A study on the optimum ratio of the ingredients in preparation of black sesame gruels (흑임자죽 재료배합비의 최적화 연구)

  • The aim of this study was to develop a standardized recipe for black sesame gruel that has been preferred for generations as a nutritional food. The method focused on optimizing the mixing ratio of the components to improve the quality of the black sesame gruels that modem consumers would like. The results are summarized as follows: The more black sesame added to the gruel, the lower its brightness was, but the redness and yellowness was higher. The amount of black sesame made a significant difference in the viscosity, color, nutty taste, bitterness, appearance and overall preference. It was highest in the overall preference when the amount of black sesame was added 33g(40% of rice weight basis). Different types of rice were added to the black sesame gruel, and it was observed that the color value of the gruel was high in its brightness, redness and yellowness when 50g(60%) of glutinous rice was added to it. The black sesame gruel was most preferred when 50g of non-glutinous rice was added. The redness value was high when 15g(18%) of non-glutinous rice flour was added. The yellowness value was high when 25g(30%) of non-glutinous rice flour was added. This observation showed significant differences in the viscosity, color, nutty taste, bitterness, appearance and overall preference. In particular, the black sesame gruel was most preferred when 50g of non-glutinous rice flour was added. The addition of 7g(9%) of salt to the black sesame gruel showed the highest brightness. The redness and yellowness was the highest when 5g(6%) of salt was added. This observation showed a significant difference in the viscosity, color, nutty taste, bitterness, appearance and overall preference. The highest preference was observed when 2.5g(3%) of salt was added. Adding more materials increased the viscosity of the black sesame gruel. With increasing temperature, the viscosity became lower, and vice versa. The intensity of sweetness and spreadability was found to be proportional to the amount of additive material. In conclusion, the optimum recipe for black sesame gruels was obtained 33g(40%) of black sesame, 50g(60%) of glutinous rice (flour), 2.5g of salt, and 500$m\ell$ of water.

A Cross-cultural Study of Influence Factors of Meat Substitutes between Korea and China (한·중 소비자들의 육류대체식품에 대한 구매의도에 미치는 영향 연구)

  • This study examined the factors influencing the purchase intention of meat substitutes. A survey was conducted on 589 consumers in Korea (297 people) and China (292 people). The perception of meat substitute foods was lower in Korean consumers than in Chinese consumers, but there was no significant difference. The purchase intention of meat substitute foods was lower in Korean consumers than in Chinese consumers (p<0.01). Korean consumers' perception of meat substitute foods was higher in males than in females (p<0.01). The purchase intention of meat substitute foods also showed the same trend (p<0.001). Chinese consumers' perception of meat substitute foods was higher in males than in females. The overall purchase intention of meat substitute foods was significantly higher in males than in females (p<0.05). The perception of meat substitute foods by Korean consumers' was higher in their 40s and 50s than in their 20s and 30s. The purchase intention of meat substitute foods was also high in their 40s and 50s (p<0.01). On the other hand, the perception of meat substitute foods by Chinese consumers' was higher in their 20s and 30s than in their 40s and 50s (p<0.01). The purchase intention of meat substitute foods also showed the same trend (p<0.01). Korean consumers' perception of meat substitute foods had a significant positive effect on the purchase intention of meat substitutes (p<0.001). Chinese consumers' perception of meat substitute foods also had a significant positive effect on the purchase intention of meat substitute foods (p<0.001). Overall, food technology neophobia has a significant negative effect on the purchase intention of meat substitutes (p<0.05).

  • 치아는 성별과 연령의 추정은 물론 혈형 검사와 유전자 검사까지 가능하게 하는 중요한 법의치과학적 자료이다 대부분 치아를 이용한 연구는 핵 DNA가 들어있는 치수에서의 연구로 치수내에는 풍부한 혈액 및 세포가 분포해 있어 핵 DNA가 다량 함유되어 있다. 그러나 순수 상아질에는 핵이 없고 따라서 핵 DNA도 없는 것으로 알려졌지만 치수내에 존재하는 핵 DNA가 상아세관을 통하여 상아질내로 침투할 가능성이 있고 실제 근관치료가 되어 있는 무수치를 감정하게 되는 경우도 있다. 본 연구에서는 이러한 치아중에서도 근관치료를 받은 무수치에서 개인식별에 활용되는 유전자가 검출되는지 여부를 확인하고자 하였다. 40개의 근관치료된 치아상아질에서 DNA출 추출하고 중합효소반응을 이용하여 증폭절편다형(Amp-FLPs)을 실시하고 X-Y homologous amelogenin gene과 STR 유전좌위 F13A01, LPL를 검색하여 다음과 같은 결과를 얻었다. 1. 40개의 근관치료된 치아중 19개에서 DNA가 추출되었다. 2. X-Y homologous amelogenin gene 검색으로 40개의 근관치료된 치아에서 21의 남자 치아중 5개, 19개의 여자치아중 7개 등 모두 12개 치아에서 성별검사가 가능하였다. 3. F13A01 유전자는 43개의 근관치료된 치아중 6개의 치아에서 검색되었으며, 4개의 대립유전자 및 5개의 유전자형을 관찰하였다. 4. LPL_유전자는 40개의 근관치료된 치아중 7개의 치아에서 검색되었으며, 3개의 대립유전자 및 3개의 유건자형을 관찰하였다. 이상의 결과를 종합하여 볼 때 근관치료된 치아상아질에서 중합효소반응을 이용한 성별검사 및 STR 유전자위의 검색은 일부 치아에서만 가능하였으나, 근관치료된 치아들도 개인식별을 위한 법의치과학적 자료로서 유용할 것으로 사료된다.

