Effect of Helianthus tuberosus Juice Mixed with Dried Bitter Melon Juice on Hypoglycemic Function in Streptozotocin-Induced Diabetic Rats (당뇨유발 흰쥐에서 돼지감자 여주 혼합즙 음용에 의한 혈당저하 효과)

  • This study was conducted to develop Helianthus tuberosus (HT) juice mixed with dried bitter melon juice and assess its hypoglycemic effect in streptozotocin (STZ)-induced diabetic rats. HT juice mixed with 5.0% dried bitter melon juice was used in this study. Male Sprague-Dawley rats were divided into four groups (eight rats per group) and drunk each sample for 4 weeks: normal water [normal control (NC) group], STZ+normal water (STZ group), STZ+HT juice (HT group), STZ+HT juice mixed with 2.5% bitter melon juice (HT2.5 group), and STZ+HT juice mixed with 5.0% bitter melon juice (HT5.0 group). HT juice was diluted to 25% in distilled water and supplied to rats. Food intake, body weight gain, and food efficiency ratio were lower in the STZ group than in the NC group. HT, HT2.5, and HT5.0 groups showed higher parameters than the STZ groups. Water intakes were higher in the STZ group than in the NC group. After 3 weeks, HT, HT2.5, and HT5.0 groups showed lower parameters than the STZ group. After 1 week, blood glucose level of the STZ group ($476.7{\pm}22.8mg/dL$) was significantly higher than those of the HT group ($376.3{\pm}25.8mg/dL$), HT2.5 group ($405.2{\pm}35.1mg/dL$), and HT5.0 group ($342.8{\pm}29.7mg/dL$). After 4 weeks, blood glucose level of the STZ group were significantly higher than those of the HT, HT2.5, and HT5.0 group. Serum insulin levels of the HT group ($3.13{\pm}0.32ng/mL$), HT2.5 group ($3.40{\pm}0.23ng/mL$), and HT5.0 group ($3.48{\pm}0.43ng/mL$) were higher than that of the STZ group ($2.72{\pm}0.53ng/mL$). These results indicate that H. tuberosus juice mixed with dried bitter melon juice helps prevent or attenuate progression of diabetes in rats with STZ-induced diabetes.

Change of photosynthetic efficiency and yield by low light intensity on ripening stage in japonica rice (등숙기의 차광 처리에 의한 광합성능 및 쌀 수량 변화)

  • Light intensity is one of the most important requirements for plant growth, affecting growth, development, survival, and crop productivity. Sunlight is the main energy source on Earth which is energy used by photosynthesis to convert light energy to chemical energy. In this study, the light use efficiency and photosynthetic characteristics of high-quality rice cultivars were evaluated after shading on ripening stage. For the study, we treated of three levels of shade (0, 50 and 70%) on rice at ripening stage and two levels of nitrogen (9 and 18 kg/10a) used three high yielding rice cultivars, such as Boramchan, Hopum, and Honong. The shade was given for the respective plots from heading up to harvesting. We were performed to determine growth survey, SPAD and chlorophyll fluorescence every 10 days interval after shading on ripening stage. At harvest stage, grain yield and yield components were determined. Results of analysis of the results representing the maximum photosynthetic efficiency of PSII, Fv/Fm, and SPAD were decreased by depending on the time at full sunlight. But shade treatments were not changed and a significant difference among cultivars did not appear. Compared with the full sunlight, shade treatments significantly delayed ripening rate and decreased rice quality of cultivated rice. Therefore, rice yield, can be reduced in proportion to the shading density is apparent, the rate of decrease was not observed difference between varieties, when protected from light 70%, and decreased to less than 50%. The adverse effects of low light intensity on the yield and yield components were not able to significantly minimize by the nitrogen level.

