Determination of Mercury in Fly Ash by Using Flow Injection Cold Vapor Isotope Dilution Inductively Coupled Plasma Mass Spectrometry

  • A method based on flow injection-isotope dilution-cold vapor-inductively coupled plasma mass spectrometry (FI-IDCV-ICP/MS) has been applied to determine trace level of mercury in fly ash. $^{200}Hg$ isotopic spike was added to 0.25 g of BCR176R fly ash and then decomposed by microwave digestion procedure with acid mixture A (8 mL $HNO_3$ + 2 mL HCl + 2 mL HF) and acid mixture B (8 mL $HNO_3$ + 2 mL $HClO_4$ + 2 mL HF) for applying IDMS. Mercury cold vapor was generated by using reductant solution of 0.2% (w/w) $NaBH_4$ and 0.05% (w/w) NaOH. The measurements of n($^{200}Hg$)/n($^{202}Hg$) isotope ratio was made using a quadrupole ICP/MS system. The accuracy in this method was verified by the analysis of certified reference material (CRM) of fly ash (BCR 176R). The indicative value of Hg in BCR 176R fly ash was $1.60{\pm}0.23$ mg/kg (k = 2). The determined values of Hg in BCR 176R fly ash by the method of FI-CV-ID-ICP/MS described in this paper were $1.60{\pm}0.24$ mg/kg (k = 3.18) and the analysis results were in well agreement with the indicative value within the range of uncertainty.

Measurement and Acceleration of Biodegradation in Soil. (토양매립에 의한 생분해도 측정 및 가속화)

  • The quantitative and rapid method for measuring the biodegradation of polymer materials in soil was developed. In this study, cellophane film was used as a model biodegradable polymer and the biodegradation was assayed by measuring the amount of glucose which was produced by a hydrolysis reaction using HCl after collecting the film from soil. Cellophane film was degraded 41.2% in 4 months during winter while it was degraded 76.5% in 2 months during summer. It means that biodegradation in soil is affected by environmental conditions. The biodegradation was also measured in an incubator (30$^{\circ}C$, humidity 50-55%) to exclude the environmental variations. Cellophane film was degraded 94% in that condition in 40 days. The biodegradation showed the first order kinetics and the rate constant was 0.067 (1/day). Acceleration of the biodegradation in soil was also studied. We added cultured soil microorganisms or nutrients such as N, P, and S into the soil. While the addition of microorganisms showed the temporary increase of rate constant, the addition of nutrients not only showed the increase of rate constant from 0.096 (1/day) to 0.21 (1/day) but also maintained the effect continuously.

Studies on the Heavy Metals in Paddy Rice and Soils in Jang-hang Smelter (제연소인근지역(製鍊所隣近地域)의 토양(土壤) 및 수도체중(水稻體中) 중금속함량(重金屬含量)에 관(關)한 조사연구(調査硏究))

  • Soils and rice plants subject to smoke from the Jang-hang Smelter were samples at two depths to assess the nature and extend of Cd, Cu, Pb, and Zn contamination, particularly with respect to distance from pollution source. Soils at east site within 1km of the center of the smelter were highly contaminated with Cd, Cu, Pb, and Zn. Metal levels in the smoke-polluted area were higher in surface soil than in subsurface soil, and decreased rapidly with distance reaching nearly background levels at 5km from pollution source. A position correlation was found between contents of total and 0.1 N HCl-extractable, or 1.0N $CH_3COONH_4$- extractable heavy metals in surface soils of pollution. Contents of heavy metals in soils were positively correlated with soil organic matter and cation exchange capacity. The contents of heavy metals in brown rice collected from the paddy field around the Jang-hang Smelter were in the range of 0.23-1.33 ppm for Cd, 2.39-6.25 for Cu, 0.95-8.32 ppm for Pb and 14.60-27.31 for Zn. The contents of Cd, Cu, and Zn in rice straw were positively correlated with those in the brown rice. Negative correlation was found between the contents of Cd, Cu, Pb, and Zn in brown rice and the distance from the source.

  • Synthetic mica $NaMg_{2.5}(Si_4O_{10})F_2 (Na-TSM)$ was treated with HCl solution and H-type exchange resin to investigate the chemical stability and the ion exchange by measuring the dissolution of chemical components and the exchanged $H^+$ ion. The replacement of $Na^+$ ion occurred in contact with HCl solution and H-type ion exchange resin at the surface of Na-TSM particles reached $Na^+$ ions at maximum value of 70~80%. $Mg^{2+}$ ion of octahedral layer became to dissolve from the pH2 solution and th amount of it dissolved in-creased almost proportional to $H^+$ ion concentration from around 0.02N $H^+$ ion equilibrium concentration. The crystalline structure of Na-TSM was destructed by dissolution of Mg2+ ion in cncentrated hydrochloric acid solution and resulted silica gel precipitation.