Effects of the Duration of Liquid Feeding on Performance and Nutrient Digestibility in Weaned Pigs

  • A total of 72 castrated, crossbred ($Landrace{\times}Yorkshire$) piglets ($5.7{\pm}0.7kg$ BW) were allotted to one of three treatments including: a dry crumbled feed fed for 40 days; liquid feed provided for 10 days followed by dry crumbled feed for 30 days; and liquid feed provided for 20 days followed by dry crumbled feed for 20 days. Liquid feed was produced fresh each day by mixing water with dry crumbled feed at a ratio of 3:1. Pigs fed liquid feed for 10 days had a higher weight gain during the first 10 days (+16.8%; p = 0.01) and over the entire experimental period (+4.9%; p = 0.07) than pigs offered dry feed. Pigs fed liquid feed for 20 days also had a higher weight gain during the period from d 0 to 10 (+12.8%; p = 0.01), from d 10 to 20 (+8.5%; p = 0.06) and from d 0 to 20 (+9.7%; p = 0.01) than pigs offered dry feed. Pigs fed liquid feed for the first 10 days had higher feed intakes from d 0 to 10 (+22.6%; p = 0.01) and from d 0 to 40 (+5.3%; p = 0.02) than pigs offered dry feed. Pigs fed liquid feed for the first 20 days had a higher feed intake from d 0 to 10 (+21.8%; p = 0.01), from d 10 to 20 (+10.6%; p = 0.06), from d 0 to 20 (+14.6%; p = 0.01) and from d 0 to 40 (+6.6%; p = 0.02) than pigs offered dry feed. Feed conversion from d 0 to 40 tended to be poorer for pigs fed liquid feed during the first 20 days (p = 0.08) indicating an increase in feed wastage with liquid feeding. Nutrient digestibility at day 10 was unaffected by dietary treatment. However, at day 30, pigs fed liquid feed for 10 days had higher digestibility of dry matter (p = 0.08), energy (p = 0.10), crude protein (p = 0.03) and neutral detergent fibre (p = 0.05) than pigs fed liquid feed for 20 days. In conclusion, liquid feeding for 10 or 20 days increased the performance of weaned pigs during the specific time period that liquid feeding occurred but there were no carry over effects into subsequent production periods.

Effects of Combined Treatment of Lactic Acid Bacteria and Cell Wall Degrading Enzymes on Fermentation and Composition of Rhodesgrass (Chloris gayana Kunth.) Silage

  • This experiment was conducted to study the effects of lactic acid bacteria (LAB) inoculation either alone or in combination with cell wall degrading enzymes on the fermentation characteristics and chemical compositions of Rhodesgrass silage. Over to 1 kg of fresh Rhodesgrass sample a treatment of inoculant LAB with or without addition of an enzyme of Acremoniumcellulase (A) or Meicelase (M) or a mixture of both enzymes (AM) was applied. The treatments were control untreated, LAB-treated (application rate $1.0{\times}10^5cfu/g$ fresh sample), LAB+A 0.005%, LAB+A 0.01%, LAB+A 0.02%, LAB+M 0.005%, LAB+M 0.01%, LAB+M 0.02 %, LAB+AM 0.005%, LAB+AM 0.01%, and LAB+AM 0.02%. The sample was ensiled into 2-L vinyl bottle silo, with 9 silages of each treatment were made. Three silages of each treatment were incubated at 20, 30 and $40^{\circ}C$ for 2-months of storage period. All silages were well preserved with their fermentation quality has low pH values (3.91-4.26) and high lactic acid concentrations (4.11-9.89 %DM). No differences were found in fermentation quality and chemical composition of the control untreated silage as compared to the LAB-treated silage. Combined treatment of LAB+cellulases improved the fermentation quality of silages measured in terms of lower (p < 0.01) pH values and higher (p < 0.05) lactic concentrations than those of LAB-treated silages. Increasing amount of cellulase addition resulted in decrease (p < 0.05) of pH value and increase (p < 0.05) of lactic acid concentration. LAB + cellulase treatments (all cellulase types) reduced (p < 0.01) NDF, ADF and in vitro dry matter digestibility of silages compared with the control untreated silages. The fermentation quality and the rate of cell wall reduction were higher (p < 0.01) in the silages treated with LAB + cellulase A than in the silages treated with either LAB+cellulase M or LAB + cellulase AM. Incubation temperature of $40^{\circ}C$ was likely to be more appropriate environment for stimulating the fermentation of Rhodesgrass silages than those of 20 and $30^{\circ}C$.