Changes in Anthocyanin Content of Aronia (Aronia melancocarpa) by Processing Conditions (물리적 처리조건 변화에 따른 아로니아(Aronia melancocarpa) 유래 안토시아닌 함량변화 특성)

  • The purpose of this study was to obtain basic data for using Aronia as a functional food material. The composition of anthocyanin was characterized and quantitated by LC-MS/MS, HPLC, and UV-VIS spectrophotometer techniques, respectively. The anthocyanin content was analyzed by temperature, time, pH, and the addition of citric acid. The UV-VIS spectrophotometer used for analysis of anthocyanin is less accurate than the LC-MS/MS method used in recent years. In the past, cyanidin-3-Glucoside was reported to be a major anthocyanin that contains Aronia. However, LC-MS/MS analysis in this study confirmed cyanidin-3-galactoside to be the major compound. The anthocyanin content of the Aronia powder began to decrease sharply at a temperature of $65^{\circ}C$ or higher when heated for 24 hours. In an aqueous solution of Aronia, the anthocyanin content was reduced by 50% at $65^{\circ}C$ for 10 hours and decreased by 85% at $85^{\circ}C$ within 10 hours. Above pH 8, the anthocyanin content was reduced by more than 50%. The results of this study will provide useful information to maintain anthocyanin content in the manufacturing process of Aronia. It could also be used to ensure the stability of anthocyanins in similar species of berries.

Fermentation of Waste Woody Biomass for the Production of Bioenergy (바이오에너지생산을 위한 목질계 폐바이오매스의 발효)

  • In this study, fermentation characteristics of waste agricultural and forest biomass for production of heat energy were focused to be used in agricultural farm households. The purpose of this study was focused on seeking practical utilization of agricultural and forest biomass wastes in agricultural farm households in the form of thermal energy by means of simple fermentation process. Fermentation process was performed in terms of different raw-materials and their mixture with different ratios. Urea, lime, and bioaids were added as fermenting aids. Moisture contents of fermenting substrates were adjusted to 55~65%. In order to optimize the fermentation process various factors, such as raw-materials, moisture contents, amount of fermenting aids, and practical measurement of hot-water temperature during fermentation were carefully investigated. The optimum condition of fermenting process were obtained from hardwood only and hardwood: softwood (50 : 50) beds. In case of hardwood only the highest temperature was recorded between 60 to $90^{\circ}C$ the lowest temperature was determined to more or less $40^{\circ}C$ and the average temperature was ranged to $50{\sim}60^{\circ}C$ and this temperature ranges were maintained up to 20~30 days. The optimum amount of additives were estimated to ca. 15 kg of urea, 20 kg of bioaids, and 10 kg of lime for 1 ton of substrate. To reach the highest temperature the optimum moisture content of fermenting substrate was proved to 55% among three moisture content treatments of 45%, 55% and 65%. The temperature of hot-water tank installed in fermenting bed of hardwood : grass (50 : 50) showed very different patterns according to measuring positions. In general, temperatures in the mid- and upper-parts of substrate piling were relative higher than lower and surface parts during 45-day fermentation process. The maximum temperature of fermenting stage was determined to $65^{\circ}C$, minimum temperature, more or less $40^{\circ}C$, and average temperature was $60^{\circ}C$. The water temperature of tank exit was ranged to $33{\sim}48^{\circ}C$ during whole measuring periods. It could be concluded that fermentation process of waste agricultural and forest biomass produces a considerable amounts of heat, averaging about $50{\sim}60^{\circ}C$ for maximum 3 months by using the heat exchanger (HX-helical type).

