수율 계수와 목적 함수의 최적 유가배양에 대한 영향

  • 1. 수율계수 함수의 영향 : 유가 배양에 있어서 수율 계수가 제한 기질 농도의 함수일 경우에 대한 연구를 수행하였다. 사용한 수율 계수 함수는 기질 농도에 대하여 선형 함수였으며 목적함수는 중간 대사 물질의 생산량이었다. 1-가) 수율 계수 함수의 기울기가 음의 값으로 적어짐에 따라 제한 기질의 공급속도가 증가되었다. 1-나) 일반적으로 초기의 얼마 동안은 회분 배양에서와 같이 공급 속도가 거의 없었으며 그후 전환점 이후에 가서야 공급 속도가 증가하였다. 1-다) 전환점 이후에서도 처음에는 대부분의 경우에 공급속도가 음의 값으로 부터 시작하여 증가하기 시작하였다. 1-라) 어떤 경우에는 배양 초기부터 공급 속도가 양의 값으로부터 시작하였으나 곧 감소하였다가 다시 증가하는 비 단조형 변화를 보였다. 1-마) 전환점 이후에서 공급 속도가 증가하는 형태에서도 처음에는 선형 증가를 보이다가 나중에 지수적 증가를 보였다. 1-바) 위와 같은 공급 속도의 변화에 따른 발효조 내의 제한 기질 농도의 변화는 초기에는 가능한 최대치를 유지하다가 전환점 이후에는 갑자기 감소하는 형태를 취하였다. 2. 목적 함수의 영향: 목적 함수가 균체량과 생성 중간 대사물질 량의 선형 결합일 때 대하여 연구하였다. 선형 결합 계수로는 균체량과 생성대사 물질의 상대적인 값어치를 취하였다. 2-가) 허용 최대 균체 농도가 제한되어 있는 상황 하에서 생성 대사 물질의 상대적인 값이 증가 할수록 목적 함수가 증가하였다. 2-나)생성대사 물질의 상대적인 값이 증가할수록 전환점의 위치가 줄어들었다. 2-다) 생성대사 물질 상대적 값의 단위 증가에 대한 목적 함수는 균체량을 감안하지 않은 야마네박사의 결과에 수렴하였다. 2-라) 공급 속도는 상대 값이 커질수록 줄어들었다. 2-마) 균체량의 시간에 대한 곡선은 상대값이 증가할수록 감소하고 전형적인 분기점 형태의 변화를 보였다.변이 뚜렷하여 이로 인한 외임파강내의 염증성병변이 뚜렷이 나타나 있으며 와우관의 특히 기저회전에서의 유모세포의 손실이 심한 것으로 보아 중이염으로 인한 골도의 고음역에서의 손실이 발생함을 알 수 있다.A group), vinclozolin, procymidone, tetradifon cypermethrin, 그리고 fenvalerate(B group)에서는 70% 이상의 회수율을 얻었고, 20%의 ethyl acetate/hexane을 사용하여 분석한 경우는 5%나 10%의 ethyl acetate/hexane을 사용하여 분석했을 때보다 좋은 결과를 나타내어 전체 16종의 농약 중 14종에서 75% 이상의 회수율을 얻었으나, alachlor와 bifenthrin은 매우 낮은 회수율을 보여서 ethyl acetate와 hexane의 혼합용매가 이들 두 농약성분의 분석에는 적합하지 않은 용출 용매임을 알 수 있었다.NO_2의$ 급성 중독증상으로서는 눈, 코를 강하게 자극하고 폐충혈, 폐수종, 기관지염, 폐염 등을 일으킨다. 만성 중독시에는 만성폐섬유와 및 폐수종을 일으킨다. (4) $오존(O_3)$ Ozone은 자동차 배기가스에서 나오는 $NO_2$ gas 및 탄화수소와 작용하여, PAN이라는 자극성 물질을 생성시키는 광학적 Smog의 주요소로 알려져 있다. 자극적인 냄새가 있음으로 불쾌감을 주고 비, 인후점막의 전조감과 두통이 오며 폐기능을 저하시키며 더욱 진행되면 폐충혈, 폐수종 등을 일으킨다 (5) Smog에 의한 건강피해 대표적인 것이 1952.12.5~12.8까지 4일간 영국 Lon-don에서 계속된 Smog사건이며, 이 사건으로 말미암아 호흡기질환 사망율이 사건전보다 사건기간 중 혹은 사건후에 5~10배의 증가율을 보였다. 이때 Smog의 주원인은 연료의 불완전 연소에 의한 연기와 이때 발생

