Formation Periods of Carpospores and Tetraspores of 16 Red Algae in Daesongri, Kyungsangnam-do, Korea (경남 대송리산 홍조 16종의 과포자 및 사분포자의 형성시기)

  • Field study on the sporogenesis periods of 16 red algae growing at the intertidal zone of Daesongri was conducted from April 1987 to March 1988, obseuing the occurence of carposporic and tetrasporic fronds. The period of Gloiopeltis furcata was in spring and summer; that of Laurencia intermedia, Laurencia undulata and Rhodymenia intricate in summer; that of Chondria dasyphylla and Hypnea charoides in autumn; that of Carpopeltis affinis, Pachymeniopsis lanceolata, Chondrus ocellatus, Awosorium polyneunm, Gymnogongrus flabelliformis, Gigartina intermedia, Champia parvula and Ceramium paniculatum in summer and autumn. Of the species, G. flabelliiformis, G. intermedia and R. intricate only matured the carposporic phase, and P. telfairiae, H charoides, C. parvula and C. paniculatum did the tetrasporic phase. But the carposporic phase of G. flabelliformis and the tetrasporic phase of C. paniculatum were not occured in Kori area where water temperature was 3 C higher than that in Daesongri. In that area they only matured the other phase. This suggests that the life history is rarely complete in nature and water temperature is a important factor influencing on the sporogenesis. Also it was observed in C. affinis, C. ocellatus and S. latiuscula that the period of one phase is much longer than that of the other phase. But in most of species with both phases, tatrasporic and caropsporic, the former phase appeared a month later than the latter phase. In addition, their biomass became maximum just before sporgenesis, and then gradually decreased with release of reproductive cells.

Direct Observations of Spawning Characteristics on the Hexagrammidae Fishes in Korean Coastal Waters Using SCUBA Diving (쥐노래미과 어류 산란특성 연구를 위한 잠수조사)

  • The spawning characteristics of Hexagrammos agrammus and Hexagrammos otakii was directly observed in coastal regions of the Yellow Sea and South Sea during the spawning seasons using scuba diving. The territorial male H. agrammus showed different body size, nuptial coloration and nest location compared to those of H. otakii, even though both species caring egg masses were found at the almost same depths. In the West sea, a relatively small guardian H. agrammus male protects 1-2 egg masses laid on a clump of red algae within his territory. The territorial males in red nuptial coloration and their egg masses were well camouflaged in their surroundings. Contrary to H. agammus, a guardian H. otakii male in yellowish nuptial coloration protects 4-8 egg masses in a nest. In addition, their nests were located on the relatively open environments such as discarded fishing nets, rock crevices and a clump of algae. The results of present study shows that even though both of two species belonged to Hexagrammidae, they have different strategies for spawning and protecting their egg masses.

UTILIZATION OF UNEXPLOITED ALGAE FOR FOOD OR OTHER INDUSTRIAL USES (미이용해조류의 이용화에 관한 연구 II. 홍조류의 carrageenan함량과 그 화학적성상)

  • Three species of Rhodophyceae namely Chondrus ocellatus, Grateloupia filicina and Gigartina tenella were collected from the coast of Haeundae, Busan, on June 33, 1975. And analysed with respect to the content of carrageenan and such chemical characteristics as the content of sulphate and 3, 6-anhydrogalactose, the solubility in potassium chloride solution. In addition, the same chemical properties were tested on the fractions separated by the different concentration of potassium chloride. Carrageenan content in Chondrus ocellatus was relatively higher than two other samples. All the samples showed more than thirty five percent. The Gigartina-carrageenan showed the highest 3,6-anhydrogalactose content among three samples of carrageenan obtained from different species, and the lowest marked from the Grateloupia-carrageenan. In comparison of the solubility of carrageenan in potassium chloride solution, the precipitation yields at 0.125M potassium chloride were marked in order of Chondrus-carrageenan, Grateloupia-carrageenan, and Gigartina-carrageenan, and the yields at 0.125 M to 2.0 M potassium chloride were in Gigartina-carrageenan, Grateloupia-carrageenan, and Chondrus-carrageenan, and the yields from the soluble fraction at 2.0 M potassium chloride were in order of Grateloupia-carrageenan, Gigartina-carrageenan, and Chondrus-carrageenan. It is noteworthy from the result of characteristics of carrageenans that, 3,6-andydrogalactose content was closely related to the solubility of carrageenans in potassium chloride solution and to the sulphate content which might affect reversibly.