Seasonal Mineral Nutrient Absorption Characteristics and Development of Optimum Nutrient Solution for Rose Substrate Culture in a Closed Hydroponic System (순환식 수경재배에서 재배시기별 장미의 무기이온 흡수특성과 적정 배양액 조성)

  • This study was performed to develop a suitable nutrient solution for standard rose substrate culture in a closed hydroponic system. 1/4, 1/2, 2/3 and 1 strength of the nutrient solution made by Japan National Institute of Vegetable and Tea Science (JNIVT) were supplied. The photosynthesis rate, quality and growth of cut flower were higher in the 1/2 and 2/3 strength of nutrient solution during high and low temperature period. Based on the above results, optimum nutrient solutions (UOS) were composed by nutrientwater (n/w) absorption ratio with 1/2S ($NO_{3^-}N$ 6.8, $NH_{4^-}N$ 0.7, $PO_{4^-}P$ 2.0, K 3.8, Ca 3.0, Mg 1.2, $SO_{4^-}S$ $1.2me{\cdot}L^{-1}$) at high temperature season and 2/3($NO_{3^-}N$ 9.7, $NH_{4^-}N$ 0.8, $PO_{4^-}P$ 2.2, K 5.0, Ca 3.9, Mg 1.5, $SO_{4^-}S$ $1.5me{\cdot}L^{-1}$) at low temperature season. The results of suitability examination showed that the EC level in newly composed nutrient solution (UOS) was more stable than other nutrient solutions due to its large amount of calcium and potassium. The growth of cut flower cultivated with UOS was higher than those of other nutrient solutions. Especially, the yield of cut flowers in UOS nutrient solution increased 1.4 times than that of other nutrient solution treatments. Consequently, the new nutrient solution investigated in this experiment was suitable for rose cultivation in a closed hydroponic system.

Solid Fermentation of Medicinal Herb Using Phellinus baumii Mycelium and Anti-thrombin and Anti-oxidation Activity of its Methanol Extract (장수상황버섯 균사체를 이용한 한약재의 고체발효 및 메탄올 추출물의 트롬빈 저해 활성과 항산화 활성)

  • To produce bioactivity-strengthen medicinal herbs, the 36 medicinal herbs which have antioxidation or blood circulation activity, were solid fermented using Phellinus baumii mycelium. Most of medicinal herbs, except Chrysanthemum indicum (flower), Zizyphus jujuba Miller (fructus), Aconitum koreanum R. Raymond (root), Magnolia denu-data (flower), and Polygonatum sibiricum Redt (root bark), showed good fermentation at $25^{\circ}C$ for 20 days under 90% of relative humidity. The poor fermentations of the herbs could be explained by lack of nutrient, structural rigidity, and the content of antifungal substance. After fermentation, the average water content of herbs were increased to $67.21{\pm}11.43%$ from $30.84{\pm}15.67%$, but the average pH and average methanol extraction ratio were slightly decreased to $11.16{\pm}7.06%$ and $4.83{\pm}0.73$ from $13.91{\pm}12.22%$ and $5.06{\pm}0.87$, respectively. The analysis of thrombin inhibition and DPPH scavenging activity of the methanol extracts of herbs showed that thrombin inhibition activities of the fermented Drynaria fortunei Kunze, Melia azedarach var. japonica, Prunus persica and Orostachys japonicus, and DPPH scavenging activities of the fermented Polygala tenuifolia, Scrophularia buergeriana, Angelica dahurica, Drynariafortunei Kunze, Cyperus rotundus, and Boschniakia rossica were increased as compared with those activities of non-fermented its cognate herbs. Our results suggest that the production of bioactivity-strengthen medicinal herbs is possible by solid fermentation of Phellinus baumii mycelium, as fermented Drynaria fortunei Kunze showed increased antioxidant and thrombin inhibitory activities than those of non-fermented herbs.