Application Effect of Organic Fertilizer and Chemical Fertilizer on the Watermelon Growth and Soil Chemical Properties in Greenhouse (유기질비료와 화학비료의 시용수준에 따른 시설수박 생육과 토양화학성의 변화)

  • BACKGROUND: Organic fertilizers in watermelon cultivation are widely used to supply nutrient and organic matter. This study was conducted to investigate the effects of application rate of organic fertilizer on the watermelon growth and soil chemical properties in greenhouse METHODS AND RESULTS: The organic fertilizers used in this experiment were mixed expeller cake (MEC) and mixed organic fertilizer (MOF). The treatments were conducted with 4 levels (1.0 N, 0.7 N, 0.5 N and 0.3 N) on the basis of soil testing nitrogen fertilization (STNF) using MEC or MOF as the basal dressing, and using chemical fertilizers (CF) as the additional dressing on the rest of STNF. These fertilizations were compared to CF 1.0 N (0.3 N as the basal and 0.7 N as the additional dressing) and non fertilization (NF). The leaf area of watermelon in treatment 0.5 N and 0.3 N using MEC or MOF was similar to CF treatment. The absorbed nutrient amounts by leaf, weight and sugar contents of fruit in the 0.5 N and 0.3 N treatments were higher than other treatments. In 0.5 N and 0.3 N treatments using MEC or MOF on the basis of STNF, soil chemical properties such as electrical conductivity (EC), available $P_2O_5$ and exchangeable K concentrations after experiment showed tendency to decreasing or similar level before experiment. CONCLUSION(s): These results suggest that the MEC or MOF application as the basal dressing at the 30~50% level of STNF and CF application as the additional dressing on the rest of STNF be best to maintain adequate nutrient of soil and to increase marketable yield for watermelon.

Effects of Soil mixtures and Soil Depths on the Growth of Zoysia japonica for the Artificial Planting Ground (인공지반의 토양조성과 토양심도가 중엽형들잔디의 생육에 미치는 영향)

  • To develope appropriate soil base for planting on the artificial ground, 9 kinds of soil types(varying the components and mixing ratios) and 3 levels of soil depths(10cm, 15cm, 20cm) were chosen. And their plant growing effects were tested and analysed from the test plant Zoysia japonica. The results of the research are as follows. 1. Among the 9 type of soil mixtures, the "sandy loam" soil type gave the worst effects on germination, disease contamination and ground covering. 2. The soil types like VSH(vermiculite20%+sand70%+humus sawdust10%), VSS(vermiculite 40%+sand 50%+humus sawdust 10%) and VS(vermiculite 70%+sand 30%), where vermiculite and sand were added to, show better germination effect promoted from the better condition of aeration and saturation. 3. The plant growing effects(leaf length and ground covering ratio) was evident under the soil types like VSH(vermiculite20%+sand70%+humus sawdust10%) and VSS(vermiculite40%+sand50%+humus sawdust10%), where organic matters were added to. 4. Vermiculite added soil types effect fast leaf decolorization on the tested Zoysia japonica plant, on the contrary to organic matter mixed soil types including SCS(sandy loam 50%+carbonized rice husk30%+sand20%) and SHS(sandy loam 50%+humus sawdust30%+sand20%) with which green leaves subsist longer. S. Soil depth effect to plant growth was found. And a favorable covering rate was accomplished even at the soil depth of 15cm - the limit soil depth for grass survival - from the soil types where organic matters were mixed to. From this result, the soil depth limit for plant survival could be said to be shall owed if appropriate soil type were based.