Antioxidative Activity of Methanolic Extracts from Seaweeds (해조류 메탄올 추출물의 항산화효과)

  • In this study, seven species of seaweeds (Sargassum horneri, Grateloupia filicina, Kjellmaniella crassifolia, Porphyra tenera, Ecklonia stolonifera, Scytosiphon lomentaria, Agarum cribrosum) from Gangwon coast were collected for antioxidative assays. The methanol soluble and insoluble (water soluble) fraction which were prepared from 75% methanol extraction were assayed by DPPH method and deoxyribose assay. The antioxidative activity of methanol soluble fraction of Agarum, Scytosiphon and Sargassum reached to 26.60%, 24.28% and 23.40%, respectively. The antioxidative activities of methanol insoluble fractions of the Kjellmaniella and Porphyra were assayed to 29.70% and 21.01% which were higher than methanol soluble fractions with the values of 9.26% and 8.66%, respectively. The results from DPPH assay and deoxyribose assay showed the methanol soluble fractions of Agarum and Sargassum have strong antioxidative activity. However the methanol insoluble (water soluble) fractions of seaweeds did not show any meaningful antioxidant activity excepted Kjellmaniella and Porphyra.

The Inhibitory Effects of Acanthopeltis japonica on Melanogenesis (새발 추출물의 멜라닌 생성 저해 효과)

  • To develop the skin whitening agent, we investigated the effects of Acanthopeltis japonica, a rhodophyta on the coast of Jeju island, on melanogenesis. Dried A. japonica was refluxed with 70 % aqueous ethanol and the extract was evaporated to dryness. To validate the activity as a depigmenting agent, various in vitro tests, polyphenol contents, and free radical scavenging activity were performed. In addition, cellular tyrosinase activity and protein expression of p-ERT, tyrosinase, TRP-1, and TRP-2 were measured in B16/F10 murine melanoma cells. A. japonica had low polyphenol contents and low free radicals scavenging activities against 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical. A. japonica suppressed cellular tyrosinase activity up to 86.9 % at $100{\mu}g/mL$ with inhibition or tyrosinase and TRP-1 expression in ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-treated B16/F10 melanoma cells. Our results suggest that inhibitory effects of A. japonica on melanogenesis are due to inhibiting the pathways involving ${\alpha}$-MSH-induced ERK activation. Therefore, A. japonica nay be useful as a skin whitening agent associated with the suppressive effect of melanotrophin-induced signaling pathway to inhibit melanin synthesis.

Taxonomic Reassessment of the Genus Polysiphonia (Rhodomelaceae, Rhodophyta): Morphology and Reproduction of Neosiphonia decumbens and N. harlandii (홍조 붉은실속 식물의 분류학적 재검토: 누은새붉은실과 가시새붉은실의 형태와 생식)

  • Neosiphonia is separated from the traditionally well known genus Polysiphonia (Rhodomelaceae, Rhodophyta) and 12 species includes in Korea. In this study, the vegetative and reproductive developments of two Neosiphonia species, N. decumbens (Segi) M.S. Kim et I.K. Lee and N. harlandii (Harvey) M.S. Kim et I.K. Lee, are reinvestigated. N. decumbens is diagnosed by following combination of the characters: plants 1-3 cm high, dwarf, decumbent, main axes indistinct, irregularly branched in dichotomous manner, and alternately or secundly ramified with wide angles above. N. harlandii is distinguished by the features: plants 4-8 cm high, usually solitary and saxicolous, erect, densely corticated at base of distinct main axes, rather irregularly alternate in branch, and with cicatrigenous branchlets developed numerously on every part of frond. The two species share typical characteristics of the genus Neosiphonia, such as a base attached by unicellular rhizoids, cut off by cross wall, pericentral cells in 4, trichoblasts moderately developed near the apex of branches, leaving persistent scar-cells, tetrasporangia arranged in a spiral series, procarps with 3-celled carpogonial branch, and spermatangial branches arising as a primary branch of trichoblast. Taxonomy of the two Neosiphonia in regard to Polysiphonia is discussed.

