Effect of soil fumigation treatment on the growth and the chemical composition of Korea ginseng, Panax ginseng C.A. Meyer (토양훈증제 처리가 2년생 인삼의 생육과 화학적 조성에 미치는 영향)

  • These experiments were conducted to investigate some effects of the soil fumigants Dow-fume MC-2, Cylone, TeloneC-17, and D-D on the growth response of the Korea ginseng(two-year old), Panax ginseng C.A. Meyer, and the chemical composition of the plants. Fumigant treatment increased growth of roots, stems, and leaves. The increase of root growth rates ranging from 2.8 to $58.7\%$ in fresh weight and 8.1 to $63.8\%$ in dry weight were observed in the 2-year old ginseng plants grown in the old ginseng soil treated with the different soil fumigants. The chemical analysis of inorganic elements such as N, P, K, Ca, Mg, Na, Fe, Mn, and Zn was made for the roots, stems and leaves of the 2-year old plants. The increases or decreases of content of the elements were observed, and their contents of each of the elements were changed with the kind of fumigants, part of the plants, and soil depth of the fumigant treatments, but their changing tendencies were not steady.

Evaluation of Phosphine Efficacy on Anoplophora chinensis and A. glabripennis in Export Bonsai (수출분재 주요 해충인 알락하늘소와 유리알락하늘소에 대한 포스핀 훈증 효과 검정)

  • Anoplophora chinensis and A. glabripennis is classified as important quarantine Cerambycidae beetles worldwide. These beetles cause serious damage on natural host plants and export bonsai trees in Korea. We evaluated efficacy of phosphine fumigant for control of two Cerambycidae beetles in bonsai. In the preliminary test to achieve CT (Concentration x time) values of phosphine to Cerambyicdae beetles larvae, we used Monochamus alternatus of larvae as an alternative insect. A 24 h phosphine fumigation with $2g/m^3$ phophine achieved 100% eggs mortality against Anoplophora chinensis and A. glabripennis. Larva of A. chinensis was also completely controlled when treated for 168-h at same dose desiccator. 100% mortality on larval stage of two beetle species were achieved both in dormancy (January) and vegetative period (February) of bonsai when treated with $2g/m^3$ phosphine. However, phosphine fumigation against vegetative period of Acer palmatum, Carpinus turczaninowii and Malus sieboldii bonsai showed slight phytotoxicity (shorten leaf and flower size and delayed flowering).

Reduction effects of N-acetyl-L-cysteine, L-glutathione, and indole-3-acetic acid on phytotoxicity generated by methyl bromide fumigation- in a model plant Arabidopsis thaliana (모델식물 애기장대에 대한 훈증제 메틸브로마이드의 약해발생 및 N-acetyl-L-cysteine, L-glutathione, indole-3-acetic acid의 약해억제 효과)

  • Understanding the phytotoxic mechanism of methyl bromide (MB), an essential fumigant during the quarantine and pre-shipment process, is urgently needed to ensure its proper use and reduce international economic losses. In a previous study, two main MB-induced toxic mechanisms such as reactive oxygen species (ROS) and auxin distribution were selected by analyzing transcriptomic analysis. In the study, a 3-week-old A. thaliana was supplied with 1 mM ROS scavengers [N-acetyl-L-cysteine (NAC) or L-glutathione (GSH)] and 1µM indole-3-acetic acid(IAA) three times every 12 h, and visual and gene expression assessments were performed to evaluate the reduction in phytotoxicity by supplements. Phytotoxic effects on the MB-4h exposed group were decreased with GSH application compared to the other single supplements and a combination of supplements at 7 days post fumigation. Among these supplements, GSH at a concentration of 1, 2, and 5mM was suppled to A. thaliana with MB-fumigation. During a long-term observation of 2 weeks after the fumigation, 5 mM GSH application was the most effective in minimizing MB-induced phytotoxic effects with up-regulation of HSP70 expression and increase in main stem length. These results indicated that ROS was a main key factor of MB-induced phytotoxicity and that GSH can be used as a supplement to reduce the phytotoxicity of MB.

