Analysis of the Benthic Nutrient Fluxes from Sediments in Agricultural Reservoirs used as Fishing Spots (낚시터로 활용중인 농업용 저수지의 퇴적물 내 영양염류 용출 분석)

  • For two agricultural reservoirs that are rented for fishing spots, benthic nutrient fluxes experiment were performed two times with two sediments from fishing-effective zone and one sediment from fishing-ineffective zone using laboratory core incubation in oxic and anoxic conditions. During benthic nutrient fluxes experiment, the changes in DO, EC, pH, and ORP in the supernatant were not significantly different between fishing-effective zone and fishing-ineffective zone, and were similar to the sediment-hypolimnetic diffused boundary layer in agricultural reservoir. Except for $NO_3{^-}-N$, more benthic nutrient fluxes of $NH_4{^+}-N$, T-P, and $PO{_4}^{3-}-P$ from sediment to hypolimnetic was measured in anoxic than in oxic conditions (p<0.05). As the DO concentration in hypolimnetic decreases, the microorganism-mediated ammonification is promoted, the nitrification is suppressed, and finally the $NH_4{^+}-N$ diffuses out from sediment to hypolimnetic. Also, the diffusion of T-P and $PO{_4}^{3-}-P$ from sediments to hypolimnetic is accelerated through the dissociation of the phosphorus bound to both organic matters and metal hydroxides. The difference in the benthic nutrient diffusive fluxes between fishing-effective zone and fishing-ineffective zone was not statistically significant (p>0.05). Therefore, it was found that fishing activities did not increase the benthic nutrient diffusive fluxes to a statistically significant level. Due to the short fishing activities of 10 years and the rate-limited diffusion of the laboratory core incubation, the contribution of fishing activities on sediment pollution is estimated to be low. No significant correlation was found between the total amount of nutrients in sediment and the benthic nutrient diffusive fluxes in both aerobic and anaerobic conditions. Therefore, nutrients input from various nonpoint sources of watersheds are considered to be a more dominant factor rather than fishing activities in water quality deterioration, and both aeration and water circulation in hypolimnetic were required to suppress the anoxic environment in agricultural reservoirs.