Radical Scavenging Activity of Ethanol Extracts and Solvent Partitioned Fractions from Various Red Seaweeds (홍조류 에탄올 추출물 및 다양한 용매 분획물의 라디칼 소거능)

  • The EtOH extracts of red seaweeds (Symphyocladia latiuscula, Chondrus ocellatus and Carpopeltis affinis) and solvent partitioned fractions were investigated for their 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and 1,1-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging effects and the total phenolic contents were correlated with ABTS and DPPH radical scavenging activities. The EtOH extracts and their solvent partitioned fractions exhibited strong ABTS and DPPH radical scavenging activities. Among the solvent partitioned fractions obtained from n-Hexane (HX), methylenchloride (MC), ethylacetate (EA), and buthanol (BuOH), the HX fraction from C. affinis showed higher radical scavenging activities than other fractions. Total phenolic contents showed significant correlation ($r^2$ = 0.709) with ABTS radical scavenging activity. The results of this study suggest that the strong radical scavenging activity of HX fraction from C. affinis is a promising natural antioxidant for healthcare products.

Industrial Applications of Saccharification Technology for Red Seaweed Polysaccharide (산업적 응용을 위한 홍조류 당화 기술)

  • Recently seaweed polysaccharides have been extensively studied for alternative energy application. Because their producing cost is high and efficiency low, their industrial applications have been limited. The main component of cell wall of red algae represented by Gelidiales and Gracilariales is agar. Red-algae agar or galactan, consisting of D-galactose and 3, 6-anhydro-L-galactose, is suitable for bio-product application if hydrolyzed to monomer unit. For the hydrolysis of algae, chemical or enzymatic treatment can be used. A chemical process using a strong acid is simple and efficient, but it generates together with target sugar and toxic compounds. In an enzymatic hydrolysis process, target sugar without toxic compounds generation. The objective of this review is to summary the recent data of saccharification by chemical and enzymatic means from red seaweed for especially focused on automobile industry.

A Study on Carpospore Release Induction Method of Agarophyton vermiculophyllum (홍조류 꼬시래기(Agarophyton vermiculophyllum)의 과포자방출 유도 방법에 대한 연구)

  • The aim of this study was to examine optimal induction method for carpospore release from Agarophyton vermiculophyllum cystocarps for seedling production. We tested the effects of environmental factors on carpospore release by using five different induction methods; spontaneous, desiccation, low temperature, desiccation+low temperature, and osmotic shock. Also, carpospores release was estimated at three temperatures (20, 25, and 30℃), and then under combinations of three day lengths (8, 12, and 16h) and two irradiances (30 and 60 μmol photons m-2 s-1), after pretreatment at desiccation+low temperature for 2 hr. The number of carpospores released was between 113 ~ 682 spores /cystocarp/day and it was maximal in the desiccation+low temperature treatment. Optimal environmental conditions for carpospore release of A. vermiculophyllum were 25℃, 16 h, and 60 μmol photons m-2 s-1. The present results suggest that massive carpospores for seedling production of A. vermiculophyllum could be obtained under a combination of 25℃, 16 h, and 60 μmol photons m-2 s-1 after pretreatment in the desiccation+low temperature.

Two Case Reports of Facial Flushing of Women in Their Twenties Treated by Sasang Constitutional Medicine (사상방을 이용한 20대 여성 안면홍조 환자 치험 2례)

  • Objectives: The purpose of this study is to report the effect of Sasang constitutional medicine and acupuncture on facial flushing of women in their twenties. Methods: Two patients who had facial flushing were treated with Sasang constitutional medicine and acupuncture. In the first case, the patient diagnosed with Taeeumin was treated by Chungpesagan-tang and Sa-Am Liver-sedating Acupuncture. In the second case, the patient diagnosed with Soeumin was treated by Palmulgunja-tang, Sa-Am Spleen-tonifying Acupuncture and Seungyangsungi-tang which is not belong to Sasang constitutional medicine. The progress of symptoms was evaluated by NRS, frequency of facial flushing, duration time of facial flushing, amount of sweating accompanied by facial flushing. Results: After treatment, the patients' symptoms of facial flushing were improved. Conclusions: These case studies show that Sasang constitutional medicine and acupuncture are effective for the patient in their twenties who had facial flushing.
