Analysis of Absorbance for Tinted Dye Absorbed into Tinted Lenses by Spectrophotometric Method (분광광도법에 의한 칼라 렌즈 내 염색제에 대한 흡광도 분석)

  • We studied absorbance of tinted dye absorbed into tinted lenses by Spectrophotometric method. Used lenses and tinted dye were CR-39 plastic optical lenses and Ons & $Lavas^{(R)}$ Blue, respectively. Absorbance values of tinted dye absorbed into tinted lenses were obtained from the differences of absorbance values between tinted lenses and CR-30 lenses. Average concentrations of tinted dye absorbed into tinted lenses were obtained from the mass differences between tinted lenses and CR-30 lenses. The relationship between absorbance and average concentration for tinted dye absorbed into tinted lenses was also confirmed to follow Beer’s law. The absorption coefficient calculated from the tangent of the calibration curve was determined to be a=0.983 $Lg^{-1}cm^{-1}$.

Extraction of Young Antler and Antler by Water, Proteases and HCl (녹용 및 녹각의 단백질 가수분해 효소 및 염산에 의한 가용화)

  • Freeze dried antler, heat dried antler, antler were extracted through processing step by water, protease and hydrochloric acid(HCl). Extraction rate of freeze dried antler at 50$^{\circ}C$ by water was 9.01%(8.82, absorbance at 280 nm), that of heat dried antler was 9.01%(4.45, absorbance at 280 nm), and that of antler was 1.10%(0.31, absorbance at 280 nm), respectively. Extraction rate of freeze dried antler by bacterial protease was 16.89%(4.50, absorbance at 280 nm), and that of heat dried antler was 17.29%(5.62, absorbance at 280 nm), and that of antler was 18.22%(0.64, absorbance at 280 nm), respectively. Extraction rate of freeze dried antler by 0.8N HCl was 72.25%(4.60, absorbance at 280 nm), that of heat dried antler was 71.14%(4.70 absorbance at 280 nm), and that of antler was 79.82% (2.80, absorbance at 280 nm), respectively. Extraction rate of freeze dried antler through three processing steps was 98.15%, that of heat dried antler was 97.35%, that of antler was 99.14%, respectively. The result of analysis by HPLC shows that high molecular pe which appears in young antler and antler extraction was changed into a small molecular peak of about 1,000 by the reaction of protease, and protein of about MW 70,000 was extracted from their remaining residue by 0.8N HCl. The above result shows that water extraction and protease extraction in the freeze dried young antler, protease extraction and HCl extraction in dried young antler, and HCl extraction in antler are most effective.

Extraction of Freeze Dried Young Antler Residue by Proteases and HCl (단백질 가수분해 효소 및 염산에 의한 녹용 각질의 추출)

  • The freeze dried young antler residue was extracted by proteases and hydrochloric acid(HCl). The young antler was extracted by water at 50$^{\circ}C$ and the residue was reacted by proteases for 5 hours at 50$^{\circ}C$. The extraction rate of its residue was 32.8%(absorbance 3.61 at 280nm) of bacteria protease, 23.8%(absorbance 0.69) of papain, and 31.2% (absorbance 2.96) of pepsin. The young antler was extracted by boiling water and the residue was reacted by proteases for 5 hours at 50$^{\circ}C$. The extraction rate of its residue was 45.0%(absorbance 3.61) of bacteria protease, 30.4%(absorbance 0.33) of papain, and 51.2% (absorbance 2.77) of pepsin. The result of HPLC analysis reveals that in 50$^{\circ}C$ water extract and boiling water extract, all high molecular peak was reduced under MW 1,000 by proteases. The result from the extract of young antler residue reacted by HCl for 5 hours at 50$^{\circ}C$ shows that its extraction rate was 45% (absorbance 0.78) in concentration of 0.1N HCl, 61% (absorbance 1.82) in 0.2N, 81% (absorbance 2.29) in 0.4N, and 82.0% (absorbance 3.28) in 2.0N. The result of HPLC analysis also reveals that in the extract by 0.8N HCl, the peak of about MW 70,000 accounted for 78% in total. Protein content of the extract by 0.8N HCl was 8.2%, and content of amino acid was 81.6%, ash was 1.3%, and mineral contents were 0.1 % of Ca, 2.3% of P, 0.8 % of Mg, 3.4% of Na, 0.002% of F by dry base.

Chlorophyll a/b Ratio as a Criterion for the Reliability of Absorbance Values Measured for the Determination of Chlorophyll Concentration (엽록소 농도 결정을 위하여 측정한 흡광도 값의 신뢰도 검정 지표로서 엽록소 a/b 비례치)

  • The Beer-Lambert law states that absorbance is proportional to the concentration of a solute in a solution at a given wavelength. This linearity works for an ideal or a 'sufficiently diluted' solution, so this linearity is often used as a criterion for the fidelity of the absorbance value measured. In this study, we used a chlorophyll (Chl) solution, isolated from rice leaves with 80% acetone to test the use of the Chl a/b ratio as an additional criterion for checking the fidelity of measured values using four different absorption spectrophotometers: Cary4E, UV-1650PC, Versamax (a microplate reader), and NanoDrop 1,000(which can handle a $4{\mu}l$ aliquot). We used Chl solutions of varying concentrations from $0.2{\mu}g/ml$ to $200{\mu}g/ml$ to measure absorbance values at 645 nm and 663 nm and checked the linearity first. The results indicated that the range of Chl concentrations that we can rely on based on the linearity was similar to the range in which the calculated Chl concentrations based on the measured absorbance values agreed with the known concentrations. However, some border cases or cases with very low Chl concentrations inside the fidelity range of Chl concentrations did not agree with the criterion that the Chl a/b ratio should not change after dilution of the Chl in the solution. These results suggest that the Chl a/b ratio is a better criterion for the reliability of the absorbance values measured for the determination of chlorophyll concentration than the criterion based on the linearity suggested by the Beer-Lambert law.