Biochemical mechanisms of fumigant toxicity by ethyl formate towards Myzus persicae nymphs (복숭아혹진딧물(Myzus persicae) 약충에 대한 에틸포메이트 훈증 독성의 생화학적 메커니즘)

  • Ethyl formate has been used for the control of insect pests by fumigation. However, there were not many reports to show its target site of fumigant toxicity on insect pests since its first use in the agricultural industry. In the present study, we showed the presumable target sites of ethyl formate fumigation in insect pests using Myzus persicae nymphs. After ethyl formate fumigation, the nymphs of this species were collected and the changes at the biochemical and molecular level were determined. The activity of cytochrome c oxidase (COX) was approximately two-fold higher after ethyl formate fumigation. In addition, the expression levels of acetylcholinesterase (AChE) decreased gradually with increasing ethyl formate concentration. These two findings suggested that COX and AChE might be the major target sites of ethyl formate fumigation. In addition to these results, the analysis of lipid content using MALDI-TOF MS/MS identified 9 phospholipids differently generated 2-fold higher in the ethyl formate-treated nymphs than that in the control nymphs, thereby leading to changes in cell membrane composition in M. persicae nymphs. Therefore, the ethyl formate fumigation caused lethal effects on M. persicae nymphs by changing COX activity, AChE gene expression, and phospholipid production.

Control of Powdery Mildew (Uncinula necator) in Vineyards by Spraying and Vapor-Action Treatments of Triazole Fungicides (Triazole계 살균제의 살포 및 훈증처리에 의한 포도 흰가루병(Uncinula nectator)의 방제)

  • 비가림재배 포도원에서 발생하는 흰가루병에 대하여 triazole계 살균제 flusilazole, myclobutanil, difenoconazole, penconazole, triflumizole, triadimefon의 방제 효과를 검정한 결과는 다음과 같다. 살균제 처리구의 이병과방율과 이병엽율은 무처리구에 비해 현저히 감소하였고, 이들의 방제가는 과방에서 약제에 따라 94.7∼97.9%, 잎에서 85.5%∼90.9%였으며 대조약제인 polyoxin B의 방제가와 유사한 정도를 보였다. 공시살균제의 보호효과는 살균제를 병발생 1주일전에 살포하는 경우, 살포 3주 후에 방제가는 약제에 따라 72.2∼90.5%였으나 4주 후에는 21.2∼41.6%로 감소하여 대조약제 polyoxin B의 52.5%보다 감소시켰으나 myclobutanil의 발아억제 효과는 약제 농도증가에 크게 영향받지 않았다. 살균제에 침지한 cheesecloth를 포도원 선반에 매달아 조사한 공시살균제의 훈증효과는 살포농도의 2배로 처리했을 때 처리점으로부터 반경 30cm이내의 포도송이에서는 약제에 따라 57.2∼71.1%의 방제가를 보였으나 전체 식물체에서는 49.1∼65.8%로서 10일 간격으로 3회 살포한 것 보다 낮은 방제가를 보였다.

  • This experiment was carried out to investigate control efficacy by cultural practice and fumigation of tacterial wilt caused by Pseudomonas solancearum in resistant tobacco line NC82 at the Korea Ginseng and Tobacco Research Institute, Eumseong Experiment Station in 1982 and 1983. The bacterial wilt of tobacco occurred severely from mid-July to last August in applicable temperature and soil humidity for increasing bacteria. Disease severity appeared low and slowly at fumigation and resistant variety treatment. Incidence of bacterial wilt in tobacco line NC82 was $44.7\~55.8\%$ being compared with susceptible variety, NC2326 and $95\~99\%$ when resistant variety, NC32 was cultivated with soil fumigation treatment. Control efficacy of cultural practices appeared low with $0.8\~20\%$ and was not different from resistant variety and fumigation treatment. Control system against bacterial wilt in flue-cured tobacco was accomplished by control efficacy over $95\%$ when resistant variety(NC82) was cultured after treatment of cultural practices (Tillering after harvest and before transplanting, stalk and root destruction, early transplanting early removal of the mulching film) and soil fumigation(Cylon).