대구지역 대기환경용량평가에 관한 연구

  • 우리나라는 산업화와 도시화의 급속한 발전으로 인한 대기오염물질배출시설, 자동차 통행량, 에너지 사용량의 증가 등으로 대기오염물질배출원의 수와 규모가 증대되어 광역도시를 중심으로 대기질이 악화되고 있다. 특히 우리나라의 수도권대기질은 선진국의 주요 도시에 비해 대기오염 상태가 좋지 않은 것으로 평가됨에 따라 정부는 대기질을 OECD선진국 수준으로 개선한다는 목표를 세우고 있으나, 지역의 대기상태 및 그 동안의 대기질 개선을 위해 이행된 정책의 효율성 측면에서 볼 때 사후적인 규제위주의 농도규제 방식으로는 급증하는 대기오염배출시설의 배출량총량 관리가 어렵고, 지자체별로 개별적인 분산관리로는 광역적으로 이동되는 대기오염물질의 관리가 불가능하다. 또한 대기오염과 상관성이 큰 에너지정책, 산업정책, 도시계획 등 관련 정책과의 통합적 접근이 어렵기 때문에 사전에 이를 예방하는 총량관리가 요구되어 진다. 총량규제란 특정지역의 기상, 지형조건 등을 이용하여 대기환경용량을 산출하고 이를 기초로 지역별 배출허용총량을 할당하여 궁극적으로는 오염원별로 대상오염물질의 삭감량을 정하는 제도로 선진국에서는 대기환경용량을 바탕으로 1970년대부터 사업장을 중심으로 배출농도 규제와 함께 총량규제를 병행 실시하고 있으며, 최근에는 자동차에도 실시하고 있다. 우리나라에서도 대기환경보전법 제9조에서 환경기준을 초과하여 사람의 건강이나 재산, 동식물의 생육에 중대한 위해를 가져올 우려가 있다고 인정되는 경우에는 동 지역 또는 특별대책지역 중 사업장이 밀집되어 있는 구역에 대하여 배출되는 오염물질을 총량으로 규제할 수 있도록 규정하고 있다. 또한, 환경부는 2003년도에 서울, 인천, 경기도내 19개시 지역을 대상으로 대기오염물질의 배출총량을 관리하는 대기오염총량제 실시를 포함한 '수도권대기질개선에관한특별법'을 제정하였고, 현재는 사업장에게 연도별 배출허용총량을 할당하고, 할당량 이내로 오염물질을 배출하도록 관리하는 사업장 대기오염물질총량관리 제도로 시행 중에 있다. 그러나 수도권대기질개선특별대책을 수립하면서 총량관리의 본격 이행 및 배출권 거래제도 도입에 대한 특별법안이 제정되고 부분적으로 시행되고 있으나, 우리나라에 총량관리를 본격 이행하는데 있어서의 필요한 준비여건은 아직 초기 단계이고, 관련 연구의 수행실적 또한 수도권에 제한되어 적은 편이다. 따라서 현재는 총량관리가 수도권에 국한하여 실시되고 있으나, 점차 타 도시까지 광역적으로 확대될 것으로 예상되는 바 이에 필요한 제반 사항들에 대한 조사 분석을 통하여 정책방향을 설정하는 더 많은 연구가 필요할 것이다. 이러한 배경에서 본 연구에서는 대구지역을 대상으로 대기오염농도 및 대기오염물질 배출량에 대한 현황조사를 실시하고, 이를 토대로 고농도가 자주 발생되거나 그러할 가능성이 높은 $NO_X$을 대상으로 대기오염기여도를 평가하고 대기확산모델을 통한 대기환경용량을 산정하였다. 대기오염농도 현황을 살펴본 결과, 대구지역의 대기오염은 $NO_2$, $SO_2$, CO는 전형적인 1차오염물질의 변화경향을 보였으며, $PM_{10}$는 봄철에 황사의 영향을 크게 받는 것으로 나타나 실제 대구지역에서 배출되는 양을 추정하기 힘든 것으로 판단된다. 또한 $NO_2$는 공업, 상업지역에서 $SO_2$$PM_{10}$는 공업지역, CO는 상업지역, $O_3$은 교외지역에서 높은 농도를 나타내는 것으로 파악되었다. 대구지역의 대기오염물질 배출량 현황은 CO가 47%, NOX가 43%로 전체 배출량의 90%를 차지하였고, 2005년 이후 $NO_X$는 감소하고 $SO_X$가 증가하는 추세이다. 또한 배출원대분류 중도로 및 비도로이동오염원에서 발생되는 선 오염원이 75%로 대구지역에서 가장 크게 기여하는 것으로 나타났다. ISCST3 대기확산모델을 이용하여 대기환경용량을 산정하기위하여, 먼저 대구지역의 대기환경용량평가는 가시적인 위해성이 높고 개선정책이 용이한 $NO_X$을 대상물질로 선정하였고, 배출량과 오염농도간의 상관도가 0.659로 높은 것으로 판단되어, 배출량을 삭감하였을 때 대기오염농도의 개선이 명확히 나타나는 것을 알 수 있었다. 다음으로, 단위격자 당 한계배출율을 알아내는 작업을 실시하여, 대구지역을 동일하게 장기환경기준 80%수준인 22.4ppb를 만족시키기 위한 한계배출율은 2.23g/s가 필요한 것으로 파악되었고, 산출한 한계배출율을 이용하여 장기환경기준치 80%수준 달성을 목표로 하는 경우의 대기환경용량을 산정하고 실제 배출량과 비교 분석하였다. 그 결과, 대구지역 전체의 환경용량은 약 3만 톤으로 실제 배출량 2만2천 톤에 약 8천 톤 이상의 여유가 있는 것으로 파악되었다. 그러나 구역별로 상이한 차이를 보였으며, 이에 따른 구역별 개선정책이 필요할 것으로 사료된다. 대기환경용량을 파악한 후 단위격자 당 한계배출율을 초과하는 대상 지역을 추출하여 삭감한 결과 초과배출량의 80%를 삭감해야 대구지역 전체에서 50ppb이하 농도가 되는 것을 알 수 있었는데, 실제로 초과배출량의 80%를 삭감하는 것은 어려움이 있다고 판단되어, 대구지역을 동일한 %율로 삭감한 결과 30% 삭감했을 때 50ppb수준을 달성하였고, 50%삭감했을때 2007년 환경기준인 30ppb수준을 달성하였다. 또한 배출원대분류 중 기여율이 높은 도로와 비도로오염원을 50%삭감한 결과 도로이동오염원의 삭감만으로도 상당한 고 배출지역의 농도저감에 효과가 있는 것으로 파악되고, 비도로오염원을 포함하여 삭감하였을 때는 대구지역 전체에서 50ppb이하로 내려가는 것을 볼 수 있었다. 따라서 향후 총량규제의 실시에 맞추어 대구지역의 실제적인 환경용량의 정확한 파악과 고배출지역에 대한 삭감방법에 관한 더 많은 연구가 필요하다고 사료된다. 이 연구 결과는 앞으로 시행될 지역총량규제에 대한 기초적인 방법론으로 활용될 수 있을 것이다.