Evaluation of Replacement Ratio of Organic Fertilizers for Basal Application of Nitrogen Fertilizer in Pot Cultivation of Rice (벼 재배 시 유기질비료의 질소 밑거름 대체량 평가)

  • The objective of this study was investigated the effect of organic fertilizer(OF) application on the grain yield, protein content, and soil chemical properties in rice cultivation to evaluate the replacement ratio of OF based on basal fertilization of inorganic(nitrogen) fertilizer, In case of soil 1 with optimum range of organic matter for rice growth, the grain yield of rice in 70% and 100% treatments of OF were higher than the other treatments. In soil 2 with higher than the optimum range of soil organic matter, the grain yield of rice of NPK, 30%, 70%, 100%, and 300% treatments of OF showed no significant difference. When the protein criteria of rice quality was below 7%, the treatment of 30%, 70%, and 100% using OF in soil 1 were satisfied with protein criteria of rice. In case of soil 1, ammonium nitrogen content, electrical conductivity, and the organic matter in 30%, 70%, 100%, 300% treatments of OF showed similar tendency to NPK treatment. These results suggested that the substitution ratio of OF 70~100% could be appropriate to increase the yield and commercial quality for rice.

Degradation of Phenanthrene and Pyrene by Burkholderia sp. D5 (Burkholderia sp. D5에 의한 phenanthrene과 pyrene 분해)

  • Burkholderia sp. D5, a polyaromatic hydrocarbons(PAHs)-degrading bacterium, was isolated from oil-contaminated soil. The bacterium could utilize phenanthrene (Phe) as a sole carbon source but could not use pyrene (Pyr). However, the strain could degrade Pyr when a cosubstrate such as yeast extract (YE) was supplemented. The PAH degradation rate of the bacterium was enhanced by the addition of other organic materials such as YE, peptone and glucose. YE was a particularly effective additive in stimulating cell growth as well as PAH degradation. When 1 g-YE/L was supplemented into the basal salt medium (BSM) with 215 mg-Phe/L, the specific growth rate (0.28 h-1) and Phe-degrading rate (29.30 μmol/L/h) were enhanced approximately ten and two times more than those obtained in the BSM with 215 mg-Phe/L, respectively. Through kinetic analysis, the maximum specific growth rate (μmax) and PAH degrading rate (Vmax) for Phe were obtained as 0.34/h and 289 ${\mu}mol$/L/h, respectively. Also, μmax and Vmax for Pyr were 0.27 h-1 and 50 ${\mu}mol$/L/h, respectively. The degradation rates for each Phe (2.20 μmol/L/h) and Pyr (2.18 μmol/L/h) were lower in mixture substrates than in a single substrate (29.30 ${\mu}mol$/L/h and 9.58 ${\mu}mol$/L/h, respectively). Burkholderia sp. D5 can degrade Phe and Pyr contained in soil, and the PAH degradation rates in soil were 20.03 ${\mu}mol$/L/h for Phe and 1.09 ${\mu}mol$/L/h for Pyr.

Resistance Mechanisms of Green Peach Aphid, Myzus persicae (Homoptera: Aphididae), to Imidacloprid (복숭아혹진딧물(Myzus persicae)의 imidacloprid에 대한 저항성 기작)