Cryopreservation of Day 3 Mouse Embryos by Vitrification (초자화동결을 이용한 제 3일째 생쥐 배아의 동결보존)

  • The use of hormonal stimulation in human in vitro fertilization and embryo transfer (IVF-ET) leads to increased production of embryos for ET. So to avoid high pregnancies and to allow conception in future, unstimulated cycles, cryopreservation of spare embryos is desirable. One of the improvement of cryopreservation methods is vitrification. We cryopreserved mouse day 3 embryos by vitrification using the three different vitrification solution (EFS40, VS11 and VS3a). EFS40 solution is consisted of 40% (v/v) ethylene glycol, Ficol170 30% (w/v) and 0.5M sucrose and VS11 is 6.0M ethylene glycol and 1.8M glycerol. And VS3a is 6.5M glycerol and 6% (w/v) BSA (bovine serum albumin). First we tested the toxicity of three vitrification solution by exposure to these solution during 3 min. After washing by thawing solution, the survival rates of each groups are 95.5%, 90.9% and 84.4% (EFS40, VS11 and VS3a). High percentages of them developed to expanded blastocyst and hatching embryos in culture 48hrs 94.2%, 97.7%, 100% and 97.4% (no treatment group, EFS40, VS11 and VS3a). So there is no significant differences among the each group. Second, after thawing of vitirfied embryos, the survival rates of each groups are 96.8% (slow freeze), 94.1% (EFS40), 85.5% (VS11) and 80.0% (VS3a, P vs. no freeze or EFS40 is 0.01). Vitrified embryos exhibited a high rate of development in vitro after 48hrs culture. The percentages of each group to blastocyst and hatching embryos are 88.7% (no freeze), 91.8% (slow freeze), 93.4% (EFS40), 87.7% (VS11) and 73.0% (VS3a, P vs. other group is 0.01). The results suggest that there is no significant differences in exposure of various vitrification solution and day 3 mouse embryos can be vitrified in solution EFS40 and VS11 by simple procedure.

Nutrient regime, N:P ratios and suspended solids as key factors influencing fish tolerance, trophic compositions, and stream ecosystem health

  • The objectives of this study were to determine the effects of stream nutrient regime, N:P ratios and suspended solids on fish tolerance/trophic compositions and stream ecosystem health, based on multi-metric model, during 2008-2013. Also, stream ecosystem health was evaluated in relation to chlorophyll-a (CHL) as a measure of algal productivity or indicators of trophic state to water chemical parameters. Total number of sampled fish species were 50 and showed a decreasing trend from 2008 to 2013. The minnow of Zacco platypus, based on the catch per unit effort (CPUE), was the most dominant species (25.9%) among the all species. Spatial heterogeneity was evident in the fish tolerance guilds that showed the dominance of sensitive species (89%) in the headwaters (S1) and the dominance of tolerant species (57%) in the urban. These conditions were directly influenced by concentrations of nutrients and organic matter (COD). The N:P ratios, as a barometer of water pollution, had a negative linear function (R2 = 0.40, P < 0.01) with CHL, and the ratios had an important role in changes of COD concentration (R2 = 0.40, P < 0.01). Under the circumstances, the N:P ratio directly influenced the relative proportions of fish trophic/tolerance compositions. According to the regression analysis of omnivore (Om) and insectivore sp. (In) on total nitrogen and total phosphorus, nitrogen had no significant influences (P > 0.05) to the two compositions, but phosphorus influenced directly the two guilds [slope (a) = -32.3, R2 = 0.25, P < 0.01 in the In; a = 40.7, R2 = 0.19, P < 0.01 in the Om]. Such water chemistry and fish trophic guilds determined the stream ecosystem health, based on the multi-metric fish model.