Accuracy Analysis of FKP for Public Surveying and Cadastral Resurvey (공공측량 및 지적재조사 사업 적용을 위한 FKP 정밀도 분석)

  • NGII (National Geographic Information Institute) has been providing VRS (Virtual Reference Station) service so that could determine precise positioning in real time since 2007. However, since the VRS service has to maintain the connected status with VRS server, the number of users who can use VRS service are limited by capacity of VRS server. To solve this problem, NGII has been providing FKP (Virtual Reference Station) service using one way telecommunication from November 1, 2012. Therefore, it is predicted that the usage of FKP service will increase in public surveying and cadastral resurveying in the future. However, the studies with respect to analysis of FKP precision for applying to public surveying and cadastral resurveying is not conducted enough. In this study, to analyse the application possibility of FKP on the public surveying and cadastral resurveying, the two kind analysis were performed. First is the analysis of accuracy according to the configuration of reference station of FKP and VRS. One is consisted of same reference stations, another is consisted of different reference stations. Second is the accuracy anlalysis of horizontal and vertical positioning acquiring VRS and FKP data in various measurement environment based on VRS regulation. Result of first study, Positioning accuracy according to the configuration of the reference stations satisfies related regulation. However, accuracy of FKP in case of different reference stations is worse than in case of same reference stations.. The result of second test shows that the horizontal precision of FKP and VRS in good measurement environment satisfy the allowed precision. However, in some case, horizontal precision of FKP and VRS in poor measurement environment exceed the allowed precision. In addition, the number of exceeding the allowed precision in the FKP is more than the VRS. The vertical precision of the VRS satisfy related work provision. In conclusion, the result of this study shows that the FKP only in open area should be used for public survey and cadastral resurvey. Therefore the additional studies with respect to the improvement of FKP precision should be conducted.