  • Resistance mechanisms in the green peach aphid (Myzus persicae) resistant to imidacloprid were investigated. Imidacloprid residues on the aphid integuments decreased slowly as time passed with no significant difference between the susceptible and resistant strains. Residue in the aphid body increased in both strains with time elapse, and was slightly more in the susceptible strain. A higher metabolic rate of imidacloprid in the resistant strain can be expected by the fact that more amount of imidacloprid were excreted in the resistant strain than in the susceptible one. The activity of AChE was higher 1.4 times in the resistant strain than in the susceptible one, and imidacloprid did not inhibit AChE at all in both strains. Piperonyl butoxide (PBO) and iprobenfos (IBP) synergized imidacloprid activity. The mixtures of imidacloprid and PBO (1 : 1 and 1 : 5) caused 69.4- and 250-fold increase of imidacloprid toxicity against the aphid. Insecticide toxicity of the mixtures of IBP and imidacloprid (1 : 1 and 1 : 5) was also increased 227 and 80.6 times. Esterase activity when $\alpha$-naphtyl butyrate and $\beta$-naphtyl acetate were used as substrates was higher in the resistance strain than in the susceptible one. This means that P450 monooxygenase and esterase are responsible for the resistance to imidacloprid in this aphid strain.

Characterization of Low-Trans Solid Fat from Canola and Fully Hydrogenated Soybean Oil by Lipase-Catalyzed Interesterification Reaction (효소적 에스테르 교환 반응 시 카놀라유와 대두극도경화유의 비율에 따른 저트랜스 고체지방의 특성)

  • Lipase-catalyzed interesterification of canola (CO) and fully hydrogenated soybean oil (FHSBO) at different weight ratios (70:30, 75:25, and 80:20) was performed in a batch type reactor to produce low-trans solid fats. Each reaction was conducted in the shaking water bath for various reaction times (1, 3, 6, 18 and 24 hr) at 70oC and 220 rpm using Lipozyme TLIM (20 wt% of total substrate) from Thermomyces lanuginosus. After 24 hr reaction, solid fat content (SFC) by differential scanning calorimetry (DSC), fatty acid and triacylglycerol (TAG) composition of low-trans solid fats were determined. SFC of the products was reduced when the content of canola oil in the reaction mixture was increased. Major fatty acids were stearic acid (C18:0), oleic acid (C18:1) and linoleic acid (C18:2). Trans fatty acid content in the low-trans solid fats showed less than 0.3 wt%. In the HPLC analysis, major TAG species showed LOO (linoleyl-oleoyl-oleoyl), OOO, POO/SOL, SOO, and SOS.

A Study on the Content of Minerals in Fortified Food (영양강화식품 중 무기질 함량 조사연구)

  • This study was done to analyze the contents of minerals and to investigate the ratio of measured values to labeled values and to analyze the ratio of calcium to other minerals in 68 specimen with minerals - fortified commercial beverages, noodles, cereals and grain products. Content of calcium, iron and zinc in samples after microwave digestion was analyzed with an ICP-OES. The measured values of calcium were ranged 82.2~293.1% of the labeled values in 38 samples composed calcium - fortified commercial beverages, noodles, cereals and grain products. The measured values of iron and zinc were ranged 83.3~301.0%, 90.1~314.1% of the labeled values in minerals - fortified commercial beverages, noodles, cereals and grain products, 42, 24 samples. The Ca : Fe ratios were 90.55 (50.55~220.64) in fruit & vegetable juice, 850.41 in fruit & vegetable beverage, 553.49 in blended beverage, 179.07 (118.37~238.01) in soy milk, 204.39(41.64~397.52) in noodle, 296.97(121.64~868.88) in fried noodle, 30.89(15.69~62.05) in cereal and 7.73(0.22~49.92) in grain product. The Ca : P ratios were 1.44(0.96~1.98) in fruit & vegetable juice, 1.92 in fruit & vegetable beverage, 1.66 in blended beverage, 4.23(2.25~7.72) in soy milk, 1.14(0.28~1.97) in noodle, 1.88(1.17~2.42) in fried noodle, 1.29(0.87~2.92) in cereal and 0.30(0.06~1.57) in grain product. The Ca : Mg ratios were 1.85(0.87~5.04) in fruit & vegetable juice, 28.72 in fruit & vegetable beverage, 2.97 in blended beverage, 5.27(2.93~9.36) in soy milk, 3.97(1.34~7.57) in noodle, 6.77(4.63~10.78) in fried noodle, 4.40(2.30~12.55) in cereal and 1.17(0.23~7.48) in grain product. These results suggest calcium contents and the ratio of calcium contents to other minerals in calcium-fortified food products should be strictly controlled. Moreover, to avoid problems with Excessive nutrition, there must be initiatives for better understanding on food labelling and nutrition for fortified food.