Studies on the Chemical Compositions of Citrus junos in Korea (한국산유자(韓國産柚子)의 화학적성분(化學的成分)에 관(關)한 연구(硏究))

  • The chemical components of Citrus junos produced in Korea were divided into two parts; common and special components respectively. In the former the relation between the physiological effects of the plant and its ripening process was observed periodically while the latter was analyzed the ripening fruits for their effective utilization as food. The results are summarized as follows: 1. The analytical result of seasonal change showed that the rind ratio was higher than the flesh ratio and on a regional basis, the rind ratio was higher in the islands than on land areas. 2. In the experiment the moisture was increased until the third period, but afterwards it was made constant. While the content of crude fat, cellulose, ash, total acid and soluble non-nitrogen material were decreased until the third period and the cotent of cellulose and total acid were continuousely redused until the last period. In con trast with the above the content of reducing sugars was increased but the content of crude fat, cellulose, ash, crude protein and soluble non-nitrogen material were increased until last period. 3. The content of vitamin C was richer in the rind than in the flesh, in the Korean species than in the Japanese. 4. Free sugars; xylose, fructose, glucose were richer in the rind than in the flesh. 5. The content of volatile organic acids was richer in the rind than in the flesh. Among them, volatile acids, acetic acid, formic and n-valeric acid were found in the rind and formic acid, acetic acid and propionic acid were deteceed in the flesh. 6. The total content of non-volatile acids was richer in the flesh than in the rind. In the kind of non-volatile acids, citric acid,glutaric acid, malic acid, tartaric acid, oxalic acid, malonic acid, succinic acid and an unknown acid were found in the rind and citric acid, malic acid, succinic acid, oxalic acid, glutaric acid and malonic acid in the flesh. 7. Three kinds of aromatic components: D-limonene, ${\alpha}-pinene$, p-cymene and seven other kinds of unknown aromatic components were detected in neutral essential oils. Among them, D-limonene seemed to be main aromatic component in the fruits. 8. From the above results it is confirmed that both rind and flesh of the ripened fruit could be utilized for food effectively, and unripened fruits are suitable for producing citric acid, ripened fruits are also useful for producing juice.

A Polarization-based Frequency Scanning Interferometer and the Measurement Processing Acceleration based on Parallel Programing (편광 기반 주파수 스캐닝 간섭 시스템 및 병렬 프로그래밍 기반 측정 고속화)

  • Frequency Scanning Interferometry(FSI) system, one of the most promising optical surface measurement techniques, generally results in superior optical performance comparing with other 3-dimensional measuring methods as its hardware structure is fixed in operation and only the light frequency is scanned in a specific spectral band without vertical scanning of the target surface or the objective lens. FSI system collects a set of images of interference fringe by changing the frequency of light source. After that, it transforms intensity data of acquired image into frequency information, and calculates the height profile of target objects with the help of frequency analysis based on Fast Fourier Transform(FFT). However, it still suffers from optical noise on target surfaces and relatively long processing time due to the number of images acquired in frequency scanning phase. 1) a Polarization-based Frequency Scanning Interferometry(PFSI) is proposed for optical noise robustness. It consists of tunable laser for light source, ${\lambda}/4$ plate in front of reference mirror, ${\lambda}/4$ plate in front of target object, polarizing beam splitter, polarizer in front of image sensor, polarizer in front of the fiber coupled light source, ${\lambda}/2$ plate between PBS and polarizer of the light source. Using the proposed system, we can solve the problem of fringe image with low contrast by using polarization technique. Also, we can control light distribution of object beam and reference beam. 2) the signal processing acceleration method is proposed for PFSI, based on parallel processing architecture, which consists of parallel processing hardware and software such as Graphic Processing Unit(GPU) and Compute Unified Device Architecture(CUDA). As a result, the processing time reaches into tact time level of real-time processing. Finally, the proposed system is evaluated in terms of accuracy and processing speed through a series of experiment and the obtained results show the effectiveness of the proposed system and method.