CHANGES IN SOME PHYSIOLOGICAL PARAMETERS OF ALBINO RATS AT DIFFERENT AMBIENT TEMPERATURES

  • Five experimental groups with five adult male rats in each, were exposed to 20, 35, 40 and $45^{\circ}C$ air temperature for 50-70 minutes, and to $50^{\circ}C$ for 30-50 minutes, respectively. Food and drinking water were not permitted during the exposure. Blood samples were obtained by heart puncture immediately after the thermal treatment. All the rats were hyperthermic (p<0.01) as compared to the controls ($20^{\circ}C$). Hyperthermia was associated with hypoglycemia which was significant (p<0.01) at 45 and $50^{\circ}C$ exposures. Plasma levels of GOT and GPT declined at 35 and $40^{\circ}C$ reaching the lowest (p<0.05) level at $45^{\circ}C$, while at $50^{\circ}C$ GOT level was elevated by 45% but GPT was normal as compared to the controls. Differences between groups were significant (p<0.01) for GOT and insignificant for GPT. Hematocrit value increased significantly (p<0.01) at 45 and $50^{\circ}C$, indicating hemoconcentration. It could be concluded that severe heat stress (45 and $50^{\circ}C$) resulted in critical hyperthermia, hypoglycemia, disturbed liver function, body dehydration, and hemoconcentration leading to death.

Efficacy of Maxforce and Avion gel baits containing fipronil, clothianidin, and indoxacarb against the German cockroach (Blattella germanica)

  • The present study evaluated the efficacy of Maxforce gel baits containing 0.01% fipronil (Maxforce), 0.05% fipronil (Maxforce Magnum), 1.0% clothianidin (Maxforce Impact), and 0.6% indoxacarb (Avion gel bait; all from Bayer Environmental Science, Clayton, CA, USA) against reference and wild strains (Mobasher, Andisheh, Tamin-e Ejtemaei, and Maskoni) of German cockroach collected in 2016. Test design and bait placement were according to World Health Organization (WHO) procedures for efficacy achieving a 95% mortality rate. Although cockroaches fed on all gel baits, gel baits containing clothianidin and indoxacarb were eaten at higher rates than gel baits containing 0.01% and 0.05% fipronil. The cumulative mortality rates reached 100% for all cockroach strains at 32, 28, 28 and 40 h after eating baits containing 0.01% fipronil, 0.05% fipronil, clothianidin, and indoxacarb, respectively. Efficacy (a 95% mortality rate) after exposure (consumption) of gel baits containing 0.05% fipronil and clothianidin was seen after 28 h, compared with 32 h for 0.01% fipronil and 40 h for indoxacarb. In conclusion, commercial gel baits have improved and may be more effective than spray formulations against German cockroaches.

Effects of Baicalein on the Bioavailability of Nicardipine in Rats

  • This study was to investigate the effect of baicalein, an antioxidant, on the bioavailability of nicardipine after orally or intravenously administered nicardipine in rats. Nicardipine was administered orally (12 mg/kg) or intravenously (4 mg/kg) with or without orally administered baicalein (0.4, 2 or 10 mg/kg) to rats. In the inhibitory effect of baicalein on CYP3A4 activity, baicalein inhibited CYP3A4 activity with $IC_{50}$ values of 9.2 ${\mu}M$. The cell-based P-gp activity test using rhodamine-123 also showed that baicalein (30-10 ${\mu}M$, p<0.01) significantly inhibited P-gp activity. Compared with the control group (given nicardipine alone), the area under the plasma concentration-time curve (AUC) was significantly (2 mg/kg, P<0.05; 10 mg/kg, P<0.01) increased by 25.9-60.0%, and the peak concentration ($C_{max}$) was significantly (10 mg/kg, P<0.01) increased by 40.0% in the presence of baicalein after orally administration of nicardipine. Consequently, the relative bioavailability (R.B.) of nicardipine was increased by 1.26- to 1.60-fold and the absolute bioavailability (A.B.) was significantly (2 mg/kg, P<0.05; 10 mg/kg, P<0.01) increased by 26.0-59.9%. Compared to the i.v. control, baicalein did not significantly change pharmacokinetic parameters of nicardipine in i.v. administration. Accordingly, the enhanced oral bioavailability of nicardipine might be mainly due to increased intestinal absorption caused by P-gp inhibition rather than to reduced elimination of nicardipine by baicalein. The increase in the oral bioavailability might be mainly attributed to enhanced absorption in the small intestine via the inhibition of P-gp and reduced first-pass metabolism of nicardipine via the inhibition of the CYP3A subfamily in the small intestine and/or in the liver by baicalein. Based on these results, nicardipine dosage should be adjusted when given concomitantly with baicalein.
