Changes in physicochemical components of Astragalus membranaceus fermented with Phellinus linteus (상황버섯균 접종 황기의 배양 중 이화학적 성분변화)

  • This study analyzed the changes in physicochemical components of Astragalus membranaceus (AM) fermented with Phellinus linteus. Moisture content, pH, total acidity, total reducing sugar content, extraction yield, free sugar content, free amino acid and isoflavonoid (calycosin, formononetin) were investigated. The moisture content was increased during fermentation with Phellinus linteus. The pH level increased while the total acidity significantly decreased during fermentation. The reducing sugar content were in the range of 0.32~0.61%. The extraction yield using water was higher than that using 80% ethanol. The major free sugars were identified as glucose, fructose, sucrose and the content of free sugars decreased through fermentation. However, the glucose and sucrose contents of the water extracts were increased. In addition, the free amino acid increased significantly during fermentation. Finally, calycosin and formononetins contents in water extracts of after 30 days of AM fermentaion with Phellinus linteus were (3.91 mg/100 g) and (1.38 mg/100 g), respectively. These results suggest that fermentation with Phellinus linteus could be used to increase the bioactivity of AM. The mycelium-fermented AM could be a valuable source of functional material and edible resource for industry.

Effect of Astragali Radix and Opuntia humifusa on Quality of Red Ginseng Drink (황기 및 천년초 첨가가 홍삼음료의 품질에 미치는 영향)

  • This study was performed to develop new functional red ginseng drinks with Astragali Radix and Opuntia humifusa. Optimum extraction conditions such as solvent property and temperature for Astragali Radix were determined by distilled water vs. ethanol (95%) ratio (0:100, 25:75, 50:50, 75:25) and 60 vs. $80^{\circ}C$. Water-soluble extracts at $80^{\circ}C$ showed higher antioxidant activities than fat-soluble extracts at $60^{\circ}C$. Viscosities of 1-2% (w/v) of Opuntia humifusa solution were similar to that of the 0.1% guar gum solution. Addtion of Astragali Radix (3% and 5%, w/v) and Opuntia humifusa (1.2%, w/v), especially, had effect on the changes of pH of the red ginseng solution(5%, w/v) during storage for 7 days. A significant difference during the storage was shown in total plate counts by addition of Opuntia humifusa (1.2%, w/v) and microorganisms were reduced by six log cycles. Significant antiproliferation effects of red ginseng (5%, w/v) solution with Astragali Radix (3% & 5%, w/v) and Opuntia humifusa (1.2%, w/v) on Colon26m-3.1 carcinoma (colorectal carcinoma) cell and U87-MG neuronale glioblastoma (brain carcinoma) cell were not observed.

  • Induction of phase II enzymes such as quinone reductase (QR) or glutathione S-transferase (GST) is considered a major mechanism of protection against initiation of carcingenesis. This study was desinged to investigate the potential of Astragali Radix Aqua-acupuncture Solution (ARAS) to induce phase II enzymes and glutathione (GSH) in murine hepatoma cells grown in microtiter plate wells. ARAS was potent inducers of QR activity. ARAS was induced about 2.6-fold at concentration of $5{\times}$. In addition, GST activity was increased with ARAS. GSH levels were increased about 1.2-fold with ARAS at concentration of $0.1{\times}$. These results suggested that ARAS may act as blocking agents against carcinogenesis by induction of phase II marker enzymes.

Removal of Cadmium and Manganese Ions Utilizing Astragalus uliginosus L.-Stem Biochar (황기 줄기 바이오차를 활용한 카드뮴과 망간 이온의 제거)

  • Astragalus uliginosus L.-stems as a by-product of oriental medicine are produced largely in a northern area of Chungbuk province. These by-products do not have any demand and thus usually discarded into the fields as a waste. In this work, a biochar was prepared from the Astragalus uliginosus L.-stem waste for recycling. The biochar was used to investigate the removal characteristics of cadmium and manganese ions dissolved in water. When adsorption equilibrium experiments were performed to treat 50 and 100 mg/L of cadmium ions, the removal efficiencies of cadmium were 100 and 95%, respectively. In addition, the maximum of adsorption amount for manganese ions in 5 h at an initial concentration of 50 and 100 mg/L was found to be as 36.1 and 37.9 mg/g, respectively. Based on the experimental results, it was found that the adsorption amount of Astragalus uliginosus L.-stem biochar for the removal of both cadmium and manganese ions was four times higher than that of the activated carbon. The surface analysis of both biochar and activated carbon samples using X-ray photoelectron spectroscopy (XPS) analysis showed that the oxygen content and O/C ratio of biochar was 2.1 and 2.4 times higher than that of the activated carbon, respectively. In order to enhance the removal capability of manganese, 50 and 100 mg/L of manganese ions were operated at different temperatures. It was observed that these equilibrium was attained in 4 h under 45 ℃ and removal efficiencies were 92 and 53%, respectively. Consequently, the experimental results can be utilized as a new removal technology for eco-friendly and economically treating cadmium and manganese ions dissolved in water.

Physicochemical Property and Antioxidative Activity of Hot-Water Extracts from Enzyme Hydrolysate of Astragalus membranaceus (황기 효소분해물 열수추출액의 이화학적 특성 및 항산화 활성)

  • To enhance the yield and bioactivity of hot-water extract from herbal medicine, Astragalus membranaceus was hydrolyzed with carbohydrases, such as ClariSEB and Fungamyl. After hot-water extracts were prepared from each hydrolysate (HW-C/F), physicochemical property, antioxidant activity and sensory property were evaluated. The solid content ($^{\circ}Brix$) of HW-C/F was higher than hot-water extract from A. membranaceus no treated enzyme (control). Although pH of HW-C/F was lower than that of the control, the acidity was higher. Lightness of Hunter's color values was increased in HW-C/F whereas redness and yellowness were decreased. The contents of reducing sugar, flavonoid and polyphenol of HW-C/F were higher than the control but the content of ascorbic acid was not different from control. The inhibitory activity of HW-C/F against lipid peroxidation was slightly higher than control, but DPPH radical scavenging, ABTS reducing, metal chelating activities were significantly increased by HW-C/F. The sensory evaluation also revealed that the sensory panelists preferred HW-C/F to that of control. Therefore, hydrolysis by carbohydrases for preparation of hot-water extract from A. membranaceus is one of the good methods to improve antioxidative activity and sensory property of hot-water extract.