Effects of Chicken Treated with Hwangki-Beni Koji Sauces on Body Weight, Serum and Hepatic Lipid Profiles of Rats Fed High Fat and High Cholesterol Diets (황기홍국소스를 처리한 계육이 고지방 및 고콜레스테롤 식이 흰쥐의 체중과 혈청 및 간 조직의 지질 함량에 미치는 영향)

  • Dietary effects of chicken treated with Hwangki-Beni Koji sauces (HBS) on serum and hepatic lipid profiles in rats fed high a fat and high cholesterol diet were investigated. Experimental plots (5 rats per each group) were divided into normal control diet group (NC), high fat and high cholesterol diet group (HFC), HFC plus 15% base sauce treated chicken supplemented diet group (HFC-BS), HFC plus 15% HBS-treated chicken supplemented diet group (HFC-HBS), and fed for 5 weeks. Feed intakes in HFC-HBS group were higher than those of HFC and HFC-BS groups, whereas body weight gain and feed efficiency ratio of the HFC-HBS group were lower than those of the HFC and HFC-BS groups. Serum alanine aminotransferase and aspartate aminotransferase activities of HFC-HBS group were significantly lower than those of HFC and HFC-BS groups. Level of serum triglyceride, total cholesterol and LDL-cholesterol in HFC-HBS group were 34.06%, 6.28% and 31.00% lower than those of HFC-BS group, respectively, whereas HDL-cholesterol level and atherogenic index in HFC-HBS group were 1.72-fold higher and 2.52-fold lower, respectively, than those of HFC-BS group. Level of hepatic total lipid, triglyceride and total cholesterol in HFC-HBS group were 9.53%, 5.25% and 22.20% lower than those of HFC-BS group, respectively. In the morphological results of liver, the hepatocytes of central vein lesion and intact hepatic cell plate are preserved well from steatosis in HFC-HBS group, whereas the hepatocytes in HFC and HFC-BS groups are distended by accumulation of multiple fat droplets, and sinusoids are occluded. In the above results, we expected that chicken treated with Hwangki-Beni Koji sauce may have functionalities of anti-obesity, anti-cholesterol and also, lowering anti-atherogenic index in high fat and high cholesterol diets.

$^{17}O$ NMR Study On Water Excharge Rate of Paramagnetic Contrast Agents ($^{17}O$ NMR 기법을 이용한 상자성 자기공명조영제의 물분자 교환에 관한 연구)

  • Purpose : The water exchange rate between bulk water and bound water is an important parameter in deciding the efficiency of paramagnetic contrast agents. In this study, we evaluated the water exchange rates of various Gd-chelates using oxygen-17 NMR technique. Material and Methods : The samples (Gd-DTPA, Gd-DTPA-BMA, Gd-DOTA, Gd-EOB-DTPA) were prepared by mixing 5% $^{17}O-enriched$ water (Isotech, USA). The pH of the samples was adjusted to physiological value [pH=7.0] by buffer solution. The variable temperature $^{17}O-NMR$ measurements were performed using Bruker-600 (14.1 T, 81.3 MHz) spectrometer. Bruker VT-1000 temperature control units were used to stabilize the temperature. The $^{17}O$ spin-spin relaxation times (T2) were measured using Carr-Purcell-Meiboom-Gill (CPMG)I pulse sequence with 24 echo trains. The variable temperature T2 relaxation data were then fitted into Solomon-Bloembergen equations using least square fit algorithm to estimate the water exchange times. Results : From the measured $^{17}O-NMR$ relaxation rates, the determined water exchange rates at 300K are $0.42{\;}{\mu}s$ for Gd-DTPA, $1.99{\;}{\mu}s$ for Gd-DTPA-BMA, $0.27{\;}{\mu}s$ for Gd-DOTA, and $0.11{\;}{\mu}s$ for Gd-EOB-DTPA. The Gd-DTPA-BMA showed slowest exchange whereas Gd-EOB-DTPA had fastest water exchange rate. In addition, it was found that the water exchange rates (${\tau}_m$) of all samples had exponential temperature dependence with different decay constant. Conclusion : $^{17}O-NMR$ relaxation rate measurements, when combined with variable temperature technique, provide a solid tool for studying water exchange rate, which is very important in investigating the detailed mechanism of relaxation enhancement effect of the paramagnetic contrast agents.
