of the transformation matrix by-1, and its corresponding transformation matrix must be applied tot he atomic coordinates of a noncentrosymmetric crystal so as to maintain the absolute configuration unchanged. The behaviour of absolute configuration caused by transforming the crystallographic unit cell was examined theoretically and experimentally on the compound (S)-(+)-4-phenyl-1-[4-aminobenzoyl) indoline-5-sulfonyl]-4,5-dihydro-2-imidazolone hydrochloride.

    Block 대체정책에 관한 연구

    • A block replacement policy using items with different reliability is discussed. We devide system unit failure modes into two modes and use less reliable unit when operating unit fails near the planned preventive replacement time. In this policy, item A has two failure modes. Mode-1 failure is removed by minimal repair, mode-2 failure by replacement. If mode-2 failure of item A happens in (0,T- $\delta$). failure item A is replaced by new item A. If mode-2 failure of item A happens in(T-$\delta$,T), failure item A is replaced by new item B. Item B should be cheaper and less durable than item A. Under this policy, we determine the preventive replacement interval T and the interval $\delta$ of item B replacement which minimize the cost rate per unit time.

    돼지의 대동맥 판막 및 심낭에서 녹색콩 알파-갈락토시다아제를 이용한 알파-갈 항원결정인자 제거 (Removal of ${\alpha}$-Gal Epitopes in Aortic Valve and Pericardium of Pig Using Green Coffee Bean ${\alpha}$-Galactosidase)

    • 배경: 최근 조직판막의 구조적 손상에 있어서 동물면역 반응이 중요한 역할을 할 가능성이 제기되면서 동물의 대표적 이종항원 물질인 알파-갈 항원결정인자에 대한 환자의 면역반응에 관한 관심이 높아지고 있다. 또한 알파-갈은 세포 표면에 존재하며 이는 녹색콩 알파-갈락토시다아제 라는 효소를 이용하여 제거할 수 있다고 알려져 있고 조직 표면의 알파-갈 항원결정인자는 Griffonia Simplicifolia의 동종렉틴 중 B4타입에 선택적으로 결합하여 이를 이용해서 염색할 수 있다고 알려져 있다. 이에 본 연구팀은 조직판막을 만드는데 많이 사용되는 돼지의 대동맥 판막 및 심낭 조직을 가지고 녹색콩알파-갈락토시다아제를 이용하여 이들 조직의 알파-갈 항원결정인자를 제거할 수 있는지 알아 보고자하였다. 대상 및 방법: 신선한 돼지의 대동맥 판막 및 심낭 조직을 0.5 unit/mL, 1.0 unit/mL, 2.0 unit/mL 농도의 녹색콩 알파-갈락토시다아제로 pH 6.5, $4^{\circ}C$에서 24시간 처리한 뒤 이를 Griffonia Simplicifolia 동종렉틴 B4 타입을 이용한 면역조직형광염색으로 염색하여, 각각의 농도에서 효소 반응 후 해당 조직의 알파-갈 항원결정인자가 얼마나 제거되는지를 형광염색의 정도로 판단하였다. 결과: 돼지의 대동맥 판막 조직의 경우 1.0 unit/mL농도의 녹색콩 알파-갈락토시다아제를 pH 6.5, $4^{\circ}C$ 에서 24시간 처리하였을 때 형광염색이 거의 되지 않을 정도로 알파-갈 항원결정인자가 제거되었고 이는 효소의 농도를 2.0 unit/mL로 증가시켰을 때에도 비슷한 양상을 보였다. 돼지의 심낭 조직의 경우 효소 처리 전의 알파-갈 염색에서도 대동맥 판막조직에 비하여 많은 양의 형광염색을 보였으며 효소 처리의 농도도 대동맥 판막의 경우보다 높은 2.0 unit/mL의 농도에서 알파-갈 항원결정인자가 제거되는 양상을 보였다. 걸론: 돼지의 대동맥 판막 조직과 심낭 조직의 알파-갈 항원결정인자는 eriffonia simplicifolia의 동종렉틴 B4를 사용한 면역조직형광염색에서 잘 염색되었으며 이를 알파-갈락토시다아제를 사용하여 제거하였을 때 각각 1.0 unit/mL, 2.0 unit/mL 농도의 녹색콩 알파-갈락토시다아제를 $4^{\circ}C$, pH 6.5의 조건에서 24시간 반응시켰을 때 효과적으로 상당량 제거할 수 있었다. 향후 돼지의 판막조직 및 심낭조직으로 만드는 조직판막의 내구성 증대에 대표적인 동물 면역항원인 알파-갈 항원결정인자의 제거가 유용한 도구가 될 수 있을 것이며 앞으로 알파-갈락토시다아제로 처리한 돼지의 조직판막에 대한 인간혈장의 항-갈 항체 및 항-갈 단클론항체를 이용한 직접적인 면역학적 연구가 필요하다.