The High Production of Cellulolytic Enzymes using Cellulosic Wastes by a Fungus, strain FJ1. (섬유소폐기물을 이용한 사상균 FJ1의 섬유소 분해효소의 고생산)

  • A filamentous microorganism, strain FJ1, was isolated from completely rotten wood for the production of cellulolytic enzymes. For the production of the enzymes, cellulolsic wastes were used as carbon sources of strain FJ1 and rice straw showed higher enzyme activities than sawdust and pulp. The activities of CMCase, xylanase, $\beta$-glucosidase, and avicelase were 2.95, 5.89, 0.45, and 0.12 unit/ml by use of rice straw, respectively. To enhance production of the enzymes, the mixture substrate of rice straw and cellulosic materials were investigated as carbon sources. The highest activities of CMCase, $\beta$-glucosidase, and avicelase were found in the mixture of rice straw (0.5%, w/v) and avicel (0.5%, w/v), and the highest xylanase was obtained at the mixture ratio of 0.71%(w/v) and 0.29%(w/v). Addition of 0.1%(w/v) peptone showed enhanced production of the cellulolytic enzymes in which the activities of CMCase, xylanase, $\beta$-glucosidase, and avicelase were 19.23, 27.18, 1.28, and 0.53 unit/ml, respectively. The production of the enzymes using rice straw was efficiently induced in the presence of avicel and pulp containing cellulose. In particular, a medium composed of rice straw (0.5%, w/v) and pulp (0.5%, w/v) yielded larger cellulolytic enzymes: CMCase 24.3 unit/ml, xylanase 38.7 unit/ml, $\beta$-glucosidase 1.5 unit/ml, and avicelase 0.6 unit/ml. The filamentous microorganism, strain FJ1 utilized various cellulosic wastes as carbon sources and will be expected as a favorable candidate for biological saccharification of cellulosic wastes.

The Manufactures and Characteristics of Chestnut Yoghurt (The Volatile Flavour Compounds and the Sensory Properties of Chestnut Yoghurt) (밤 Yoghurt의 제조와 특성 (밤 Yoghurt의 휘발성 향미성분 및 관능적 특성))

  • The effects of the addition of raw and dried chestnut with levels of 1%($T_1$),2%($T_2$),3%($T_3$) and 4%($T_4$) in skim milk substrate on the volatile flavour compounds and sensory properties of yoghurt fermented with the mixed cultures of YC-380 and ABT-4 during fermentation and storage period were investigated. In all treatments, the contents of acetaldehyde and acetone were detected at 2 hrs. and 1 hr. of fermentation, respectively and decreased with the storage period. The contents of ethanol and diacetyl were detected in all treatments at 3 hrs. and 4 hrs. of fermentation, respectively and increased significantly(p<0.05) with the storage period. The contents of volatile flavour compounds of treatments were increased gradually with decreasing the level of addition of raw and dried chestnut, and increased in order of fermented with YC-380 and ABT-4. The contents of volatile flavour compounds of raw chestnut yoghurt were slightly high compared to those of dried chestnut yoghurt. The taste, flavour and texture of $T_1$ were slightly higher than those of all treatments immediately after fermentation and during the storage period. The scores of sensory evaluation of treatments except $T_1$ were lowered significantly(p<0.05) with increasing the level of addition of raw and dried chestnut. The quality of both of flavour and texture, and taste were superior to chestnut yoghurt fermented with YC-380 and ABT-4, respectively. Generally the scores of sensory evaluation of dried chestnut yoghurt were slightly high compared to those of raw chestnut yoghurt. From the results mentioned above, the addition of raw and dried chestnut at 1%(w/v) level in skim milk substrate were suitable for volatile flavour compounds and sensory property of raw and dried chestnut yoghurt.