Changes in physicochemical components of Astragalus membranaceus fermented with Phellinus linteus (상황버섯균 접종 황기의 배양 중 이화학적 성분변화)

  • This study analyzed the changes in physicochemical components of Astragalus membranaceus (AM) fermented with Phellinus linteus. Moisture content, pH, total acidity, total reducing sugar content, extraction yield, free sugar content, free amino acid and isoflavonoid (calycosin, formononetin) were investigated. The moisture content was increased during fermentation with Phellinus linteus. The pH level increased while the total acidity significantly decreased during fermentation. The reducing sugar content were in the range of 0.32~0.61%. The extraction yield using water was higher than that using 80% ethanol. The major free sugars were identified as glucose, fructose, sucrose and the content of free sugars decreased through fermentation. However, the glucose and sucrose contents of the water extracts were increased. In addition, the free amino acid increased significantly during fermentation. Finally, calycosin and formononetins contents in water extracts of after 30 days of AM fermentaion with Phellinus linteus were (3.91 mg/100 g) and (1.38 mg/100 g), respectively. These results suggest that fermentation with Phellinus linteus could be used to increase the bioactivity of AM. The mycelium-fermented AM could be a valuable source of functional material and edible resource for industry.

Physicochemical properties and protease activities of microencapsulated pineapple juice powders by spray drying process (분무건조공정을 이용한 파인애플 착즙액 미세캡슐 분말의 물리화학적 특성 및 protease 활성)

  • The physicochemical properties and protease activities of spray-dried pineapple juice powders were investigated. The pH, soluble solids, and protease activity of the pineapple juice were pH 5.43, $12.8^{\circ}Brix$, and 4.82 unit/mL, respectively. The optimum pH and temperature of the protease activity from pineapple juice were pH 7.0 and $50^{\circ}C$, respectively. The microencapsulation of pineapple juice was achieved using maltodextrin and alginic acid through spray-drying. The L value and moisture content of the spray-dried powder were higher than those of the freeze-dried powder. The particle size of the freeze-dried powder ($501.57{\mu}m$) was higher than that of the spray-dried powder ($42.58-53.32{\mu}m$). The water absorption and water solubility of the powders were 0.41-0.87, and 90.45-99.76%, respectively. When compared, the protease activities were found to be in the following order : FD (1,297.47 unit/g) > SD-MA-1 (692.08 unit/g) > SD-MA-2 (664.66 unit/g) > SD-MA-3 (642.65 unit/g) > SD-M (633.51 unit/g). In the in vitro dissolution study measurements were conducted for 4 hr in pH 1.2 simulated gastric fluid and pH 6.8 simulated intestinal fluid, using a dissolution tester at $37^{\circ}C$ in 50 rpm. The protease survival of the 3.74-15.69% microencapsulated pineapple juice powders improved with an increase in the treatment concentration of alginic acid.

Quality characteristics of fermented vinegar prepared with the detoxified Rhus verniciflua extract (무독화 옻 추출물로 제조한 발효식초의 품질 특성)

  • In this study, vinegar was produced using urushiol-free fermented Rhus verniciflua extract to create a lacquer with added value. The effect of manufacturing conditions on the quality of vinegar using detoxified R. verniciflua extract for fermentation was investigated. The acidity of the vinegar for inoculations with various liquid starter contents was 4.8~4.9%, and it was similar among all treatment groups. The acidity of vinegar was higher when the initial alcohol content was high. The acetic acid yields were 82.8%, 84.4%, 77.7%, and 69.5%, and the maximum yield was observed when the initial alcohol content was 6%. For acetic acid fermentation using different amounts of detoxified R. verniciflua extracts, the acidity of the vinegar with the extract after fermentation was 5.3~5.9%. However, the acidity of vinegar without the extract was 5.5%. The intensity of the brown color was high for vinegar without the extract. Hunter's L values were high for vinegar with an extract content of 2%. Acetic acid (53.3~65.8 mg/mL) was the predominant acid. Arginine ($190.3{\sim}333.3{\mu}g/mL$), proline ($125.6{\sim}290.8{\mu}g/mL$), alanine ($126.1{\sim}270.9{\mu}g/mL$), and glutamic acid ($159.0{\sim}262.4{\mu}g/mL$) were the predominant amino acids in detoxified R. verniciflua vinegar.