A Study on the Heavy Metal Contamination of paddy Soil in the Vicinity of the Seosung Pb-Zn Mine (서성 연-아연광산 주변 농경지 토양의 중금속 오염 연구)

  • Fifty seven soil samples were collected from the paddy soil filled with tailings in the vicinity of the Seosung Pb-Zn mine. Those samples were analyzed for As, Cd, Co, Cr, Cu, Pb, and Zn in order to investigate heavy metal pollution levels in the paddy soil. Analyses of the soil samples were carried out using Inductively Coupled Plasma Atomic Emission Spectrometry(ICP-AES) . Paddy soils show pH range from 6.55 to 8.26. X-ray diffraction analyses of the paddy soil indicate that the soils consist predominantly ankerite, siderite, quartz, mica, and clay minerals with minor amounts of amphibole and chlorite. The mineral composition of the waste rocks consists of massive galena, sphalerite, and minor amounts of pyrite, arsenopyrite, chalcopyrite, calcite, siderite, Pb-sulfosalt, and marcasite. The paddy soils were significantly contaminated by heavy metals(average concentrations, As: 334.4 ppm, Cd: 37.6 ppm, Co: 15.7 ppm, Cu: 214.1 ppm, Pb: 4,612 ppm, and Zn: 4,468 ppm).

Analysis of Isoflavone Contents of Soybean By-products with Acid Hydrolysis Method (산 가수분해시 가열방법과 시간 및 추출조건에 따른 대두가공 부산물의 이소플라본 함량 변화)

  • To establish a rapid and effective method for the analysis of soy isoflavone which is known to have lots of variation in derivatives of glucoside, conversion rate from isoflavone conjugates to its aglycones, and decomposition of conversed aglycones were investigated with various acid hydrolysis conditions. A number of heating conditions for acid hydrolysis including heating at convection oven $(105^{\circ}C)$, water bath $(95^{\circ}C)$, heating block $(120^{\circ}C)$, and hot plate $(120^{\circ}C)$ were applied. Acid hydrolysis in heating block with reflux was chosen as the best heating condition. From the stability test of isoflavone aglycone during acid hydrolysis, genistein, daidzein, and glycitein did not show any significant changes in their contents for 60 min of hydrolysis. Ten to thirty milligram of sample per 1 mL HCl was the best ratio of sample to acid. As conclusion, acid hydrolysis for 60 min after addition of 15 mL HCl solution to 0.5 g soybean, and then fill up to 50 mL with methanol, followed by HPLC analysis was set as a final analysis method. From this method, isoflavone contents expressed as total aglycone of feed meal was the highest with content of $1288.5{\mu}g/g$ followed by those of dehulled meal.

A Study of EPMA Analysis for Nitric Acid Insoluble Alloys (질산불용성 합금의 EPMA분석)

  • Ternary and quaternary alloys composed of Mo-Ru-Rh-Pd were prepared and their crystal structure and quantitative chemical compositions were analysed by XRD, WDX, EDX and ICP/AES. The results of X-ray diffraction studies showed that the alloys were crystallized in hexagonal close packing, well known as ${\varepsilon}$-phase with $P6_3/mmc$ of space group. The optimum accelerating voltage for Zr~Cd(40~48) analyzed by EPMA using PET crystal was found to be 15 kV and the linear regression coefficient(R) of atomic number and X-ray intensity was approximately 0.998 without Tc standard specimen. The WDX results of alloys of Mo and Pd by linear regression equations were detected to be 0.1% lower compared to WDX analysis results using standard specimen, while Ru, Rh were detected 3% higher. These alloys were completely dissolved in mixed acid of 12.5 mL HCl and 1 mL $HNO_3$, at $220^{\circ}C$ for 22 hours using autoclave with PTFE vessel. There was no reprecipitating phenomnon when diluted 100 times with 1N-HCl. The results of ICP/AES analysis deviated less than 4% comparing with those of normal WDX analysis.

Effect of Jichul-hwan on Gastric Motility in Conscious and Gastric Wall Injured Rats (지출환(枳朮丸)의 흰쥐 위(胃) 운동성(運動性)에서 정상 상태 및 위벽 국소손상 후에 대한 효능)

