해양홍합 유래 바이오-접착소재 개발 동향

  • 잠재적인 생체의료용 소재로서 홍합접착단백질은 그 동안 커다란 관심을 받아왔었다. 홍합접착단백질은 환경친화적인 수중접착제로써 강하면서도 유연하게 다양한 물질들의 표면에 접착하며 수분에 강하고 인체에 무해하며 생분해되는 특성을 가지고 있다. 여러 가지의 홍합접착단백질들이 홍합으로부터 발견되어 그 특성들이 연구되었으며 홍합의 접착기작에 대한 생화학적 지식들이 축적되어 왔다. 또한 이렇게 가능성이 높은 해양홍합유래의 바이오-접착소재를 현실화하려는 많은 노력들이 시도되어 왔다. 본 고에서는 홍합접착단백질의 기능적 생산에 초점을 맞추어 다양한 개발 접근방법들의 진행들의 추이를 정리하였다.

Effects of Hot Water Extracts of Domestic Blue Mussel and New Zealand Green Lipped Mussel on Alcohol Metabolizing Enzymatic, DPPH Radical Scavenging, and Angiotensin Converting Enzyme Inhibitory Activities (국산 홍합과 뉴질랜드 초록입 홍합 열수 추출물의 알코올분해효소 활성에 미치는 영향 및 DPPH 라디칼 소거능과 Angiotensin Converting Enzyme 저해 활성)

  • The physiological activities of cultivated Korean blue mussel (Mytilus edulis) and New Zealand green-lipped mussel (Perna canaliculus) were analyzed and compared. Both hot water extracts of blue mussel flesh (BMF) and green-lipped mussel flesh (GMF) showed increased activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). BMF showed increased ADH and slightly decreased ALDH activities compared to GMF. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity of BMF was higher than that of GMF at the same concentration. BMF and GMF showed similar inhibitory activity against angiotensin converting enzyme at a concentration of 30 mg/mL. These results suggest that cultivated Korean blue mussel has similar physiological activity with New Zealand green-lipped mussel.

  • 홍합감시(Mussel Watch) 연구의 일환으로 1988년과 1989년에 남한 연안의 24곳에서 홍합(Mytilus)속에 속하는 종들을 채집하였다. 채집된 표본들의 패각의 형태 특성과 분포를 조사 분석한 결과 3종-홍합, 진주담치(M. edulis galloprovincialis), Mytilus sp. 로 밝혀졌다. 이 3종은 패각의 형태에서 뚜렷한 차이를 보이고 있으며, 지리분포와 서식지 수직분포에서도 나름대로의 특성을 보여주고 있다. 지리분포에서는 수온이 수직분포에서는 종간 경쟁이 분포의 제한 요인인 것 같다.

  • 홍합은 수중 환경에서 다양한 표면에 강하게 붙어서 자라는 수중 생물이며, 이러한 홍합의 특이적인 접착능력은 많은 연구자들에게 관심을 받고 있다. 홍합의 접착력에 관여하는 화학적 작용기를 모방한 폴리도파민/폴리노르에피네프린 표면 개질 기술은 자연의 홍합이 가지는 특성을 그대로 가지고 있어 수용액 상의 조건에서 표면의 성질에 관계없이 거의 모든 표면에 뛰어난 접착력을 나타낸다. 이러한 자연 모방 표면 개질 기술은 다양한 생체/에너지 재료, 신소재 복합 재료 등의 개발에 응용되고 있으며, 다양한 분야에서 활용될 수 있다.

Preparation and Evaluation of Physiological Activity of White Bread Containing Korean Blue Mussel (국산 양식 홍합 함유 식빵의 제조 및 생리활성 평가)

  • White breads containing Korean blue mussel (Mytilus edulis) powder were prepared and characterized. WB (white bread without blue mussel) and four different MBs (white breads containing blue mussel; number in front of MB means added % of blue mussel powder per wheat flour) were prepared by the straight dough method. With addition of blue mussel to bread, lightness decreased, whereas redness and yellowness increased. Addition of blue mussel did not significantly affect specific volumes of breads. DPPH and ABTS radical scavenging activities significantly increased with increasing blue mussel content. Addition of blue mussel to breads also increased alcohol dehydrogenase and acetaldehyde dehydrogenase activities. In the sensory test, 1MB acquired relatively high points for taste, flavor, texture, and preference. The results indicate that blue mussel can be applied to white bread to improve physiological functions without reduction of physicochemical characteristics.