Effect of pectinase treatment on extraction yield and physicochemical properties of Aronia juice (Pectinase 처리가 아로니아의 착즙 수율에 미치는 영향)

  • The main objective of this study was to investigate the effects of various commercial pectin enzyme treatments on juice yield from Aronia fruits (Aronia melanocarpa) as well as changes in physicochemical properties such as pH, total acid, reducing sugar, soluble solid, total anthocyanin, total phenolic compounds, and DPPH radical scavenging activity. Different types and reaction conditions of pectinase were also investigated in order to improve extraction yield of Aronia juice. The optimal conditions of enzyme treatment were 0.1% of concentration (w/w) at $50^{\circ}C$ for 120 min. Among enzymes used in this study, extraction yield with Rapidase Press L treatment from Aronia juice was the highest and resulted in a significant increase in juice from 51.0 to 69.1%. Rapidase C80 MAX showed 68.83% extraction yield while Plantase TCL showed 66.70% extraction yield. Reducing sugar and soluble solid contents increased after enzyme treatment. Total acids also slightly increased after enzyme reaction. No significant difference was observed in pH regardless of pectin enzyme treatment. However, enzyme treatment resulted in an increase in total phenolic compounds, total anthocyanin, and DPPH radical scavenging in Aronia juice compared to the juice prepared juice.

Protective Effect of a Herbal Preparation (HemoHIM) on the Self-Renewal Tissues and Immune System against γ-Irradiation (방사선에 대한 생약복합조성물(HemoHIM)의 재생조직 및 면역계 방호 · 회복촉진 효과)

  • In our previous study, a novel herb mixture (HIM-I) of Angelim gigas radix, Cnidium officinale rhizoma, and Paeonia japonica radix was developed to protect the intestinal and immune systems and promote its recovery against radiation damage. In this study, a new herbal preparation (HemoHIM) with the high immune modulating activity was developed from HIM-I. HIM-I was fractionated into ethanol fraction (HIM-I-E) and polysaccharide fraction (HIM-I-P). And HemoHIM was prepared by adding HIM-I-P to HIM-I. The protective activities against $\gamma$ -irradiation were compared among HemoHIM, HIM-I and the fractions. HemoHIM and HIM-I significantly decreased the radiation-induced DNA damage in vitro, and scavenged hydroxyl radicals in a dose-dependent manner. HemoHIM showed similar activity to HIM-I. In vitro proliferation assay with mouse lymphocytes and bone marrow cells showed that HIM-I-P was remarkably higher than HIM-I and HIM-I-E in cell proliferating activity. HemoHIM showed higher activity than HIM-I and this might be associated with the higher polysaccharide content. The in vivo protective effects of HemoHIM and HIM-I were investigated in $\gamma$-irradiated mice. HemoHIM increased the surviving intestinal crypts to a similar extent compared with HIM-I. In contrast, HemoHIM appeared to be more effective than HIM-I in endogenous spleen colony formation assay. The recovery of white blood cells and lymphocytes in irradiated mice were significantly enhanced by the administration of HemoHIM. Also HemoHIM administration prolonged the survival of irradiated mice. These results showed that the novel herbal preparation, HemoHIM, effectively protected the self-renewal tissues and immune system, and promoted the survival of irradiated mice. Moreover, in comparison with HIM-I, HemoHIM maintained similar activity in the reduction of oxidative damage of self-renewal tissue but exhibited the higher activity in protection and proliferation of immune and hematopoietic cells. These results suggested that HemoHIM might be more effective than HIM-I in immune modulation as well as radioprotection.