  • Background & Objective : Jichul-hwan(JCH) has been used for the treatment of functional dyspepsia, regarded as a gastric dysmotility disease. We investigated the effects of JCH on gastric motility and its mechanisms of action in rats. Methods : The gastric wall was injured by tracting a part of stomach body in rats. Gastric emptying was measured after administration of normal saline(NS) or JCH in normal rats and gastric wall injured rats. To evaluate the mechanism of JCH under delayed gastric emptying conditions, normal rats were treated with atropine sulfate(1mg/kg, s.c.), quinpirole HCl(0.3mg/kgg, i.p.), $NAME(N^{G}-nitro-L-arginine$ methyl ester, 75mg/kg s.c.) and cisplatin(10mg/kg, i.p.). The gastric slow waves were measured for 30 minutes before and after administration of each solution(NS, JCH). Results : JCH 110.1mg/kg improved gastric emptying for 2 hrs(p=0.014). JCH 110.1mg/kg improved gastric emptying in the gastric wall injured rats(p=0.001). Under the delayed gastric emptying, JCH 110.1mg/kg improved gastric emptying in the group treated with atropine $sulfate(1.83{\pm}0.96$ vs $8.43{\pm}8.46$, p=0.003), but aggravated it with quinpirole $HCl(4.7{\pm}2.9$ vs $1.61{\pm}2.09$, p=0.021). Administration JCH 110.1mg/kg increased EGG power in rats. Conclusions : JCH stimulates gastric motility through the cholinergic pathway, so we expect that it would be effective in the treatment of dysmotility-like functional dyspepsia with low activity of vagus nerve.

Determination of Sulfur Requirement to Adjust pH of Alkaline Soil by Buffer Curve Method (알칼리성 토양 pH 교정시 완충곡선법을 이용한 황 시용량 결정)

  • To determine application rate of elemental sulfur to adjust pH of alkaline soil, buffer curve method was investigated. The elemental sulfur required to control pH 8.3 to pH 6.3 by buffer curve calculation was treated in two soils of silty loam and sandy loam, and the sulfur-mixed soils were moistened with 50% of water holding capacity during incubation of 6 weeks at $30^{\circ}C$. Soil pH was lowered with incubation and reached to target point after 4 weeks of incubation, and elemental sulfur was oxidised entirely to sulfate. This means that buffer curve has the accuracy to determine sulfur application rate in alkaline soil. However it is estimated that application rate of sulfur should be carefully determined in the field scale. Excess application of elemental sulfur resulted in extremely low soil pH and caused the hinderance of lettuce growth by nutritional imbalance and ion toxicity. To simplify the determination procedure of sulfur requirement, buffer curve method by addition of 0.1N-HCl solution as unit of mL was developed, it was compared with theroutine methods which diluted $H_2SO_4$ solution and $Ca(OH)_2$ are added as cmolc per kg soil to adjust each pH step. Buffer capacities, cmolc kg $soil^{-1}$ $pH^{-1}$, calculated from two buffer curves were not significantly different. The result indicates that buffer curve method by 0.1N-HCl can be used to adjust high pH of alkaline soil.

Bioequivalence of Traline Tablet to Zoloft® Tablet (Sertraline HCI 50 mg)

  • Sertraline HCl, (1S-cis)-4-(3, 4-dichloro-phenyl)-1, 2, 3, 4-tetrahydro-N-methyl-l-naphthalenamine hydrochloride, is a potent and selective serotonin reuptake inhibitor which is used in the treatment of depression and obsessivecompulsive disorders. The purpose of the present study was to evaluate the bioequivalence of two sertraline HCl tablets, Traline tablet (Myungin Pharm. Co. Ltd.) and Zoloft$^{(R)}$ tablet (Pfizer Inc.), according to the guidelines of the Korea Food and Drug Administration (KFDA). The in vitro release of sertraline from the two sertraline HCl formulations was tested using KP VIII Apparatus II method with various dissolution media. Twenty four healthy Korean male volunteers, $23.50{\pm}1.74$ years in age and $64.09{\pm}7.10\;kg$ in body weight, were divided into two groups and a randomized $2{\times}2$ crossover study was employed. After a single tablet containing 50 mg as sertraline HCl was orally administered, blood samples were taken at predetermined time intervals and the concentrations of sertraline in serum were determined using an online columnswitching HPLC method with UV/Vis detection. The dissolution profiles of two formulations were similar in all tested dissolution media. The pharmacokinetic parameters such as $AUC_t$, $C_{max}$ and $T_{max}$ were calculated, and computer programs (Equiv Test and K-BE Test) were utilized for the statistical analysis of the parameters using logarithmically transformed $AUC_t$, $C_{max}$ and un-transformed $T_{max}$. The results showed that the differences between two formulations based on the reference drug, Zoloft$^{(R)}$ tablet, were 0.04, 3.26 and -1.29% for $AUC_t$, $C_{max}$, and $T_{max}$, respectively. There were no sequence effects between two formulations in these parameters. The 90% confidence intervals using logarithmically transformed data were within the acceptance range of log0.8 to log1.25. Thus, the criteria of the KFDA bioequivalence guideline were satisfied, indicating Traline tablet was bioequivalent to Zoloft$^{(R)}$ tablet.