Measurement of Cell Concentration of Fungal Broth Using Optical Density -Characteristics of Pellets- (흡광도를 이용한 곰팡이 배양액의 세포농도 측정 -Pellet의 특성에 관한 연구-)

  • A new method that considers the pellet sedimentation characteristics for fungal cell concentration measurement was developed using optical density. Appropriate mixing of the pellet suspension almost homogeneously was tried to prevent the sedimentation of the pellet by a small magnetic bar in cuvette, giving a stable optical density. The linear relationship between optical density and the dry cell weight was obtained. However, different curved lines were observed according to the pellet size. Optical density couldn't be detectable in the size range of $355{\mu}m$above. It was concluded from the result that the use of optical density for measuring cell concentration in fungal broth became possible by considering the characteristics.

pH Measurement Using Isosbestic Point of Indicator (지시약의 등흡광점을 이용한 용액의 pH 측정)

  • The methyl red stock solution was added to sample solution and the absorbance values of this solution were measured at the maximum absorption wavelengths of the acidic form, the basic form, and the isosbestic point of the indicator. From the measurements of absorbance, the pH of this solution was calculated. The range of absorbance at the isosbestic point was maintained within 0.1 ${\sim}$ 0.3. The error of pH measurement was within ${\pm}0.08$ pH unit in the pH range of sample solution of $pK_I{\pm}1$.

Studies on the Near Infrared Diffuse Reflection Spectroscopy for Meat Components (근적외선 반사분광분석법에 의한 정육 및 그 구성성분의 반사 spectra에 관한 연구)

  • This study was conducted to determine the effect of the physical and chemical factors on the absorbance when the chemical component of food was rapidly and effectively analyzed with near infrared reflection spectrophotometer. The absorbance was measured within the wavelength range of 1100 and 2500m and at the various sample temperatures. The absorbance was greatly increased with the rising sample temperature. When the grinding time of samples prolonged, the absorbance was decreased. The absorption peaks occurring in the near infrared spectra of meat, meat protein, pork fat and water were attributed to the overtone and combination vibration of C-H, N-H, O-H or C=O bond within the macromolecules of each sample. The absorbance was lower for meat protein than other meat components. Pork fat was characterized by the ${\cdot}CH_2{\cdot}$ absorption peak at the wavelength of 1700 and 2300 nm and water by the relatively high absorption peak at the wavelength of 1450 and 1930 nm. As a result, the absorbance of near infrared reflection spectrum of meat was affected mainly by fat and water in meat.

  • 수용액에 첨가된 나노다이아몬드는 용매상 내에서 안정된 분산 특성을 나타낸다. 이런 분산 특성은 일정 시간동안 유지되지만, 농도와 시간에 따라 분산특성이 떨어지면서 일부 다이아몬드들의 침강이 나타난다. 이런 분산 및 침강 특성을 정량화하기 위하여 나노다이몬드 수용액의 흡광특성을 비교 분석하였다. 파장에 따른 나노다이아몬드의 흡광도는 농도에 상관없이 350 nm까지 상승하다가 점차적으로 완만하게 감소하고 있다. 또한 300nm 파장에 대한 흡광도는 나노다이아몬드의 농도에 따라 거의 직선적으로 증가하는 것으로 나타났다.

  • The efficiency of Photocatalytic Oxidation Process were investigated for the treatment of Aquatic Humic Substances (AHS). In UV-only system, pH 7-9 was the optimum pH range for TOC removal, and alkali range was the optimum pH for absorbance decrease. In UV/$TiO_2$ system, the optimum $TiO_2$ dosage was 50ppm and over 50ppm of $TiO_2$ dosage was not effective for removal of AHS. In UV/$H_2O_2$ system, optimum $H_2O_2$ dosage was 20mM, when over 20mM dosage, removal of TOC (Total Organic Carbon) and absorbance was decreased. Radical scavenger affected on the photo-oxidation of AHS. Removal rate of TOC and absorbance was decreased by addition of carbonate ions and TOC removal was more effected than that of absorbance.

펌프-촉침 방법을 이용한 피코초 순간 흡광 속도분광기 제작

  • A picosecond transient absorption kinetic spectrometer using a pump-probe method is presented and compared with other methods of picosecond transient absorption measurements. This constructed kinetic spectrometer detects the transient transmittance of sample using a lock-in amplifier as a function of delay time between pump and probe pulses generated from a picosecond mode-locked cw dye laser. Typical transient absorption and ground state bleach recovery kinetic profiles measured with this spectrometer are shown. Excellent kinetic curves of transient absorption or ground state bleach recovery may be obtained at single wavelengths with this spectrometer.