Study on Biochemical Pollutant Markers for Diagnosis of Marine Pollution X. Changes in Oxygen Radicals and Their Scavenger Enzymes of the Mussel (Mytilus coruscus) in the South Sea (해양오염의 진단을 위한 생화학적 오염지표에 관한 연구 X. 남해산 홍합 (Mytilus coruscus)의 산소 라디칼 및 제거효소의 변화)

  • This study was designed as a part of efforts to investigate the biochemical pollutant markers for diagnosis of marine pollutions by changes in oxygen radicals and their scavenger enzymes of the mussel (Mytilus coruscus) in South Sea of Korea. Protein contents in muscle of cultured mussel in South Sea were remarkably lower (4-14%, respectively) than those of wild mussel in Pohang of East Sea. Superoxide radical activities in muscle of cultured in South Sea were significantly higher 82∼138% than those of wild mussel in Pohang. Hydroxyl radical formations in muscle of cultured mussels in South Sea were significantly 9∼25% higher than those of wild mussels in Pohang. Superoxide dismutase (SOD) activities in muscle of cultured mussels in South Sea were significantly 16∼28% lower than those of wild mussels in Pohang. It is believed that significantly decrease of protein contents in muscle, remarkable increases of superoxide radical and hydroxyl radical in muscle of cultured mussels of South Sea may be used as a biochemical pollutant markers for diagnosis of marine pollutions. These results suggest that near-coastal water as well as neritic water of the south sea might be affected by pollutant.

  • 해양생명체인 홍합(mussel)은 접착단백질(adhesive protein)을 생산, 분비함으로써 홍합 자신을 바다 속의 바위와 같은 젖은 고체표면에 단단히 부착할 수 있도록 하여 강한 파도의 충격이나 바닷속의 부력 효과에 대하여도 저항할 수 있다. 홍합접착단백질은 현재 알려진 어떠한 화학합성 접착제보다도 강력한 자연적인 접착제로 알려져 있으며 대부분의 에폭시 수지보다 두 배 정도의 커다란 인장강도를 지니고 있으면서도 휘어질 수 있는 유연성을 가지고 있는 혁신적인 물질이므로 접착제 산업계에서 매우 커다란 관심을 끌고 있다. Attachment of Hard Shelled Mussel, Mytilus coruscus and Blue Mussel, Mytilus edulis (홍합 Mytilus coruscus과 지중해담치 Mytilus edulis의 부착 비교)

  • In this study, the reattachment processes of small and medium size of spot of Mytilus coruscus and M. edulis were observed. The small spats (Shell length, SH, $3{\sim}7mm$) of hard shelled mussel, M. coruscus showed 100% reattachment rate within 7 minutes after the detachment of byssus thread and 100% survival rate. While large spats (SH 25 mm) showed 85% reattachment rate within 5 hours and 100% survival rate for 24 hours. The reattachment rate of M. edulis (SH 30 mm) was higher than that of M. coruscus (SH, 28 mm). The thread consisted of three identifiable structures; adhesive disc, adhesive thread and adhesive root. The adhesive disk and adhesive thread of M. coruscus were larger and thicker than those of M. edulis, whereas the thickness of adhesive root was the other way round. Further studies are required to identify the relationship between the structure of byssus thread and attachment ability. This study suggests that the sizes of spats could be an important factor in determining the timing of removing and reattaching mussel seedling for aquaculture or releasing to the sea.

Effect of Hot Water Extracts of Blue Mussel and Several Plants on Alcohol Metabolizing Enzymatic, Antioxidant, and Angiotensin Converting Enzyme Inhibitory Activities (홍합과 여러 식물의 열수 추출물의 알코올분해효소에 미치는 영향과 항산화 및 ACE 저해 활성)

  • For the development of a hangover soup containing blue mussel, 11 kinds of hot water extracts were prepared - A (mistletoe); B (shepherd's purse); C (arrowroot); D (bean sprout); E (oriental raisin); F (blue mussel); G (blue mussel and mistletoe); H (blue mussel and shepherd's purse); I (blue mussel and arrowroot); J (blue mussel and bean sprout); and K (blue mussel and oriental raisin). Extract C showed the highest effect for increasing the activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH); however, the addition of blue mussel did not provide a synergy effect on extract C. Other than the arrowroot-containing extracts (C and I), extract H showed relatively higher ADH ($237.4{\pm}1.7%$) and ALDH ($136.5{\pm}2.1%$) activities. Moreover, extract H showed the highest DPPH radical scavenging activity ($93.9{\pm}0.1%$) and angiotensin I converting enzyme (ACE) inhibitory activity ($42.7{\pm}1.6%$). The combination of blue mussel with shepherd's purse had a synergic effect on its ADH and ACE inhibitory activities.

Lipid Oxidation in Shellfish under the Different Conditions of Drying (패류의 건조조건에 따른 지질산화)

  • This study was carried out in order to investigate oxidative deterioration during dehydration at $40^{\circ}C,\;50^{\circ}C$ and $60^{\circ}C$ of sea mussel and baby clam. Moisture content was decreased with drying temperature and time. Sea mussel was dehydrated more rapidly than baby clam that had Harder muscle tissue. Both samples were not reached to Aw 0.62 in case of 10 hrs drying at $40^{\circ}C$, But it reached within 8 hrs in sea mussel and 10 hrs in baby clam at $50^{\circ}C$, respectively. Even if $60^{\circ}C$ could speed up drying, it caused to form more free fatty acid, peroxide, thiobarbituric acid and brown pigments. Lipophilic brown pigment was 10 times higher than hydrophilic and actively increased in all samples. fluorescence intensity was also increased with drying temperature and time. Particularly, it was higher sea mussel than baby clam more or less.