Quality Characteristics of Sikhe Prepared with the Roots Powder of Doraji (Platycodon grandiflorum A. DE. Candolle) (도라지 분말을 첨가한 식혜의 품질 특성)

  • In this study, we investigated the quality characteristics of Sikhye, a traditional Korean drink, made with various amounts (0.5, 1, 2, and 3%) of powder from the roots of Doraji (RPD), for different storage periods (1, 3, 5, 7, 10, 13, or 15 days). The pH and Hunter's L value of Sikhye decreased with higher amounts of added RPD, while saccharinity as well as "a" and b values increased. During storage, pH and Hunter's L value of Sikhye tended to decrease while "a" and b values increased. The saccharinity of Sikhye with 0.5% and 1% RPD reached its maximum values on the tenth storage day and continuously decreased during the storage period. In contrast, the saccharinity of Sikhye with 2% and 3% RPD increased during the storage period, and had its maximum saccharinity on the fifteenth storage day. During the storage of Sikhye the total microbial cell count, typically $3.19{\sim}0.35{\times}10^2$ CFU/mL on the third storage day, was undetected in Sikhye with 3% RPD. The total microbial cell count of Sikhye without RPD increased to $1.91{\times}10^9$ CFU/mL by the fifteenth storage day, while Sikhye with 0.5, 1, 2, and 3% RPD added contained $8.43{\times}10^8$, $9.77{\times}10^7$, $2.10{\times}10^7$, and $7.21{\times}10^6$ CFU/mL, respectively. Thus, the total cell count was lower with higher additions of RPD. In sensory tests, the Sikhye control, or with 0.5% and 1% RPD added, didn't show significant differences in the texture of rice granules, plumpness of rice granules, floating of rice granules, taste, flavor, and sweetness values. The overall preference was highest (5.38) for Sikhye with 0.5% RPD, followed by the control, 1% RPD, 2% RPD and 3% RPD. In conclusion, according to preference tests, we recommended the addition of 0.5% or 1% RPD in Sikhye.

Antioxidant and Angiotensin Converting Enzyme I Inhibitory Activities of Extracts from Mulberry (Cudrania tricuspidata) Fruit subjected to Different Drying Methods (건조방법에 따른 꾸지뽕열매 추출물의 항산화활성과 Angiotensin Converting Enzyme I 저해활성)

  • The physiological functionalities of 70% ethanol extracts (EE) from Cudrania tricuspidata fruit (ECFD, EE of C. tricuspidata subjected to freeze-drying; ECHD, EE of C. tricuspidata subjected to heat air drying; ECID, EE of C. tricuspidata subjected to infrared drying) were investigated. Yields of freeze-dried powders of ECFD, ECHD, and ECID were 51.50%, 53.91%, and 56.14%, respectively. Color $L^*$, $a^*$, $b^*$, and $H^{\circ}$ values of ECHD and ECID decreased. Dried ECHD and ECID had relatively higher contents of total polyphenolics and flavonoids than ECFD. Electron donating abilities at a concentration of 10 mg/mL (w/v) were in order of ECID (62.37%) >ECHD (80.17%)>ECFD (77.80%). Reducing powers ($OD_{700}$) of ECFD, ECHD, and ECID were 0.75, 1.70, and 1.89, respectively. Additionally, ABTS radical scavenging ability of ECID was the highest with a value of 62.37% at a concentration of 10 mg/mL (w/v). Nitrite scavenging activities of ECFD, ECHD, and ECID at a concentration of 10 mg/mL (w/v) were 28.76%, 30.69%, and 41.64%, respectively. SOD (superoxide dismutase)-like activities at 5 mg/mL (w/v) were in the order ECFD (891.93 mUnits)>ECHD (723.02 mUnits)>EFID (611.97 mUnits). Whereas ferrous ion chelating activity of ECFD (52.36%) and ECID (47.16%) was significantly higher than that of ECHD (30.04%). ACE inhibitory and xanthine oxidase (XO) inhibitory activities of ECHD and ECID at a concentration of 1 mg/mL (w/v) were higher than those of ECFD. In conclusion, we provided experimental evidence that extracts of pre-dried C. tricuspidata exhibit increased physiological functionalities. Further, infrared drying technique is the best method for enhancement of antioxidant activity of C. tricuspidata fruit.