  • The differential enhanced degradation of cis- and trans-1,3-D was observed in the previous two studies performed by several researchers. This study was initiated to investigate the involvement of microorganisms in the differential enhanced degradation of the chemicals. As expected, microorganisms were responsible for the enhanced degradation. A mixed bacterial culture capable of degrading 1,3-D was isolated from an enhanced soil sample collected from a site treated with 1,3-D. Similar to the enhanced soil, the mixed culture degraded trans-1,3-D faster than cis-1,3-D. This mixed culture could not utilize cis- and trans-1,3-D as a sole source of carbon for growth. Rather, a variety of second substrates were evaluated to stimulate the differential enhanced degradation of the two isomers. As a result, the mixed culture degraded cis- and trans-1,3-D only in the presence of a suitable second substrate. Therefore, it appeared that the degradation of cis- and trans-1,3-D was a cometabolic process. Second substrates that had the capacity to stimulate the degradation included soil leachate, tryptone, tryptophan, and alanine. Other substrates tested. including soil extract. glucose, yeast extract and indole, failed to stimulate the degradation of the two isomers. The mixed culture was composed of four morphologically distinctive colonies on L-agar plates.

Control of Soilborne Fungal Diseases on Muskmelon by Soil Disinfestation in Consecutively Cultivated Fields (토양살균에 의한 멜론 연작재배지 뿌리썩음병 방제)

  • This study was carried out to determine the causal agents of soil-borne fungal diseases that pose a threat to the muskmelon production in Cheong Yang, Korea and to investigate the potential effects of hot water drenching and three fumigant (metam sodium, dazomet and methyl bromide) on these diseases. As the agents of the diseases, Monosporascus cannonballus, Didymella sp., Fusarium sp., Phytophthora sp., were detected. Hot water and the fumigants were treated on two successive cropping seasons of melon. Soil temperature was measured at 0, 10, 20 and 30 cm soil depth. In 2005, soil sterilization by hot-water was more effective significantly to control of the diseases than by fumigant. yield was the highest in hot-water sterilized plot as $39\;ton{\cdot}ha^{-1}$. Dazomet ($50\;g/m^2$) treated plot was followed as $23\;ton{\cdot}ha^{-1}$. In 2006, hot water sterilized plot showed higher yields than non-treated plots ($14.8\;ton{\cdot}ha^{-1}$). But the other three fumigant contained Dazomet ($50\;g/m^2$) were no difference (P<0.05) in yield.

  • The effect of calcium cyanamide (China-made) soil fumigation on the growth of the ginseng pathogen Rhizoctonia, Pythium), soil microbes, and seed germination of lettuce and radish was investigated. At twice the recommended level (2S0-ppm $CaCN_2$), the growth of Rhizoctonia and Pythium, and the seed germination, were not inhibited. Rather, the effective level was 10,000 ppm. The powder form was more effective than the granular form in inhibiting pathogen growth and seed germination. The lettuce seed was also more sensitive than the radish seed. Calcium cyanamide appearedto decrease the fungi population and to increase Actinomycetes in the soil.

  • Overwintering striped rice borer, Chilo suppressalis W. larvae, pupae and adults in rice straw handicrafts were exposed to different doses and periods of phostoxin in gas chambers(ave.temp. 27C). The following results were obtained; 1. For 100 percent mortality, larvae required 48 hours, pupae 24 hours and adults only 8 hours of exposure. 2. The insecticidal effect of tile fumigant was more dependent upon the length of exposure than the dosage of fumigant.
